Discovery of molecular chaperone has stimulate cell biologists and thus made it possible to re-examine the processes whereby proteins achieve and maintain their functional conformations within living cells. the term ‘Molecular chaperone’ was first coined to describe one particular protein involved in the assembly of nucleosomes, but the term has now been extended to describe the function of a wide variety of proteins that assist protein transport across membranes, folding of nascent polypeptide, the assembly and disassembly of oligomeric structures, and the recovery or removal of proteins damaged by various environmental stresses including heat shock. Progress of molecular chaperone research is still limited by the lack of 3-dimensional structural information and detailed interacts with taget proteins in the cell. However, several laboratories around the world are attempting to extend our knowledge on the functions of molecular chaperone, and such efforts seem justified to finally provide the answers to the most burning questions shortly.

Numerical calculation was applied to supersonic under-expanded jets, and compared with the results of a linear theory and other experiments. TVD difference scheme was employed to solve 2-dimensional and axisymmetric inviscid Euler equation. This paper aims to explore the effects of angle of divergence and design Mach number of nozzle on the structure of under-expanded jets. The angle of divergence was varied from 0 to 20 deg. The results show that the length of the first cell of the under-expanded jets decreases and Mach disk generates at lower nozzle pressure ratio, if the angle of divergence or design Mach number of nozzle increases. The distance from the nozzle exit to Mach disk in 2-dimensional jets becomes much larger than that of axisymmetric jets, and the widths of the jet boundary and the barrel shock wave are also larger than that of axisymmetric jets. Calculation results indicate that the configuration of the under-expanded jets is strongly dependent on the nozzle pressure ratio.

Experiments are performed to investigate the detailed structure of underexpanded twin jet impinging on a perpendicular flat plate. The major parameters, such as nozzle operating pressure and nozzle spacing, are varied to create different jet flow fields resulted from the complicated interactions of the twin jets. From the surface pressure measurements and shadowgraphs taken by schlieren optical system, the jet structure is strongly dependent on the nozzle operation pressure and the spacing. The results obtained show that the closer nozzle spacing may induce to decrease the diameter of the Mach disk within the first shock cell in the underexpanded twin jet. With the increasing nozzle operating pressure and decreasing the nozzle spacing, a new shock wave appears at the entrainment region between the two jets, due to the enhancement of mixing effect of the both jets. The closer nozzle spacing makes the overall impinging pressure level higher, while severe pressure oscillation along the axis of symmetry. Furthermore it is recommended the wider spacing to obtain higher thrust under the present experimental conditions.
In our previous study [14], we found that heat-shock exposure did not stimulate the neutral trehalase activity in Sacchromyces cerevisiae KNU5377, but did in ATCC24858. Consequently, the trehalose content in KNU5377 became 2.6 times higher than that in ATCC24858. Because trehalose has been shown to stabilize the structure and function of some macromolecules, the present work was focused to elucidate the relationship between trehalose content of these strains and thermal stabilities of whole cells, through differential scanning calorimetry (DSC), and to predict critical targets calculated from the hyperthermic cell killing rates. These analyses showed that the prominent DSC transition of both strains gave identical $T_m$ (transition temperature) values in exponentially growing cells, and that the $T_m$ values of critical targets was about $3^{\circ}C$ higher in KNU5377 than in ATCC24858. Both heat-shocked KNU5377 and ATCC24858 cells displayed similar shifts in their DSC transition profiles. On the other hand, the $T_m$ value of the critical target of KNU5377 was decreased by $2.1^{\circ}C$, which was still higher than ATCC24858 showing no changes. In view of these results, the intrinsic thermotolerance of KNU5377 did not appear to result from the stability of entire cellular components, but rather possibly from that of particular macromolecules, including critical targets, even though it should be investigated in more details. Although the trehalose levels in heat-shocked cells are significantly different, as described in our previous study [14], the overall pattern of thermal stabilities and their predicted critical targets in two heat-shocked strains seemed to be identical. These data suggest that the trehalose levels examined before and after heat shock of exponentially growing cells are not closely correlated with the stabilities of whole cells and/or critical targets in both yeast strains.
High temperature requirement A (HtrA) and its homologues constitute the HtrA familiy proteins, a group of heat shock-induced serine proteases. Bacterial HtrA proteins perform crucial functions with regard to protein quality control in the periplasmic space, functioning as both molecular chaperones and proteases. In contrast to other bacterial quality control proteins, including ClpXP, ClpAP, and HslUV, HtrA proteins contain no regulatory components or ATP binding domains. Thus, they are commonly referred to as ATP-independent chaperone proteases. Whereas the function of ATP-dependent chaperone-proteases is regulated by ATP hydrolysis, HtrA exhibits a PDZ domain and a temperature-dependent switch mechanism, which effects the change in its function from molecular chaperone to protease. This mechanism is also related to substrate recognition and the fine control of its function. Structural and biochemical analyses of the three HtrA proteins, DegP, DegQ, and DegS, have provided us with clues as to the functional regulation of HtrA proteins, as well as their roles in protein quality control at atomic scales. The objective of this brief review is to discuss some of the recent studies which have been conducted regarding the structure and function of these HtrA proteins, and to compare their roles in the context of protein quality control.
In this paper, incompressible and compressible flow characteristics around the butterfly valve have been investigated. In order to simplify the problem, a flat disk valve with various valve disk angles and pressure ratios is considered in the present calculations. It was found that as the disk angle increases, the stagnation point on the front surface of the disk moves to the center of the surface and the inflow velocity decreases. The maximum flow velocity occurs at the downstream of throat because of the formation of vents contracta. As the pressure ratio decreases, compressibility effects increase and the jet formed between the throttle body wall and the disk edge becomes supersonic. This flow also builds up as a shock cell structure. The increase of disk angle and pressure ratio makes the mass flow at the inlet decrease, while the increase of disk angle and the decrease of pressure ratio make the pressure loss coefficient increase.

By extending one-dimensional ZND detonation structure analysis model, a simple model for two-dimensional oblique detonation wave structure analysis is presented by coupling Rankine-Hugoniot relation and chemical kinetics for oblique shock wave and oblique detonation wave. Base on this study, two-dimensional fluid dynamics analysis is carried out to investigate the detailed unsteady structure of oblique detonation waves involving triple point, transverse waves and cellular structures. CFD results provide a deeper insight into the detailed structure of oblique detonation waves, and the simple model could be used as a unified design tool for hypersonic propulsion systems employing oblique detonation wave as combustion mechanism.

A 25kW-class polymer electrolyte membrane (PEM) fuel cell system has been developed for the propulsion of a leisure boat. The fuel cell system was designed to satisfy various performance requirements, such as resistance to shock, stability under rolling and pitching oscillations, and durability under salinity condition, for its marine applications. Then, the major components including a 30kW-class PEM fuel cell stack, a DC-DC converter, a seawater cooling system, secondary battery packs, and balance of plants were developed for the fuel cell system. The PEM fuel cell stack employs a unique design structure called an anodic cascade-type stack design in which the anodic cells are divided into several blocks to maximize the fuel utilization without hydrogen recirculation devices. The performance evaluation results showed that the stack generated a maximum power of 31.0kW while maintaining a higher fuel utilization of 99.5% and an electrical efficiency of 56.1%. Combining the 30-kW stack with other components, the 25kW-class fuel cell system boat was fabricated for a leisure. As a result of testing, the fuel cell system reached an electrical efficiency of 48.0% at the maximum power of 25.6kW with stable operability. In the near future, two PEM fuel cell systems will be installed in a 20-m long leisure boat to supply electrical power up to 50kW for propelling the boat and for powering the auxiliary equipments.

Three-dimensional flow analysis is implemented to investigate the flow through transonic axial-flow compressor rotor(NASA R67) and to evaluate the performances of Abid's low-Reynolds-number k-$\omega$ and Baldwin-Lomax turbulence models. A finite volume method is used fur spatial discretization. The equations are solved implicitly in time by the use of approximate factorization. The upwind difference scheme is used for inviscid terms and viscous terms are approximated with central difference. The flux-difference-splitting method of Roe is used to obtain fluxes at the cell faces. Numerical analysis is performed near peak efficiency and near stall. The results are compared with the experimental data for NASA R67 rotor. Blade-to-Blade Mach number distributions are compared to confirm the accuracy of the code. From the results, it is concluded that Abid'k-$\omega$ model is better for the calculation of flow rate and efficiency than Baldwin-Lomax model. But, the predictions for Mach number and shock structure are almost the same.
