In this study the effects of water extracts of endocarps and seeds of omija(Schizandra chinensis Baillon)on alcohol metabolism in rats were investigated. 25% alcoholic solution(v/v,0.75g/200g B.W.)and 40% alcoholic solution(v/v,0.80g/200g B.W.)were orally administered to rats for 2 hours. The levels of metabolites and enzyme activities both on serum and liver were not changed by acute oral adminiatration of 25% and 40% alcoholic solution. Blood alcohol levels were significantly lowered by treatment of water extracts of endocarps and seeds of omija in rats administered with 25% and 40% alcohol. In 25% alcohol treated group, serum GPT level was decreased but hepatic G-6-P DH and pyruvate levels were increased. In 40% alcohol treated group, water extracts of endocarps and seeds of omija resulted in a significant decreased in serum levels of FFA, GPT but increased in serum glucose level. Hepatic levels of cytosolic protein, glycogen and pyruvate were increased by water extracts of omija parts in 40% alcohol treated group.

Objectives: In most DEHP exposure assessment studies, single spot urine sample was used. It could not compare the exposure level among studies. Therefore, we are going to represent the necessity of selection of proper sampling time of spot urine for assessing the environmental DEHP exposure, and the association urinary DEHP metabolites with steroid hormones. Methods: We collected urine and plasma from 25 men. The urine sampling times were at the end of the shift (post-shift) and the next morning before the beginning of the shift (pre-shift). Three metabolites of DEHP {mono(2-ethylhexyl) phthalate [MEHP], mono-(2-ethyl-5-hydroxyhexyl)phthalate [MEHHP], and mono(2-ethyl-5-oxohexyl)phthalate [MEOHP]} in urine were analyzed by HPLC/MS/MS. Plasma luteinzing hormone, follicle stimulating hormone, testosterone, and $17{\beta}$- estradiol were measured at pre-shift using a ELISA kit. A log-transformed creatinine-adjusted urinary MEHP, MEHHP, and MEOHP concentration were compared between the post- and pre-shift. The Pearson’s correlation was calculated to assess the relationships between log-transformed urinary MEHP concentrations in pre-shift urine and hormone levels. Results: The three urinary metabolite concentrations at post-shift were significantly higher than the concentrations in the pre-shift (p<0.0001). The plasma hormones were not significantly correlated with log-transformed creatinine - adjusted DEHP metabolites. Conclusions: To assess the environmental DEHP exposure, it is necessary to select the urine sampling time according to the study object. There were no correlation between the concentration of urinary DEHP metabolites and serum hormone levels.
Objectives: Medicinal plants are vital sources of bioactive compounds that are useful for the treatment of patients with snake bites or are indirectly applicable for boosting the effects of conventional serum therapy. These plants are being used traditionally by local healers and tribes for the treatment of patients with snake bites and therefore can be used as an alternative against snake envenomation. Scientifically, using the secondary metabolites of plants to neutralize venom enzymes has an extra benefit of being based on traditional knowledge; also, the use of such metabolites for the treatment of patients with snake bites is cheaper and the treatment can be started sooner. Methods: All the available information on various secondary metabolites exhibiting venom neutralizing ability were collected via electronic search (using Google books, Pubmed, SciFinder, Scirus, Google Scholar, and Web of Science) and articles of peer-reviewed journals. Results:Recent interest in different plant has focused on isolating and identifying of different phytoconstituents that exhibit Phospholipase A2 activity and other venom enzyme neutralizing ability. In this support convincing evidence in experimental animal models are available. Conclusion: Secondary metabolites are naturally present, have no side effect, are stable for a long time, can be easily stored, and can neutralize a wide range of snake enzymes, such as phospholipase A2, hyaluronidase, protease, L-amino acid oxidase, 5'nucleotidase, etc. The current review presents a compilation of important plant secondary metabolites that are effective against snake venom due to enzyme neutralization.
The effect of dietary pectin, tangerine pulp meal, propionate, lactate or fumarate on cholesterol (C) and triacylglycerol(TG) levels in the serum and liver, and the effect of pectin on dietary C absorption were studied in a series of three experiments. Mature female Sprague Dawley rats were fed a control diet or diets containing 5% pectin, 5% tangerine pulp meal, 3% propionate, 3% lactate 3% fumarate, or 10% pectin. Serum total C levels were lower(p<0.05) in rats fed the diet containing 5% pectin than in control rats after a 4-week feeding period(93.8 vs 119.2mg/100mL). Serum HDL, LDL+VLDL C levels were not different among diet groups. Liver total C level was also lower(p<0.05) in rats fed the diet containing 5% pectin than in control rats, but liver TG level was not influenced by diet. Dietary propionate, lactate or fumarate did not reduce serum C, indicating that propionate is not a regulator of serum C. However, dietary pectin(10%) increased fecal excretion of dietary C(or its metabolites) more than 70% over a control value. Our data indicate that dietary pectin reduces serum and liver C levels by increased fecal secretion of dietary C, but not by its fermentation product propionate or other gluconeogenic substrates. Fifth one Nili-Ravi dairy buffaloes in their last two months of gestation, were selected in NWFP Pakistan. Rectal palpation was carried out fortnightly, until the occurrence of first estrus. Ovulation was confirmed per rectum and milk progesterone levels (MPL). Body condition score (BCS)was recorded weekly. Milk samples were collected weekly for MPL and blood samples fortnightly for metabolites analysis. Milk yield was recorded and samples were collected for milk fat. The buffaloes calving during the normal breeding season (NBS, August to January)had short (p<0.01) postpartum estrus interval of $55.95{\pm}4.90$ days versus $91.15{\pm}11.61 $ days in the buffaloes calving during the low breeding season (LBS, February to July). MPL in the LBS calvers remained lower than NBS calves (p<0.01). The incidence of silent ovulation was higher during LBS as compared to NBS (70.6% versus 29.4%). MPL showed a pattern opposite to atmospheric temperature. In NBS calvers serum glucose levels were higher (p<0.01) and magnesium levels were lower (p<0.01) than LBS calvers. Higher serum urea was found in summer and spring than that autumn and winter (p<0.05). The reproductive performance in buffaloes calving in the LBS coincided with a low BCS (p<0.01). Fat corrected milk production (FCM) was higher in NBS than LBS (p<0.01) calvers.
Thirty new born crossbred (Bos taurus${\times}$Bos indicus) calves, divided randomly in a $3{\times}2$ factorial design, were fed calf starters containing one of three protein sources i.e., groundnut cake (GN), cottonseed meal (CS) and meat and bone meal (MB) along with either raw (M) or gelatinized maize (MG) for 90d. Milk was fed upto 56d of age. Green oats and respective calf starters were offered from 14d of age onwards ad lib. Clinical profile of serum suggested significantly (p<0.05) higher albumin and lower alanine aminotransferase activity due to CS feeding. Alklaine phosphatase activity varied significantly (p<0.05) among dietary treatments showing interaction between protein and starch sources. Inclusion of gelatinized maize resulted in significantly higher concentration of serum globulin (p<0.05) and alkaline phosphatase activity (p<0.01). reduced (p<0.05) ruminal pH was accompanied by a significant decrease (p<0.01) in $NH_3-N$ concentration in the strained rumen liquor (SRL) of MG fed calves. Ruminal amylase activity was lower (p<0.05) on MG diets. Alanine aminotransferase activity in the rumen exhibited a significant (p<0.01) interaction between protein and starch sources. While feeding of CS significantly (p<0.01) reduced alanine aminotransferase activity, inclusion of thermally processed maize reduced (p<0.01) both aspartate and alanine aminotransferase activities in the rumen. The overall blood picture was similar among treatments, whereas rumen metabolites especially enzyme activities, seems to be altered with source of degradable protein an starch.
This experiment was conducted to determine the effect of nattokinase (NK) additives on milk production and composition, and blood metabolites in dairy cows. The two kinds of nattokinase with high fibrinolytic activity were produced by two strains of bacteria, Bacillus amyloliquefacines (NK1) and Bacillus subtilis (NK2). Total fifteen Holstein cows (average $1.83{\pm}0.37$ parity; average milk yield $23.2{\pm}3.2$ kg/d) were randomly assigned to three treatments (5 animals per treatment). Cows were fed TMR supplemented with 0g, 100g and 100g for control, NK1 and NK2 treatment, respectively for 4 weeks. Milk yield was significantly higher (p<0.05) for NK1 (22.89 kg/d) than for control (21.07 kg/d) and NK2 (21.36 kg/d). Somatic cell counts in NK treatments were significantly lower than that in control group (58,000 vs. 21,000 and 35,000 cells/ml, control vs. NK1 and NK2). Serum ALT levels in all treatment were similar to the range of 32.00~35.83 IU/L, but AST levels in NK1 (85.67 IU/L) was significantly decreased compared with those in control and NK2 (121.67 and 117.67 IU/L respectively). Serum T-CHO levels in NK1 (145.33 mg/dl) was significantly decreased (p<0.05) compared with that in control (179.00 mg/dl) and NK2 (176.17 mg/dl). This finding showed that NK1 additives could possibly have a positive effect in lactation performance of mid-lactation dairy cows by increasing milk yield, reducing somatic cell count, improving liver function and decreasing cholesterol in blood.
Effects of water extracts of endocarps and seeds of omija(Schizandra chinensis Baillon)on alloxan and CCl4 induced liver injuries were determined by measuring the contents of metabolites and enzyme activities both in liver and serum in male Sprague-Dawley rats. The treatment with water extracts in parts of omija showed low levels of glucose, GOT, LDH and FFA of serum but high level of hepatic G-6-P DH in alloxan treated rats. CCl4 treatment with water extracts of parts of omija resulted in low levels of GOT, LDH, UN of serum and hepatic pyruvate but hepatic cytosolic protein has been increased.
Excretory-secretory products of Toxocara canis larvae have been considered as a major functional antigen in immune responses against toxocariasis. We studied ultrastructural localization of T. canis second-stage larval antigen using a seropositive human serum under immunogold electron microscopy. High-density gold particles were observed in the secretory cells, excretory duct, intestinal epithelium, and cuticle of the larval worm sections. The distribution of the positive reactions in the larval worms suggests that the nature of the antigen is excretory-secretory antigen including waste metabolites and secretory enzymes.
Twelve adult male goats, comprising of six castrated and six intacts, (2.5-3 years; $24.4{\pm}0.62kg$) were randomly but evenly divided into two groups ($I_0$ and $I_{100}$) and fed conventional concentrate mixture along with Leucaena leucocephala leaf meal (100 g/head approx.), the latter to supply 50 per cent of the crude protein (CP) requirements. The $I_{100}$ group was provided with supplemental iodine as potassium iodide solution at 0.1 mg/day/animal. Wheat straw was provided ad libitum as sole source of roughage during the experimental period of 105 d. Blood samples were collected at the begining (0 d) and thereafter at 30, 60 and 90 d of experimental feeding. The study revealed that the serum glucose level was significantly higher (p<0.01) in $I_{100}$ group as compared to $I_0$. Haemoglobin, packed cell volume and serum concentrations of total protein, albumin, globulin, calcium, inorganic phosphorus and alkaline phosphatase did not show significant differences as a result of iodine supplementation. Though the serum levels of triiodothyronine ($T_3$) were comparable between the two groups, that of thyroxine ($T_4$) increased significantly (p<0.001) in the $I_{100}$ group. The $T_3:T_4$ ratio was also similar between both the groups. The study indicated that the adverse effect of Leucaena feeding on thyroid gland could possibly be alleviated by provision of extra iodine. However, this needs further confirmation using long duration studies.
