Multiple Shoot Formation from Cotyledonary Nodes of Soybean Cultivars (대두 품종에 따른 자엽절에서의 다신초 형성)

  • For the plant regeneration of soybean (Glycine me L. Merr.), the shoot formation rate, optimal medium and tissue conditions were examined using Korean soybean cultivars. Among the parts of seedling, a node that includes one cotyledon showed the highest shoot formation rate among other tissues. Half-strength B5 medium was more efficient than full strength medium. Formation rates of pair shoots (1 to 2 shooting) were higher in the when benzyl adenine was supplemented. The formation rates of multiple shoots, that is, 4 to 5 in shooting, were high when thidiazuron was supplemented. Multiple shoot was de novo formed in cutting side of cotyledonary node. The effective concentration of thidiazuron for shoot induction treatment was 2 mg/L. Among the 27 cultivars, multiple shoot formation rates were high in the 11 cultivars including 'Heugcheongkong, and pair shoot formation rates were high in the 16 cultivars including 'Malikong'.

Growth Characteristics of Teratoma Shoot derived from Crown Gall Callus of Nicotiana tabacum cv. NC2326 (연초 Crown Gall Callus 유래 Teratoma Shoot의 생장특성)

  • The present study was conducted to obtain some basic information on the shoot formation from crown gall callus and the characteristics of teratoma shoot derived from crown gall induced by inoculation of Agrobacterium tumefaciens C58. Crown gall callus could be continuously cultured on the phyohormone free basic medium. The growth of crown gall callus was inhibited when BA were added to the cultural media. Shoot formation from crown gall callus fail to be initiated except teratoma shoot which induced on the phytohormone free medium after several subculture on rare occasions. Teratoma shoot could not form root and grow as normal shoot. Addition of BA to cultural media was not effective for shoot elongation, reduction in multiple shoot formation, but IBA was somewhat effective for shoot elongation of teratoma shoot, never for root formation.

Effects of Medium Components and Growth Regulators on Callus Development and Shoot Regeneration from Shoot Explants of Black Locust (Robinia pseudoacacia)

  • Effects of growth regulators and medium components were tested for shoot multiplication and callus growth from shoot explants of black locust. During shoot multiplication, callus growth at the cut end of shoot explants proceeded shoot development. The basal callus growth seemed to be a function of both mineral components and cytokinin supplemented in the medium. Maximum callus growth was induced by 0.5${\mu}$M BAP and the callus growth decreased as the level of BAP increased. Positive correlations were found between basal callus growth, and shoot multiplication and growth. Shoot multiplication was greatest on BSM medium (black locust shoot culture medium) supplemented with 1 $\mu$M BAP. With medium containing high nitrogen content, both shoot multiplication and growth were significantly enhanced. A new BRM medium was the most effective for rooting of black locust among three rooting media tested.

Growth Properties of Central and Peripheral Ramets in a Zoysia sinica's Clone

  • A natural, tidal-flat clone of Zoysia sinica was studied to compare ramet growth properties in central area with in peripheral area. In new stolon and rhizome, internode length, weight, shoot height and weight, and spike production were monitored on July 25, 2004. The weight/height rate of shoot between stolon and rhizome, the shoot/stolon (or rhizome) rate in weight between central and peripheral area were not different. However, other properties differed in stolon from rhizome or between central and peripheral area significantly differed: 1. The rhizome in central area had a larger node number, shorter internode length, higher shoot height, larger shoot biomass, and higher rate of non-shoot nodes than that in peripheral area. 2. The stolon in central area had a smaller node number, shorter internode length, and smaller biomass than that in peripheral area. 3. In the same area, the rhizome had a larger node number (except for central area), shorter internode, higher shoot height, larger shoot biomass, higher rate of non-shoot node, and higher rate of node having over two shoots than the stolon. No relationship could be found between shoot size and spike production in shoot on vertical rhizome (lower node of old shoot).

  • The experiment was conducted to identify the optimal in vitro propagation condition for P. lactiflora Pall. Through apical shoot tip and axillary shoot tip culture of winter bud. When apical shoot tip and axillary shoot tips excised from winter bud were cultured on MS medium supplemented with various concentrations of plant growth regulators, all the apical shoot tips elongated regardless of the composition of the medium but axillary shoot tips responded differently. Shoot of 'Uisong' local cultivate was well elongated in the medium containing 0.01mg/L NAA. Frequency of shoot formation and subsequent shoot growth in axillary shoot tip culture were promoted in the medium containing 0.01 mg/L NAA and 5.0mg/L zeatin. 30% of the elongated shoots were vigorously rooted on the medium containing 0.1mg/L NAA with vermiculite as a support medium.

Optimized Shoot Induction and Histological Study of in vitro Cultured Korean Soybean Cultivars

  • Soybean is the one of recalcitrant legume species for shoot induction. Shoot regeneration via direct organogenesis was investigated in five soybean cultivars, 'Dawon', 'Pungsan', 'Daewon', 'Taekwang' and 'Chongdoo 1' by using cotyledonary node explants. Out of 5 soybean cultivars, an efficient shoot regeneration condition was developed in the two soybean cultivars, 'Dawon' and 'Pungsan'. When various kinds of plant growth regulators with different concentration were estimated, the optimum medium condition for shoot induction in both soybean cultivars was MS + B5 vitamin supplemented with BA at concentration 2 mg/L. In addition, shoot formation efficiency was increased with 97.09% and 93.88% by the pretreatment of BA onto the explants before in vitro culture in both cultivars. Shoot induction in 'Dawon' cultivar was originated from epidermal tissue and sub-epidermal layers when histological changes were investigated under shoot regeneration after culturing cotyledonary node segments on shoot induction medium for 0 to 21 days. Especially, cell dedifferentiation was observed from parenchyma cells to meristematic cell in 3-day cultured segments.

  • To enhance the shoot induction efficiency from nodal stem of yooza, the culture conditions such as basal medium, carbohydrate source, solidifying agent and the optimum concentration of plant growth regulators for shoot induction were investigated. The nodal explants were cultured better on MS medium than on MT, SH, B5 or W media in considering of shoot induction rate and mean shoot length. Solidifying agent in medium was better with 0.8% agar than with 0.3% agar, 1.2% agarose or 0.2% gelrite. Carbohydrate source in shoot induction medium was efficient with 30 g/L sucrose. The optimum concentrations of plant growth regulators were determined that 0.1 mg/L NAA as auxin was effective on the shoot induction, and 1.0 mg/L BAP as cytokinin induced multiple shoots efficiently. Shoot induction was the most effective on MS medium supplemented with 4 mg/L $GA_3$ in yooza.

The Effects of Growth Regulators and Medium Strength on the Shoot and Bud Formation from the Shoot Apex of Chinese Yam (Dioscorea opposita Thunb)

  • Plantlet regeneration from the shoot apex was studied in three different genotypes of the chinese yam (Dioscorea opposita Thunb) cv. Jnagma and Danma, Dunggunma. The effects of plant growth regulators and inorganic salts concentration of the culture medium on bud induction and shoot growth were examined. The combinations of 0.2 mg/L BAP + 0.2mg/L kinetin, 0.01mg/L NAA + 0.2 mg/L kinetin and a single treatment of 0.2mg/L BAP were equally effective for bud and shoot formation from the shoot apices in the three cultivars. Auxin (2,4-D, NAA) treatment enhanced calli formation from the cultured apices. Also, the shoot apices of the cv. Dunggunma produced more callus and buds on the culture medium (MS) containing 0.05mg/L NAA and 0.5-1.0mg/L SAP. Lower salt strength of medium inhibited shoot elongation but did not have much effect on the shoot and bud induction from the shoot apices. These results will be useful to obtain disease-free plants of the Chinese yam.

  • These studies were conducted to investigate effects of phytohormone and activated carbon on the growth and rooting of teratoma shoots induced from Nicotiana tabacum cv. NC2326 transformed by Aerobacterium tumefaciens C58. GA was effective for shoot elongation and reduction of multiple shoots from teratoma shoot, however, leaves of teratoma shoot cultured on the medium with GA were pointed. ABA was also effective in promoting shoot elongation, but was not for reduction of multiple shoots. Teratoma shoot cultured on the medium with 1 n activated carbon promoted shoot elongation and inhibited the number of shoots differentiated, but was grown as abnormal shoot. Addition of 1% activated carbon and 0.5mg/l BA to culture media was effective for shoot elongation and reduction of multiple shoot and for formation of round leaves as normal leaves. Though these shoots were inoculated on the rooting medium, they could not from roots but formed multiple shoots. Boric acid, myo-inositol and sucrose were also ineffective on the rooting of teratoma shoots.

Multiple Shoot Formation by Apical Meristem Culture in Ipomoea batatas Poir. (고구마 정단분열 조직배양에 의한 多芽體 형성)

  • In sweet potato cultivars 'Mokpo #29' and 'Sanchunza', shoots from extplants were formed 100% on the MS medium with 0.1 ㎎/L NAA and 2.0 ㎎/L BA after 30 days of culture and roots produced from the base of stem at frequencies of 66.7% ('Mokpo #29') and 69.2% ('Sanchunza'), respectively, The media with 0.5∼4.0 ㎎/L BA were produced the greatest frequency of multiple shoot and the most of shoots developed rapidly into normal plantlets with rooting within 60 days of culture. Whereas the cultivar 'Keumsi' failed to produce normal shoot multiplication on the medium with cytokinins alone because of callusing of adventitious shoots. When single shoots with 1 to 2 nodes were excised from the multiple shoot or shoots covered with callus devoid of root and transferred to MS medium with 4.0 ㎎/L BA or kinetin. Host divided shoots showed the callus induction at the stem base and it was enable to obtain regenerated plantlets with shoot and root normally.
