Cloning and Characterization of the Lactococcus lactis subsp. lactis ATCC 7962 pts HI Operon

  • The ptsH and ptsI genes of Lactococus lactis subsp. lactis ATCC 7962 (L. lactis 7962), encoding the general proteins of phosphotransferase system (PTS) components, HPr and enzyme I, respectively, were cloned and characterized. A 1.3 kb PCR product was obtained using a primer set that was hybridized to the internal region of the L. lactis 7962 pts HI genes and then subcloned into a low-copy number vector, pACYC184. The 5' upstream and 3' downstream region from the 1.3 kb fragment were subsequently clone using the chromosome walking method. The complete ptsHI operon was constructed and the nucleotide sequences determined. Two ORFs corresponding to HPr (88 amino acids) and enzyme I (575 amino acids) were located. The ptsHI genes of L. lactis 7962 showed a very high homology (84-90%) with those genes from other Gram-positive bacteria. A primer extension analysis showed that the transcription started at either one of two adjacent bases upstream of the start codon. Using a Northern analysis, two transcripts were detected; the first, a 0.3 kb transcript corresponding to ptsH and the second, a 2 kb transcript corresponding to ptsH and ptsI. The transcription level of ptsH was higher than that of ptsI. The concentration of the ptsH transcript in cells grown on glucose was similar to that in cells grown on lactose, yet higher than that in cells grown on galactose. The ptsI transcript was scarcely detected in cell grown on lactose or galactose. The ptsI transcript was scarcely detected in cells grown on lactose or galactose. The results of a sequence analysis and Northern blot confirmed that the ptsH and ptsI genes of L. lactis 7962 were arranged in an operon like other known ptsHI genes and the expression of the ptsHI genes was regulated at the transcriptional level in response to the carbon source.

Safety Evaluation of Water-soluble Chromium in Makeup Products (메이크업 화장품에서 수용성 크롬의 안전성 평가)

  • There is now a growing concern about the possible toxicity of heavy metals in cosmetics. Heavy metals can be used as cosmetic ingredients or may be present as low level impurities in some of the raw materials. Chromium derivatives are used as pigments in cosmetics. Chromium is essential and toxic trace elements. Chromium may cause skin allergy. However, the regulations related to cosmetic products give no limit values for Chromium. Hexavalent chromium is significantly more toxic than trivalent chromium. Hexavalent chromium may present a carcinogenic risk at high concentrations. Therefore, it is important to consider oxidation state of chromium when analyze chromium. The purpose of this study is to determine the concentrations of water-soluble trivalent and hexavalent chromium in samples of makeup products, and to assess the safety of cosmetics on the basis of animal sensitization tests using guinea pig. The present study of chromium in 48 makeup products of 12 manufacturers provides a basis for assessing safety of makeup products. Water-soluble hexavalent chromium was not detected in any product. Water-soluble trivalent chromium was detected in only 9 eye shadows out of 48 makeup products, and could not be quantified 3 out of 9 eye shadows. The highest level of water-soluble trivalent chromium was about 10 mg/kg in spite of 90,000 mg/kg of total chromium. The results of animal sensitization tests show that 200 mg/kg of trivalent chromium and 5 mg/kg of hexavalent chromium have no harmful effect. No cross-reaction among these metals was found. Accordingly, the concentrations of water-soluble chromium in makeup products seemed to be safe. The overall results indicate that chromium in cosmetics probably have no significant toxicological effects. However, It is necessary to set guidelines on the maximum permissible concentration of water-soluble chromium in cosmetics.

Development of Molecular Diagnosis Using Multiplex Real-Time PCR and T4 Phage Internal Control to Simultaneously Detect Cryptosporidium parvum, Giardia lamblia, and Cyclospora cayetanensis from Human Stool Samples

  • This study aimed to develop a new multiplex real-time PCR detection method for 3 species of waterborne protozoan parasites (Cryptosporidium parvum, Giardia lamblia, and Cyclospora cayetanensis) identified as major causes of traveler's diarrhea. Three target genes were specifically and simultaneously detected by the TaqMan probe method for multiple parasitic infection cases, including Cryptosporidium oocyst wall protein for C. parvum, glutamate dehydrogenase for G. lamblia, and internal transcribed spacer 1 for C. cayetanensis. Gene product 21 for bacteriophage T4 was used as an internal control DNA target for monitoring human stool DNA amplification. TaqMan probes were prepared using 4 fluorescent dyes, $FAM^{TM}$, $HEX^{TM}$, $Cy5^{TM}$, and CAL Fluor $Red^{(R)}$ 610 on C. parvum, G. lamblia, C. cayetanensis, and bacteriophage T4, respectively. We developed a novel primer-probe set for each parasite, a primer-probe cocktail (a mixture of primers and probes for the parasites and the internal control) for multiplex real-time PCR analysis, and a protocol for this detection method. Multiplex real-time PCR with the primer-probe cocktail successfully and specifically detected the target genes of C. parvum, G. lamblia, and C. cayetanensis in the mixed spiked human stool sample. The limit of detection for our assay was $2{\times}10$ copies for C. parvum and for C. cayetanensis, while it was $2{\times}10^3$ copies for G. lamblia. We propose that the multiplex real-time PCR detection method developed here is a useful method for simultaneously diagnosing the most common causative protozoa in traveler's diarrhea.

APPLICATION OF TIME-OF-FLIGHT NEAR INFRARED SPECTROSCOPY TO WOOD

  • In this study, the newly constructed optical measurement system, which was mainly composed of a parametric tunable laser and a near infrared photoelectric multiplier, was introduced to clarify the optical characteristics of wood as discontinuous body with anisotropic cellular structure from the viewpoint of the time-of-flight near infrared spectroscopy (TOF-NIRS). The combined effects of the cellular structure of wood sample, the wavelength of the laser beam λ, and the detection position of transmitted light on the time resolved profiles were investigated in detail. The variation of the attenuance of peak maxima At, the time delay of peak maxima Δt and the variation of full width at half maximum Δw were strongly dependent on the feature of cellular structure of a sample and the wavelength of the laser beam. The substantial optical path length became about 30 to 35 times as long as sample thickness except the absorption band of water. Δt ${\times}$ Δw representing the light scattering condition increased exponentially with the sample thickness or the distance between the irradiation point and the end of sample. Around the λ=900-950 nm, there may be considerable light scattering in the lumen of tracheid, which is multiple specular reflection and easy to propagate along the length of wood fiber. Such tendency was remarkable for soft wood with the aggregate of thin layers of cell walls. When we apply TOF-NIRS to the cellular structural materials like wood, it is very important to give attention to the difference in the light scattering within cell wall and the multiple specular-like reflections between cell walls. We tried to express the characteristics of the time resolved profile on the basis of the optical parameters for light propagation determined by the previous studies, which were absorption coefficient K and scattering coefficient S from Kubelka-Munk theory and n from nth power cosine model of radiant intensity. The wavelength dependency of the product of K/S and n, which expressed the light-absorbing and -scattering condition and the degree of anisotropy, respectively, was similar to that of the time delay of peak maxima Δt. The variation of the time resolved profile is governed by the combination of these parameters. So, we can easily find the set of parameters for light propagation synthetically from Δt.

  • The research described here was undertaken with the aim of monitoring, optimizing and ultimately controlling the production of heterofermentative microbes used as starters in the salami industry. The use of starter cultures in the fermented meats industry is a well-established technique used to shorten and standardize the ripening process, and to improve and control the organoleptic quality of the final product. Starter cultures are obtained by the submerged cultivation of suitable microorganisms in stirred, and sometimes aerated, fermenters where monitoring of key physiological parameters such as the concentration of biomass, substrates and metabolites suffers from the general lack of real-time measurement techniques applicable to aseptic processes. In this respect, the results of the present work are relevant to all submerged fermentation processes. Previous work on the application of on-line NIR spectroscopy to the lactic acid fermentation (Dosi et al. - Monreal NIR1995) had successfully used a system based on a measuring cell included in a circulation loop external to the fermenter. The fluid handling and sterility problems inherent in an external circulation system prompted us to explore the use of an in-line system where the NIR probe is immersed in the culture and is thus exposed to the hydrodynamic conditions of the stirred and aerated fluid. Aeration was expected to be a potential source of problems in view of the possible interference of air bubbles with the measurement device. The experimental set-up was based on an in-situ sterilizable NIR probe connected to the instrument by means of an optical fiber bundle. Preliminary work was carried out to identify and control potential interferences with the measurement, in particular the varying hydrodynamic conditions prevailing at the probe tip. We were successful in defining the operating conditions of the fermenter and the geometrical parameters of the probe (flow path, positioning, etc.) were the NIR readings were reliable and reproducible. The system thus defined was then used to construct and validate calibration curves for tile concentration of biomass, carbon source and major metabolites of two different microorganisms used as salami starters. Real-time measurement of such parameters coupled with the direct interfacing of the NIR instrument with the PC-based measurement and control system of the fermenter enabled the development of automated strategies for the interactive optimization of the starter production process.

  • Internet, which has been developed as a new exchange revolution, forms a huge virtual exchange market, and the innovative electronic commerce has completely broken off the way of existing goods distribution. This study begins with an awareness of the importance of customer retention to keep winning over the competition in internet shopping mall. In order to explain of the customer retention between individual and internet shopping mall, the study introduces first a satisfaction on shopping followed by an awareness of the importance of customer retention, and looks into a formation process of trust, satisfaction, and relationship orientation occurred by the offer of valuable convenience to customers. The study also explores the influence on shopping value by the characteristics with which internet shopping mall can bear, unfold by a cause and effect relationship the degree of shopping satisfaction, trust, and relationship orientation, and inquires a question to find out how to fuse the characteristics for internet retention. Therefore, this study has the following purposes: After examining prior research for the characteristics of internet shopping mall, it presents a possibility to connect shopping value with customer retention in light of theoretical system on characteristic elements derived from emotional and utilitarian perspectives. In order to achieve the purposes, the characteristics of internet retailing shop included site design, virtual reality, web awareness, customer concern, merchandise search, information supply, product value, and transaction system. Hypotheses were set up for the relationship with these characteristics and substantially analyzed. To prove this research, we analyzed collected data in which customers had experienced in shopping at internet shopping mall and discussed strategic current issues about its analytic results.

Development of a Hospital Service-based Costing System and Its Application (병원서비스별 원가분석모형의 개발과 적용)

  • The managerial environment of hospitals in Korea characterized by low levels of medical insurance fees is worsening by increasing government regulations as to the utilization of medical services, rising costs of labor, material, and medical equipments, growing patient expectations concerning the quality of services, and escalating competitions among large hospitals in the market. Hospitals should seek for their survival strategies in this harsh environment and they should have information about costs of their products in doing so. However, it has not been available due to the complexity of the production process of hospital services. The objectives of this study were to develop a service-based cost accounting model and to apply the developed model to a study hospital to obtain cost information of hospital services. A model commonly used for the job-order product cost accounting in the manufacturing industry was modified for the use in hospitals in Korea. Actual costs, instead of standard costs, incurred to produce a unit of services during a given period of time were estimated in the model. Data required to implement the model included financial information, statistics for the allocation of supportive cost center costs to final cost centers, statistics for the allocation of final cost center costs to services, and the volume of each services charged to patients during a study period. The model was executed using data of a university teaching hospital located in Seoul for the fiscal year 1992. Data for financial information, allocation statistics fo supportive service costs, and the volume of services, most of them in electronic form, were available to the study. Data for allocation statistics of final cost center costs were collected in the study. There were 15 types of evaluation and management service, 2, 923 types of technical service, and 2, 608 types of drug and material service charged to patients in the study hospital during the fiscal year 1992. Labor costs of each of seven types of pesonnel, material costs of 611 types of drugs and materials, and depreciation costs of 212 types of medical equipments, miscellaneous costs, and indirect costs incurred in producing a unit of each services were estimated. Medical insurance fees for basic services such as evaluation and management of inpatients and outpatients, injection, and filling prescriptions, and for operating procedures were found to be set lower than costs. Infrequent services which use expensive medical equipments showed negative revenuse as well. On the other hand, fees for services not covered by the insurance such as CT, MRI and Sonogram, and for laboratory tests were higher than costs. This study has a significance in making it possible for a hospital to obtain cost information for all types of services which produced income based on all types of expenses incurred during a given period of time. This information can assist the management of a hospital in finding an effective cost reduction strategy, an efficient service-mix strategy under a given fee structure, and an optimum strategy for within-hospital resource allocations.

The Study on the Costume of Parhae through the Exchange for Tang and Ancient Japan - focused on the Exchanged Items- (대당$\cdot$대일본과의 교류관계를 통해 본 발해의 복식문화 연구 - 교류시 품목을 중심으로 -)

  • This study is consider·ed about the relation or costume among Parhae, ancient Japan and Tang and the culture of this time commonly was influenced by Tang;s culture. The recorded items, which were exchanged from Parhae to Tang and ancient Japan, are the pelt of marten, seal, bear and tiger skin or Ammowhaha(암모화) Gu(구) and so on. Other items from Tang and ancient Japan to Parhae are various clothes and material like silk and so on. This Parhae's clothes made with leather obtained through hunting were the main product of Parhae and had been greatly famous in Tang and ancient Japan. At that time, Tang and ancient Japan had been only dependent on the agricultural life but had not been accustomed to the hunting life, and thereupon, the people in Tang and Nara Era had difficulties to get leather clothes. Accordingly, the leather products stood for the rich, and the people who purchased the leather products from Parhae were recognized as the rich. On the other hand, The prince and the royal family in Parhae were dispatched to Tang as Sooktwui(숙위). They paid a tribute to Tang and In return for it, Tang gave to Sookwui as well as to Parhae's envoys various items and clothes such as Po(포), belt(대), gold and silver Eodae(어대) and textile like a type of silk. These clothes were considered as the formal uniform like Kwanbok(관복) of Parhae. Specially, the time when Sookwui was sent to Tang rot-responded with the time when Tang gave formal uniform to Parhae's envoys. Therefore, we can confirm that the sending of Sookwui influenced to form uniform system of Parhae. In this exchange relation, there were several duplicated items showed, for example Po(포)-belt(대), Eui set(의일습), Eoeui(어의), Jobok(조복), Go(고). These items in Parhae were appeared in process of exchanging from Tang to Japan and from Tang to Parhae. At that time, the exchanges between Tang and Parhae were frequently active much more than those between Tang and Japan, and the acceptance of the culture from Tang was easier in Parhae than Japan in consideration of the geographical location. Therefore, those clothes could be understood as the items already used in Parhae and affected from Tang. These items in Parhae were appeared in process of exchanging from Tang to Japan and from Japan to Parhae. As a result, it is expected that the costumes, which Parhae took from Japan, are the same as those of Tang.

Microstructural Realization of SD400 Rebar by Developing Tempcore Simulation Apparatus (템프코어 냉각모사 장치 개발을 통한 SD400 철근 미세조직 구현)

  • The cooling process referred to as Tempcore has been applied to produce a high-strength rebar. Excellent rebar with strength and weldability can be manufactured from mild steel without the addition of alloying elements by using the Tempcore process. However, there are limitations to evaluating the effect of various chemical compositions and cooling conditions within a site facility. In this study, we developed an apparatus to simulate the Tempcore process and obtained microstructures with a hardened surface layer, an intermediate region and a soft inner core. The experimental apparatus has been equipped with a cooler set that is the same as the site facility and consists of a pump line that supplies pressure of 12-13 bar and flow rate of up to $50m^3/h$. In accordance with the simulation result of steel grade SD400 that requires more than 400 MPa of yield strength, both the hardened area ratio and the hardness with respect to each cooling depth were found to agree well with the product.

  • This study would be research for correlations between ethical factors and company's competitiveness of small and medium enterprises thorough recognizing of important company ethic and approaching to them theoretically. This study has collected all information which small and medium enterprises around Seoul and Kyounggi province and employing more than 5, which 134- survey are used totally. Study reveals that firstly customer and social responsibility provide Positive effects on the organization Dust, secondly CEO's views serve those positive effects as well, thirdly it is not preyed that ethical capacity of company have a positive effect on organization trust. Fourthly, organization trust has good effects on competitiveness of product and price. This results lead to some suggestions as follows. First of all, company should serves systemized value to customers and their societies. For that, it is necessary to analyse their needs and to reflect them into corporate management to build close relationship with customers and reliable marketing activities, efforts to enhance corporate images. This chain of work can make employees feel proud themselves. Secondly, supervisors including CEO should set the pace with high-consensus for ethical behaviour. Thirdly, ethical factors should be improved for improving organization trust simultaneously. Every customer, no matter they are inner or outer, should have highly required ethical understandings for their networks. Furthermore, they have to enhance organization trust and to strengthen company competitiveness through stimulating employee's courages and contribution.
