  • The importance of wild plant resources along with the development of high biotecyhnology is highlighted for exploitation of new materials which can make the added value. The goal of this study is providing fundamental data bases for developing new materials through the analyses of morphological characteristics, antibiotics and molecular cytogenetics in Baegseon (Dictamnus dasycarpus Turcz.). Baegseon has several characteristics; there are two types of flower color, pink and white, the seed germination starts about February 20th, the maximum flowering season is around May 17th in southern Korea, the growth period is about 60 days. The number of chromosomes are 2n=2x=36, the size of chromosomes in metaphase is $4.2{\sim}8.1{\mu}m$. The amount of 2C nuclear DNA is 1.93pg, and there is no variation of genome size amoung varieties. The extraction juice of baegseon young roots has the excellent antibiotic activity against the mold (Botrytis cinerea).

FISH Karyotype and GISH Meiotic Pairing Analyses of a Stable Intergeneric Hybrid xBrassicoraphanus Line BB#5

  • xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed. Lilium longiflorum 'Charm' as a F1 Hybrid for Pot Plant (종자번식 일대잡종 분화용 나팔나리(Lilium longiflorum) 'Charm' 육성)

  • Lilium longiflorum 'Charm' as a $F_1$ hybrid cultivar was released by crossing inbred line '$L_2$-14' and '$L_2$-21' which were obtained from 5 self crosses originated from 'Nellie White', 'Ace' and 'Hinomoto'. The growth and flowering characteristics were evaluated in a greenhouse maintained at a minimum of $13^{\circ}C$ at night during winter in 2006 and 2007. The flower of 'Charm' is white color and horizontal-facing. The flower number of a plant and its diameter is 7.4 and 16.5 cm with 24.5 ornamental(flowering) days. The plant height is 60.3 cm with 70.3 number of leaves. The stem diameter and internode length is 1.2 cm and 1.1 cm, respectively, meaning the plant is compact and sturdy. And the number of seed per a capsule is 251.1. The results of these evaluation, therefore, suggest that seedling Lilium longiflorum 'Charm' can be used as a pot plant due to its short stems, many number of flowers per plant, long ornamental period, strong growth habit with many leaves and thick stem diameter.

Development of a Phalaenopsis (P. Blume) Cultivar, 'Yellow Cream' with Striped Yellow Flower (황화 스트라이프 대륜계 호접란 신품종 '옐로우 크림' 육성)

  • A cultivar 'Yellow Cream' of Phalaenopsis(P. Blume) was developed by cross breeding and line selection at the Flower Research Institute, Gyeongnam ARES, from 2000 to 2008. Characteristic trials for this cultivar were carried out three times from 2006 to 2008. 'Yellow Cream' was developed from a crossing between P. 'Sogo Firework' and P. 'Sogo Gold'. 'Yellow Cream' exhibits light yellow flower color with pink stripe and favorable flower shape. Individual flowers are formed with parallel arrangement and have long life with more than 55 days. 'Yellow Cream'(Grant No.3232) was registered to the Korea seed and variety Service(KSVS) for commercialization in 2010 and suitable for the cultivation under greenhouse conditions in Korea.

  • Wild ginseng production is increasing due to forest recovery for last 30 years. Total number of Symmani (traditional mountain ginseng digger) was 558 in 2001. Provincial distribution of Symmani in 2001 was highest in Kangwon $(32\%),$ next in Choongbook $(21\%)$ and least in Jeonnam $(0.7\%)$ and Kyoungnam $(0.9\%).$ Age distribution of Symmani was $33\%\;for\;fourties,\;32\%$ for fifties and $20\%$ for sixties. There were 8 persons in eighties. Symmanies are still keeping traditional ritual for mountain god serving clothes of colored ribbons and foods. Increased production induced open market system from underground dealing of mountain ginseng. Korea Mountain Ginseng Association established mountain ginseng assessment committee with professional Symmanies in 2001. From September to November in 2001, 987 roots were requested for quality assessment to the committee and 476 roots $(48\%)$ were passed and graded and others were rejected. Highest frequency of rejection was foreign origin. Pass rate was highest $(74\%)$ in Choongnam suggesting best place for quality. Number of collected roots in each province was positively correlated (p=0.05) with number of Symmanies. There are 3 quality groups of mountain ginseng, Heaven (pure natural), Earth (from seeding of wild ginseng) and Man (from seeding or seedling of wild ginseng with slight environmental modification). The relationship between price and age was polynomial in high quality root, Heaven, Earth and seed long head of Man group, and linear in low quality group, seedling long head of Man. The best one in 2001 was 26 g, 124 years old and sold with 109 million won. Quality criteria are age, shape, weight, color and healthy outlook. Fine roots are criteria for health status of roots and taproot is criteria for efficacy and called as medicine barrel. The implication is that ginsenosides have rarely been experienced for efficacy. The quality criteria of cultivated ginseng were originated from those of mountain ginseng. It is unique for mountain ginseng that only fresh one can be on market. Since quality criteria of mountain ginseng must be based on the efficacy experience it is well expected that present criteria might almost be established at the age of Shinnong Materia Medica.

Major Morphological Characteristics of Safflower(Carthamus tinctorius L.) Accessions (홍화 수집종의 주요 형태적 특성)

  • Morphological characteristics of 16 collected safflower (Carthamus tinctorius L.)accessions were studied at Gyongbuk A.T.A. Uisong Medicinal Plant Experiment Station in 1998. As for plant height, 5 lines were tall(more than 131cm), which were mainly foreign safflower. Middle and small were 5 and 6 accessions, respectively. Number of branch per plant was 11∼13 in 2 accessions of domestic safflowers while foreign safflowers have more than 50 branches per plant. Leaf shapes were oblong(3 lines), lanceolate(10 lines) and broadly linear(3 lines). There were three types of leaf margin shape which were repand(7 lines), serrate(4 lines) and parted serrate(5 lines). Flower color was white(1 line), yellow(13 lines) and red orange(2 lines). As for flower head shape, 14 lines were conical and 2 lines were flattened. There were thorns on bract of flower head in 11 lines but in 5 lines there were not. Domestic safflower lines had seeds of conical shape. Contrastively, seed shapes of foreign saffower lines were oval.

Antioxidant, Anti-inflammatory, Anti-obesity and Estrogen-like Activities of Soybean Pod Extracts (콩깍지 추출물의 항산화·항염·항비만 및 에스트로겐 유사활성 평가)

  • In this study, soybean pods of 45 soybean landraces (or varieties) were classified as yellow (19 samples), black (23 samples), or black in green (3 samples) based on soybean seed coat color. Total polyphenol and flavonoid contents were measured, and antioxidant, anti-inflammatory, anti-obesity, and estrogen-like activities were assessed. Total polyphenol and flavonoid content ranges were 24.13-108.03 mg GAE/g and 3.31-72.02 mg CE/g, respectively, and were highest in the black group followed by the yellow group and were least in the black in green group, while ABTS and DPPH activities followed the order black in green > black > yellow. Estrogen-like and estrogen receptor-α activity ranges were 29.06-35.58 pg/mL and 7.05-10.13 pg/mL and were followed the order yellow > black > black in green and black in green > yellow > black, respectively. Nitric oxide (NO) inhibitory and UCP-1 activities followed the same order as estrogen receptor-α activities. Our findings suggest that soybean pods are excellent sources of antioxidants and high-quality functional materials.

Marker Assisted Development and Characterization of Beta-Carotene Rice

  • Beta-carotene producing transformants were produced in the background of 'Nagdongbyeo', a Japonica rice cultivar. Introgression of the carotenoid locus in the transformant, PAC4-2 into the elite cultivar 'Ilpumbyeo' was started. To initiate a backcrossing program, we surveyed 220 SSR markers and found that 38% of them were polymorphic between 'Ilpumbyeo' as a recurrent parent and the PAC4-2 as a recipient parent. The selection strategy comprising foreground and background selection was employed. First, foreground selection was practiced in $BC_1$, $BC_2$, and $BC_3$ generations using the transgene specific PCR-based marker in addition to visual scoring of the seed color. Marker-based background selection combined with phenotypic selection was employed from $BC_3F_2$ to $BC_3F_4$ generations. Blast search indicated that the transgene PAC4-2 was located between SSR markers, RM6 and RM482. 240 $BC_3F_3$ and 63 $BC_3F_4$ lines were evaluated for four agronomic traits including days to heading. Most of the lines were similar to Ilpumbyeo in agronomic traits evaluated. The percentage of PAC4-2 genome ranged from 4% to 21% with a mean of 12.5%, which was higher than the expected for an unselected $BC_3$ backcross population. This could be explained by the fact that two genes for beta-carotene and the stripe virus resistance were targeted in this study. We selected 10 representative $BC_3F_5$ lines from 63 $BC_3F_4$ lines based on agronomic traits and carotenoids content. The selection strategy would be appropriate for the introgression of beta-carotene gene in a breeding program.

Fruit Yield and Quality Evaluation of Sweet Pepper (Capsicum annuum L.) F1 Hybrids Derived from Inbred Lines

  • Sweet pepper inbred lines (KNU1003, KNU1006, KNU1007, KNU1009, KNU1015, KNU1017 and KNU2006) developed at Kangwon National University (KNU) through conventional means, inbred lines (5AVS1, 5AVS2, 5AVS3, 5AVS5, 5AVS7 and 5AVS8) collected at Rural Development Administration (RDA) and inbred lines (SP12, SP27 and SP14) derived from anther culture were used as female parents and anther culture derived homozygous lines (SP9, SP10, SP14, SP24, SP25, SP27, SP30, SP32, SP34, SP38, SP43, SP45 and SP51) were used as male parents to produce $F_1$ hybrids. A total of 37 $F_1$ hybrids were evaluated for fruit yield and quality characters in summer season, 2007. Variation in fruit number, fruit weight, fruit yield per plant and fruit volume was observed among the $F_1$ hybrids. Superiority on yield over standard/commercial varieties were differed among $F_1$ hybrids. Hybrid $5AVS8{\times}SP45$ exhibited highest heterosis over Special (16.5%) and Fiesta (24.7%). Fruit quality characters (fruit length, fruit width, pericarp thickness, total soluble solid, fruit shape and fruit color) were varied among the $F_1$ hybrids. Fruit number, fruit weight and fruit volume per plant were correlated with fruit yield. Based on the standard heterosis expressed by the hybrids and quality characters evaluation, $KNU1017{\times}SP27$, $5AVS1{\times}SP43$, $5AVS5{\times}SP27$, $5AVS8{\times}SP45$, $SP12{\times}SP38$ and $SP27{\times}SP25$ hybrids were found to be superior over commercial cultivars and are selected. Inbred lines of these hybrid combinations can be used to produce $F_1$ hybrid seed for commercial production.

A Study on the Development and Consumer Preference of the Soup·Stew HMR New Products (탕·찌개류 HMR 신제품 개발을 위한 소비자 기호도 연구)

  • This paper is a study on the development and consumer preference of the soup stew HMR new products, and the main contents are as follows. A total of nine products were studied, including three developed products and six market products. For this study, the demographic characteristics, the awareness and attitude of HMR products were investigated. In addition, the consumer panel evaluated overall liking, appearance, color, flavor, and taste using the nine-point recertification scale, while the strength assessed viscosity, sweetness, saltiness, sourness, and Umami. Familiarity, health degree, Purchase intention and Recommendation intention were investigated and the reasons for preference and non preference were analyzed by multi-response method. The consumer preference analysis indicates that the product is competitive among existing products and that it would be desirable to improve after identifying the causes of non-preferred factors.
