  • Cytochrome P450 14${\alpha}$-sterol demethylase enzyme (CYP51) is the target a of azole type antifungals. The azole blocks the ergosterol synthesis and thereby inhibits fungal growth. A three-dimensional (3D) homology model of CYP51 from Candida albicans was constructed based on the X-ray crystal structure of CYP51 from Mycobacterium tuberculosis. Using this model, the binding modes for the substrate (24-methylene-24, 25-dihydrolanosterol) and the known inhibitors (fluconazole, voriconazole, oxiconazole, miconazole) were predicted from docking. Virtual screening was performed employing Structure Based Focusing (SBF). In this procedure, the pharmacophore models for database search were generated from the protein-ligands interactions each other. The initial structure-based virtual screening selected 15 compounds from a commercial available 3D database of approximately 50,000 molecule library, Being evaluated by a cell-based assay, 5 compounds were further identified as the potent inhibitors of Candida albicans CYP51 (CACYP51) with low minimal inhibitory concentration (MIC) range. BMD-09-01${\sim}$BMD-09-04 MIC range was 0.5 ${\mu}$g/ml and BMD-09-05 was 1 ${\mu}$g/ml. These new inhibitors provide a basis for some non-azole antifungal rational design of new, and more efficacious antifungal agents.

  • Actin binding proteins play key roles in cell structure and movement particularly as regulators of the assembly, stability and localization of actin filaments in the cytoplasm. In the present study, a cDNA clone encoding an actin bundling protein named as AhABP was isolated from Acanthamoeba healyi, a causative agent of granulomatous amebic encephalitis. This clone exhibited high similarity with genes of Physarum polycephalum and Dictyostelium discoideum, which encode actin bundling proteins. Domain search analysis revealed the presence of essential conserved regions, i.e., an active actin binding site and 2 putative calcium binding EF-hands. Transfected amoeba cells demonstrated that AhABP is primarily localized in phagocytic cups, peripheral edges, pseudopods, and in cortical cytoplasm where actins are most abundant. Moreover, AhABP after the deletion of essential regions formed ellipsoidal inclusions within transfected cells. High-speed co-sedimentation assays revealed that AhABP directly interacted with actin in the presence of up to $10{\mu}M$ of calcium. Under the electron microscope, thick parallel bundles were formed by full length AhABP, in contrast to the thin actin bundles formed by constructs with deletion sites. In the light of these results, we conclude that AhABP is a novel actin bundling protein that is importantly associated with actin filaments in the cytoplasm.

  • We build the web document recommending agent system which offers a certain amount of web documents to each user by monitoring and learning the user's action of web browsing. We also propose a method of query expansion using the Korean thesaurus. The queries to search for new web documents generate a candidate set using the Korean thesaurus. We extract the words which are mostly correlated with the queries, among the words in the candidate set, by using TF-IDF and mutual information. Then, we expand the query. If we adopt the system of query expansion, we can recommend a lot of web documents which have potential interests to users. We thus conclude that the system of query expansion is more effective than a base system of recommending web-documents to users.

  • As an effort to search new bacterial biocontrol agents against pepper anthracnose, we screened myxobacteria, which might inhibit the growth of Colletotrichum acutatum, the agent of that plant disease. When 93 myxobacterial strains including 59 Myxococcus spp. and 34 Corallococcus spp. were tested against C. acutatum ACYSJ001 on agar plates, 10 strains identified as the genus Myxococcus significantly obstructed the growth of C. acutatum, whereas the majority of strains belonging to the genus Corallococcus did not demonstrate any counteractive effect. Such results have indicated that the strains of the genus Myxococcus have a high potential to play roles of biocontrol agents for control of pepper anthracnose. These also have revealed that the strains of the genus Myxococcus could be used as excellent microbial resources for screening novel antifungal substances.

  • Recently semantic web document is produced and added in repository in a cloud computing environment and requires an intelligent semantic agent for analytical classification of documents and information retrieval. The traditional methods of information retrieval uses keyword for query and delivers a document list returned by the search. Users carry a heavy workload for examination of contents because a former method of the information retrieval don't provide a lot of semantic similarity information. To solve these problems, we suggest a key word frequency and concept matching based semantic clustering model using hadoop and NoSQL to improve classification accuracy of the similarity. Implementation of our suggested technique in a cloud computing environment offers the ability to classify and discover similar document with improved accuracy of the classification. This suggested model is expected to be use in the semantic web retrieval system construction that can make it more flexible in retrieving proper document.

  • Solar variability is widely known to affect the interplanetary space and in turn the Earth's electromagnetical environment on the basis of common periodicities in the solar and geomagnetic activity indices. The goal of this study is twofold. Firstly, we attempt to associate modes by comparing a temporal behavior of the power of geomagnetic activity parameters since it is barely sufficient searching for common peaks with a similar periodicity in order to causally correlate geomagnetic activity parameters. As a result of the wavelet transform analysis we are able to obtain information on the temporal behavior of the power in the velocity of the solar wind, the number density of protons in the solar wind, the AE index, the Dst index, the interplanetary magnetic field, B and its three components of the GSM coordinate system, $B_X$, $B_Y$, $B_Z$. Secondly, we also attempt to search for any signatures of influence on the space environment near the Earth by inner planets orbiting around the Sun. Our main findings are as follows: (1) Parameters we have investigated show periodicities of ~ 27 days, ~ 13.5 days, ~ 9 days. (2) The peaks in the power spectrum of $B_Z$ appear to be split due to an unknown agent. (3) For some modes powers are not present all the time and intervals showing high powers do not always coincide. (4) Noticeable peaks do not emerge at those frequencies corresponding to the synodic and/or sidereal periods of Mercury and Venus, which leads us to conclude that the Earth's space environment is not subject to the shadow of the inner planets as suggested earlier.

  • We evaluated the inhibitory effects of extracts and components of Geranium thunbergii on aldose reductase (AR) and galactitol formation in rat lenses with high levels of galactose as a part of our ongoing search of natural sources for therapeutic and preventive agents for diabetic complications. The inhibitory effects of water, methanol and ethanol extracts of G. thunbergii on rat lens AR (RLAR) were determined. Comparing inhibitory effects of various solvent extracts, ethanol extract showed RLAR inhibitory activity ($IC_{50}$ values, 5.24 and $6.39{\mu}g/m{\ell}$, respectively). The ethanol extract was fractionated to chloroform, ethyl acetate and water. Of these, the ethyl acetate fraction from ethanol extract of G. thunbergii exhibited RLAR inhibitory activity ($IC_{50}$ value, $2.64{\mu}g/m{\ell}$). In order to identify the bioactive components of ethyl acetate soluble fraction of ethanol extract from G. thunbergii, eight compounds, namely gallic acid (1), protocatechuic acid (2), p-hydroxybenzoic acid (3), brevifolin carboxylic acid (4), geraniin (5), ellagic acid (6), kaempferol-3-O-arabinofuranosyl-7-O-rhamnopyranoside (7), kaempferitrin (8) were isolated. The isolates were subjected to in vitro bioassays to evaluate their inhibitory activity on RLAR and galactitol formation in rat lenses. The ellagic tannins (5, 6) and flavonoid (7) exhibited strong inhibitory effects on RLAR. Also, these three compounds (5, 6 and 7) suppressed galactitol accumulation in rat lens under high galactose conditions, demonstrating the potential to prevent galactitol accumulation exo vivo. These results suggest that the extracts and components of G. thunbergii are a promising agent in the prevention or treatment of diabetic complications.
