피부 마이크로바이옴의 연구 동향 분석 (Analysis of Research Trends in the Skin Microbiome)

  • 피부에는 박테리아, 바이러스, 곰팡이를 포함하는 마이크로바이옴이 인체세포와 공존하고 있으며, 최근 피부 및 화장품 분야에서 피부 미생물 구성물과 피부 건강, 질병과의 연관성에 대해 많은 연구가 이루어지고 있을 뿐만 아니라 마이크로바이옴 화장품의 개발이 활발한 상황이다. 피부 마이크로바이옴은 매우 다양하며, 피부에서 구성비나 서식하는 부분이 다르지만 다양한 방법으로 상호작용을 통해 피부의 영양을 공급하거나 경쟁자인 병원균의 증식을 제한하고 있지만, 피부 마이크로바이옴과 병원균의 균형이 깨지면 면역 항상성의 파괴에 기여하고 피부 질환의 발병으로 이어질 수 있다. 이에 따라 본 논문은 우리 피부에 공생관계인 피부 마이크로바이옴의 역할과 연구 동향에 대해 파악하고, 마이크로바이옴 균형에 도움이 되는 바이오틱스 소재에 관한 산업동향을 고찰함으로써 미래의 큰 성장 가능성을 가진 피부 마이크로바이옴 시장의 중요한 자료로 사용될 수 있을 것으로 사료된다.

3차원 배양 인공피부를 활용한 마이크로바이옴 추출물의 선정 및 이를 함유한 에멀젼 제형의 피부주름개선 임상 평가 (Effect of Skin Wrinkle Reduction Using Emulsions with Microbiome Extracts Selected by 3D Human Skin Equivalents)

  • 마이크로바이옴의 화장품 응용에 대한 관심이 높아지고 마이크로바이옴 추출물의 복합적인 효능에 의한 피부 개선 연구가 중요해지고 있다. 본 연구에서는 세포 및 3차원 배양 인공피부를 이용한 in vitro 시험법을 통하여 마이크로바이옴 중 피부 주름 개선 효능이 뛰어난 Bifidobacterium bifidum (B. bifidum)을 선정하였고, 이의 추출물을 함유한 화장품의 피부주름개선 효능평가를 진행하였다. 이후, 비피다발효여과물 원료의 세포독성 및 항산화, 항염 및 주름개선 유효성 시험을 실시하였다. 세포독성이 없고 효능이 우수한 5% (v/v) 농도의 비피다발효여과물이 함유된 에멀젼을 제조하였고 placebo 에멀젼와 비교하여 총 21 명 여성을 대상으로 2 주간 임상시험을 수행하였다. Antera 3D를 사용하여 눈가 부위의 주름을 기기 평가하였다. 주름 개선도는 placebo 에멀젼 사용군 대비하여 주름 감소율(%)에서 5.6% 차이를 보였다. 이러한 3차원 배양 인공피부를 활용한 마이크로바이옴 추출물 선정 및 이를 함유한 화장품의 효능 연구는 다양한 효능을 가지는 마이크로바이옴 화장품의 개발 및 메커니즘 연구에 응용될 수 있을 것이라 기대된다.

The Microbiome-Immune Axis Therapeutic Effects in Cancer Treatments

  • During the last decades, research and therapeutic methods in cancer treatment have been evolving. As the results, nowadays, cancer patients are receiving several types of treatments, ranging from chemotherapy and radiation therapy to surgery and immunotherapy. In fact, most cancer patients take a combination of current anti-cancer therapies to improve the efficacy of treatment. However, current strategies still cause some side effects to patients, such as pain and depression. Therefore, there is the need to discover better ways to eradicate cancer whilst minimizing side effects. Recently, immunotherapy, particularly immune checkpoint blockade, is rising as an effective anti-cancer treatment. Unlike chemotherapy or radiation therapy, immunotherapy has few side effects and a higher tumor cell removal efficacy depend on cellular immunological mechanisms. Moreover, recent studies suggest that tissue immune responses are regulated by their microbiome composition. Each tissue has their specific microenvironment, which makes their microbiome composition different, particularly in the context of different types of cancer, such as breast, colorectal, kidney, lung, and skin. Herein, we review the current understanding of the relationship of immune responses and tissue microbiome in cancer in both animal and human studies. Moreover, we discuss the cancer-microbiome-immune axis in the context of cancer development and treatment. Finally, we speculate on strategies to control tissue microbiome alterations that may synergistically affect the immune system and impact cancer treatment outcomes.

Lactobacillus plantarum APsulloc 331261 발효 용해물의 피부 미생물 조절 효과 (Lactobacillus plantarum APsulloc 331261 Fermented Products as Potential Skin Microbial Modulation Cosmetic Ingredients)

  • 피부에는 여러 종의 미생물들이 군집을 이루며 서식하고 있고, 서로 상호작용을 하며 균형을 유지하고 있다. 하지만, 여드름, 건선 및 아토피 피부염과 같은 질병 상태에서는 피부 미생물의 균형이 깨져 건강한 피부와는 다른 미생물 군집 구성을 보인다. 피부 미생물 군집 구성의 조절은 이러한 피부 질환에 대한 잠재적인 치료 방법이 될 수 있다. Quorum sensing (QS)은 세포간 전달인자로, QS을 조절하면 유해균의 바이오필름 형성이나 효소 분비등을 억제할 수 있고, 이 역시 피부 미생물 군집 구성에 영향을 준다. 이번 연구에서는 아모레퍼시픽의 특허 성분인 유산균 발효 용해물(Lactobacillus plantarum APsulloc 331261, KCCM 11179P 이용)의 QS억제능력 및 피부 유익균과 피부 유해균의 생장에 미치는 영향을 확인하였다. 유산균 발효 용해물 10 ㎍/mL 처리시, QS에 의해 제어되는 Chromobacterium violaceum의 자주색 안료 생산을 대조군 대비 27.4% 감소시켰다. 또 피부 유익균인 Staphylococcus epidermidis의 생장을 대조군 대비 12% 증가 시켰고, 피부 유해균인 Pseudomonas aeruginosa의 생장을 38.5% 감소, 바이오필름 형성을 17.7% 감소시켰다. S. epidermidis를 같은 속(genus)에 포함되어 있는 대표적인 피부 유해균 Staphylococcus aureus와 함께 공배양 하였을 때 S. epidermidis의 생장은 대조군 대비 134% 증가시켰고, S. aureus의 생장은 13% 억제시켰다. 이러한 결과들을 종합하였을 때 L. plantarum APsulloc 331261을 이용한 발효 용해물은 피부 미생물의 균형을 조절해 줄 수 있는 화장품 원료로 유용하게 사용 될 수 있다고 사료된다.

신규 Sporichthyaceae Bacterium Strain K-07 배양액의 피부장벽, 보습 및 항염증 활성 (Novel Sporichthyaceae Bacterium Strain K-07 Skin Barrier, Moisturizing and Anti-inflammatory Activity)

  • 피부의 생태계는 미생물에게 다양한 형태의 서식처를 제공하며, 광범위한 미생물들이 살고 있다. 숙주인 사람은 이들과 공생관계를 이루고 있으며, 이들은 숙주에 많은 긍정적인 영향을 미친다. 피부에 분포하는 미생물들의 다양한 대사물질은 피부세포에 영향을 미치고, 피부장벽 기능, 노화방지 및 항염증에 광범위한 효능을 나타내는 것으로 알려져 있다. 본 연구에서는 사람의 피부에서 신규한 Sporichthyaceae bacterium strain K-07을 분리하였고, 해당 미생물의 16S rRNA 분석결과 Sporichthya속의 미생물과 상동성이 93.4%인 것으로 신규속(genus)으로 확인되었다. 그리고 신규로 분리된 K-07 배양액을 처리하였을 때, HaCaT cell에 어떤 변화가 나타나는지에 대한 분석을 실시하였다. 분리된 신규 미생물 strain K-07의 16S rRNA sequence 분석결과 상동성이 93.4% 이하로 확인되었고, 보고되지 않은 새로운 종으로 확인되었다. Filaggrin, cluadin1, claudin4, ${\alpha}SMase$, 및 CerS3 / HAS3 및 aquaporin3 / IL-6 및 TNF-${\alpha}$ / TSLP 및 TARC를 대상으로 strain K-07 배양액을 처리하여 변화를 관찰하였다. 그 결과 filaggrin, cluadin1, claudin4, ${\alpha}$SMase, 및 CerS3 / HAS3 및 aquaporin3에서는 음성 대조군 대비 우수한 증가 효과가 나타남을 확인하였다. 그리고 IL-6 및 TNF-${\alpha}$ / TSLP 및 TARC에 대해서도 우수한 억제능을 나타내는 것을 확인하였다. 결론적으로 신규 미생물 strain K-07 배양액은 피부 장벽활성 증진에 매우 효과적인 작용을 하며, 염증 억제에 우수한 효능을 나타내는 것으로 확인되므로 피부에 효과적인 소재로 사용될 수 있을 것이다.

개 아토피 피부염에서 3종 프로바이오틱스 복합제의 임상 효능 평가 (Clinical efficacy of L. plantarum, L. reuteri, and Ped. acidilactici probiotic combination in canine atopic dermatitis)

  • Canine atopic dermatitis (CAD) is an inflammatory and pruritic skin disease with a genetic predisposition, characterized by allergic sensitivity. It is known for its distinctive clinical features, including a high recurrence rate and chronic progression. To manage CAD, medications such as steroids and immunosuppressants are commonly used, but consideration should be given to the potential resistance and side effects associated with long-term use. In order to reduce these risks, various adjunctive factors are currently under consideration. One of these adjunctive agents, probiotics have shown effectiveness in regulating atopic dermatitis by modulating immune responses, as demonstrated in several recent studies. In this study, a substance combining three probiotics-L. plantarum, L. reuteri, and Ped. Acidilactici-was used in patients diagnosed with CAD, and its clinical effects and safety were evaluated. The trial involved four groups: a group receiving conventional treatment for atopic dermatitis (A), a group prescribed low-dose probiotics (B), a group prescribed high-dose probiotics (C), and a group prescribed topical probiotics (D). For assessment, the Canine Atopic Dermatitis Extent and Severity Index (CADESI), Trans-Epidermal Water Loss (TEWL) test, gut microbiome, and serum IgE test were conducted. As a result, the CAD severity index (CADESI-4) significantly decreased in the probiotics groups (B & C). In the serum total IgE test, the groups consuming probiotics showed a significant difference, while the group using topical probiotics (D) did not exhibit a significant change. Also, the TEWL test showed improved scores in the probiotics groups (B & C). Therefore, L. plantarum, L. reuteri, and Ped. Acidilactici probiotic combination could be considered as an effective adjunctive treatment, especially for atopic patients with moderate to severe skin lesions.

Influence of Panax ginseng formulation on skin microbiota: A randomized, split face comparative clinical study

  • Background: Skin microbiota is important for maintenance of skin homeostasis; however, its disturbance may cause an increase in pathogenic microorganisms. Therefore, we aimed to develop a red ginseng formulation that can selectively promote beneficial bacteria. Methods: The effects of red ginseng formulation on microorganism growth were analyzed by comparing the growth rates of Staphylococcus aureus, S. epidermidis, and Cutibacterium acnes. Various preservatives mixed with red ginseng formulation were evaluated to determine the ideal composition for selective growth promotion of S. epidermidis. Red ginseng formulation with selected preservative was loaded into a biocompatible polymer mixture and applied to the faces of 20 female subjects in the clinical trial to observe changes in the skin microbiome. Results: Red ginseng formulation promoted the growth of S. aureus and S. epidermidis compared to fructooligosaccharide. When 1,2-hexanediol was applied with red ginseng formulation, only S. epidermidis showed selective growth. The analysis of the release rates of ginsenoside-Rg1 and -Re revealed that the exact content of Pluronic F-127 was around 11%. The application of hydrogel resulted in a decrease in C. acnes in all subjects. In subjects with low levels of S. epidermidis, the distribution of S. epidermidis was significantly increased with the application of hydrogel formulation and total microbial species of subjects decreased by 50% during the clinical trial. Conclusion: We confirmed that red ginseng formulation with 1,2-hexanediol can help maintain skin homeostasis through improvement of skin microbiome.

Superficial and Deep Skin Preparation with Povidone-Iodine for Ventriculoperitoneal Shunt Surgery : A Technical Note

  • Objective : Ventriculoperitoneal (VP) shunt surgery is a common and effective treatment for hydrocephalus and cerebrospinal fluid disorders. Infection remains a major cause of morbidity and mortality after a VP shunt. There is evidence that a deep skin flora microbiome may have a role to play in post-operative infections. In this technical note, we present a skin preparation technique that addresses the issue of the skin flora beyond the initial incision. Methods : The patient is initially prepped, as standard, with. a single layer of 2% CHG+70% isopropyl alcohol. The novel stage is the 'double incision' whereby an initial superficial incision receives a further application of povidone-iodine prior to completing the full depth incision. Results : Of the 84 shunts inserted using the double-incision method (September 2015 to September 2016), only one developed a shunt infection. Conclusion : The double incision approach to skin preparation is a unique operative stage in VP shunt surgery that may have a role to play in reducing acute shunt infection.

Effects of cooling systems on physiological responses and intestinal microflora in early gestating sows exposed to high-temperature stress

  • This study was conducted to investigate the effect of cooling systems on reproductive performance, body temperature, blood metabolites, and the intestinal microbiome in early gestating sows exposed to high ambient temperature. In total, 39 pregnant sows (Landrace × Yorkshire; 2 parities) were randomly assigned to and maintained in the following three treatment groups (13 sows per group) over days 0 to 35 of pregnancy: (i) air cooling (AC; 26.87 ± 1.23℃), (ii) water-drip cooling (WC; 28.81 ± 0.91℃), and (iii) a lack of cooling with heat stress (HS; 30.72 ± 0.70℃). Backfat thickness was measured before and after HS. Feces were collected on day 0 and 35 d of the trial for microbiome analysis, whereas blood was taken at day 35 of pregnancy and analyzed. Reproductive performance and physiological responses were identified at day 35. Respiration rate along with rectal and skin temperatures were lower (p < 0.05) in the AC group than in the HS and WC groups. Serum blood urea nitrogen values were increased (p < 0.05) in the WC group compared with those measured in the AC and HS groups. Triiodothyronine was found at greater levels (p < 0.05) in the AC than in the HS group. Reproductive performance was not affected by the cooling systems. At the phylum level, fecal pathogenic Spirochaete and Euryarchaeota were found in higher numbers (p < 0.05) in all groups after HS. Similarly, at the genus level, the amount of Treponema was greater (p < 0.05) in all groups after HS. In conclusion, our results suggest that AC or WC can ameliorate or mitigate the adverse effects of HS on the physiological parameters of pregnant sows reared under high temperatures.

Breast abscess caused by Staphylococcus aureus in 2 adolescent girls with atopic dermatitis

  • Atopic dermatitis (AD) is a chronic inflammatory skin disease in children. Patients with AD experience a high rate of colonization of the skin surface by Staphylococcus aureus. Because of a skin barrier defect, there is a potential risk of staphylococcal invasive infection in patients with AD. Here, we present 2 cases of breast abscess caused by S. aureus in 2 adolescent girls with severe AD. Methicillin-sensitive S. aureus was identified from the breast abscess material. They were treated with appropriate antibiotics, however surgical drainage of the abscess was needed in case 1. Identical strains were found from the breast abscess material as well as the lesional and the nonlesional skin of the patients through matrix-assisted laser desorption/ionization time-of-flight analysis. We characterized the differential abundance of Firmicutes phylum in patients' skin in microbiota analysis. In particular, S. aureus, a member of Firmicutes, differed significantly between the lesional and the normal-appearing skin. Our cases demonstrate the potential severity of bacterial deep tissue infection in AD and the dysbiosis of skin microbiota may be involved in inflammation in AD.
