Objectives : The Skin is composed of multiple layers, including the epidermis, dermis, and hypodermis. It provides a vital barrier structure that protects vertebrates from external environmental antigens, solvents, ultraviolet light, microorganisms, toxins, and weather conditions. Although several biological effects of Mori Follium have been reported, beneficial effects of Mori Follium in skin health remain unclear. In this study, we prepared water extract of Mori Follium (MLE) and evaluated the effects on melanin accumulation and expression levels of skin fibril-related proteins.Methods : The cytotoxicities of MLE in B16F10 melanoma and human skin fibroblasts (HSF) were examined by MTT assay. Inhibitory effect of MLE on the α-MSH- and IBMX-induced melanosis in B16F10 melanoma was examined. The expression levels of fibronectin, collagen 1α2, and CCN2 in MLE-treated HSF were analyzed by reverse transcription-polymer chain reaction (RT-PCR) and western blotting.Results : The MLE treatment for 24 h did not affect to the B16F10 and HSF at concentrations of 1, 10, 50, 100, 200, 400 and 800 ㎍/ml. The MLE treatment for 72 h significantly and dose dependently suppressed melanin accumulation in B16F10 melanoma. In addition, the MLE treatment up-regulated expression levels of skin fibril-related genes such as fibronectin, collagen 1α2, and CCN2 in HSF. Our western blot analysis revealed MLE-induced up-regulation of skin fibril-related genes required the activation of CCN2 protein.Conclusions : In conclusion, these findings suggest that the MLE could be used in development of cosmetic natural material of maintaining healthy skin.
Background: Panax ginseng is regarded as one of the best compounds for promoting health, and it has been used traditionally as a medicinal herb. Recently, Korean Red Ginseng (RG) has been shown to protect skin from aging and wrinkling; it can also relieve atopic dermatitis and allergy symptoms. This study aimed to evaluate RG's effects on the regeneration of the full-thickness skin wounds in rat. Methods: Full-thickness skin wounds were generated in rats, and then RG was administered either orally or topically. The wound-healing effects of RG were investigated by assessing wound size, mRNA expression patterns of genes related to wound healing, histological staining, and measurements of lipid, moisture, and elasticity in skin tissues. Results: The wound size was smaller, and tissue regeneration rate was faster in the RG-treated group than that in the control group on days 15 and 20 after initiating treatment. On postoperative day 20, skin lipid and moisture content had increased significantly in the RG-treated group. Significant increases in the gene expression levels of transforming growth $factor-{\beta}1$ and vascular endothelial growth factor were found in the RG group during the early stages of wound healing. Matrix metalloproteinase-1 and matrix metalloproteinase-9 showed significant increases in gene expression levels on day 20. Conclusion: The results suggested that RG may promote healing of full-thickness skin wounds in rats. They also provided basic insights into the effects of RG on skin regeneration, supporting its use as a dressing material for wound treatment and its development as a functional food.

It is well-documented that decreased antioxidant defense system by ultraviolet(UV) irradiation is the most important reason to induce the skin aging, especially photoaging. Chlorogenic acid(CA), a nonflavonoid catecholic compound, is present in the diet as part of fruits, tea, coffee and wine and has been reported to have anti-inflammatory, antimutagenic and anticarcinogenic activities. In this study, we examined the effects of CA on the UV -induced photoaging. Firstly, we investigated the protective effect of CA on antioxidant defense system in HaCaT human keratinocytes after UV irradiation treatment. UV irradiation decreased antioxidant defence enzyme activities of superoxide dismutase, catalase and GSH contents, which were restored by CA. To elucidate the effect of CA, 1% of CA and vehicle were applied to human buttock skin before and after UV irradiation (2MED). CA prevented UV -induced matrix metalloproteinase-1 mRNA expression and procollagen mRNA depression. And CA also increased CD1a(Langerhans cell) expression significantly. Our results suggest that CA has protective effects on UV -induced photoaging by increasing cellular antioxidant defense system. Therefore, CA may be a useful anti-aging agent for cosmetic purpose.

In 2022, the domestic production performance of functional cosmetics in South Korea reached 4.6 trillion won, accounting for 33.85% of the total cosmetics production. The number of functional cosmetics reviewed increased by about 7.5% from the previous year, totaling 974 items. Especially with the increasing importance of the skin barrier function due to skin sensitivity caused by various environmental pollutants, domestic cosmetic companies are showing interest in the development of new ingredients and products related to this area. This study aims to analyze academic research trends related to in vitro experiments for the development of cosmetics improving the skin barrier, to provide practical information for the cosmetic industry. The findings are as follows: Academic research mainly focused on the efficacy of natural ingredients in improving the skin barrier, but there is a significant lack of quantitative accumulation of research. For the development of skin barrier-improving cosmetic ingredients, efficacy evaluation indicators were set, including hyaluronic acid production, expression of filaggrin gene, loricrin, formation of cornified envelope (CE), and expression of ceramide synthesis enzyme genes. Moreover, effective cosmetic ingredients for improving the skin barrier included lemongrass and perilla leaf extracts, flavonoids, Lactococcus lactis subsp. lactis, Exosomelike Nanovesicles derived from apple callus, Eleutherococcus sessiliflorus, Acanthopanax sessiliflorus, Eleutherococcus gracilistylus, Acer okamotoanum extracts, Aloe vera adventitious root extract, ethanol extract of Aruncus dioicus, and organic solvent fraction of Dracocephalum argunense.
Ultraviolet (UV) irradiation is well known to cause human skin aging and skin cancer through activation of matrix metalloproteinases (MMPs) which are responsible for the degradation of collagen, an extracellular matrix component. However, the molecular mechanisms of UV-induced MMP expression are yet to be defined. The aim of this research was to investigate the dynamic changes of the level of total prolactin receptor (PRLR) mRNA and the short form prolactin receptor (S-PRLR) mRNA in skin of cashmere goats from the initiation of cashmere fibre growth to active growth. Eighteen half-sib wethers were allocated randomly to two groups. Melatonin implants were used in order to initiate growth of cashmere fibre before the normal time and reduce blood plasma prolactin (PRL) concentration. Real-time reverse transcription quantitative polymerase chain reaction (real-time PCR) was used to determine PRLR mRNA expression levels of skin from June to November. The results showed that, in Chinese Inner Mongolia cashmere goats, there were seasonal variations in expression of total PRLR mRNA in skin with levels decreasing from June to October. Synchronously, the cashmere fibre growth rate gradually increased during this period, but the expression levels of S-PRLR mRNA did not decrease along with seasonal variation from initiation to active growth of cashmere fibre. These results suggest that expression levels of S- PRLR mRNA might be involved in the process of cashmere growth. It was also possible that the change of alternative splicing of PRLR occurred in the skin of cashmere goats from proanagen to anagen.

Chitinase-3-like-1 (CHI3L1) is known to induce inflammation in the progression of allergic diseases. Previous our studies revealed that 2-({3-[2-(1-cyclohexen-1-yl)ethyl]-6,7-dimethoxy-4-oxo-3,4-dihydro-2-quinazolinyl}sulfanyl)-N-(4-ethylphenyl)butanamide (K284-6111; K284), the CHI3L1 inhibiting compound, has the anti-inflammatory effect on neuroinflammation. In this study, we investigated that K284 treatment could inhibit the development of atopic dermatitis (AD). To identify the effect of K284, we used phthalic anhydride (5% PA)-induced AD animal model and in vitro reconstructed human skin model. We analyzed the expression of AD-related cytokine mediators and NF-κB signaling by Western blotting, ELISA and quantitative real-time PCR. Histological analysis showed that K284 treatment suppressed PA-induced epidermal thickening and infiltration of mast cells. K284 treatment also reduced PA-induced release of inflammatory cytokines. In addition, K284 treatment inhibited the expression of NF-κB activity in PA-treated skin tissues and TNF-α and IFN-γ-treated HaCaT cells. Protein-association network analysis indicated that CHI3L1 is associated with lactoferrin (LTF). LTF was elevated in PA-treated skin tissues and TNF-α and IFN-γ-induced HaCaT cells. However, this expression was reduced by K284 treatment. Knockdown of LTF decreased the expression of inflammatory cytokines in TNF-α and IFN-γ-induced HaCaT cells. Moreover, anti-LTF antibody treatment alleviated AD development in PA-induced AD model. Our data demonstrate that CHI3L1 targeting K284 reduces AD-like skin inflammation and K284 could be a promising therapeutic agent for AD by inhibition of LTF expression.

Ultraviolet B (UVB) is a primary environmental factor that induces adverse effects on skin such as photoaging, skin burn and cancer. UVB also increases the expression of $11{\beta}$-hydroxysteroid dehydrogenase type 1 ($11{\beta}-HSD1$), which converts inactive cortisone to active cortisol in response to a variety of stressors in target tissues. Thus, we have screened new herbal extracts that suppress $11{\beta}-HSD1$ expression induced by UVB in human dermal fibroblasts. We also investigated whether Trapa japonica (TJ) extract and its fractions inhibit UVB-induced photoaging in Hs68 cells and 3D skin model. Results showed that TJ extract inhibited the increase of $11{\beta}-HSD1$ expression in UVB-exposed Hs68 cells significantly. TJ extract and its fractions not only inhibited $11{\beta}-HSD1$ expression, but also suppressed the increase of matrix metalloproteinases (MMP-1, 3, 9) and proinflammatory cytokines (IL-6, 8) in UVB-irritated Hs68 cells. TJ extract also inhibited MMP-1 expression in UVB-irritated 3D skin model. In addition, TJ extract recovered UVB-induced decreases of epidermal thickness and PCNA-positive cells in 3D skin model. Taken together, these results suggest that TJ extract and its fractions inhibit UVB-induced skin photoaging by interfering with $11{\beta}-HSD1$ and MMPs.

Skin basement membrane (BM) is a specialized structure that binds dermis and epidermis of the skin and plays an important role in maintaining skin structure. Structural change and destruction of BM is reported to appear due to UV exposure and aging, which may contribute to skin aging including wrinkle formation and a decrease in elasticity of the skin. One of the key components of the BM is laminin-332 (LN-332), and is a major contributor to epidermal-dermal attachment. In this study, we elucidated the effects of Meladrium firmum hexane fraction (MFHF) on LN-332 expression in HaCaT, a human keratinocyte cell line. Quantitative real-time PCR (RT-PCR) and immunoblot analysis revealed that MFHF induced upregulation of LN-332 gene and protein expression. Next, cells were treated with p38 MAPK inhibitor (SB202190) prior to MFHF treatment to analyze the signaling pathway contributing to LN-332 expression. The mRNA and protein levels of LN-332 expression were suppressed completely by pretreatment with p38 MAPK inhibitor. Furthermore, MFHF also increased the mRNA level of collagen type VII and integrin ${\alpha}6$ of skin BM component. These results collectively suggest that MFHF may have potential as an effective agent to stimulate the synthesis of BM components, and could be used to improve phenomenon of skin aging ascribed to the structural and functional impairments of BM in aged human skin.
BACKGROUND/OBJECTIVES: Ultraviolet B (UVB) irradiation on skin can induce production of reactive oxygen species (ROS), which cause expression of matrix metalloproteinases (MMPs) and collagen degradation. Thus, chronic exposure of skin to UVB irradiation leads to histological changes consistent with aging, such as wrinkling, abnormal pigmentation, and loss of elasticity. We investigated the protective effect of the standardized green tea seed extract (GSE) on UVB-induced skin photoaging in hairless mice. MATERIALS/METHODS: Skin photoaging was induced by UVB irradiation on the back of Skh-1 hairless mice three times per week and UVB irradiation was performed for 10 weeks. Mice were divided into six groups; normal control, UVB irradiated control group, positive control (UVB + dietary supplement of vitamin C 100 mg/kg), GSE 10 mg/kg (UVB + dietary supplement of GSE 10 mg/kg), GSE 100 mg/kg (UVB + dietary supplement of GSE 100 mg/kg), and GSE 200 mg/kg (UVB + dietary supplement of GSE 200 mg/kg). RESULTS: The dietary supplement GSE attenuated UVB irradiation-induced wrinkle formation and the decrease in density of dermal collagen fiber. In addition, results of the antioxidant analysis showed that GSE induced a significant increase in antioxidant enzyme activity compared with the UVB irradiation control group. Dietary supplementation with GSE 200 mg/kg resulted in a significant decrease in expression of MMP-1, MMP-3, and MMP-9 and an increase in expression of TIMP and type-1 collagen. CONCLUSIONS: Findings of this study suggest that dietary supplement GSE could be useful in attenuation of UVB irradiation-induced skin photoaging and wrinkle formation due to regulation of antioxidant defense systems and MMPs expression.
