고추역병 길항미생물 Bacillus subtilis AH18과 Bacillus licheniformis K11의 토양미생물 생태에 미치는 영향 (Effects of Phytophthora Blight-antagonistic Microorganisms Bacillus subtilis AH18 and Bacillus licheniformis K11 on the Soil Microbial Community)

  • 고추역병에 대한 길항미생물로 선발된 Bacillus subtilis AH18과 Bacillus licheniformis K11의 토양미생물 생태에 미치는 영향을 조사하기 위하여 이들 미생물을 단독 또는 복합으로 토양에 처리한 후 토양미생물상의 변화를 관찰하였다. 토양미생물 생태의 변화는 토양에서 인지질 지방산을 추출한 후 전체 지방산과 지표 지방산의 상대적 비율로 분석하였다. 전체적 인지질 지방산 구성과 각 지표 지방산 분석에 의해서 B. subtilis AH18 처리구는 무처리 대조구와 완전히 구분되었다. 전체 토양미생물상을 B. subtilis AH18 처리가 바꾸었고 세균 비율을 감소시켰으며, 곰팡이세균 비율을 높였으며, 그램음성균/그램양성균의 비율을 낮추었으며, 혐기성균/호기성균의 비율은 증가시켰다. 또한 불량환경 적응성을 의미하는 cy19:0/18:$1{\omega}7c$ 지방산비율과 단불포화지방산/포화지방산 비율도 B. subtilis AH18 처리구가 증가시켰다. 이에 반하여 B. licheniformis K11 처리나 두 미생물의 동시 처리는 전술한 토양미생물 지표에 큰 영향을 미치지 않았다. 따라서 지방산 분석을 토양미생물상 분석에서 B. subtilis AH18의 단용처리는 미생물을 처리하지 않은 대조구에 비하여 토양미생물상을 뚜렷이 바꾸었지만, B. subtilis AH18가 B. licheniformis K11와 혼용되었을 때는 토양미생물상 변화에 미치는 영향이 적었다. 따라서 이들 두 미생물을 동시에 처리할 경우 고추역병 방제와 토양미생물 생태 보존의 두 목적을 동시에 이룰 수 있을 것으로 기대된다.

Improvement of PCR Amplification Bias for Community Structure Analysis of Soil Bacteria by Denaturing Gradient Gel Electrophoresis

  • Denaturing gradient gel electrophoresis (DGGE) is one of the most frequently used methods for analysis of soil microbial community structure. Unbiased PCR amplification of target DNA templates is crucial for efficient detection of multiple microbial populations mixed in soil. In this study, DGGE profiles were compared using different pairs of primers targeting different hypervariable regions of thirteen representative soil bacteria and clones. The primer set (1070f-1392r) for the E. coli numbering 1,071-1,391 region could not resolve all the 16S rDNA fragments of the representative bacteria and clones, and moreover, yielded spurious bands in DGGE profiles. For the E. coli numbering 353-514 region, various forward primers were designed to investigate the efficiency of PCR amplification. A degenerate forward primer (F357IW) often yielded multiple bands for a certain single 16S rDNA fragment in DGGE analysis, whereas nondegenerate primers (338f, F338T2, F338I2) differentially amplified each of the fragments in the mixture according to the position and the number of primer-template mismatches. A forward primer (F352T) designed to have one internal mismatch commonly with all the thirteen 16S rDNA fragments efficiently produced and separated all the target DNA bands with similar intensities in the DGGE profiles. This primer set F352T-519r consistently yielded the best DGGE banding profiles when tested with various soil samples. Touchdown PCR intensified the uneven amplification, and lowering the annealing temperature had no significant effect on the DGGE profiles. These results showed that PCR amplification bias could be much improved by properly designing primers for use in fingerprinting soil bacterial communities with the DGGE technique.

인삼 유묘 뿌리썩음병 진전에 따른 토양군별 특성 (Characteristics of Soil Groups Basd on the Development of Root Rot of Ginseng Seedlings)

  • Based on the principal component analysis (PCA) of Richards' parameter estimates, ginseng field soils were grouped as the principal component 1 (PC1) and the principal component 2 (PC2). The microflora and physico-chemical characteristics of each soil group were compared to elucidate soil environmental factors affecting the disease development of root rot of ginseng seedling. Among 3 soil groups by PC1, there were differences in the populations of total fungi (TF) and Cylindrocarpon plus Fusarium (C+F), and the population ratio of Cylindrocarpon plus Fusarium to total fungi or total bacteria (C+F/TF, C+F/TB) in rhizoplane of ginseng seedlings, the population of total actinomycetes (TA) and the population ratio of total Fusarium to total actinomycetes (Fus/TA) in soil, and soil chemical properties (EC, NO3-N, K, Mn, ect.). Among 4 soil groups by PC2, there were differences in TF, C+F, TB, C+F/TF and C+F/TB in the rhizoplane, Trichoderma plus Gliocladium (T+G) in soil, and P2O5 content in soil. Especially, EC, NO3-N, K, K/Mg and Mn were positively correlated to PC1, and TA was negatively to PC1; however, TF, C+F, TB, C+F/TF and C+F/TB in the rhizoplane were significantly correlated to PC2 positively. On the other hand, microbes in the rhizoplane were not significantly correlated to the stand-missing rate (SMR), although TA and Fe/Mn were negatively correlated, and pH and Ca were positively correlated to SMR.

Relationships between Biodegradation and Sorption of Phenanthrene in Slurry Bioremediation

  • Bioremediation of hazardous hydrophobic organic compounds, such as polycyclic aromatic hydrocarbons (PAHs), is a major environmental concern due to their toxic and carcinogenic properties. Due to their hydrophobicity, the hydrophobic organic compounds are mainly associated with the soil organic matter or nonaqueous-phase liquids. A major question concerns the relationships between biodegradation and sorption. This work develops and utilizes a non- steady state model for evaluating the interactions between sorption and biodegradation of phenanthrene, a 3-ring PAH compound, in soil-slurry systems. The model includes sorption/desorption of a target compound, its utilization by microorganisms as a primary substrate existing in the dissolved phase and/or the sorbed phase in biomass and soil, oxygen transfer, and oxygen utilization as an electron acceptor. Biodegradation tests with phenanthrene were conducted in liquid and soil-slurry systems. The soil-slurry tests were performed with very different mass transfer rate: fast mass transfer in a flask test at 150 rpm, and slow mass transfer in a roller-bottle test at 2 rpm. In the slurry tests, phenanthrene was degraded more rapidly than in liquid tests, but with a similar rate in both slurry systems. Modeling analyses with several hypotheses indicate that a model without biodegradation of compound sorbed to the soil was not able to account for the rapid degradation of phenanthrene, particularly in the roller bottle slurry test. Reduced mass-transfer resistance to bacteria attached to the soil is the most likely phenomenon accounting for rapid sorbed-phase biodegradation.

토양세균 군집의 대사 다양성과 16S rDNA의 제한효소 지문분석에 의한 유전적 다양성의 비교 (Comparison of metabolic diversity by sole carbon source utilization and genetic diversity by restriction patterns of amplified 16S rDNA (ARDRA)in soil bacterial communities.)

  • BIOLOG GN microplate를 이용한 유일탄소원의 이용능 비교를 통한 대사적 유사성과 16S rDNA 의 PCR 증폭산물의 제한효소 지문 분석에 따른 유전적 유사성을 5종의 식생토양에 따른 토양미생물 군집을 대상으로 비교하였다. 16S rDNA를 증폭하여 제한효소 지문을 분석한 결과, 토양으로부터 직접 추출하여 증폭한 토양세균 군집의 16S rDNA의 유전적 유사도는 BIOLOG GN microplate를 이용한 대사적 구조와 일치하는 경향을 보았다. 그러나 배양된 종속영양세균 군집의 다양성과는 유전적 유사도가 매우 낮게 나타났다.

해안간척지토양의 생물학적 토성개량에 관한 연구 5 (Biological improvement of reclaimed tidal land soil (V))

  • Chenges in respiration of the soils collected from the reclaimed tidal soil in Chogi-ri, Kanghwa Island and treated with organic matters are presented. The measurement of the respiration for the smaples, which were incubated for 0,2,4 and 5 weeks, were carried out by using Warburg's respirometer. While the respirations of the samples added by organic matters were increased 6.7 - 28.0 times compared with that 0 week during the incubation in case of 5-year soil, the respiration in case of 3-year soil were increased 3.3-11.8 times. Thus, the effect of adding organic matters on the respiration of the experimented soils, as this indicates, was much higher for the soil of 5-year area than that of 30-year area. And for the organic matters Salicornia was most effective and then Suaeda and Oryzae. The samples treated with Salicornia and Suaeda showed their highest respiration rate at the 4th week, but the one with Oryzae was measured to increase progressively during 5 weeks experimented. Regarding the salinity, content of organic matters and number of bacteria, in each intact soil experimented, 5-year soil samples had much poor habitat then 30-year soil for the activity of soil microorganisms, but according to the result mentioned above, it is firmly believed that the addition of organic matters on the saline soil is one of the best means to change the reclaimed tidal land into arable land with less time duration.

Soil Microorgnisms Comparison that Exist to Around of Upland and Wild Codonopsis lanceolata and Microorganism Investigation Affecting to Aromatic Substances

  • We investigated the microbial populations and viable cell counts of Codonopsis lanceolata from uncultivated and cultivated soil in the suing. The microbial populations and viable cell counts from both types of soils were also investigated simultaneously. It had existed 10 more kinds of microorganisms in uncultivated than those in cultivated. The total viable cell counts of C. laneolata from uncultivated soil, especially in the upper zone, were 9.7$\times$10$^{6}$ CFU/g. However, the C. lanceolata from cultivated soil was 4.2$\times$10$^{6}$ CFU/g. (omitted)

  • The study was carried out to evaluate the characteristics of biodegradation by Pseudomonas putida G7 in soil column. The reactor system was used to investigate mass transfer of VOCs as Toluene in a column of unsaturated soil. Determination of the fate of VOCs in unsaturated soil is necessary to evaluate the feasibility of natural attenuation as a VOCs remediation strategy. The objective of this study was to develop a mechanistically based mathematical model that would consider the interdependence of VOC transport, microbial activity, and sorptive interactions in a moist, unsaturated soil. Because the focus of the model was on description of natural attenuation, the advective VOCs transport that is induced in engineered remediation processes such as vapor extraction was not considered. It can be concluded that the coefficient for gas liquid mass-transfer was found to be a key parameter controlling the ability of bacteria to VOCs. Finally, it appeared that bioremediation technology of VOCs which are difficult to be decomposed by chemical methods.

Effect of microorganism on engineering properties of cohesive soils

  • This paper presents the study of the effect of microorganism Bacillus pasteurii on the properties such as Atterbergs' limit and unconfined compressive strength of cohesive soils. The results of this study reveal that the liquid limit and plasticity index for all clay soils decreased and the unconfined compressive strength increased. Decrease in plasticity index is very high for Kuttanad clay followed by bentonite and laterite. The unconfined compressive strength increased for all the soils. The increase was high for Kuttanad soil and low for laterite soil. After 24 h of treatment the improvement in the soil properties is comparatively less. Besides the specific bacteria selected Bacillus pasteurii, other microorganisms may also be taking part in calcite precipitation thereby causing soil cementation. But the naturally present microorganisms alone cannot work on the calcite precipitation.

L-글루타민산 생산균 Brevibacterium lactofermentum의 Bacteriophag에 관한 연구 (Studies on the Bacteriophages of Brevibacterium lactofermentum)

  • Many industrial processes those employ bacteria are subjected to phage infestations. In L-glutamic acid fermentions using acetic acid, the phage infestations of the organisms have been recently recognized. In efforts to elucidate the sources of phage contamination involved in the abnormal fermentation, a series of study was conducted to isolate the phages both from the contents of abnormally fermented tanks and the soil or sewage samples from the surroundings of a fermentation factory, to define major charateristics of the phage isolates, and finally to determine the correlation between the phage isolates and temperate phages originating from the miscellaneous bacterial species isolated from the soil or sewage samples. The results are summarized as follows; 1) All phages were isolated from the irregular fermentation tanks and soil or sewage samples, and they were designated as phage PR-1, PR-2, PR-3, PR-4, PR-5, PR-6, and PR-7, in the order of isolation. These PR-series phages were proved to be highly specific for the variant strains of Br. lactofermentum only, namely, phage PR-1 and PR-2 for Br. lactofermentum No. 468-5 and phage PR-3~PR-7 for Br. lactofemrentum No. 2256. By cross-neutralization test, the 7 phagescould be subdivided into 3 groups, i. e., phage PR-I and PR-2 the first, phage PR-3, PR-4, PR-5, PR-6 the second, and the phage PR-7 the third. 2) The 7 phages were virulent under the experimental conditions. They produced plaques with clear and relatively sharp margins without distinct halo. The mean sizes of plaques were 1.5mm in diameter for phage PR-1 and PR-2, and 1. Omm for phages PR-3~PR-7. Double layer technique modified by Hongo and described by Adams, was applied to assay of the PR-series phages. The factors influencing the plaques were as follows;young age cells of host bacteria cultured for 3-6 hours represented the largest number and size, optimum was pH 7.0, incubation temperature was $30^{\circ}C$, and agar concentration and amount of overlayer medium were 0.6% and 0.2ml, respectively. 3) PR-series phages were stable in 0.05M tris buffer and 0.1M ammonium acetate buffer solution. The addition of $5{\times}10^{-3}M$ magnesium ion effectively increased the stability. Thermostability experiments indicated that PR-series phages were stable at the teinperture between $50^{\circ}{\sim}55^{\circ}C$ in nutrient medium, $45^{\circ}{\sim}50^{\circ}C$ in buffer solution. However, the phages mere completely inactivated at 603C and 65$^{\circ}$C within 10 minutes. The phages were stable at the range of pH6~9 in nutrient medium and of pH 8-9 in buffer solution, respectively. Exposure of the phages to UV for 25, 60 and 100 seconds resulted in the complete loss of infectivily, respectively. 4) Electron microscopy showed that PR-series phage particles exhibited rather similar morphology, differing in the size All of PR-series phages had a multilateral head and had a simple long tiil about three to five times long as compared with head. By the size, phage PR-1 and PR-2, PR-3, PR-4, PR-5, and PR-6 and PR-7 were classified into same groups, respectively. The head and tail size of phage PR-1, PR-5, PR-5(T) and PR-7 were 85nm, 74nm and 235nm and 350mm, and 72nm and 210nm, respectively. 5) Nucleic acids of PR-series phages were double stranded DNA. The G+C contents of phage PR-1, PR-5 and PR-7 were 56.1, 52.9 and 53.7, respectively. The values of G+C contents derived from the $T_m$ were in agreement with the chemically determined values. 6) PR-series phages effectively adsorbed on their host bacteria at the rate of more than 90% during 5 min. K value for phage PR-1, PR-5 and PR-7 were calculated to be $6{\times}10^9 ml$ per minute, respectiveky. The pH of the medium did effect adsorption rate, but both temperature and age of host cells did not. Generally, optimum adsorption condition of phages seemed to be almost same as optimum growth conditions of host bacteria. 7) In one-step growth experiments, the latent periods at $30^{\circ}C$ for PR-1, and PR-7 were about 70, 50 and 55 min, respectively. The corresponding average burst size was 200, 70 and 90, respectively. Lpsis period according to the multiplicity of infection and a phage series. In case of m. o. i. 100, strain No. 2256 (PR-5) and No. 468-5(PR-1) failed to grow and turbidity decreased after 50 and 70min, respectively. 8) In the lysate of a plaque purified phage PR-5 infected bacteria, there observed 2 types ofphage particles, i. e., phage PR-5 and PR-5 (T) of similar morphology but differing at the length of phage tail, and phage tail like particles. The phage taillike particles could be divided into 4 types by the length. Induction experiments of Br. lactofermentum with UV irradiation, mitomycin C or bacitracin treatment produced neither phage PR-5 (T) or phage tail-like particles. 9) No lysis occured when the growth of 7 strains of miscellaneous bacteria, isolated from soil and sewage samples, were inoculated with either phage PR-5 (T) or phage tail-like particles the inoculation of phage PR-5 pellet resulted in the growth inhibition of the orgainsms in the spot test. The lysates obtained from 3 miscellaneous soil derived bacteria following mitomycin C treatment the growth of Br. lactofermentum, but did not lyze the bacterium.
