Characteristics of a Double-Tube Structure for the Hydraulic WIM Sensor

  • A new hydraulic tube structure for WIM sensor of a new generation is presented in this paper. The double-tube structure has been developed in order to improve the performance of the hydraulic load cell. The double-tube structure hydraulic element could be reduced by 46% in pressure changes according to temperature compared to a single-tube structure. In addition to the nonlinearity can be reduced by 67.19% at the same load condition. The hydraulic load cell shows an excellent linearity and measurement accuracy as the result of the static load test.

MHC Multimer: A Molecular Toolbox for Immunologists

  • The advent of the major histocompatibility complex (MHC) multimer technology has led to a breakthrough in the quantification and analysis of antigen-specific T cells. In particular, this technology has dramatically advanced the measurement and analysis of CD8 T cells and is being applied more widely. In addition, the scope of application of MHC multimer technology is gradually expanding to other T cells such as CD4 T cells, natural killer T cells, and mucosal-associated invariant T cells. MHC multimer technology acts by complementing the T-cell receptor-MHC/peptide complex affinity, which is relatively low compared to antigen-antibody affinity, through a multivalent interaction. The application of MHC multimer technology has expanded to include various functions such as quantification and analysis of antigen-specific T cells, cell sorting, depletion, stimulation to replace antigen-presenting cells, and single-cell classification through DNA barcodes. This review aims to provide the latest knowledge of MHC multimer technology, which is constantly evolving, broaden understanding of this technology, and promote its widespread use.

Vision-based Automatic System for Non-contact Measurement of Morphometric Characteristics of Flatfish

  • This paper introduces a vision-based automatic system (VAMS) for non-contact measurement of morphometric characteristics of flatfish, such as total length (TL), body width (BW), height (H), and weight (W). The H and W are simply measured by a laser displacement and a load cell, respectively. The TL and BW are measured by a proposed morphological image processing algorithm. The proposed algorithm cans measurement, when the tail of flatfish is deformed, and when it is randomly oriented. In the experiment, the average and maximum measurement errors were recorded, and standard deviations and coefficients of variation (CVs) for the measurements were calculated. From those results, when flatfish the TL measurements had an average of 266.844 mm, a standard deviation of 0.351 mm, a CV of 0.131%, and a maximum error of 0.87 mm with straightened flatfish ($TL_A$ : 267 mm, $BW_A$ : 141 mm), and when flatfish of different sizes were measured, the errors in the TL and BW measurements were both about 0.2 %. Using a single conveyor, the VAMS can process up to 900 fishes per hour. Moreover, it can measure morphometric characteristics of flatfish with a TL of up to 500 mm.

  • Oldenlandia diffusa, Cremastra appendiculata and Fritillaria thunbergii are widely distributed in the Korea, China and Japan, and has been used in traditional medicine for various diseases, such as pharyngolaryngitis, tonsillitis, goiter and stomach ulcer. However, the anti-cancer activities in human breast cancer have not been clearly elucidated yet. In this study, it was compared the in vitro cytotoxic effects of single and complex treatment of O. diffusa, C. appendiculata and F. thunbergii. We treat human breast cancer MCF-7 cells with O. diffusa, C. appendiculata and F. thunbergii. And we evaluated viability, growth inhibition, morphological changes, apoptotic bodies formation, measurement of the cell cycle and formation of DNA fragmentation of these cells. It was found that single treatment of O. diffusa could inhibit the cell proliferation in human breast cancer MCF-7 cells. However, complex treatment of O. diffusa, C. appendiculata and F. thunbergii is weakly or not affect the cell proliferation of MCF-7 cells. And anti-proliferative effects of O. diffusa in MCF-7 cells was associated with G1 arrest of cell cycle. These findings suggest that O. diffusa may be a potential chemotherapeutic agent for the control of human breast cancer cells and further studies will be needed to identify the molecular mechanisms.

Optimization of Expression, Purification, and NMR Measurement for Structural Studies of Syndecan-4 Transmembrane Region

  • Syndecan-4 is a transmembrane heparan sulfate proteoglycan, which is a coreceptor with integrins in cell adhesion. To get better understand the mechanism and function of Syndecan-4, it is critical to elucidate the three-dimensional structure of a single transmembrane spanning region of them. Unfortunately, it is hard to prepare the peptide because syndecan-4 is membrane-bound protein that transverse the lipid bilayer of the cell membrane. Generally, the preparation of transmembrane peptide sample is seriously difficult and time-consuming. In fact, high yield production of transmembrane peptides has been limited by experimental adversities of insufficient yields and low solubility of peptide. Here, we demonstrate experimental processes and results to optimize expression, purification, and NMR measurement condition of Syndecan-4 transmembrane peptide.

  • $CdS_{0.69}Se_{0.31}$ single crystal grown by sublimation method. Hall effect measurement were carried out by the Van der Pauw method. The measurement values under the temperature were found to be carrier density $n=1.95{\times}10^{23}m^{-3}$, Hall coeffcient $RH=3.21{\times}10^{-5}m^3/c$, conductivity ${\sigma}=362.41{\Omega}^{-1}m^{-1}$, and Hall mobility ${\mu}=1.16{\times}10^{-2}m^2/v.s.$ Heterojunction solar cells of $n-CdS_{0.69}Se_{0.31}/p-Cu_{2-x}S_{0.69}Se_{0.31}$ were fabricated by the substitution reaction. The open-circuit voltage, short-circuit currint density, fill factor and power conversion efficiency of $n-CdS_{0.69}Se_{0.31}/p-Cu_{2-x}S_{0.69}Se_{0.31}$ heterojunction solar cell under $80mW/cm^2$ illumination were found to be 0.41V, $19.5mA/cm^2$, 0.75 and 9.99%, respectivity.

고체 전해질을 이용한 새로운 전지에 관한 연구 (A Study on Solid Electrolyte for New Type Cell)

  • 새로운 전지의 개발을 위하여$ AgI-Ag_8S_3SO_4$system들을 만들어 이에 대한 전기전도도 측정, 상 연구 및 그 구조를 조사하였다. X-ray powder diffraction method로 20mole% $ Ag_8S_3SO_4$-AgI system의 구조가 단사정계임을 규명하였고 DTA method로 단일상을 이룸을 확인하였다. 4-probe method를 적용시켜 전기전도도를 측정한 결과 20mole% $ Ag_8S_3SO_4$-AgI system이 순수한 $ Ag^+ $이온성 couduct임을 밝힐 수가 있었다.

단일층 다결정 실리콘 Flash EEPROM 소자의 제작과 특성 분석 (Fabrication and Characteristic Analysis of Single Poly-Si flash EEPROM)

  • In this paper, we propose the single poly-Si Flash EEPROM device with a new structure which does not need the high voltage switching circuits. The device was designed, fabricated and characterized. From the measurement results, it was found that the program, the erase and the read operations worked properly. The threshold voltage was 3.1 V after the program in which the control gate and the drain were biased with 12 V and 7 V for $100{\mu}S$, respectively. And it was 0.4 V after the erase in which the control gate was grounded and the drain were biased with 11 V for $200{\mu}S$. On the other hand, it was found that the program and the erase speeds were significantly dependent on the capacitive coupling ratio between the control gate and the floating gate. The larger the capacitive coupling ratio, the higher the speeds, but the target the area per cell. The optimum structure of the cell should be chosen with the consideration of the trade-offs.

Molecular Tension Probes to Quantify Cell-Generated Mechanical Forces

  • Living cells generate, sense, and respond to mechanical forces through their interaction with neighboring cells or extracellular matrix, thereby regulating diverse cellular processes such as growth, motility, differentiation, and immune responses. Dysregulation of mechanosensitive signaling pathways is found associated with the development and progression of various diseases such as cancer. Yet, little is known about the mechanisms behind mechano-regulation, largely due to the limited availability of tools to study it at the molecular level. The recent development of molecular tension probes allows measurement of cellular forces exerted by single ligand-receptor interaction, which has helped in revealing the hitherto unknown mechanistic details of various mechanosensitive processes in living cells. Here, we provide an introductory overview of two methods based on molecular tension probes, tension gauge tether (TGT), and molecular tension fluorescence microscopy (MTFM). TGT utilizes the irreversible rupture of double-stranded DNA tether upon application of force in the piconewton (pN) range, whereas MTFM utilizes the reversible extension of molecular springs such as polymer or single-stranded DNA hairpin under applied pN forces. Specifically, the underlying principle of how molecular tension probes measure cell-generated mechanical forces and their applications to mechanosensitive biological processes are described.
