Objectives: This study was carried out to concrete the concept of Hand Gworeum Skin referred in Suwen of Huangdi Neijing. Methods: The Hand Gworeum Meridian was labeled with latex in the body surface of the cadaver, subsequently dissecting a superficial fascia and muscular layer in order to observe internal structures. Results: Skin histologically encompasses a common integument and a immediately below superficial fascia, this study established the skin boundary with adjacent structures such as relative muscle, tendon as its compass. The realm of the Hand Gworeum Skin is as follows: The skin close to the nipple on the 4th intercostal space, the interceps of biceps brachii muscle, the cubital surface at ulnad of bicipital aponeurosis, the anterior surface of the forearm, between flexor carpi radialis and palmaris longus(from wrist crease to 5chon above), the palm between the 3rd and 4th metacarpals on the cross part with the palm crease, the radiod from the middle finger nail(or the end of middle finger). The realm of the Hand Gworeum Skin is situated on between Hand Taeeum Skin and Hand Soeum Skin in front of arm. Conclusion: The realm of Hand Gworeum Skin from the anatomical viewpoint seems to be the skin area outside the superficial fascia or the muscle involved in the pathway of the Hand Gworeum Meridian vessel, Collateral Meridian vessel, and Meridian muscle, being considered adjacent vessels or nerves at the same time.

Ultraviolet (UV) B irradiation induces the production of matrix metalloproteinases (MMPs), which are responsible for the degradation or synthesis inhibition of collagenous extracellular matrix in connective tissues, causing skin photoaging. In this study, we examined the inhibitory effect of MMP-1 expression of yam extract in tumor necrosis factor-alpha (TNF-${\alpha}$)-stimulated human dermal fibroblast neonatal (HDFn) cell and preventive effect of UVB-induced damage in hairless mice skin. The synthesis of procollagen and the release of MMP-1 in HDFn cells were measured by EIA kit and MMP-1 assay kit, respectively. UVB radiation was applied to the backs of the mice three times a week for 8 weeks. Mice were randomly divided into three groups, and were topical application with the Dioscorea batatas (DB, 6%) or vehicle. Reduction of TNF-${\alpha}$-induced procollagen synthesis was increased by DB (50 ug/ml), which was higher than positive control group (TGF-${\beta}$). Also, pre-treatment of HDFn cells with DB inhibited TNF-${\alpha}$-induced release of MMP-1. In vivo study, we found that preventive effect of DB against UV-induced epidermal thickness. DB suppressed the expression of MMP-3 and MMP-13 induced by UVB irradiation. Our results show that DB have preventive effect of UV-induced skin damage in hairless mice.

To investigate whether aqueous extract from Aconitum koreanum (AEAK) effects in the process of melanin synthesis, the expression of tyrosinase-related proteins (TRPs) by immunohistochemical methods were performed in ultraviolet B (UVB) irradiated skin of guinea pig. The irradiation of UVB (60 mJ/day) was performed for 3 days and treated with AEAK for 15 days. About the color evaluation, the visual scores of UV B irradiated guinea pig with AEAK treatment were slightly lower than those in the UV B alone irradiated ones. At day 15 after UVB exposure, immunohistochemical analysis for TRPs expression were performed. The intensive expression of tyrosinase was mainly observed over epidermis with skin appendage and in the cells of dermis. Slight increase of these reaction was induced in response to UVB in the spinous and granular layer of epidermis, but similar expression in the AEAK treated guinea pig as normal one. The TRP-1 and TRP-2 expression were not detected in the skin of normal guinea pig. But intensive expression for TRP-1 and TRP-2, especially TRP-2, induced by UV B irradiation in the cells of dermis. These expressions were decreased in the AEAK treated guniea pig. Collectively, these results suggest that AEAK has a potential to inhibit synthesis through regulation of TRPs expression in the skin of guinea pig, but better understanding the function of AEAK, more research should be done in the effects of AEAK on the function of TRPs in melanogesis.
Objectives This study is conducted to evaluate skin fat barrier formation of Hataedock using the Coptis japonica & Glycyrrhiza uralensis extract. Methods The 3-week-old NC/Nga mice were divided into 3 groups: control group (Ctrl), Hataedock-treated group that uses the Coptis japonica & Glycyrrhiza uralensis (CGT) extract, and Hataedock-treated group that uses Bifidobacterium (BBT). After 2 weeks, changes in immunohistochemicals, and skin-lipid-barrier regulators were observed for the effects of Hataedock. Results In CGT group, loricrin-positive reaction has been increased by 231%, along with involucrin-positive reaction by 90%, filaggrin-positive reaction by 143%, and ASM-positive reaction by 341% in the stratum corneum. Conclusions Hataedock, using the extract of Coptis japonica & Glycyrrhiza uralensis, increased the expression of proteins promoting keratinocyte differentiation. This leads into conclusion that Hataedock may increase the keratinocyte formation and function which promotes skin barrier formation.
The skin is the largest organ of the integument system whose surface is closely related with many physiological and pathological conditions. Various methods are used to understand the structural and functional status of human skin. We would like to present usefulness of scanning electron microscopic (SEM) observation of skin replica and its significance of training module for a novice. The silicon replicas from several regions of the body (hand, finger, forearm, lip, and face) were casted by applying Exafine$^{(R)}$ mixture. The positive replicas were prepared by applying EPON 812 mixture on negative silicon replicas. Some of the negative silicon replicas were cut with a razor blade and surface profiles were observed. The negative and positive replicas were coated with platinum and were observed under the scanning electron microscope. We could investigate the detailed structures of the human skin surface without any physical damage to the subject. The positive replicas depicted real surface structure of the human skin vividly. The cross sectional view of the negative silicon replicas provided surface profile clearly. The scanning electron microscopic observation of the human skin replicas would be useful to study skin surface structures and to evaluate medical and esthetical applications.
Skin flap necrosis remains a major complication of reconstructive surgery. Euterpe oleracea Mart., popularly known as "acai berry" contains hydroxybenzoic acid, antioxidant polyphenolics and anthocyanins. These and other compounds within the acai berry confer anti-inflammatory and anti-oxidative effects. In this current study, we evaluated the protective effect of acai berry extracts on survival of random-pattern skin flaps in a murine model by histologic analysis. ICR mice were subjected to skin elevation surgery and orally administered acai berry extract (100 mg/kg) daily for 7 days. Tissues were stained with hematoxylin-eosin or Masson's trichrome to observe tissue integrity and collagen deposition. In addition, $TGF-{\beta}$ and VEGF was stained by immunofluorescence to determine anti-inflammatory cell infiltration and neovascularization, respectively. We found a decrease in inflammatory cell infiltration and increase in collagen deposition in the acai berry extract treated mice compared to control mice. Immunofluorescence staining reveal a higher number of $TGF-{\beta}$ positive cells and enhanced VEGF staining in the acai berry extract treated mice. The results from this study indicate that oral uptake of acai berry extract can promote healing and survival of surgical skin flaps in mice providing an augmentative therapeutic approach to enhancing skin flap survival.
Objectives The purpose of this study is to confirm the effect of alleviating inflammation through creating skin fat barrier from Scutellaria baicalensis extract. Methods Four-week-old Balb/C mice were divided into four groups: control group (Ctrl), lipid barrier eliminated group (LBEG), dexamethasone (DM) treated group after lipid barrier elimination (DMTG), and Scutellaria baicalensis (SB) treated group after lipid barrier elimination (SBTG). Scutellaria baicalensis extract were administered for 5 days after removal of the fat barrier. Changes in skin condition, improvement of the fat barrier, and relief of inflammation were observed in each group. Results Compared to LBEG and DMTG, pathological skin damage and tissue changes were less in SBTG, and transepidermal water loss (TEWL) and pH were also significantly reduced. Filaggrin was also significantly increased in SBTG. KLK7, PAR-2, and TSLP in SBTG also showed significant reduction compared to the LBEG and DMTG. Conclusions Scutellaria baicalensis extract restores skin barrier and relieves inflammation through the creation of skin fat barrier. This means that the Scutellaria baicalensis extract can regulate Th2 differentiation through the creation of the epithelial fat barrier.
Objectives After inducing dermatitis in 6-week-old mice, we tried to find out the effects of recovery in the damaged skin by administering genistein and palmitoylethanolamide (PEA) Methods The 6-week-old mice were divided into the control group (Ctrl), dermatitis causing group (AcDE), genistein-administered group after the onset of dermatitis (GsT), and PEA-administered group after the onset of dermatitis (PEAT). Seven mice were assigned to each group. Changes in the skin barrier were observed after three days of administration following the onset of dermatitis. Results In the GsT and PEAT, there was less skin damage compared to the AcDE, and the lowest skin damage showed in the GsT. The intensity of CB1 and CB2 expression was increased by 64% (CB1) and 39% (CB2) in the GsT, and 38% (CB1) and 28% (CB2) in the PEAT compared to the AcDE. The E-catherin positive reaction was decreased in the AcDE, while the E-catherin positive reaction was increased in the GsT (76%) and PEAT (34%). The p-JNK positive reaction was increased in the AcDE, while the p-JNK positive reaction was decreased in the GsT (60%) and PEAT (39%). Toxic Hepatopathy was not observed in the liver tissue of Genistein administered 3OGsT, and PEA administered 3OPEAT. Conclusions Genistein has recovery effect on dermatitis damage through active induction of the endocannabin system (ECS).

This study is designed to investigate the protective effects of fermented Red Ginseng (FRG) against photoaging in vitro and in vivo. UVB was irradiated to the human keratinocyte HaCaT cell and dorsal skin of SKH-1 mice for the induction of photoaging. After treatment of non-fermented red ginseng (NRG), fermented red ginseng (FRG), and fortified fermented red ginseng (FFRG) to the UVB irradiated HaCaT cell, ROS production and activity of MMP-9 were examined by DCFC-DA assay and gelatin zymographic assay respectively. UVB irradiated SKH-1 mice were treated with NRG, FRG, and FFRG via oral(300 mg/Kg B.W./day) and topical($100{\mu}{\ell}/mouse/day$) route.All of NRG, FRG, and FFRG had significantly reduced the intracellular ROS production elicited by UVB, among them FRG slightly more reduced the ROS production than NRG and FFRG. FFRG had slightly more reduced the MMP-9 activity in UVB irradiated HaCaT cells than NRG and FFRG in high dose. Oral and topical treatment of NRG, FRG, and FFRG had decreased the expression of matrix metalloproteinase-2, -3, and -9 in dorsal skin of UVB irradiated mice. Among them, inhibitory effect of FRG on the expression of MMP-2 was apparent. We speculate that FRG has therapeutic potentials on the UVB irradiated photoaging.
