Benzo[a]pyrene (B[a]P) is a polycyclic aromatic hydrocarbon and ubiquitous environmental toxin with known harmful effects to human health. Abnormal phenotypes of keratinocytes are closely associated with their exposure to B[a]P. Resorcinol is a component of argan oil with reported anticancer activities, but its mechanism of action and potential effect on B[a]P damage to the skin is unknown. In this study, we investigated the effects of resorcinol on B[a]P-induced abnormal keratinocyte biology and its mechanisms of action in human epidermal keratinocyte cell line HaCaT. Resorcinol suppressed aryl hydrocarbon receptor (AhR) activity as evidenced by the inhibition of B[a]P-induced xenobiotic response element (XRE)-reporter activation and cytochrome P450 1A1 (CYP1A1) expression. In addition, resorcinol attenuated B[a]P-induced nuclear translocation of AhR, and production of ROS and pro-inflammatory cytokines. We also found that resorcinol increased nuclear factor (erythroid-derived 2)-like 2 (Nrf2) activity. Antioxidant response element (ARE)-reporter activity and expression of ARE-dependent genes NAD(P)H dehydrogenase [quinone] 1 (NQO1), heme oxygenase-1 (HO-1) were increased by resorcinol. Consistently, resorcinol treatment induced nuclear localization of Nrf2 as seen by Western analysis. Knockdown of Nrf2 attenuated the resorcinol effects on ARE signaling, but knockdown of AhR did not affect resorcinol activation of Nrf2. This suggests that activation of antioxidant activity by resorcinol is not mediated by AhR. These results indicate that resorcinol is protective against effects of B[a]P exposure. The mechanism of action of resorcinol is inhibition of AhR and activation of Nrf2-mediated antioxidant signaling. Our findings suggest that resorcinol may have potential as a protective agent against B[a]P-containing pollutants.

The purpose of this study was to provide an economical and easy preparation method for recombinant human epidermal growth factor (rhEGF) without the need for an expensive enzyme to cleave the fusion part. However, the N-terminal fusion part is still useful for affinity chromatography. The hEGF is an important hormone in cell growth and proliferation in humans, and many studies on the expression and purification of this protein have been reported. In the present study, the hEGF gene was designed to be optimized with the E. coli codon usage preference and to contain Asn-Gly at the N-terminus of the protein. The gene was inserted into pRSET_A, an E. coli expression vector, and transformed into E. coli BL21 (DE3). The recombinant fusion protein was successfully co-expressed with pG-Tf2, a chaperone vector, in E. coli and purified by Ni-NTA column chromatography. The rhEGF was then released by hydroxylamine treatment and confirmed by SDS-PAGE. ELISA analysis showed that the activity of the free rhEGF was more than 92% similar to that of commercial EGF. The biological activity of the rhEGF was confirmed by a cell proliferation test with human skin fibroblasts.

This study focuses on the preparation of acetaminophen (AP) imprinted functional biomaterials for a transdermal drug delivery using mung bean starch (MBS), polyvinyl alcohol (PVA), sodium benzoate (S) as a crosslinking agent, glycerol (GL) as a plasticizer, and melanin (MEL) as a photothermal agent. The prepared AP imprinted biomaterials were characterized using FE-SEM and their physical properties were evaluated. The photothermal effect and AP release property for functional biomaterials were examined with the irradiation of near infrared (NIR) laser (1.5 W/cm2). When the NIR laser was irradiated on functional biomaterials with/without the addition of MEL, the temperature of MEL added biomaterial increased from 25 ℃ to 41 ℃, whereas the biomaterial without MEL increased from 25 ℃ to 28 ℃. Results indicate that there is the photothermal effect of prepared biomaterial with the addition of MEL. Based on the results, AP release properties were evaluated using standard buffer solutions and artificial skin. It was found that AP release rates of MEL added AP loaded biomaterials were 1.2 times faster than those of MEL non-added AP loaded biomaterials when irradiating with NIR laser. We envision that the developed functional biomaterials can be utilized for an acute pain-killing treatment.
Various dietary pigments are added to processed foods to improve their sensory and commercial properties. In this study, autoclave sterilization (121℃ for 15 min at 15 psi) was performed on 34 food pigments, and changes in their color stability and antioxidant activity were analyzed. The autoclaving process drastically reduced the peak color intensities of water-soluble paprika and beet red (BR) by ~90%. Turmeric oleoresin (TO), water-soluble β-carotene, and grape skin color were also unstable and showed a remaining color intensity of 45-60%. The colors of all the synthetic pigments tested were stable under this process. The scavenging activities of BR and paprika against ABTS, DPPH, and AAPH radicals decreased significantly, whereas those of TO were enhanced after the autoclaving treatment. The results suggest that the chemical and bioactive properties of certain dietary pigments are affected by the autoclaving process, and this phenomenon should be considered during food processing.

Objectives : The objective of this study is to predict how future infectious diseases will affect healthcare utilization patterns within korean medicine hospitals and, specifically, departments of ophthalmology, otolaryngology and dermatology of Korean medicine. Methods : We retrospectively analyzed medical records of patients who visited Kyung Hee University Korean Medicine Hospital during Pre COVID-19(from March 1st, 2018 to Feb 29th, 2019) and Post COVID-19(from March 1st, 2019 to Feb 28th, 2022). The data was analyzed by visit type and diagnosed disease code. Results : 1. After the COVID-19 break-out, the number of first visit patients increased but those of revisit patients, total patients, and revisit/first visit ratio decreased in all departments. 2. In Kyung Hee University Korean Medicine Hospital, the decrease rate of the number of total patients diagnosed with diseases of respiratory system(J00-J99) was the highest, while that for diseases of circulatory system(I00-I99) was the lowest. 3. In the department of Ophthalmology and Otolaryngology of Korean Medicine, the decrease rate of the number of total patients with nose disease was the highest, followed by eye disease and ear disease. 4. In the department of Dermatology of Korean Medicine, the number of total patients with dermatitis remained consistent, while the number of total patients with skin appendage disease showed relatively high decrease rate. Conclusions : Severe or chronic diseases showed relatively low decrease rate of the number of total patients. While mild diseases or diseases with high probability of exposure to COVID-19 infection during treatment showed high decrease rate of the number of total patients.

Earthing, caused by direct skin contact with the Earth's surface, is used to reduce the symptoms of inflammation (fever, fever, swelling and pain). However, there is little evidence to support the anti-inflammatory effects of earthing mattresses. Therefore, this study was conducted to investigate whether anti-inflammatory effect of earthing mattress using an in vivo animal model. The anti - inflammatory effect was evaluated by measuring ear thickness and foot volume in 12-O-tetradecanoylphorbol-13 acetate (TPA) - induced ear edema and carrageenan - induced paw edema model, respectively. Balb/c mouse in carrageenan paw edema model showed significant anti - inflammatory effect in the group treated with earthing mattress for 4 hours or 24 hours for 3 days. For females, the anti-inflammatory effect was greater when the earthing mattress was added to the mattress than the mattress alone treatment. From the above results, it was found that the female responds more to the effect of the earthing as well as the mattress effect. In addition, when the male and female Balb/c mice were exposed to mattresses and earthing mattresses for 24 h for 3 days, respectively, the mattress and earthing mattresses showed significant inhibition of IL (Interleukin)-1β levels compared to the control. In the TPA ear edema model, Balb/c mouse showed significant anti - inflammatory effect in the group treated with the earthing mattress for 4 hours or 24 hours for 3 days. Both males and females showed more anti-inflammatory effects when they were exposed to earthing mattresses with mattresses added to the mattresses. From the above results, it was found that both male and female respond to the effect of earthing as well as the mattress effect in the TPA ear edema model. In conclusion, in this study, we have verified that earthing mattress shows inhibitory effects on TPA and carrageenan-induced inflammation. From these results, it is suggested that the anti-inflammatory effect can be expected by applying the earthing mattress to patients suffering from inflammatory diseases. However, there is a need to pinpoint exactly how the earthing mattress relieves inflammation, and further research is needed to investigate the mechanism.
The leaves, stems, seeds, and roots of Dendropanax morbifera have been used since ancient times as folk medicines for the treatment of headaches, skin diseases, infectious diseases, and other ailments. This study investigated the antioxidant, alcohol metabolism, and hepatoprotective effects of D. morbifera leaf and stem water extracts. The total polyphenol content of the D. morbifera leaf and stem water extracts was 49.56 mg tannic acid equivalent (TAE)/g, and the 1,1-diphenyl-2-picrylhydrazy (DPPH) radical scavenging activity of the D. morbifera leaf and stem water extracts was 84.09% at 1,000 ㎍/ml concentration. The effects of D. morbifera leaf and stem water extracts on alcohol metabolism were determined by measuring the generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). The ADH and ALDH activities of D. morbifera leaf and stem water extracts were increased in a dose-dependent manner at 37.68% and 41.67%, respectively, at a 1,000 ㎍/ml concentration. The D. morbifera leaf and stem water extracts showed significant protective effects against tacrine-induced cytotoxicity in HepG2 cells at 50 ㎍/ml. Based on our results, we concluded that D. morbifera leaf and stem water extracts may be used as major pharmacological agents, such as antioxidants, alcohol metabolism, and anti-hepatitis remedies.

Objectives : The objective of this study is to identify the characteristics of outpatients and the frequently diagnosed diseases in dermatology of Korean medicine through periodic research and comparison with previous studies. Methods : We analyzed medical records of new outpatients who visited department of dermatology at Kyung Hee university Korean medicine hospital between January 1, 2018 and December 31, 2022. We analyzed the patient demographic characteristics and the frequency of diagnosed diseases. Then we compared these results with previous studies in 2000, 2007 and 2016. Results : 1. Average number of patients and proportion of male patients have increased from 1996 to 2022. 2. From 1996 to 2022, there was a decrease in the proportion of patients aged 0-39 and an increase in the proportion of patients aged over 40. 3. The distribution proportions of eczema, atopic dermatitis, and rosacea during 2018-2022 have increased compared to those during 2011-2015. 4. The distribution of diseases according to age appears to vary based on the epidemiological characteristics of the diseases. 5. The distribution of diseases according to gender also appears to vary based on the epidemiological characteristics of the diseases, as well as cosmetic issues related to the affected areas. Conclusions : Considering the sustained trend of a high proportion of non-infectious inflammatory diseases since 1996, it is important to enhance treatment capacity through continuous research.
Objectives : The purpose of this study is to understand the temperature characteristics depending on the thickness and material of the needle used with the Electronic Warm Acupuncture Device (EWAD). Methods : We controlled experimental environment and measured temperature changes of a silicon phantom in which K-type thermocouples were inserted at depths of 2, 7 mm. EWAD perfomed with acupuncture needles of various thicknesses (0.25×60 mm, 0.40×60 mm, and 0.50×60 mm) and materials (Gold 0.40×50 mm). We set non-needle (only heated with EWAD skin heater) group as a control group. Results : The maximum temperature and temperature changes of 0.40 mm, 0.50 mm needle group were significantly higher than the non-needle group. The highest temperature range in all needle groups was 0.50 mm needle group (41.44±0.31℃). However, the 0.25 mm needle group was not significantly different from the non-needle group. Maximum temperature of gold needle group was significantly higher than stainless steel needle group. Temperature changes of gold needle group were higher than stainless steel group at the depth of 7 mm. Conclusions : It was found that needle thickness and material of acupuncture had an effect on the temperature of the EWAD. When performing EWAD treatment, consideration of thickness and material of acupuncture is needed. Future research is needed using phantoms that can reflect actual clinical situations and better mimic the human body.

Hesperetin(HT) is a potent antioxidant flavonoid aglycone derived from hesperidin(HD). The antioxidant, anti-inflammatory, and antimicrobial activities of HT and its cyclodextrin(CD) inclusion complexes were compared in vitro. HT was prepared by enzymatic hydrolysis of HD, and HT/CD complexes were prepared using 𝛽-cyclodextrin(𝛽-CD) and hydroxypropyl-𝛽-cyclodextrin(HP-𝛽-CD) by solvent co-evaporation method. The solubility of the HT/HP-𝛽-CD inclusion complex increased 93.5-fold compared to HT, and the solubility of HT/𝛽-CD increased 22.5-fold. The HT/HP-𝛽-CD inclusion complex showed a similar effect as HT on radical scavenging activity in antioxidant assays, whereas the HT/𝛽-CD inclusion complex showed slightly lower activity than HT. Cytotoxicity was low in the following order; HT/HP-𝛽-CD, HT/𝛽-CD, and HT in murine macrophage RAW264.7 cells. Treatment with HT and HT/CD inclusion complexes reduced the levels of inflammatory mediators such as nitric oxide(NO), tumor necrosis factor-𝛼(TNF-𝛼) and interleukin-6(IL-6) in the cells. HT and HT/HP-𝛽-CD inclusion complex were more effective than HT/𝛽-CD inclusion complex at relatively low concentrations. Inhibitory effects were tested on skin-pathogenic bacteria, Staphylococcus aureus and Pseudomonas aeruginosa, and they showed an antimicrobial effect on S. aureus in the order of HT = HT/HP-𝛽-CD > HT/𝛽-CD, but they did not show any significant inhibitory effect on P. aeruginosa. In conclusion, HT, the aglycone form of HD, and its CD inclusion complexes showed various biological activities. HT/HP-𝛽-CD inclusion complex, which is the highly soluble form of HT, showed relatively higher activity compared to HT/𝛽-CD inclusion complex.
