Seed Characteristics and Germination Properties according to Change of Cone Production in Pinus densiflora Stands (소나무 우량 임분의 구과 생산량에 따른 종자 및 발아 특성)

  • This study was carried out to obtain the several data for improving the seed supply in Pinus densiflora. Seed characteristics and germination properties according to change of cone production from 2003 to 2006 were analyzed in three plus stands of P. densiflora. P. densiflora showed good production at interval of two years in the periodicity of seed production. Seed potential and percent of filled seed (PFS) among the major seed characteristics showed positive relations to the change of cone production, whereas percent of developed seed (PDS) and seed efficiency showed opposite trend. As a result of regression analysis between seed characteristics and cone production, seed potential ($R^2=0.682$, p<0.01) and PFS ($R^2=0.161$, non-significant) presented positive correlation with cone production while PDS ($R^2=0.418$, p$R^2=0.308$, non-significant) presented negative correlation. In germination properties percent of germination (PG) showed similar tendency to cone production, while mean germination time (MGT) showed opposite tendency. As a result of regression analysis between germination characteristics and cone production, PG showed positive correlation, whereas MGT showed negative correlation. Therefore, we could expect high germination percentage and short period for germination when we harvested cones at good cone production time.

The Effect of Isoflavone and/or Grape Seed Oil Supplementation on Blood Lipid Profiles and Bone Strength in Ovariectomized Female Rats (이소플라본과 포도종실유의 보충이 난소절제 흰쥐의 혈중지질 패턴 및 골격강도에 미치는 영향)

  • The study was performed to investigate the effects of isoflavone and/or grape seed oil supplementation on serum and liver lipid profiles and bone strength in ovariectomized female rats. Female Spraque-Dawley rats were assigned into 4 groups of ovariectomized (O), isoflavone (0.085 mg/100 g b.w/day) in ovariectomized rats, grape seed oil in ovariectomized rats and isoflavone and/or grape seed oil in ovariectomized rats. After 8 weeks, biochemical profiles of serum, liver and bone were analyzed. Total food intakes, body weight gains and FER (food efficiency ratio) were not statistically significantly different among groups. Total cholesterol, triglyceride and LDL-cholesterol levels in serum were decreased by fed of isoflavone and/or grape seed oils. However, crude lipid and total cholesterol contents in liver were not affect of isoflavone and/or grape seed oil. The hepatic glutathione contents were increased by isoflavone and/or grape seed oil fed. The hepatic glutathione-S-transferase activity in isoflavone and/or grape seed oil supplemented groups were higher than that O group. Bone (scapular and femur bone) dry weight, femur of max weight and bending strength were no significant difference among groups. Our finding suggest that isoflavone/grape seed oils might have potential role for serum lipid profiles improvement and bone strength in vivo.

Cone and Seed Characteristics among Different Cone Abundance Classes in a Seed Orchard of Pinus koraiensis (잣나무 채종원에서 구과 착과수에 따른 구과 및 종자특성)

  • This study was conducted to examine the relationship between cone abundance and cone/seed characteristics to improve seed quality and seed orchard management of Pinus koraiensis. The data was collected from P. koraiensis seed orchards at Chungju in 2007. That year was a rich year of cone harvest and the average number of cone per tree was 2.5 times much than that of normal year. We collected the cones from all trees in the sample plot of the seed orchard and investigated morphological characters such as cone length, cone width and cone weight. According to cone abundance per tree, the samples were divided into eight classes and analysed by cone analysis method. There is a decreasing tendency of number of fertile scale, seed potential, percent of filled seeds, seed weight per cone and seed efficiency, when the cone number per tree is increased. Additionally, the implications of the results to seed orchards management of P. koraiensis were discussed.

Development of a Plum (Japanese Apricot) Seed Remover for Multipurpose Plum Flesh Processing

  • Purpose: Japanese Apricot, a type of plum, has various medicinal and economical applications. Plums are quite popular worldwide, but their deseeding remains a serious impediment to their processing. Therefore, a plum (Japanese Apricot) seed remover (PSR) was developed that can use various types of cutters according to the purpose of the plum processing, and its performance was evaluated. Methods: The proposed PSR, which allows multipurpose cutters, namely, zero-, two-, and four-blade cutters, to be installed, was first designed and manufactured. To identify appropriate parameters related to the cutting pressure, plums were harvested from three regions during three harvesting periods, and their geometrical and mechanical properties were measured. After application of the parameters related to the cutting pressure, a performance test was carried out on both fresh and frozen plums by identifying the ratios of the flesh recovery, seed recovery, seed breakage, deseeding efficiency, and machine efficiency. Results: The results show that, using the proper calculation of the processing parameters, 100% deseeding efficiency was facilitated regardless of the type of cutter used. However, in the case of a four-blade cutter, there are significant differences in the flesh recovery ratio according to the plum setting angle. Between the fresh and frozen plums, all cutters showed a significantly better flesh recovery ratio for the case of fresh plums. Conclusions: This machine will advance the plum processing technology, and eventually help the plum industry flourish.

Treatment Time and Concentration of GA3 for Increasing Efficiency of the Lettuce Seed Production for Lettuce Breeding and Selection (상추 육성 및 선발을 위한 채종 효율증진을 위한 GA3 처리시기 및 농도)

  • To increasing efficiency of the seed production through GA3 treatment, treatment of GA3 20mg/ℓ at its fourteen leaf stage had an effect on 21% increasing seed yield at leaf lettuce (cv.'Ttugseomjeokchugmyeon') in seed production from 50.9ℓ/10a to 61.7ℓ/10a, and 'GA3 50mg/ℓ at its eight leaf and fourteen leaf stage had a effect on 14% increasing the seed yield of head lettuce (cv.'Urake') at seed production from 14.2ℓ/10a to 21.1ℓ/10a reducing rate of dead plant in field. Especially, it will be recommended for planting after June for seed production of head lettuce owing to decreasing rate of dead by soft rot and stem rot.

Optimum Operating Conditions of a Vacuum Nozzle Seeder (진공노즐식 파종기의 최적 작동조건)

  • For maximum seeding efficiency of a nozzle type seeder, the performance of the nozzle should be considered sufficiently. This study was carried out to investigate the optimum operating conditions of a seeder attached the vacuum nozzle which was modified syringe needle acting on the plug seedling tray and the seed plate. Such operating factors as the hole diameter of the nozzle (d), the distance from the nozzle tip to the bottom plate of seed hopper(D) the absorbing air pressure of the nozzle tip(P) the bounding height of seed from the vibrated bottom plate of seed hopper and the seeding speed were selected based on the weight of a grain of seed(W). The treated materials were pepper seed as the flat type, cucumber seed as the oval type and radish seed as the spherical type. The optimum operating conditions of the experimental seeder were revealed as follows: 1. The height of the seed bounding from the bottom plate of seed hopper and the distance from nozzle tip to bounded seed were 5 mm and 0.5 mm at all seeds. The hole diameter of the nozzle and the absorbing pressure for pepper seed, cucumber seed and radish seed was 0.45 mm, 0.65 mm. 0.65mm and 39.2 kPa, 88.3 kPa, 58.8 kPa, respectively. 2. The absorbing pressure P was represented as P=η.4W/$\pi$d$^2$ where η was 100. The seeding speed using a 128 cell tray was 2.4 cm/s which was same transfer as 2.5 trays per minute. 3. The maximum seeding rate in case of the pepper seed was 97% the cucumber seed was 95% and the radish seed was 100% under the optimum operating conditions of the seeder.

Efficiency for increasing seed oil content using WRINKLED1 and DGAT1 under the control of two seed-specific promoters, FAE1 and Napin

  • Seed storage oils are essential resources for not only human and animal diets but also industrial applications. The primary goal of this study was to increase seed oil content through comparative analysis of two seed-specific promoters, AtFAE1 from Arabidopsis Fatty Acid Elongase 1 gene and BnNapin from Brassica napus seed storage protein gene. AtWRI1 and AtDGAT1 genes encoding an AP2-type transcription factor and a Diacylglycerol Acyltransferase 1 enzyme, respectively, were expressed under the control of AtFAE1 and BnNapin promoters in Arabidopsis. The total seed oil content in all transgenic plants was increased by 8-11% compared with wild-type seeds. The increased level of oil content in AtWRI1 and AtDGAT1 transgenic lines under the control of both promoters was similar, although the activity of the BnNapin promoter is much stronger than that of AtFAE1 promoter in the mature stage of developing seeds where storage oil biosynthesis occurs at a maximum rate. This result demonstrates that the AtFAE1 promoter as well as the BnNapin promoter can be used to increase the seed oil content in transgenic plants.

Cone Characteristics and Seed Quality among Harvest Times in the Clonal Seed Orchard of Larix kaempferi (낙엽송 클론 채종원에서 구과 채취시기에 따른 구과특성 및 종자품질)

  • Harvest time is one of the most important determining factors of seed quality, especially for species that produce seeds over irregular and long-term periods, such as Larix kaempferi. A cone collection plan must be established to increase seed production efficiency and stable mass production. We investigated seed qualities such as seed efficiency, germination rate, and T50 (germination speed), with 7 or 8 cone collection times at a clonal seed orchard of L. kaempferi in Chungju between 2021 and 2022. A multivariate analysis was then performed for the collected data. In early August, decreases in the moisture contents and browning of cones were observed. These were followed by a decrease in germination rate, with a peak at the end of September, but no clear trend was observed. The later the cones were harvested, the better the seed vigor (T50). However, the seed yield and efficiency decreased owing to increases in seed scattering and the number of insect-damaged seeds. As a result, the optimal time of seed harvest for the seed orchard was in early August. To produce uniform seedlings, insect damage must be reduced through timely control and harvest cones in early September. This shows that the degree of browning and moisture content of cones can be used as indicators of the timing of cone collection in L. kaempferi seed orchards.

Improving Efficiency and Effectiveness of Disinfection by Soaking Seeds before Rice Seed Disinfection (벼 종자소독 전 침종에 따른 소독효과 및 효율성 향상)

  • Seed disinfection is a necessary process to decrease Bakanae disease in rice and is carried out at 30℃ to increase disinfection efficiency. However, the length of sprout after seed disinfection is not suitable for sowing, so additional processes such as seed soaking is needed. For this reason, this study was conducted to develop the efficient and effective seed disinfection method. When the seed disinfection finished after soaking seeds in cold water for 2 days, length of sprout was about 1.0 mm and sowing can be done without additional processes. In addition, the incidence of Bakanae disease was greatly reduced by 64% compared to conventional method. In order to find out why the newly developed method (disinfection after soaking) is more efficient than conventional method, soaked and not soaked seeds were stained using methylene blue staining. It was found that soaked seeds were more easily stained with about 50% of internal and external endosperm stained. Therefore, pesticide can be absorbed easily into internal endosperm of soaked seeds and efficiently destroy the spore of pathogen. As a results of this study, newly developed method will be useful as it can improve efficiency of agricultural work and also effectively decrease Bakanae disease compared to conventional method.

Extraction of Resveratrol Containing Grade Seed Oil with Supercritical Carbon Dioxide (초임계 이산와탄소를 이용한 Resveratrol 함유 포도씨유 추출)

  • Grape seed oil made by press or organic solvent extraction does not contain resveratrol, a bioactive compound. Supercritical carbon dioxide could extract oil containing resveratrol from grape seed. The extraction efficiency was mainly dependent on the water content in grape seed. More resveratrol was contained in the oil extracted with un-dried grape seed. No resveratrol was extracted with dried grape seed. Time course changes of grape seed oil extraction also resulted that resveratrol could be extracted by supercritical carbon dioxide with the positive influence of water.
