Selenium-containing peptide (Selenium peptide) was produced by autolysis of Saccharomyces cerevisiae which was cultured in inorganic selenium-supplemented medium. Selenium peptide showed antioxidant activity and protective effects on UVB irradiated human fibroblast. Minimal toxicity of selenium peptide was observed whereas selenium nitrate exhibited cell toxicity as low as $10^{-9}\;M$. Selenium peptide also increased human fibroblast growth, procollagen type I and also decreased MMP-1 (matrix metalloprotease-1). This result showed the potential of selenium peptide as a nontoxic antioxidant.
Background: Selenium is one of the trace minerals whose deficiency is known to lead to complications of female reproduction. The identified gaps in researches regarding selenium and pregnancy include optimizing the dosage of selenium supplementation, timing of supplementation, finding the best form and type of selenium, and selenium administration combined with other antioxidants. Hence, this study was conceptualized to address one of the identified gaps, that is, to find out the best timing of selenium administration around the time of pregnancy. Specifically, this study aimed to assess the effects of maternal Selenium-supplementation, administered at various stages of periconception period, on murine blastocyst morphology, percent occurrence of good quality blastocysts, and implantation status. Methods: ICR female mice were randomly assigned into the unsupplemented group (Group I) receiving basal diet without selenium, and treatment groups given with $3.0{\mu}g$ selenium-supplement per day during pregestation only (Group II), pregestation-throughout-gestation (Group III) and gestation only (Group IV). Both blastocyst morphology and implantation status were assessed. Results: The morphometric measurements of blastocysts appeared to be unaffected by selenium-supplementation at different stages of periconception. Selenium-supplementation at pregestation only (Group II) and gestation only (Group IV) produced higher percent occurrence of good quality blastocysts and lower percent pre-implantation loss than Group III. Among all the treatment groups, Group III (Selenium-supplementation during pregestation-to-gestation) yielded the lowest quality blastocysts and highest percent pre-implantation loss. Conclusion: Maternal selenium-supplementation during pregestation and gestation stages of the periconception period yielded a high percent occurrence of good quality blastocysts and pre-implantation success.

Purpose: We conducted this study to study the clinical correlation between the characteristics of gastric cancer and serum selenium and zinc levels. Materials and Methods: The following data were measured in the baseline serum selenium and zinc levels of 74 patients with curative gastrectomy subsequent to confirmed gastric cancer, from March 2005 to August 2012. Results: Among the 74 gastric cancer patients, 53 patients were male. Mean serum selenium and zinc levels were $118.7{\pm}33.1$ ug/L and $72.2{\pm}24.3$ ug/dl, respectively. Seven patients (9.5%) showed lower selenium level, and 33 patients (44.6%) showed lower zinc level. Serum Selenium level was $99.1{\pm}31.8$ ug/L in cardia cancer group (10 cases) and $121.8{\pm}32.4$ ug/L in non-cardia cancer group (64 cases)(P=0.044). According to tumor gross type, zinc level was $78.7{\pm}29.6$ ug/dl in early gastric cancer (33) and $66.9{\pm}17.8$ ug/dl in advanced gastric cancer (41) (P=0.064). Conclusions: The serum selenium level was highly correlated with the location of gastric cancer. The serum zinc level was lower in advanced gastric cancer.

Recently, amorphous selenium is used as x-ray conversion material for flat-panel x-ray detector. In this paper, we investigated the effect of breakdown under high voltage and leakage current in PN-type multi-layer structure based on p-type a-Se and n-type conductive thin film. Experimental results show that the multi-layer based detector reduced leakage current because n-type CeO2 conductive layer prevent from hole injection into a-Se layer from collection electrode, Also, the breakdown voltage was improved by dielectric layer between a-Se and top electrode.

Objective: Placental oxidative stress is known to be a factor that contributes to pregnancy failure. The aim of this study was to determine whether selenium could induce antioxidant gene expression and regulate invasive activity and mitochondrial activity in trophoblasts, which are a major cell type of the placenta. Methods: To understand the effects of selenium on trophoblast cells exposed to hypoxia, the viability and invasive activity of trophoblasts were analyzed. The expression of antioxidant enzymes was assessed by reverse-transcription polymerase chain reaction. In addition, the effects of selenium treatment on mitochondrial activity were evaluated in terms of adenosine triphosphate production, mitochondrial membrane potential, and reactive oxygen species levels. Results: Selenium showed positive effects on the viability and migration activity of trophoblast cells when exposed to hypoxia. Interestingly, the increased heme oxygenase 1 expression under hypoxic conditions was decreased by selenium treatment, whereas superoxide dismutase expression was increased in trophoblast cells by selenium treatment for 72 hours, regardless of hypoxia. Selenium-treated trophoblast cells showed increased mitochondrial membrane potential and decreased reactive oxygen species levels under hypoxic conditions for 72 hours. Conclusion: These results will be used as basic data for understanding the mechanism of how trophoblast cells respond to oxidative stress and how selenium promotes the upregulation of related genes and improves the survival rate and invasive ability of trophoblasts through regulating mitochondrial activity. These results suggest that selenium may be used in reproductive medicine for purposes including infertility treatment.

The development of digital x-ray detector has been extensively progressed for the application of various medical modalities. In this study, we introduce a new hybrid-type x-ray detector to improve problems of a conventional direct or indirect digital x-ray image technology, which composed of multi-layer structure using a CaWO$_4$ phosphor and amorphous selenium (a-Se) photoconductor. The leakage current of our detector was found to be ∼180 pA/cm$^2$ at 10 V/m, which was significantly reduced than that of a single a-Se detector. The x-ray sensitivity was measured as the value of 4230 pC/cm$^2$/mR at 10 V/m. We found that the parylene thin film between a CaWO$_4$ phosphor and an a-Se layer acts as an insulator to prevent charge injection from indium thin oxide (ITO) electrode into an a-Se layer under applied bias.
Purpose: This study was aimed to assess selenium and zinc status in female collegiate athletes and their relationship with dietary intake. Methods: Female collegiate athletic groups of judo and aerobics, and healthy sedentary collegiate females were recruited for this study and their serum selenium and zinc contents were measured by the neutron activation analysis (NAA) method. In addition, the dietary intake of subjects was measured using the two days 24-hour recall method. Results: Serum selenium in judo athletes was $10.7{\mu}g/dl$, which was significantly lower than that of aerobic athletes ($12.2{\mu}g/dl$), but not different from that of the sedentary group ($11.4{\mu}g/dl$). Additionally, serum zinc levels were $96.1{\mu}g/dl$ and $90.2{\mu}g/dl$ in aerobic and judo athletes, respectively, which did not differ significantly. Moreover, dietary selenium and zinc intake of the athletic groups did not differ significantly from that of the sedentary female group. Overall, 33.3% of the serum selenium concentration variation was explained by the intake of vitamin E, selenium and saturated fatty acids, while 14.7% of the serum zinc level variation was explained by the intake of saturated fatty acids. The strongest dietary indicator for serum selenium and zinc levels was saturated fatty acids intake. Conclusion: Judo athletes appear to have lower selenium status than aerobic athletes, suggesting different body selenium status according to sport type. To maintain body selenium and zinc levels, the dietary intake of saturated fatty acids should be decreased.
Many defined cell culture media were formulated over 3() years ago and may be deficient in certain micronutrients whose essentiality has only subsequently been recognised. The objective of this study was to evaluate whether alpha-minimal essential medium (MEM) supplemented with 10% foetal bovine serum contained sufficient selenium for optimal activity of the selenium containing enzymes cytosolic glutathione peroxidase (cGPx) and phospholipid hydroperoxide glutathione peroxidase (PHGPx) in cultured Chinese hamster ovary (CHO) cells. Additionally, the effect of zinc deficiency and metallothionein (MT) overexpression on cGPx and PHGPx activity was studied. The addition of 100 nM of selenous acid to the culture medium increased cGPx expression by 10-fold and PHGPx by about 2-fold in both wild-type CHO-K1 cells and CHO-K1 cells overexpressing mouse MT-1. Zinc deficiency had no significant effect on enzyme activity, but cells overexpressing mouse MT-1 had higher levels of cGPx activity. Zinc deficiency decreased cell survival but overexpression of MT-1 was partially protective, probably because its presence in quantity favoured the uptake, sequestration and cellular retention of any remaining zinc. This study demonstrates that selenium in complete alpha-MEM is insufficient for optimal cGPx and PHGPx activity and may compromise the cellular response to oxidative stress.
This study was conducted to determine the total amount and type of seleniwn (Se) in the Se-enriched mushroom and its spent mushroom composts (SMC), and to investigate the metabolism in relation to Se accwnulation in the mushroom. Mushrooms, Flammulina velutipes, used in this study were grown for 60 days by adding 2 rng of inorganic Se (Na2Se03) per kg of mushroom composts (MC) on as-fed basis and were compared with normal mushrooms grown on the non Se-supplemented Me. Total Se contents for Se-treated mushrooms were significantly increased (P < 0.0001) by 20-fold (4.51 $\mu$/ g of dry) compared to Se-untreated (0.23 $\mu$/ g of dry). On the contrary, organic Se ratio was significantly lower (P < 0.0001) in the Se-treated mushroom (72.3 %) than the Se-untreated one (100 %, not analytically detected of inorganic Se). Se distribution upon a length in the Se-treated mushrooms was the highest in the bottom part (6.86 $\mu$/ g of dry) near to MC, and top and middle parts were significantly lower (3.71 and 3.01 $\mu$/ g of dry, respectively; P < 0.001) than the bottom. In the SMC from Se-treated mushrooms, the significant amount of Se (5.04l1g/g of dry) was remained, but that from the Se-untreated mushrooms was significantly low (P$\mu$ / g of dry. Se-treated SMC showed a high ratio of organic Se (65.67 %), suggesting that the significant amount of inorganic Se in the SMC was converted to organic Se by mushroom mycelia. Prior to mycelia inoculation in the mushroom culture, the sterilization of MC brought approximately 18% of Se loss in the MC. Apparent and net accumulation rates (%) for Se into mushrooms were 14.81 and 10.14 %, respectively, resulting from the Se volatilization into the air via metabolic process of mushroom itself. The result of this study shows that inorganic Se addition to MC for mushroom improved the organic Se contents in the mushroom and SMC. This study showed the possibility that Se in Se-enriched mushroom and SMC could be utilized as Se sources of food for human as well as feed for livestock.

This study was conducted to investigate the effects of feeding seleniferous whole crop barley silage (WCBS) on the growth performance, blood and carcass characteristics, and tissue selenium deposition infinishing Hanwoo steers. A total of 20 growing Hanwoo steers were selected and assigned to one of the following feed groups: control (0.1 ppm Se), T1 (0.3 ppm Se), T2 (0.9 ppm Se), and T3 (0.9 ppm of inorganic Se). Five steers were allocated to each treatment group, and the trial lasted for 90 days. All experimental diets combined seleniferous and/or non-seleniferous WCBS up to a ratio of 30%. For the T3 diet, 0.9 ppm Se in the form of sodium selenite was added to the control diet. Dietary level and type of selenium did not affect feed intake and body weight gain. Blood total lipid and triglyceride concentrations were significantly (p<0.05) lower in the T2 group than in control. Blood immunoglobulin G concentration increased significantly (p<0.05) with increasing levels of dietary selenium; concentrations were significantly (p<0.05) higher in T2 and T3 than in control. Blood selenium concentration was the highest (p<0.05) in T2. No significant difference was observed in dressing rate, ribeye area, marbling score, meat color and fat color. Further, no association was found between levels and chemical form of dietary selenium and quality and quantity of meat. To the contrary, tissue selenium content in kidney, liver, and hind leg increased significantly (p<0.05) with increasing levels of selenium; however, feeding inorganic selenium did not introduce a significant increase in tissue selenium content of finishing Hanwoo steers. The results indicated that the selenium present in seleniferous WCBS was an effectively absorbable selenium source, suitable for increasing tissue selenium content in beef. Approximately 100 g of hind leg muscle from steers fed on the T2 diet met approximately 69% of the recommended daily selenium requirements.
