Observation for drying non-uniformity of allium vegetables using NIR spectroscopy (근적외 분광법을 이용한 양념 야채의 건조 불균일성 관찰)

  • In this study, near-infrared spectroscopy was used to observe the drying non-uniformity of vegetables such as spring onions, onions, and garlic, which are commonly used for seasoning. For the warm-air convection drying method, the vegetables showed drying non-uniformity, which is due to the unevenness of the wind temperature and humidity depending on the height and position of the drying tray. The second derivative spectra between the vegetable samples with different drying degrees were compared. The peak at around 1,390~1,400 nm, which is assigned to weak hydrogen bonds of water, was changed during drying whereas the peak near 1,420 nm, which represents strong hydrogen (H-) bonds of water, was not changed, indicating that water with weak H-bonds evaporates first during drying, and that water with strong H-bonds remains after drying. The hyperspectral NIR imaging technique combined with principal-component analysis made it possible to discriminate the dried vegetables according to their drying degree.

Evaluation of Firmness and Sweetness Index of Tomatoes using Hyperspectral Imaging

  • The objective of this study was to evaluate firmness, and sweetness index (SI) of tomatoes (Lycopersicum esculentum) by using hyperspectral imaging (HSI) in the range of 1000-1400 nm. The mean spectra of the 95 matured tomato samples were extracted from the hyperspectral images, and the reference firmness and sweetness index of the same sample were measured and calibrated with their corresponding spectral data by partial least squares (PLS) regression with different preprocessing method. The results showed that the regression model developed by PLS regression based on Savitzky-Golay (S-G) second-derivative preprocessed spectra resulted in better performance for firmness, and SI of tomatoes compared to models developed by other preprocessing methods, with correlation coefficients (rpred) of 0.82, and 0.74 with standard error of prediction (SEP) of 0.86 N, and 0.63 respectively. Then, the feature wavelengths were identified using model-based variable selection method, i.e., variable important in projection (VIP), resulting from the PLS regression analyses and finally chemical images were derived by applying the respective regression coefficient on the spectral image in a pixel-wise manner. The resulting chemical images provided detailed information on firmness, and sweetness index (SI) of tomatoes. Therefore, these research demonstrated that HIS technique has a potential for rapid and non-destructive evaluation of the firmness and sweetness index of tomatoes.

Development of Diagnostic Strip for Determining Paraquat in Urine (뇨중 파라쿼트(paraquat)측정용 진단 시험지의 개발)

  • Paraquat is an effective and widely used herbicide, but it is also very toxic to humans. It is well-known that urine paraquat concentration is one of the most important prognostic indicator for paraquat-poisoning. Quantitative analysis of paraquat, however, are not generally used in clinical laboratories. In this work, a new test strip to detect paraquat concentration using sodium dithionite in urine was developed. Using these second-derivative method, the test strip prepared in $0.5{\%}$ borate buffer (pH 8.0), 0.25 M $Na_2S_2O_4,\0.1~0.8{\%}$ PVP, and $1{\%}$ decanol showed not only better color reaction but also an excellent application possibility to be used in automatic analyzer.

Vascular Responses to Acupuncture at the Kidney Jeonggyeok in Diabetic and Normal Groups Assessed by the Second Derivative of the Finger Photoplethysmogram Waveform (가속도맥파로 평가한 당뇨군과 정상군의 신정격 자침으로 인한 혈관변화)

  • 목적 : 신정격 자침이 당뇨군과 정상군의 혈관변화에 미치는 영향을 알아보기 위해 가속도맥파를 이용한 실험을 설계하였다. 방법 : 20명의 당뇨군과 20명의 정상군에서 신정격 자침 이전과 이후, 2회에 걸쳐서 Cardio Peri SA6000을 이용하여 가속도맥파를 측정하였다. 가속도맥파의 b/a, c/a, d/a 및 연령지수를 통해 혈관상태를 평가하였다. 결과 : 1. 당뇨군과 정상군의 신정격 자침 이전 가속도맥파 측정에서 당뇨군이 정상군보다 b/a값(p=0.000), 연령지수(AI)(p=0.000)는 유의하게 높았으며, c/a값(p=0.000), d/a값(p=0.000)은 유의하게 낮았다. 2. 당뇨군에서 신정격 자침 이후 가속도맥파의 b/a값(p=0.004), 연령지수(p=0.012)는 유의하게 낮아졌으며, c/a값(p=0.026)은 유의하게 높아졌다. 3. 정상군에서 신정격 자침 이후 가속도맥파의 b/a값(p=0.025)은 유의하게 낮아졌다. 결론 : 신정격 자침이 당뇨로 인해 유발된 혈관의 탄력도 감소 및 노화를 다소 회복시킬 수 있는 것으로 사료된다.

Retrospective Evaluation of Heptaplatin Toxicities in Patients with Advanced Gastric Cancer (말기 암환자에 투여한 Heptaplatin의 신독성에 대한 후향적 평가)

  • Heptaplatin, a new platinum derivative, has several contradicting reports on the nephrotoxicity. Therefore, the aim of this study is to compare the toxicities of heptaplatin-containing regimens in the chemotherapy. This study was performed retrospectively on seventy-seven patients with advanced gastric cancer who did not receive chemotherapy within the last 1 months before taking of heptaplatin- or cisplatin-containing chemotherapy. The 38 patients among total patients was received heptaplatin-containing regimens (26 with SEF regimens: heptaplatin/epirubicin/5-FU, 12 with SF regimens: heptaplatin/5-FU) and the rest 39 patients was received cisplatin-containg regimens (11 with CEF regimens: cisplatin/epirubicin/5-FU, 28 with ELF regimens: epirubicin/leucovorin/5-FU). Before and after the chemotherapy serum creatinine (Scr) and proteinuria were measured by urine stick test in all patient groups. Also Scr was measured a day before the second cycle and did not vary significantly between groups. However Scr on cycle 3 were significantly higher in SEF and SF groups. In case of proteinuria, it was more frequent on cycle 1 in heptaplatin/5-FU group. Proteinuria before and after on cycle 2 was not different between the two cisplatin -containing groups, but was more frequent in heptaplatin-containing groups. The reason why the Scr measured was not so different could be because we excluded the patients who received only one cycle of heptaplatin and changed the regimen due to signs of nephrotoxcity. As the results nephrotoxicity such as protienuria was appeared to be more frequent with heptaplatin-treated patients. It suggests that the clinical consequences of the toxicity need to further evaluation and also the modalities to prevent or minimize nephrotoxicity of heptaplatin should be studied for future utilization of the drug.

  • This paper introduces a heave compensation system for offshore crane when it subjected to unexpected disturbances such as ocean waves, tidal currents or winds and their external force. The dynamic model consists of a crane which is considered to behave in the same manner as a rigid body, a hydraulic driven winch, an elastic rope and a payload. To keep the payload from moving upwards and downwards, PD(Proportional-Derivative) control was applied by using linearization. In order to achieve a better performance, the sliding mode control and the nonlinear generalized predictive control algorithm was applied according to the time-delay. As a result, the oscillating amplitude of the payload was reduced by the control algorithm. Considering the time-delay involved in the system to be one second, nonlinear generalized predictive controller with a robust controller was a suitable control algorithm for this heave compensation system because it made the position of te payload reach the desired position with the minimum error. This paper presented a control algorithm using the robust control and its simulation results.

Measurement of Lipid Content of Compost in the fermentation Process using Near-Infrared Spectroscopy

  • Near infrared spectroscopy (NIRS) was applied to determination of the lipid content of compost during compost fermentation of tofu(soybean-curd) refuse. The reflected rays in the wavelength range between 800 and 2500 nm were measured at 2 nm intervals. The absorption of lipid observed at 4 wavelengths, 1208, 1712, 2312 and 2352 nm on the second derivative spectra. To formulate a calibration equation, a multiple linear regression analysis was carried out between the near-infrared spectral data and on the lipid content in the calibration sample set (sample number, n=60) obtained using a Soxhlet extraction method. The calibration equation for prediction of lipid, the value of the multiple correlation coefficient (R) was 0.975 when using the wavelengths of 1208 and 1712nm. To validate the calibration equation obtained, the lipid content in the validation sample set (n=35) not used for formulating the calibration equation were calculated using the calibration equations, and compared with the values obtained using the Soxhlet extraction method. Good agreement were observed between the results of the Soxhlet extraction method and those values of the NIRS method. The simple correlation coefficient (r) and standard error of prediction (SEP) were 0.964 and 0.815 %, respectively. Then, the NIRS method was applied to a compost fermentation in which the time course the lipid content were measured and good results were obtained. The study indicates that NIRS is a useful method for process management of the compost fermentation of tofu refuse.

  • Yeast, Kurtzurnanomyces sp. I-11, produces biosurfactant, mannosyl erythritol lipid (MEL), from soya oil. The properties of biosurfactant MEL include low-toxicity and high biodegradability. MEL provides new possibilities for a wide range of industrial applications, especially food, cosmetic, pharmaceutical fields and chemicals for biotechnology. In the fermentation process, techniques of measuring and controlling substrates and products are important to obtain high productivity with optimum concentrations of substrate and product in the culture broth. The measurement system for the concentrations of soya oil and MEL in the fermentation process was developed using near-infrared spectroscopy (NIRS). Soya oil and MEL in the culture broth were extracted with ethyl acetate and NIR spectra was carried out between the second derivative NIR spectral data at 1312 and 2040 nm and MEL concentrations obtained using a thin-layer chromatography with a flame-ionization detector (TLC/FID) method. A calibration equation for soya oil was results of the validation of the calibration equation, good agreement was observed between the results of the TLD/FID method and those of the NIRS method for both constituents. NIR method was applied to the measurement of the concentrations of MEL and soya oil in the practical fermentation and good results were obtained. The study indicates that NIRS is a useful method for measurement of the substrate and product in the glycolipid fermentation.

Inactivation of Brain Succinic Semialdehyde Reductase by o-Phthalaldehyde

  • Succinic semialdehyde reductase was inactivated by o-phthalaldehyde. The inactivation followed pseudo-first order kinetics, and the second-order rate constant for the inactivation process was 28 $M^{-1}s^{-1}$ at pH 7.4 and $25^{\circ}C$. The absorption spectrum ($\lambda_{max}$ 337 nm) and fluorescence excitation ($\lambda_{max}$ 340 nm) and fluorescence emission spectra ($\lambda_{max}$ 409 nm) were consistent with the formation of an isoindole derivative in the catalytic site between a cysteine and a lysine residue approximately about 3 $\AA$ apart. The substrate, succinic semialdehyde, did not protect enzymatic activity against inactivation, whereas the coenzyme NADPH protected against o-phthaladehyde induced inactivation of the enzyme. About 1 isoindole group per mol of the enzyme was formed following complete loss of enzymatic activity. These results suggest that the amino acid residues of the enzyme participating in a reaction with o-phthalaldehyde are cysteinyl and lysyl residues at or near the NADPH binding site.

Vasorelaxation Study and Tri-Step Infrared Spectroscopy Analysis of Malaysian Local Herbs

  • Objectives: The aim of this paper is to investigate the activities of Malaysian local herbs (Clinacanthus nutans Lindau, Strobilanthes crispus, Murdannia bracteata, Elephantopus scaber Linn., Pereskia bleo, Pereskia grandifolia Haw., Vernonia amygdalina, and Swietenia macrophylla King) for anti-hypertensive and vasorelaxant activity. An infrared (IR) macro-fingerprinting technique consisting of conventional fourier transform IR (FTIR), second-derivative IR (SD-IR), and two-dimensional correlation IR (2D-correlation IR) analyses were used to determine the main constituents and the fingerprints of the Malaysian local herbs. Methods: The herbs were collected, ground into powder form, and then macerated by using three different solvents: distilled water, 50% ethanol, and 95% ethanol, respectively. The potentials of the extracts produced from these herbs for use as vasorelaxants were determined. Additionally, the fingerprints of these herbs were analyzed by using FTIR spectra, SD-IR spectra, and 2D-correlation IR spectra in order to identify their main constituents and to provide useful information for future pharmacodynamics studies. Results: Swietenia macrophylla King has the highest potential in terms of vasorelaxant activity, followed by Vernonia amygdalina, Pereskia bleo, Strobilanthes crispus, Elephantopus scaber Linn., Pereskia grandifolia Haw., Clinacanthus nutans Lindau, and Murdannia bracteata. The tri-step IR macro-fingerprint of the herbs revealed that most of them contained proteins. Pereskia bleo and Pereskia grandifolia Haw. were found to contain calcium oxalate while Swietenia macrophylla King was found to contain large amounts of flavonoids. Conclusion: The flavonoid content of the herbs affects their vasorelaxant activity, and the tri-step IR macro-fingerprint method can be used as an analytical tool to determine the activity of a herbal medicine in terms of its vasorelaxant effect.
