Rheological Properties of Cement Paste Mixed with Aqueously Dispersed Single-Walled Carbon Nanotubes (Single-Walled 탄소나노튜브 수용액 혼입 시멘트 페이스트의 유변학적 특성)

  • Single walled carbon nanotube (SWCNT) has been used as a material for reinforcing various advanced materials because it has superior mechanical properties. However, pure SWCNT that does not have any functional group has a hydrophobic character, and exists as bundles due to the strong Van der Waals attraction between each SWCNT. Due to these reasons, it is very difficult to disperse SWCNTs in the water. In this work, in order to use SWCNT for production of cementitious composites, SWCNT was first dispersed in water to make an aqueous solution. Sodium deoxycholate (DOC) and Sodium dodecyl sulfate (SDS) were chosen as surfactants, and the dosage of DOC and SDS were 2wt% and 1wt%, respectively. Sonication and ultracentrifugation were applied to separate each SWCNT and impurities. Using such processed SWCNT solutions, cement paste was prepared and its shear stress vs. strain rate relationship was studied. The yield stress and plastic viscosity of cement paste were obtained using Bingham model. According to the results in this work, cement pastes made with DOC and SDS showed similar rheological behavior to that of air entrained cement paste. While cement paste made with DOC 2 wt.% SWCNT solution showed similar rheological behavior to that of plain cement paste, cement paste made with SDS 1 wt.% SWCNT solution showed different rheological behavior showing much less yield stress than plain cement paste.

  • Thermostable $\beta$-mannanase from Bacillus sp. YA-14 was purified by acetone precipitation, CM-cellulose, Sephadex G-100 and hydroxyapatite column chromatography from culture supernatant. The final enzyme preparation appeared to be homogeneous on sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). $\beta$-Mannanase appeared to be a monomeric protein with a molecular weight of 67, 000 daltons. The optimal pH and temperature of the enzyme reaction were pH 6.0 and $75^{\circ}C$ , respectively. The enzyme was stable at a pH range of 6.0 to 9.0 and at temperatures between 45 and $85^{\circ}C$. The kinetic constants of $\beta$-mannanase as determined with a galactomannan (locust bean) as substrate were a Vmax of 25 unit/ml and a Km of 1.1 mg/ml. The enzyme had only limited activity on galactomannan substrate. It was suggested that mg $\beta$-mannanase activity is limited by the number of branched $\alpha$-galactose residues.

Bacteriocin with a Broad Antimicrobial Spectirum, Produced by Bacillus sp. Isolated from Kimchi

  • An antimicrobially active bacterium which was identified as Bacillus brevis, was isolated from kimchi. The antimicrobial activity was found against various Gram-positive and Gram-negative bacteria including some pathogens food-spoilage microorganisms, and some yeast strains. The antimicrobial activity was especially strong against Bacillus anthracis and Shigella dysenteriae. The strong activity was observed during an early stationary phase in the culture when incubated at $37^{\circ}C$ with initial medium pH of 6.8. The antimicrobial activity was found to be stable at $90^{\circ}C$ for 30 min and in the pH range of 3-11, and it was insensitive to organic solvents including acetone, acetonitrile, ethanol, and methanol. Analysis of the bacterocin on tricine-sodium dodecyl sulfate-polyacrylamide gel suggested a molecular mass of approximately 4.5-6.0 kDa. The antimicrobial substance was characterized as a bacteriocin, because of its proteinaceous nature and low molecular weight. The bacteriocin could potentially be used as a food preservative, because of its thermostable property and broad antimicrobial spectrum.

Purification and Properties of $\gamma$-Glutamyl Transpeptidase from Bacillus sp. KUN-17

  • $\gamma$-Glutamyl transpeptidase ($\gamma$-GTP; EC 2.3.2.2) present in the culture filtrate of Bacillus sp. KUN-17 was purified 400-fold through a consecutive procedure including organic precipitation and column chromatography. The enzyme has an estimated molecular weight of 70, 000 and consists of hetero-subunits with molecular weights of 42, 000 and 22, 000. In vitro optimal conditions for those transfer and hydrolysis reactions appeared to be pH 7.0 at $50^{\circ}C$ and pH 8.4 at $40^{\circ}C$, respectively. The denatured enzyme recovered most of its $\gamma$-GTP activity by removing detergents such as sodium dodecyl sulfate (SDS) or urea with dialysis. The enzyme showed higher affinities against a number of amino acids as $\gamma$-glutamyl acceptors than glycylglycine in the following order: L-valine, L-methionine, L-glutamic acid or L-as-paragine, L-alanine. Also, it was shown that L-glutamine was the most suitable $\gamma$-glutamyl donor for the transfer reaction among those tested. Amino acids generally inhibited the enzyme activity for the transfer reaction, but not for the hydrolysis reaction.

  • The $\alpha$,$omega$-diacrylate poly(dimethylsiloxane) (DA-PDMS) containing vinyl ester of versatic acid/vinyl acetate (Veova-10/VAc) was prepared by emulsion copolymerization of (DA-PDMS), Veova-10 (with VAc), and auxiliary agents at $85^{\circ}C$ in the presence of ammonium peroxodisulfate (APS) as an initiator. Sodium dodecyl sulfate (SDS) and nonylphenol ethylene oxide-40 units (NP-40) were used as anionic and nonionic emulsifiers, respectively. The resulting copolymers were characterized by using Fourier transform infrared spectroscopy (FT-IR). Thermal properties of the copolymers were studied by using thermogravimetric analysis(TGA) and differential scanning calorimetry (DSC). The morphology of copolymers was also investigated by scanning electron microscopy (SEM) and then the effects of variables such as temperature, agitation speed, surfactant kinds, molecular weights, initiator, and DA-PDMS concentrations on the properties of the silicone-containing Veova-10/VAc emulsions were examined. The calculation of monomer conversion versus time histories indicates that by increasing the DA-PDMS concentration the polymerization rate and the number of polymer particles decrease, respectively.

Quality characteristics and protein digestibility of Protaetia brevitarsis larvae

  • Herein, the in vitro protein digestibility of lyophilized Protaetia brevitarsis larvae flour with and without defatting using 70% ethanol was compared with beef loin. Proximate analysis showed that the defatted larvae contained the highest protein content (p < 0.05). The viable counts of total aerobic bacteria, Escherichia coli, and coliform bacteria decreased significantly after defatting the larval samples with 70% ethanol (p < 0.05). Measurement of α-amino group content and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) revealed higher amounts of low molecular weight proteins in the larvae compared to beef loin (p < 0.05). After in vitro digestion, the degree of protein hydrolysis of the digesta was higher for both larvae samples compared to beef loin (p < 0.05). No change was observed in the in vitro larval protein digestibility after defatting. These results highlight the excellent protein digestibility of P. brevitarsis larvae with high protein content. Defatting insect flour with 70% ethanol could enhance microbial safety while maintaining excellent protein digestibility.

  • Ornithogalum showing mosaic symptoms were collected from the isolated field of National Plant Quarantine Service in Sengrimmyon of Kyungnam province. Electron microscopic examination of negatively strained preparation was filamentous particle of 740nm in lenght. Indirect-ELISA determined that the virus was serologically related to potyvirus. A single major protein band of Mr 30,000 was observed after sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Indicator plant test showed masaic, necrotic local lesion and sunken areas in leaves of Nicotiana clevelandii and Tetragonia expansa, while the others of indicator plants did not infect. An enzyme-aided purification protocal was used, which eliminated a highly viscous mucilage from extracts of the Omithogalum. Total RNA extracted from infected Omithogalum leaves were amplified of 411b.p fragment in reverse transcription (RT)-PCR when primers specilic for the coat protein gene. An isolate of Omithogalum mosaic virus (OrMV) of the genus Potyvirus was identified as the casual agent of the disease on the basis of electron microscopic, biological and serological reaction.

  • Poly (vinyl acetate) (PVAc) was used as a precursor of PVA/PVAc (skin/core) bicomponent. In order to investigate the possibility of PVA particles for electrical applications, PVA/PVAc particles were produced with an emulsifier, SDS (Sodium Dodecyl Sulfate) and an initiator, V-50 (2,2'-azobis(2-amidinopropane)digydrochloride). In this study, we investigated the electrical property of PVA/PVAc (skin/core) particles. The hydroxyl group of the PVA/PVAc (skin./core) was confirmed by the analysis of PVAc and PVA/PVAc (skin/core) using Fourier Transform Infrared Spectroscopy (FT-IR). The zeta-potential of the PVA/PVAc (skin/core) and PVAc has similarity; however, charge control agent (CCA) treated PVA/PVAc (skin/core) particles has lower zeta-potential than untreated PVA/PVAc particles. The zeta-potential (negative values) of the PVA/PVAc (skin/core) were enhanced in proportion to the increased concentration of CCA.

Comparison of Meat Quality Characteristics of Wet- and Dry-aging Pork Belly and Shoulder Blade

  • The physicochemical characteristics and oxidative stability of wet-aged and dry-aged pork cuts were investigated at different aging periods (1, 7, 14 and 21 d). Samples were assigned into four groups in terms of shoulder blade-wet aging (SW), shoulder blade-dry aging (SD), belly-wet aging (BW), and belly-dry aging (BD). SD showed significantly higher pH at 21 d of aging than the other samples. Wet-aged cuts had significantly higher released water (RW) %, lightness ($L^*$) and shear force compared to the dry-aged meats. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis showed greater degradation of proteins for dry-aged cuts than the wet-aged cuts. At the end of aging, wet-aged cuts showed significantly lower thiobarbituric acid-reactive substances (TBARS) value than the dry-aged samples, indicating higher oxidative stability for wet-aged pork cuts. However, dry-aging led to higher degradation of proteins resulting in increased water-holding capacity (WHC) and decreased shear force value.

Quantitation of CP4 5-Enolpyruvylshikimate-3-Phosphate Synthase in Soybean by Two-Dimensional Gel Electrophoresis

  • Changes of CP4 5-enolpyruvylshikimate-3-phosphate synthase (CP4 EPSPS) in the glyphosate-tolerant Roundup Ready soybean were examined using purified CP4 EPSPS produced in cloned Escherichia coli as a control. CP4 EPSPS in genetically modified soybean was detected by twodimensional gel electrophoresis (2-DE) and identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and electrospray ionization tandem mass spectrometry (ESI-MS/MS) with databases. CP4 EPSPS in soybean products was resolved on 2-DE by first isoelectric focusing (IEF) based on its characteristic pI of 5.1, followed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) based on its molecular mass of 47.5 kDa. We quantified various percentages of soybean CP4 EPSPS. The quantitative analysis was performed using a 2D software program on artificial gels with spots varying in Gaussian volumes. These results suggested that 2-DE image analysis could be used for quantitative detection of GM soybean, unlike Western blotting.
