Cosmetic Application Using Skin Whitening and Anti-microbial effects of Ethyl Acetate and n-Butanol Fractions from Eruca sativa (Eruca sativa 에칠아세테이트와 부탄올 분획물의 미백 및 항균효과를 이용한 화장품 응용연구)

  • Eruca sativa, called arugula, is a perennial plant in the Brassicaceae family, an edible plant commonly used in Italian cuisine. To study as a cosmetic material application E. sativa was extracted with 70% ethanol (ES). Then ES was fractionated using n-hexane, chloroform, ethyl acetate, n-butyl alcohol and water (EHex, EEA, ECHCl3, EBuOH and EDW). EEA showed mushroom tyrosinase inhibitory activity. ES, EEA and EBuOH showed inhibition of tyrosinase activity. As a result, ES is expected to have skin whitening efficacy. ES was applied to 0.05, 0.1% the toner and emulsion formulation to test the stability. The anti-microbial activity of eight bacteria and fungi including Staphylococcus aureus and Propionibacterium acnes which cause dermatitis and acne was evaluated. EEA showed effects in all of microorganisms. The toner and emulsion containing ES with 0.05, 0.1% were passed in the challenge test. At -20, 4, 25, 55 ℃ and daylight, there was no significant change on pH, viscosity for 4 months. However, emulsions had phase separation phenomenon at 55 ℃, so the base formulation needs improvement. In addition, through the skin penetration test, EEA penetrated 0.058% in 6 hr, predicting the clinical efficacy. This means that E. sativa can contribute whitening agent and the synergistic effect of preservatives.

  • To develope new natural substances for whitening agent, Baeckji(Angelica dahuria;BK), Seasin(Asarum sieboldii;SS). Baecjgangjam(Bombycis corpus;BGJ) and Mynkanbang(MKB) were selected and extracted by hot water and 70% EtOH, respectively. Hydrothermal and ethanolic extracts of BK, SS, BGJ and MKB were evaluated for anti-oxidative effect, tyrosinase activity, melanogenesis of B16 melanoma cells and changes in level of tyrosinase expression by using Western blotting. All hydrothermal and ethanolic extracts showed scavenging activities of free radicals against 1,1-diphenyl-2-picrylhydrazyl(DPPH) but no inhibitory effect on tyrosinase activity. Ethanolic extracts of BK, SS, and BGJ and especially highly, those of MKB inhibited production of melanin in B16 melanoma cells and were able to reduce the level of tyrosinase expression in B16 melanoma cells. These results suggest that ethanolic extracts of BK, SS, BGJ and MKB can be an effective whitening agent from natural plant.

Whitening effect of novel peptide mixture by regulating melanosome biogenesis, transfer and degradation

  • Peptides are short chain of amino acids linked by peptide bonds. They are widely used as effective and biocompatible active ingredients in cosmetic industry. In this study, we developed novel peptide mixture and identified its anti-pigmentation effect on melanocytes and keratinocytes. Our results revealed that peptide mixture inhibited melanosome biogenesis through the regulation of microphthalmia-associated transcription factor, a key factor of melanogenesis in melanocytes. And we observed that peptide mixture inhibited melanosome uptake through the reduction of protease-activated receptor 2, a phagocytosis-related receptor in keratinocytes. Furthermore, peptide mixture activated autophagy system resulting in degradation of transferred melanosomes in keratinocytes. The anti-pigmentation effect of multi-targeting peptide mixture was assessed in a human skin equivalent model (MelanoDerm). Melanin contents in epidermal layer were significantly decreased by topical treatment of peptide mixture, suggesting that it can be applied as a novel cosmetics material having a whitening function.

Depigmentation activity of Kakamseosiokyong-san (가감서시옥용산의 미백효과에 관한 연구)

  • Objective : The aim of this study was to investigate the skin-whitening effect of Kakamseosiokyong-san Method : We investigated that the extracts of Kakamseosiokyong-san inhibit activity of tyrosinase, the enzyme which converts 3-(3,4-dihydroxyphenyl)alanine to dopachrom in the biosynthetic process of melanin. the UV absorbance of the extracts in the UV - A region and UV - B region was measured by UV scanning. the effect of extracts on cell viability and melanin production in cultured B16 mouse melanoma cells was measured, and cytoprotective effects of extracts on PC12 cells injured by hydrogen peroxide was measured by MTT assay Results: The extracts of Kakamseosiokyong-san inhibited activity of tyrosinase. The extracts not only showed inhibitory effects on melanin production in cultured B16 mouse melanoma cells, but also exhibited cytoprotective effects on PC12 cells injured by hydrogen peroxide, but did not showed an absorbance in the UV - A region and UV - B region. Conclusion: These results suggest that Kakamseosiokyong-san inhibit melanin biosynthesis which is involved in hyperpigmentation and could be used as a whitening agent for the skin.

The Anti-Wrinkle and Whitening Effect of Extracts of Castanea crenata Inner Shell (밤부산물(율피)의 주름개선 및 미백 효과)

  • Aging in humans is inexorable and inescapable. The progressive decrease of physiological capacity and the reduction of the ability to cope with environmental stresses lead to increased susceptibility and vulnerability to human disease. Recently, in the cosmetic industry, many researchers have paid considerable attention to delaying or improving the symptoms of skin aging. Since the early 1990`s, there have been various challenges in developing cosmeceutical products which have strong anti-aging effects, and this has been an important issue in the cosmetic industry. Meanwhile, development of anti-aging cosmetics supported by biochemical activities in the skin has been researched. Castanea crenata inner Shell solvent extracts were investigated for anti-wrinkle and whitening effects, in order to apply it as a functional ingredient for cosmetic products. For anti-wrinkle effect, elastase inhibition activity of Castanea crenata inner shell acetone extract (CA) was 51.0% at a concentration of 100 ug/ml. The collagenase inhibition activity of CA and Castanea crenata inner shell ethanol extract (CE) was 96.4%, 94.3% at a concentration of 50 ug/ml. The tyrosinase inhibitory effect, which is related to skin-whitening, was 47.2% and 45.8% in CA and CE at a concentration of 500 ug/ml. All these results suggest that Castanea crenata inner shell can be effectively used as a cosmeceutical ingredient for the prevention of wrinkles.

Study on the whitening effect and deterrent effect on gene expression of MMP-1 in wheat sprout extracts (밀싹 추출물이 MMP-1의 유전자 발현 억제효과 및 미백효과에 관한 연구)

  • This study set its purpose on to specify whether it is possible to use wheat sprout extracts for source of cosmetics, and its biological activity. Cellular experiment was conducted to apprehend toxicity of wheat sprout extracts and through study on the whitening and aging activity of skin cell. As a result, it is appeared that wheat sprout extracts have weak toxicity for HDF, B16F10 cells. Also, it is appeared that wheat sprout extracts have weak deterrent effect on melanin biosynthesis, but since the extracts impede expression of MMP-1, which is induced UV ray, the extracts hold down the aging effect by UV rays. Hence the extracts are effective on preventing wrinkles caused by UV ray and aging. Therefore, this study is expected to be utilized usefully, if the wheat sprout extracts are used as source of functional cosmetics from the perspective of preventing skin-aging in the future.

Studies on Skin Whitening Effects of Resorcinol Dipentyl Ether (레조시놀다이펜틸에터의 피부 미백 효능 평가)

  • The objective of this study was to investigate skin whitening effect of Resorcinol dipentyl ether [1,3-di(pentyloxyl)benzene] by in vitro experiments. Resorcinol dipentyl ether was prepared by alkylation of resorcinol with 1-bromopentane. The reaction products were confirmed by NMR, MS and other analytical equipments. In order to evaluate the skin safety of resorcinol dipentyl ether, the cytotoxicity of the cells constituting the skin (keratinocyte, melanocyte, fibroblast) was analyzed and similar cell viability was observed in all cell lines as compared with the control group. Inhibition of extracellular melanin synthesis effect of resorcinol dipentyl ether was approximately 65.75% at $20{\mu}g/mL$ and inhibition of intracellular melanin synthesis effect of resorcinol dipentyl ether was approximately 53.89% at $20{\mu}g/mL$. The real-time PCR and western blot analysis of mRNA expression and protein expression of tyrosinase, TRP-1, and TRP-2 related to melanogenesis revealed that melanin inhibitory effect of resorcinol dipentyl ether was inhibited from the transcription stage respectively. Finally, this study suggested applicability of Resorcinol dipentyl ether [1,3-di(pentyloxyl)benzene] as a whitening functional cosmetic new material.

The Anti-melanogenic Effect of Whitening Agent Containing Arbutin and ${\beta}-Glucosidase$ (알부틴과 베타-글루코시다제를 함유하는 미백 화장품의 항멜라닌 효과)

  • Arbutin is a glycosylated hydroquinone found at high concentration in certain plants capable of surviving extreme and sustained dehydration. It has been reported to have an inhibitory effect of melanogenesis and to be hydrolyzed easily to yield glucose and hydroquinone by ${\beta}-glucosidase.$ While hydroquinone also has an anti-melanogenic effect, however, is carefully used as a topical whitening agent because of side effects. The present study was undertaken to examine the inhibitory effect of an whitening agent containing arbutin and ${\beta}-glucosidase$ on UV radiation induced pigmentation in human skin. Experimental subjects were UVB-irradiated on the back. UVB-irradiated areas were assigned to three groups: arbutin and ${\beta}-glucosidase$ treated group, vehicle control, and no-application control. Arbutin and ${\beta}-glucosidase$ treatment inhibited pigmentation by 50.17 percent, compared with the controls (N : 10: P<0.05). These results suggest that the whitening agent containing arbutin and ${\beta}-glucosidase$may be used as an agent to inhibit melanin formation induced by UV radiation.

Whitening and Anti-aging Activities of Soluble Silkworm Gland Hydrolysate (수용성 누에 실샘 가수분해물의 미백 및 항노화 효과)

  • Silkworm-derived silk materials which contain 45 to 55% protein (18 amino acids, including 8 essential amino acids) have free radical-scavenging activities. In the present study, we investigated the whitening and anti-aging effects of silkworm gland under various conditions with B16F1 melanoma cells and human dermal fibroblasts. To evaluate the toxicity of soluble gland hydrolysate (SGH), mouse-derived B16F1 melanoma cells were treated with 0.2~5 mg/mL, cytotoxicity was not observed at all concentrations. The tyrosinase and elastase activities were significantly decreased as dosage levels of SGH increased. In addition, cell death was decreased by treated with SGH and antioxidative activity was the most effective in the SGH treatment. These findings suggest that SGH may be used as a potential functional biomaterial in having the skin whitening effect and anti-aging activity by inhibition of tyrosinase and elastase activities.

Effect of Ailanthi Radicis Cortex Extracts on Melanogenesis

  • Melanogenesis refers to the biosynthesis of melanin pigment in melanocytes. Melanogenesis is controlled by the intra- and extracellular environments. In the present study, to develop a new whitening agent, it was investigated the antioxidant activity and the inhibitory effect of Ailanthi Radicis Cortex extract on tyrosinase activity and on melanogenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of ethylacetate fraction from Ailanthi Radicis Cortex was higher than that of arbutin. The ethylacetate fraction showed scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide anion radicals in a dose dependent manner. The highest inhibitory activity of melanogenesis was also in ethylacetate fraction ($40.0{\pm}5%$ at the concentration of $400{\mu}g/ml$). This study demonstrates that the Ailanthi Radicis Cortex extract might be used to be a potential agent for skin whitening.
