Seed disinfection is a necessary process to decrease Bakanae disease in rice and is carried out at 30℃ to increase disinfection efficiency. However, the length of sprout after seed disinfection is not suitable for sowing, so additional processes such as seed soaking is needed. For this reason, this study was conducted to develop the efficient and effective seed disinfection method. When the seed disinfection finished after soaking seeds in cold water for 2 days, length of sprout was about 1.0 mm and sowing can be done without additional processes. In addition, the incidence of Bakanae disease was greatly reduced by 64% compared to conventional method. In order to find out why the newly developed method (disinfection after soaking) is more efficient than conventional method, soaked and not soaked seeds were stained using methylene blue staining. It was found that soaked seeds were more easily stained with about 50% of internal and external endosperm stained. Therefore, pesticide can be absorbed easily into internal endosperm of soaked seeds and efficiently destroy the spore of pathogen. As a results of this study, newly developed method will be useful as it can improve efficiency of agricultural work and also effectively decrease Bakanae disease compared to conventional method.
The seeds of Elaeocarpus serratus, a tropical underutilized fruit tree are characterized by hard seed coat and consequent poor water uptake and low germination. To improve the regeneration through seeds, various parameters such as viability of seeds, water uptake, and effect of seed mass on germination and pretreatments were performed using a completely randomized design (CRD). Tetrazolium (TZ) test was conducted using fresh, mature seeds revealed $50{\pm}2.56%$ mean viability. Seeds of different weight classes showed similar pattern of water uptake and the saturation level was achieved at 60 hrs of soaking. Seeds belong to weight class 2.6-3.5g were germinated ($12.5{\pm}1.26%$) with $175{\pm}1.75days$ (d) of mean time taken for germination (MTG). Germination capacity of seeds varied significantly among different populations and Varkala population gave $12.5{\pm}1.1%$ germination with $174.6{\pm}2.5d$ MTG. Among various seed treatments, mechanical scarification was superior in germination and significant reduction in MTG ($p{\leq}0.05$). The mechanical scarification by complete removal of seed coat resulted in $49.2{\pm}1.52%$ germination within a short period of time ($9.52{\pm}0.89d$ MTG). However, the complete removal of seed coat without damaging to embryo is a difficult task. An alternate treatment (Mechanical scarification II) by making cracks on nut faces vertically followed by soaking in distilled water for 24 hrs gave $48.4{\pm}1.73%$ germination with significantly reduced MTG ($12.14{\pm}0.56d$) over unsoaked, untreated control ($6.5{\pm}1.84%$ germination and $197.18{\pm}1.79d$ MTG; $p{\leq}0.05$). This treatment (Mechanical scarification II) is therefore recommended for E. serratus seeds as it can adopt easily and can achieve 7 fold increases in germination over control. The recorded germination through mechanical scarification is in tune with realized viability percentage of the seeds.

We examined the effects of soaking, saturation, and aging conditions of raw cowpea seeds, on the yield and growth responses of cowpea sprouts. The absorption caused rapid growth for the first two hours of soaking, then the growth slowed. The longer the soaking period (varied from 1 to 6 hours), the lower the germinability, yield ratio, and lateral root output became. A five-day saturation (moist $96{\pm}1%$, $20^{\circ}C$) treatment led to higher germinability, yield ratio, and lateral root output than no treatment and one- or three-day saturation treatment. High-temperature aging treatment led to lower germinability and yield ratio compared to no such treatment. Taking these findings into account, the optimal treatment conditions of raw cowpea seeds are a five-day saturation (moist $96{\pm}1%$, $20^{\circ}C$) treatment followed by cleaning and growing.
To find out the effects of humidification and hardening of rice (Oryza sativa L. cv. Ilpumbyeo) seeds on the germination, both normal and artificially aged seeds with 60% germination rate were humidified at 40, 60, and 80% RHs and $25^{\circ}C$ for five weeks or hydrated for 6, 12, 18, and 24 hours at $25^{\circ}C$ from one to five cycles for hardening. Relative humidity and duration of humidification did not affect the germination rate of normal seeds, while reduced the time to get 50% germination rate ($T_{50}$) by 1.0~1.6 days compared to that of normal seeds at $25^{\circ}C$. Aged seeds humidified at 40 and 60% RHs did not affect the germination rate and $T_{50}$ regardless of relative humidity and duration of treatment, while at 80% RH, the germination rate decreased and $T_{50}$ increased significantly with the duration of humidification. Hardening of normal seeds reduced $T_{50}$ by 0.7~1.1 days without changes in the germination rate. However, the germination rate and $T_{50}$ of aged seeds soaked in water for 6, 12, and 18 hours were similar regardless of soaking/drying cycles, while hardening of artificially aged seeds with 1~4 cycles in 24 hours soaking increased the germination rate by 11-16% and reduced $T_{50}$ by 1.4~2.0 days.
Quantity of protein, amino acid, and sugar leaked from seeds was greater as the viability of seeds was dropped by the time elapsed of seed aging treatment. In the seeds with the artificial aging treatment for 20 days, 35.8 mg of protein was leaked on the 4th day after soaking, which was 6.9 times higher than that of control. Leakage of amino acid was also higher from low quality seeds treated with the aging treatment. In the seeds with the aging treatment for 20 days, $36.5{\mu}g$ fig of sugar was leaked on the 4th day after soaking, which was 2.8 times higher than that of control. The leakage of inorganic compound was higher from the low quality seeds, and leakage of total protein, amino acid, and sugar. According to .the quantity of leakage, water soluble compounds, which can be used for the assessment of seed quality without any destruction, were protein and potassium. Germination rate and percentage of seeds were dropped with the seed aging treatment, and the seed viability could be recovered by priming treatment. This phenomenon was very clear when the low quality seeds were germinated at low temperature.
The study is aimed to understand effects of ethephon(lmg/1), ABA(0.lrng/l), BA(0.lmg/l). and NAA (0.lmg /1) soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw'. These experiments are carried in vitro and their results are summarized as the followings ; In case of ethephon (lmg /1) soaking treatment of seed, the early germination of tall fescue 'Arid' and perennial ryegrass 'Dandy' was good without the soaking times. creeping bentgrass 'Penlinks' and kentucky bluegrass 'Nasaw' obtained the best results at 12 hr. soking treatment, and the longer the soaking time, the higher the germination percentage. The early germination percentage of tall fescue 'Arid' and perennial ryegrass 'Dandy' increased more highly at ABA(0. tmg /1) soaking treatment than at the control. The germination percent-age of creeping loentgrass 'Penlink' and kentucky bluegrass 'Nasaw' had respectively the best results at 6hr. and l2hr. soaking treatment. The early germination percentage of tall fescue 'Arid' and kentucky bluegrass 'Nasaw'increased more highly at BA (0.lmg /1) soaking treatment than at the control, and especially had the best results in 6hr. soaking treatment. perennial ryegrass 'Dandy' had the best results at l0min. soaking treatment, and the longer the soaking time, the lesser the germination percentage. The early germination of perennial ryegrass 'Dandy'. creeping bentgrass 'Penlinks' and tall fes- cue 'Arid' increased more highly at NAA (0.lmg /1) soaking treatment than at the control, and especially had the best results at l2hr. soaking treatment. This study is part of research to develop the technology for managing major medicinal crops after harvest. We studied the optimal germination conditions of $Cynanchum$$wilfordii$ (Maxim.) Hemsl. The mean germination time (MGT) of $C.$$wilfordii$ seeds was higher after soaking for 4 days after storage at $5^{\circ}C$ for 8 weeks, than with germination at $25^{\circ}C$ (3 days). However, the germinative energy (GE) decreased as the number of days soaking increased. The greatest germination rate ($83.3{\pm}8.8%$) was at $25^{\circ}C$ with no soaking of seeds stored at $-20^{\circ}C$ for 8 weeks. Based on these results, we characterized the germination conditions of a major medicinal crop.
This study was carried out to develop an effective seed propagation method for Thalictrum rochebrunianum var. grandisepalum (H. Lev.) Nakai by analyzing seed dormancy types and germination characteristics. Seeds were collected between September to October at Gangwon province, and well-selected seeds were used while being dry-stored at 4±1℃. The seed size ranged 4.52 × 1.58 ㎜ and the weight of thousand seeds were 1,603.5 ± 0.02 ㎎. The moisture content was 7.2%. Seeds were achene type, and morphology characters showed an elliptical shape and rough texture, and light brown in color. Moist-chilling treatment was conducted for dormancy breaking because the seeds had an undeveloped embryo of liner type. The embryo had developed during a moist-chilling period, constantly, and fully developed in 10 weeks. Consequently, it seemed to be non-deep complex or intermediate complex type of morphophysiological dormancy, and embryo dormancy was broken by wet-chilling for 10 weeks. After 10 weeks of wet-chilling treatment, seed germination began. Germination percentage was higher in dark condition raher than light condition and recorded the maximum at 25℃ in the dark (16.3%). A pre-soaking treatment with a combined plant growth hormones promoted germination and shortened T50. Specifically, seed germination of 84.5% was achieved by pre-soaking of seeds with a combined solution of 500 ㎎/L GA3 and 10 ㎎/L kinetin for 24 h after a wet-chilling treatment for 10 weeks. Thus the effect of plant growth hormones coupled with chilling temperature on seed breaking dormancy provide asubsequent growth of seedlings for successful plantation.

Background : The aim of the present study was to identify an effective physicochemical control method to reduce Fusarium species infestation in adlay (Coix lacryma-jobi L.) before and after harvesting. Methods and Results : We observed that prochloraz emusifiable concentrate and hexaconazol prochloraz emusifiable concentrate strongly inhibited the mycelial growth of 10 Fusarium species. Strong growth inhibitions and cell lysis were observed following treatment with 4% NaOCl solution. The total number of fungi detected were lower follwing treatment with thiophanatemethyl triflumizole wettable powder ($1.1{\times}10^4CFU/g$), hexaconazol prochloraz emulsifiable concentrate ($1.2{\times}10^4CFU/g$), carboxin thiram dustable powder ($1.6{\times}10^4CFU/g$) and prochloraz emulsifiable concentrate ($1.7{\times}10^4CFU/g$) than in the non-treated control ($7.7{\times}10^4CFU/g$). The reduction of Fusarium fungi varies with the concentration and soaking time of NaOCl solution. Fungal detection was not observed after soaking in NaOCl solution for 24 h and harmful effects were not observed for plant growth by NaOCl after soacking for 6 - 12 h. Conclusion : Soaking seed for 6 - 12 h in 4% NaOCl could be an effective method of disinfectant treatment for the control of Fusarium fungi in adlay seeds.
