서해산 바지락 (Ruditapes philippinarum)의 토사 배출 조건 (Sand Elimination in Shortnecked clam, Ruditapes philippinarum, Harvested from Western Coast of Korea)

  • 서해안에서 생산되는 주요 패류인 바지락의 토사 배출률을 최적화하기 위하여 토사 배출에 관여하는 환경인자의 영향과 토사 배출에 따른 해수의 pH 및 용존 산소의 변화, 그리고 바지락에 대한 세균학적 조사, 폐사율 및 토사 잔존유무를 검토하였다. 바지락의 토사 배출에 있어서 해수 온도의 영향을 보면 수온 $23^{\circ}C$에서 가장 양호하여 48시간 후에 $97.9\%$가 배출되었으며, 염분농도는 $32.9\~40\%_{\circ}$에서 가장 양호하여 48시간 후에 $94.7\~97.1\%$가 배출되었다. 또한 pH의 영향을 보면 pH 9.0에서 가장 많은 양이 배출되어 48시간 후에 $98.0\%$가 배출되었으며, 자연해수인 pH 7.9와 큰 차이는 보이지 않았다. 바지락은 해수를 유수시키지 않고 12시간마다 해수만 교환한 시험구에서는 48시간 경과후 $2\~24\%$의 폐사체가 발생하였으나, 유수 시킨 시험구에서는 48시간까지 폐사체가 발생하지는 않았다. 모든 시험구에서 36시간 이상 경과시 대부분의 토사가 배출되었다. 바지락의 토사 배출을 위한 최적 해수 공급 조건은 바지락에 대한 해수 비율이 3,000과 4,000 L/shellfish $m^3$일 때에는 해수를 150L/min./shellfish $m^3$ 이상, 6,000과 8,000 L/shellfish $m^3$일 경우에는 해수를 100L/min./shellfish $m^3$ 이상 공급하는 것이었다.

Bandsharing Values and Genetic Distances of Two Wild Shortnecked Clam, Ruditapes philippinarum Populations from the Yellow Sea Assessed by Random Amplified Polymorphic DNAs-Polymerase Chain Reaction

  • Genomic DNAs were extracted from the muscle of twenty-two specimens of two shortnecked clam, Ruditapes phifippinarum populations collected in Anmyeondo and Seocheon. Genetic differences within and between populations were analysed by random amplified polymorphic DNAs-polymerase chain reaction (RAPD-PCR) using twenty arbitrary decamer primers. Out of 20 primers, 6 generated a total of 1,111 major and minor RAPD bands from individuals of two sites, producing approximately 4.2 average polymorphic bands per primer in individuals from Anmyeondo and ranging in size from less than 50 to larger than 1,500 base pairs (bp). The electrophoretic analysis of RAPD products amplified showed moderate levels of similarity among the different individuals in Seo-cheon population. The average bandsharing values (BS value) of the samples within population from Anmyeondo ranged from 0.155 to 0.684, whereas it was 0.143∼0.782 within population from Seocheon. The average BS value between individuals No. 13 and No. 14 from Seocheon was 0.782 which was higher than that of those from Anmyeondo. The single linkage dendrogram resulted from three primers (OPA-08, -09 and -20), indicating six genetic groupings composed of group 1 (No.4, 8 and 10), group 2 (No. 18), group 3 (No.2, 5 and 7), group 4 (No. 1, 3, 6, 9, 11, 12, 13, 14, 15 and 17), group 5 (16, 19 and 20) and group 6 (No. 21 and 22). In the Seocheon population, the individual No. 18 clustered distinctly from the others of this population. The observed genetic distance between the two populations from Anmyeondo and Seocheon was more than 0.209 (0.247 and 0.275). The shortest genetic distance (0.094) displaying significant molecular differences was between individuals No. 13 and No. 14. Especially, the genetic distance between individuals No. 22 and the remnants among individuals in two geographical populations was highest (0.275). This result illustrated that individual No.22 is distinct from other individuals within two shortnecked populations. The different geographical features of two sites may have caused the genetic diversity in two shortnecked clam populations.

The RAPD profiles obtained with DNAs of two populations from Anmyeondo and Seocheon, respectively, were considerably different (0.20 and 0.51, respectively). The varying degrees of difference among populations amy also be of relevance to the restricted hybridization of wild bivalve. Besides gene mapping and breeding applications, PCR-RAPD systems could be very useful for the rapid certification and quality control of seed production and for every projects based on PCR amplification of specific bivalve DNA fragments.

Morphological Variation and Genetic Relationship among Populations of the Shortnecked Clam Ruditapes philippinarum Collected from Different Habitats

  • The characteristics of the populations of shortnecked clam (Ruditapes philipinarum) originated from three different seed-production sites, Hadong, Kochang and Ulsan along the coast of Korea, were analysed by the morphological differences and the random amplified polymorphic DNA (RAPD) profiles. The morphology of the shell and survival rate for each population were also investigated 13-months after transplantation to a farming site in Shinsung. The morphology of the populations from Hadong and Kochang showed significant differences (P<0.05), but one year after transplantation to Shinsung, the morphology of these three populations was no longer significantly different (P>0.05). The template DNA for RAPD was efficiently extracted from the digestive diverticula of the clams. Up to 13 of amplified fragments were detected using arbitrary primers. Within the species of R. philipinarum, the genetic similarities ranged from 0.196 to 0.259. The populations from Hadong and Ulsan showed the highest similarity. The survival rates of the populations from Hadong $(69.4\%)$ and Ulsan $(63.8\%)$ were higher than that from Kochang $(41.7\%)$ 13-months after transplantation. From the RAPD analysis, it could be used as one of the primary criterion in determining which shellfish populations among various seed-production sites tend to be genetically similar and more adaptable and transplantable to a farming site.

  • Shortnecked clam is a commercially important mollusks species, which is distributed all over the Yellow Sea. Consequent of the rapid increase in seed production, there is a need to understand the genetic composition of wild shortnecked clam populations in order to evaluate exactly the latent genetic effects induced by seed production operations. Many genetic and molecular researches were made because RAPD-PCR is a simple and rapid method for determining genetic diversity and similarity in various organisms with the advantage that no prior knowledge of the genome under research is needed (Fischer et at., 2000). Population characteristics of the bivalve Ruditapes philippinarum from Cheju Island coasts, Korea

  • A comparative analysis of size and age structures of coastal subfossil shell assemblages of the shortnecked clam Ruditapes philippinarum from open and protected bays of Cheju Island (Korea) was carried out. On the whole, taking into account the damage of small fragile shells, size and age structures of the shell assemblages corresponded to the classical curve of bivalve population distribution when its mortality diminishes with age increase up to a certain threshold. It was found that shell samples from open bays of the western, southern and eastern coasts included shells of smaller and younger individuals (L ${\leq}$ 40 mm, ${\leq}$ 4 years) than samples from the eastern protected bay (L ${\leq}$ 54.5 mm, ${\leq}$ 6 years). Evidently, strong wave activity was the reason for a short life-span of the clams from the open areas. Growth was investigated retrospectively by annual growth rings on the shells. Growth rates of the clams from the various coasts of Cheju Island differed. However, growth rates of the clams from different biotopes at the same (eastern) side of the Island were similar. Shell height/length and width/length ratios statistically significantly increased with the clam age increase. Most likely, the reason for such shell shape alteration is that more conglobated individuals more survive being more energy-optimal than oblong specimens.

  • Gonadal developmint, age and growth of Ruditapes philippinarum were investigated using samples from the intertidal zone of Simpo on the coast of Kimje, Korea, which were collected onthly for one year from Februaty 1993 to January 1994.Ruditapes philippinarum is diecious in sex. The gonads are located between the subregion of the midintestinal glands and reticular connective tissue of the foot. The ovary is composed of a number of ovarian sacs, and the testis is composed of numerous seminiferous tubules. The clam spawns once a year from early June to darly October, and the main spawning occurred between July and August when the water temperature went above 23$^{\circ}C$. Ripe oocytes are about 65-70${\mu}{\textrm}{m}$ in diameter. Gonadal phases of this species can be divided into five successive stages; multiplicative(February to March), growing (April to May), mature(Aprilto Septimber), spent(June to October), and degenerative and resting(july to March). Spawning is closely related to the sea water temperature. Based on the monthly variations of marginal index (MI')of the shell, it was suggested that the annual ring mark formation occurred in March once a year and took approximately 8 months (0.67 year) for first ring to be formed on the shell. The relationship between the shell length(SL) and the total weight (TW) was represented by nonlinear equation; TW=1.208 x 10/ sup -4/ S $L^{3.158}$, and also in the relationship be-tweenthe shell length (SL) and the shell height(SH), the shell length and the shell width (SW) were represented by the linear equations; SH=0.726 SL-0.483, SW=0.542 SL-0.803.Growth curves for shell length and total weight fitted to von Bertalanffs equation were expressed as: S $L_{t}$ =68.34(1- $e^{0.221}$(t+0.418)) T $W_{t}$ =75.16(1- $e^{0.221}$(t=0.418))$^{3.158/3.158}$

한산도 진두해역 양식 바지락의 자원생태학적 연구 (Ecological Study of Shortnecked Clam Ruditapes philippinarum from the Jindu Coast of Hansan Island, Korea)

  • 양식생물의 최적생산을 위해서는 적극적 관리 외에도 환경 개선과 지속적으로 최대생산을 위하여 수산자원학적 관리방안도 함께 강구되어야 한다. 따라서 양식장의 생산성 향상을 위하여 어느 시기에 어느 정도의 사이즈의 생물을 선택적으로 채취하여야 할 것인가는 양식생산 측면에서 아주 중요한 문제가 될 수 있다. 이를 위하여 통영시 한산면 진두연안의 바지락양식장의 양성중인 바지락자원의 자원학적 특성치로부터 가입당 생산량 모델을 구현하였고, 이 모델을 통해 현재의 양식생물 이용현황을 파악하고자 하였다. 현재의 어업형태에 따른 $t_c$와 F를 적용하면, 가입당 생산량은 3.46 g에 해당하였지만, $t_c$의 경우 3.48 세까지, F의 경우 0.9295/yr까지 증가시키면 약 5% (3.63 g)의 생산증대효과를 볼 수 있는 것으로 예상되었다. 그러나 채취시기를 연장함에 따른 사육소요기간의 장기화 및 자연사망계수의 변동 가능성을 고려한다면 현재의 어업행위가 가장 적합한 것으로 판단된다.
