  • In 2004, average temperature was higher, and rainfall was less than those of previous year, The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Seedling diseases, leaf blast, sheath blight and bacterial blight of rice, phytophthova blight, virus diseases and anthracnose of pepper, and sudden wilt syndrome and powdery mildew of chinese melon grown in plastic greenhouse were severe. Especially, sheath blight and bacterial blight of rice occurred two times higher than those of previous year, Panicle blight of rice decreased about 4 times, compared with the previous year, presumed that the lower rainy days, rainfall and RH suppressed spread of the fungal pathogens to panicles of rice. Lower rainfall during mid- and late Aug caused three-times less occurrence of phytophtora blight of red-pepper than that of the previous year, Another diseases surveyed occurred similar or less than those of the previous year.

Purification and Properties of Trehalase from Rhizoctonia solani (Rhizoctonia solani가 생산하는 Trehalase의 정제 및 특성)

  • Nonregulatory trehalase has been purified from mycelia of Rhztoctonia solani. a pathogen of rice sheath blight. Purification procedures involved sonification, gel filtration and fast protein liquid chromatography. Purity was confirmed by isoelectric focusing with silver staining. The purified trehalase was estimated to have a molecular weight of 54,000 and pI point of 5.1. Maximal activity was observed at pH 5.4 and temperature $45^{\circ}C$. The purified trehalase exhibited on apparent Km for trehalose of 3.11 mM and a Vmax of 105.3 $\mu mol min^{-1}\times mg^{-1}$. Validamycin, a commercial antibiotics of rice sheath blight, was a non-competitive inhibitor of Rhzzoctoniu solani trehalase.

Preventive Effect of Ebelactone B, an Esterase Inhibitor on Rice Sheath Blight Caused by Rhizoctonia solani

  • Two types of Rhizoctonia solani esterases induced by cutin hydrolysate were partially purified by ammonium sulfate precipitation and gel filtration. The esterase I with hydrolyzing activity toward both ${\rho}-ni-trophenyl$ butyrate and ${\rho}-nitrophenyl$ palmitate and the esterase II with hydrolyzing activity toward only ${\rho}-ni-trophenyl$ butyrate were inhibited by ebelactone B, an esterase inhibitor produced by actinomycetes with $IC_{50}$ values of 0.01 and $0.09{\;}\mu\textrm{g}/l$, respectively. Spraying on rice seedling with ebelactone B at a concentration of $30{\;}\mu\textrm{g}/ml$ completely suppressed infection by R. solani. Ebelactone B could not protect the wounded rice seedling and did not show any inhibitory effect on the mycelial growth at a concentration of 1 mg/ml. These results indicate that ebelactone B, an esterase inhibitor protects rice plants from infection with R. solani by inhibition of penetration, not through fungitoxic or fungicidal effect.

In Vivo Antifungal Activities of 57 Plant Extracts Against Six Plant Pathogenic Fungi

  • Methanol extracts of fresh materials of 57 plants were screened for in vivo antifungal activity against Magna-porthe grisea, Corticium sasaki, Botrytis cinerea, Phyto-phthora infestans, Puccinia recondita, and Blumeria graminis f. sp. hordei. Among them, seven plant extracts showed disease-control efficacy of more than 90% against at least one of six plant diseases. None of the plant extracts was highly active against tomato gray mold. The methanol extracts of Chloranthus japonicus (roots) (CjR) and Paulownia coreana (stems) (PcS) displayed the highest antifungal activity; the CjR extract controlled the development of rice blast, rice sheath blight, and wheat leaf rust more than 90%, and tomato gray mold and tomato late blight more than 80%. The PcS extract displayed control values of more than 90 % against rice blast, wheat leaf rust, and barley powdery mildew and more than 80% against tomato gray mold. The extract of PcS also had a curative activity against rice sheath blight and that of CjR had a little curative activity against rice blast. On the other hand, the extract of Rumex acetocella roots reduced specifically the development of barley powdery mildew. Further studies on the characterization of antifungal substances in antifungal plant extracts are underway and their disease-control efficacy should be examined under greenhouse and field conditions.

Disease Assessment in Transgenic Rice (CPPO06) Resistant to the Protox-inhibiting Herbicide (Protox 저해 제초제 내성 형질전환 벼(CPPO06)에서의 병 발생 조사)

  • The rice diseases occurring on the transgenic rice plant carrying protox gene (CPPO06) was assessed and compared with other varieties of rice such as Dongjin, Chucheong, Ilpeum, and Onnuri in the fields located in Cheongwon of Chungbuk, Suwon of Gyeunggi, and Gwangju of Chonnam. In the field of Cheongwon, the diseases such as blast, leaf spot, sheath blight and Fusarium blight were observed. False smut were observed only in the field of Suwon, where the ratio of diseased plants was 0.28% in CPPO06 and 0.37% in Onnuri, respectively. In the field of Gwangju, leaf spot caused by Cochlioborus miyabeanus was the most severely occurring disease among rice diseases mentioned above. Fusarium blight occurred in all the 3 fields, which were more severe in CPPO06 plants treated with or without oxadiazon as the herbicide. Except for Fusarium blight, there was no significant difference in the rice diseases as blast, leaf spot, sheath blight and false smut between CPPO06 and other rice varieties.

Isolation and Identification of Myxobacteria KR025 and Searching of Their Bioactive Compounds (점액세균 KR025의 분리 동정 및 생리활성물질의 탐색)

  • Fifty isolates of myxobacteria were isolated from soils from several areas in Korea during 1996-1997 and bioactivity against plant pathogenic fungi of these isolates was examined. A myxobacterial isolate KR025 showed good antifungal activities against Pyricularia oryzae, Cryphonectria parasitica, Colletotrichum lagenarium, and C. gloeosporioides but did not against Rhizoctonia solani, Fusarium oxysporum and Pythium ultimum. The bacterium was identified as Myxococcus fulvus based on morphological and physiological characteristics. Antifungal substances were extracted from culture broth and bacterial cell of Myxococcus fulvus KR025 by ethyl acetate. Antifungal substance of Myxothiazole (100 ${\mu}{\textrm}{m}$/ml) produced by Myxococcus fulvus KR 025 controlled 97.0% rice blast, tomato late blight, wheat leaf rust, and barley powdery mildew and showed 45.0 and 82.6% disease control of rice sheath blight and cucumber gray model, respectively.

Control Efficacy of Phloretin Isolated from Apple Fruits Against Several Plant Diseases

  • In the course of a searching natural antifungal compounds from plant sources, we found that the methanol extract ($3,000\;{\mu}g/ml$) of Malus domestica fruits had potential of control against rice blast (Magnaporthe grisea) and tomato late blight (Phytophthora infestans). Under bioassay-guided purification, we isolated phloretin, a phenolic compound, with in vivo antifungal activity against M. grisea. By 1-day protective application of phloretin ($500\;{\mu}g/ml$), the compound strongly inhibited the disease development of M. grisea and P. infestans on rice and tomato seedlings, respectively. And red pepper anthracnose caused by Colletotrichum coccodes also was moderately suppressed. However, rice sheath blight (Rhizoctonia solani AG1), and barley powdery mildew (Blumeria graminis f. sp. hordei) were hardly controlled. In addition, the compound showed in vitro antifungal activity against some plant pathogenic fungi including Phytophthora capsici, Alternaria panax, Sclerotinia sclerotiorum, R. solani AG4, and M. grisea. This is the first report on the antifungal activity of phloretin against plant pathogenic fungi.

Ecological Studies on the Transition of Sheath Blight of Rice in Korea (한국(韓國)에서의 벼 잎집무늬마름병 발생변동(發生變動)에 관(關)한 생태학적(生態學的) 연구(硏究))

  • In an attempt to obtain a basic information to develop an effective integrated system of controlling sheath blight of rice in Korea, the transition of this disease, the variation of cultural characters and pathogenicity of the pathogen, environmental conditions affecting the disease outbreak and varietal resistance have been investigated. 1. Rice sheath blight which has been minor disease in the past was widely spread, especially since 1971. This disease has promptly spread all over the country and infected 65.2% of total rice growing area in 1976. Various factors are considered to be related to such transition of this disease. Above all, increace of application of nitrogenous fertilizer, early season and earlier cultivation of rice, introduction of more susceptible "Tongil" varieties etc. must be important factors influencing the outbreak of this disease. 2. Great variations in cultural characteristics-such as mycelial growth rate, color of the medium, amount of the aerial mycelium, shape and color of the sclerotia- and in the pathogenicity of isolates of the pathogen, Thanatephorus cucumeris Dank were observed. The optimum temperature for mycelial growth also varied with isolates, from $25^{\circ}C$ to $30^{\circ}C$. There were not necessarily any correlation between curtural characteristics and pathogenicity of isolates of Thanatephorus cucumens. 3. Mycelial grow th of isolates of Thanatephorus cucumens on the PDA medium were correlated with the air temperatures of the region where the isolates were collected. The isolates from the regions with high temperature grew well on PDA medium at $35^{\circ}C$ than those from the region with low temperature, on the other hand, the isolates from the regions with the low temperature grew well on the same medium at $12^{\circ}C$ than those from the regions with high temperature. 4. Pectin polygalacturonase (PG) and cellulase (Cx) were most active on the 3rd day after inoculation on the leaves of rice plant with Thanatephorus cucumeris, whereas pectin methylestrase (PE) was most active on the 4th day after inoculation. Relationship between the activities of PE of isolates and the strength of pathogenicity of isolates was obtained, but PG and cellulase activities were not correlated with pathogenicity of isolates. 5. The tolerence of sclerotia from in-vitro culture to low temperature varied with their water content, the dried cultural sclerotia were more tolerent than wet ones, Dried cultural sclerotia maintained almost 100% germinability for 45 days at $-20^{\circ}C$, whereas wet sclerotia lost viability at $-5^{\circ}C$. The germination ratio of the sclerotia after overwintering changed from 18% to 70% according to the water content of the test paddy fields and the ratio was low in wet paddy condition. 6. To investigate the host range of this fungi in and near paddy field, 17 weeds were inoculated with fungi. The lesions of sheath blight disease was obserbed on Sagittaria trifolia L., Echinochloa crusgalli P. Beauv., Monochoria vaginal is Presl, Polygonum Hydropiper L., Eclipta prostrata L., Digitaria sanguinalis Scapoli. 7. When the level of nitrogen applied was doubled over standard level, total nitrogen content in rice sheath increased, ami when silicate was applied, starch content in rice sheath decreased, inducing the rice plants more susceptible to sheath blight disease. Increased dressing of potash ferilizer reduced the incidence of sheat blight disease. 8. The percentage of infected stems in the early period increased more in the narrow hill plot than in the wide hill plot, but in the late period this tendency was inversed; the percentage of infected stems as well as severity in the wide hill plot increased more compared to the narrow hill plot, and the disease severity in the one plant per hill plot was also low. The number of stems in the wide hill plot was more than the number of stems in the narrow hill plot. This indicates that the microclimate, such as the relative humidity, in the narrow hill plot was more favorable for the development of this disease. 9. There was a high negative correlation between the disease severity of varieties to the sheath blight and the maturity of the varieties, that is, the early varieties were more susceptible than the late ones, and much-tillering varieties usually showed more infection than less tillering varieties. 10. No relationship was obtained between the percentage of infected stems in the early period and the severity after heading, whereas a distinct relationship was obtained between former and latter after Aug. 10.

Comparative Molecular Similar Indice Analysis on Fungicidal Activity of N-phenyl-O-phenylthionocarbamate Derivatives against Rice Sheath Blight and Phytophthora Blight (벼잎집무늬마름병균 및 고추역병균에 대한 N-Phenyl-O-phenyl-thionocarbamate 유도체들의 살균활성에 관한 비교분자 유사성 분석)

  • Comparative molecular similarity indice analysis (CoMSIA) models on the fungicidal activities of N-phenyl substituents (X) in N-phenyl-O-phenylthionocarbamate derivatives against rice sheath blight (Rhizoctonia solani: RS) and phytophthora blight (Phytophthora capsici: PC) were derived. Also, the characterizations of H-bonds between substrates and ${\beta}-tubulin$ were discussed quantitatively. It was revealed that, from the contour maps of CoMSIA models, the H-bond acceptor field contributed the most highly to fungicidal activity for two fungi in common. It is predicted that the selectivity in the fungicidal activity between two fungi is caused by results from the roles of H-bond donor disfavor functional groups in RS and H-bond acceptor disfavor functional groups in PC when these two groups induced at meta- and para-position on the N-phenyl ring. And also, if the substituents (X) are steric disfavor group, negative charge favor groups are introduced at the metaposition in RS and H-bond acceptor group is introduced at the para-position in PC, the antifungal activity against two fungi will be likely able to be increased.
