The characteristics of seed production in an Adonis multiflora (Ranunculaceae) population

  • A natural population of Adonis multiflora, a spring ephemeral herb growing in temperate deciduous forests, was studied to determine the seed production characteristics. Plant size, flowering time, and seed number were monitored from February 2009 to May 2011 in main growing season (i.e., from March through May). The biomass rates of the shoot and the root in the A. multiflora population were 22-24% and 76-78%, respectively, and the biomass of the root was proportional to that of the shoot. The flowering rate was 60% in the plants with 1 to 2 g of shoot biomass, and 100% in the plants with >2 g of shoot biomass. In the plants with root biomass between 4 and 6 g, the flowering rate was 43% and, in the plants with the root biomass over 8 g, it was 100%. The shoot biomass was a better predictor of the flower production probability than the root biomass. The number of flowers and seeds was closely correlated to shoot biomass at 1% significance level. The size of the plant that produced seed excessively instead of the shoot biomass in one year typically decreased in the next year and vice versa. The flowering time and its duration were closely related to the number of faithful seeds but not to that of total seeds. The number of faithful seeds was proportionate to flowering duration and inversely proportionate to flowering time (year day, YD). In a plant, the number of faithful seeds noticeably decreased with the inflorescence (i.e., order of flower in a plant), and this difference between the two successive flowers was significant at the 1% level between the first and the third flower in 2009 and 2011 but not between the third and the fourth. However, the number of total seeds was mostly similar in the first through the fourth flower for all three years.

Evaluation of Cancer Chemopreventive Potential of Various Grape Shoot Extracts and Refined Materials Using in vitro Bioassay Systems

  • Since reactive oxygen species, prostaglandins, and nitric oxide are closely involved in various pathological conditions anti play important roles in the initiation, promotion, and progression of carcinogenesis, agents that modulate the production or activity of them might be considered as cancer chemopreventive agents. In the present study, we evaluated chemopreventive potential of some grape shoot extracts and their refined materials using various in vitro assay systems. As a result, both grape shoot extracts and refined materials possessed effective radical scavenging activities about 70~80% at the concentration of 500 ${\mu}$g/ml, and especially the Sheridan shoot extract showed the most potent 1, 1-diphenyl-2-picrylhydrazyl radical scavenging activity that was similar to resveratrol. In addition, refined materials from grape shoot extracts suppressed lipopolysaccharide-induced nitric oxide production in macrophage cells, anti refined materials from Kyoho and Campbell shoot extracts exhibited similar inhibitory activities with $IC_{50}$ value of 224 ${\mu}$g/ml and 285 ${\mu}$g/ml, respectively. In addition, at the concentration of 50 ${\mu}$g/ml, all of refined materials inhibited cell proliferation against various human cancer cells about 30~40% compared to control. These findings suggest that grape shoot extract and their refined materials might be useful sources for the development of chemopreventive agents and/or functional foods.

Cryopreservation of in Vitro Grown Shoot Tips of Sweet Potato (Ipomoea batatas L.) by the Encapsulation-Vitrification Method

  • Sweet potato (Ipomoea batatas L.) shoot tips grown in vitro were successfully cryopreserved by encapsulation-vitrification. Encapsulated explants are very easily manipulated, due to the relatively large size of the alginate beads, and a large number of samples can be treated simultaneously. In this study, the effects of sucrose preculture, cryoprotectant preculture, and post-warm recovery media on regrowth, following liquid nitrogen (LN) exposure, were investigated to establish an efficient encapsulation-vitrification protocol for sweet potato. Shoot tips of plants grown in vitro were precultured in 0.3 M sucrose for 2 d before encapsulation. Encapsulated shoot tips were pre-incubated in liquid MS (Murashige and Skoog) medium containing 0.5 M sucrose for 16 h, before preculturing in sucrose-enriched medium (0.7 M sucrose) for 8 h. Shoot tips were osmoprotected with 35% plant vitrification solution 3 (PVS3) for 3 h, before being dehydrated with PVS3 for 2 h at $25^{\circ}C$. The encapsulated and dehydrated shoot tips were transferred to 2 mL cryotubes, suspended in 0.5 mL PVS3, and plunged directly into liquid N. High levels of shoot formation were obtained for the cv. Yeulmi (65.7%) and Yeonwhangmi (80.3%). The regrowth rates of cryopreserved samples in Yeulmi (78.9%) and Yeonwhangmi (91.3%), following culture on ammonium-free MS medium for 5 d, were much higher than those cultured on standard MS medium (65.7% and 80.3%, respectively). This encapsulation-vitrification is a promising method for the long-term preservation of sweet potato.

  • A plant regeneration system using shoot basal callus of in vitro cultured black locust(Robinia pseudoacacia L.) was established. Shoot basal callus was induced on MS medium supplemented with BA, or NAA, and mere more proliferated on BA containing medium than NAA containing medium at both light and dark conditions. Shoot basal callus was induced during shoot multiplication procedure. Two types of callus, green colored callus and whitish-yellow colored callus, were cultured on mMS medium containing 2.0 mg/l BA and 0.5 mg/l NAA. Green colored callus showed the shoot regeneration ability while whitish-yellow callus failed to regenerate shoot and died. Regenerated shoot were rooted on hormone-free ${\frac{1}{2}}MS$ medium within 2 weeks.

Development of Plant Regeneration and Genetic Transformation System from Shoot Apices of Sorghum bicolor (L.) Moench

  • Development of efficient plant regeneration and genetic transformation protocols (using the Particle Inflow micro-projectile Gun and the shoot-tips as target tissue) of Sorghum bicolor (L.) Moench in terms of expression of the reporter gene, $\beta$-glucuronidase(uidA) is reported here. Two Indian cultivars of sorghum were used in the study, viz. M-35-1 and CSV-15. Plant regeneration was achieved from one-week-old seedling shoot-tip explants via multiple-shoot-clumps and also somatic embryos. The multiple-shoot-clumps were produced on MS medium containing BA (0.5, 1.0 or 2.0 mg/$L^{-1}$), with biweekly subculture. Somatic embryos were directly produced on the enlarged dome shaped expansive structures that developed from shoot-tip explants (without any callus formation) when cultured on MS medium supplemented both with BA (0.5, 1.0 or 2.0 mg/$L^{-1}$) and 2,4-D (0.5 mg/$L^{-1}$). Whereas each multiple-shoot-clump was capable of regenerating more than 80 shoots via an intensive differentiation of both axillary and adventitious shoot buds, the somatic embryos were capable of 90% germination, plant conversion and regeneration. The regenerated shoots could be efficiently rooted on MS medium containing 1.0mg/$L^{-1}$ IBA and successfully transplanted to the glasshouse and grown to maturity with a survival rate of 92%. The plant regeneration efficiency of both the genotypes were similar. After the micro-projectile bombardment, expression of uidA gene was determined by scoring blue transformed cell sectors in the bombarded tissue by an in situ enzyme assay. The optimal conditions comprising a helium pressure of 2200 K Pa, the target distance of 11 cm with helium inlet fully opened and the use of osmoticum have been defined to aid our future strategies of genetic engineering in sorghum with genes for tolerance to biotic and abiotic stresses.

  • The effects of cytokinin and putrescine on adventitious shoot induction from leaf explant of Rhodiola sachalinensis A. Bor were investigated. Among cytokinin used in this experiment, BA was more effective on adventitious shoot induction and shoot elongation than kinetin. Especially, 1 mg/L BA was the best to increase adventitious shoot induction (71%) and shoot elongation (3.0 mm). In addition, 100 mM putrescine in MS medium with 1 mg/L BA was higher in adventitious shoot induction (93%) and shoot elongation (3.8 mm) than single treatment of 1 mg/L BA. Adventitious shoots induced in this experiment rooted on 1/2 MS medium and acclimated over 95% on composed soil (peatmoss:sand=1:1).

Plant Regeneration via Organogenesis from Leaf and Stipule Segments of Strawberry (Fragaria ananassa Duch.) (딸기의 잎과 탁엽 절편체로부터 기관형성을 통한 식물체 재생)

  • Plant regeneration via organogenesis from leaf and stipule explants of micropropagated shoots of strawberry (Fragaria $\times$ ananassa cv. Suhong) was achieved. Leaf and stipule explants were detached from shoot-tip cultured shoots and cultured on MS medium with various combinations of BA and NAA under light or dark condition. Shoot regeneration from leaf explant was observed after 3 weeks in culture and was good at the high ratio of BA and NAA among various combination treatments. The highest shoot regeneration frequency from leaf explants was obtained with 1.0 mg/L BA and 0.1 mg/L NAA, in which 31.1% shoot regeneration frequency(1.7 shoots per leaf explant) was yielded. In case of stipule explants, shoot regeneration was largely affected by plant growth regulators during incubation under dark condition for initial 4 weeks but not under continuous light condition. The combination treatment with 2.0 mg/L BA and 0.1 mg/L NAA showed the most excellent shoot regeneration from stipule explants, where 44.4% regeneration frequency(4.0 shoots per explants) obtained. Regenerated shoots were rooted on MS medium with 0.1 mg/L NAA after shoot elongation, and the plantlets regenerated were transferred to soil mixtures with vermiculite and perlite for acclimation.

Thidiazuron-induced Shoot Formation of Hibiscus syriacus L. 'Honghwarang' by Suspension Culture (Thidiazuron이 무궁화 '홍화랑' 품종 액체 현탁 배양시 신초형성에 미치는 영향)

  • This study was conducted to determine the optimum cultural condition and method for in vitro mass production of Hibiscus syriacus L. 'Honghwarang'. When callus induced in MS solid medium supplemented with 0.01 mg/L TDZ was cultured in liquid medium containing 0.01mg/L TDZ, callus growth and shoot primordia formation was most effective. Formed shoot primordia were regenerated into shoot in MS or 1/2 MS medium of growth regulator-free condition. Effects of mesh size, shaking speed on callus and shoot primordia formation were examined after 5 weeks. Callus and shoot primordia formation was formed most effectively at 10 mesh and 80 rpm shaking speed in liquid medium.

Analysis of Kickers' Strategic Actions for Finishing in Penalty Shoot-outs (승부차기 상황에서의 키커의 골 결정 전략행동 분석)

  • The present study aims to analyze kickers' strategic actions for finishing in penalty shoot-outs among youth soccer players ultimately to increase success rates of shoot-outs and to score a victory. Subjects were experienced 271 soccer players attending high schools located around the capital area. To determine any difference in strategic actions for finishing in penalty shoot-outs among the subjects, frequency analysis or chi-square was used for the analysis. The analysis found the following points: First, the success or failure rates of kickers in penalty shoot-outs were found to depend on subjects' experiences as soccer players. Second, in terms of kickers' shooting qualities, they were found to shoot on goal regardless of shooting positions. Third, an important factor to enhance the shoot-outs probability was found to be a strategic action through the eyes. Fourth, in view of the strategic actions prior to kickers' shooting, goalies were found to check out kickers' positions first and estimate the directions and qualities of shooting via kickers' body actions and eye lines.

In Vitro Multiple Regeneration from Cotyledons and Hypocotyls of Impatiens

  • In vitro shoot regeneration from cotyledon and hypocotyl explants derived from germinating mature Impatiens seeds. The induction of organogenetic tissue was also influenced by the cotyledon and hypocotyl. Multiple shoot induction was higher in hypocotyl than in the cotyledon explant with Thidiazuron and a NAA medium.
