Chronophin activation is necessary in Doxorubicin-induced actin cytoskeleton alteration

  • Although doxorubicin (Dox)-induced oxidative stress is known to be associated with cytotoxicity, the precise mechanism remains unclear. Genotoxic stress not only generates free radicals, but also affects actin cytoskeleton stability. We showed that Dox-induced RhoA signaling stimulated actin cytoskeleton alterations, resulting in central stress fiber disruption at early time points and cell periphery cortical actin formation at a later stage, in HeLa cells. Interestingly, activation of a cofilin phosphatase, chronophin (CIN), was initially evoked by Dox-induced RhoA signaling, resulting in a rapid phosphorylated cofilin turnover leading to actin cytoskeleton remodeling. In addition, a novel interaction between CIN and $14-3-3{\zeta}$ was detected in the absence of Dox treatment. We demonstrated that CIN activity is quite contrary to $14-3-3{\zeta}$ binding, and the interaction leads to enhanced phosphorylated cofilin levels. Therefore, initial CIN activation regulation could be critical in Dox-induced actin cytoskeleton remodeling through RhoA/cofilin signaling.

Increased Antioxidative Activities against Oxidative Stress in Saccharomyces cerevisiae KNU5377 (산화 스트레스 대한 Saccharomyces cerevisiae KNU5377의 항산화 활성의 증가)

  • Oxidative stress is a consequence of an imbalance of the defense system against cellular damage generated by reactive oxygen species (ROSs) such as superoxide anions (menadione; MD). Most organisms have evolved a variety of defense systems to protect cells from adverse conditions. In order to evaluate stress tolerance against oxidative stress generating MD, comparative analyses of antioxidant capacity, or free radical scavenger ability, were performed between S. cerevisiae KNU5377 (KNU5377) and three wild-type S. cerevisiae strains. In a medium containing 0.4 mM MD, the KNU5377 strain showed higher cell viability and antioxidant ability, and contained higher levels of trehalose, superoxide dismutase, thioredoxin system, glucose-6-phosphate dehydrogenase, and some heat shock proteins. The KNU5377 strain also produced a lower level of oxidative stress biomarker than the other three yeast strains. These results indicate that S. cerevisiae KNU5377 has a higher level of tolerance to oxidative stress due to the increased expression of cell rescue proteins and molecules, thus alleviating cellular damage more efficiently than other S. cerevisiae strains.

The relationship between the stress in college life, vulnerability to depression, and smartphone overuse among undergraduate students majoring in public health (보건계열 학부생의 대학생활 스트레스, 우울 취약성 및 스마트폰 과다사용의 관련성)

  • purpose: This study aims to understand the relationship between stress in college life, vulnerability to depression, and smartphone overuse among undergraduate students majoring in public health. Methods: The subjects of the study were 320 students who were selected through convenience sampling from the department of emergency medical technology, physical therapy, and public health and administration attending G University in Gwangju. The study was carried out from October 22 to 26, 2012. For final analysis, 266 copies were used. Statistical analysis was done using SPSS WIN 12.0. Results: Excessive use of smartphones had a negative effect on stress in college life and vulnerability to depression. The stress of college life had an effect on students' sense of value, their friends of the same sex, and job. Depressive vulnerability had an effect on a students' dependency. Conclusion: For the prevention of smartphone overuse, programs to prevent college life stress and vulnerability to depression should be developed. Counselling programs for the groups should be made available to the students.

Factors Influencing Quality of Life in a Serious Case : the sixth Korea National Health and Nutrition Examination Survey (중증질환자 삶의 질에 영향을 미치는 요인 : 제6기 국민건강영양조사 이용)

  • Objectives: The purpose of this study was to identify the quality of life of a serious case and to analyze the factors influencing them. A serious disease were limited to cancer, cerebrovascular disease, and cardiovascular disease. Methods: This study used original data from the 6th National Health and Nutrition Survey (2013~2015) and finally extracted 385 persons diagnosed with cancer, cerebrovascular disease and cardiovascular disease among adults over 20 years old. Linear logistic regression analysis was conducted to identify factors affecting the quality of life of this study subjects. Results: The quality of life scores of the subjects were $13.92{\pm}1.54$ points (Highest possible=15). Regression analysis showed that quality of life decreased as age increased, lower income level, lower education level, stress, depression, suicidal ideation. The explanatory power (R2) of the analysis model was 0.170. Conclusions: The results of this study showed that age, income level, education level, stress, depression, and suicidal ideation affect the quality of life of a serious case in the community.

A Study of Low Cycle Fatigue Characteristics of 11.7Cr-1.1Mo Heat Resisting Steel with Mean Stress (Mean Stress를 고려한 11.7Cr-1.1Mo강의 고온저주기 피로특성에 관한 연구)

  • The Low cycle fatigue behavior of 11.7Cr-1.1Mo heat-resisting steel has been investigated under strain-controlled conditions with mean stresses at room temperature and $300^{\circ}C$. For the tensile mean stress test, the initial high tensile mean stress generally relaxed to zero at room temperature, however, at $300^{\circ}C$ initial tensile mean stress relaxed to compressive mean stress. Low cycle fatigue lives under mean stress conditions are usually correlated using modifications to the strain-life approach. Based on the fatigue test results from different stain ratio of -1, 0, 0.5, and 0.75 at room temperature and $300^{\circ}C$, the fatigue damage of the steel was represented by using cyclic strain energy density. Total strain energy density considering mean stress indicated well better than not considering mean stress at $300^{\circ}C$. Predicted fatigue life using Smith-Watson-Topper's parameter correlated fairly well with the experimental life at $300^{\circ}C$.

Parenting Stress in Mothers of School-aged Children : Effects of Ego-resilience, Social Support and School Grades of Children (학령기 자녀를 둔 어머니의 양육스트레스 : 자아탄력성, 사회적지지 및 자녀성적의 영향)

  • This study aimed to examine the effects of ego-resilience and social support on parenting stress perceived by mothers of school-aged children. The effects of demographic variables(including school grades of children) on the mothers' parenting stress were also investigated. The main results were as follows. First, ego-resilience of mothers was found to have the greatest predictive power on mothers' parenting stress. The more ego-resilient the mothers were, the less parenting stress was perceived. Second, social support also played an important role in alleviating maternal parenting stress. Emotional support of spouses was particularly effective in reducing mothers' parenting stress. Third, the only demographic variable that influenced mothers' parenting stress was their children's academic achievements. Finally, mothers perceived high parenting stress in relation to their children's school grades, while they perceived relatively less parenting stress concerning their children's temperament or the relationship with their children. The implications of these results were discussed.

Factors and Countermeasures for Middle School Students' School Related Stress (중학생의 학교 스트레스 요인과 대처방식)

  • This research was conducted to identify the factors that cause school related stress among middle school students, and to develop countermeasures in order to prevent the rebellious acts of middle school students that are on the rise and to pursue after effective guidance measures. To achieve these research objectives, 856 questionnaires were distributed to the two middle schools, situated in Goyang-si, Gyeonggi Province, and these questionnaires were analyzed, using the SPSS statistical package program. The conclusions were as follows; 1. Perceived factors for School Related Stress Overall average of the perceived degree school related stress factors was a level that goes beyond the normalcy. Stress on the grade was the highest Among these, the average of question items that addressed decrease in grade was the highest of all items followed by class factors, teacher-related factors, friend-related factors, and environmental factors. Accordingly, this research demonstrated that the students feel pressured by the university entrace examination and a school life that is focused on grades. The teacher-related factors and class factors were higher than the average, and others were relatively low. 2. School Related Stress according by Individual All the factors for school related stress were statistically significant difference according to the gender, and teacher-related factors, class factors, and environmental factors were statistically significant difference according to their school years. Their grade made their grade factors statistically significant difference, and the number of friends produced a statistically significant difference result on the teacher-related factors. Wheather they had extra classes or not made a statistically significant difference result on the class factors and environmental factors, and especially the time of doing physical exercises produced statistically significant difference on the environmental factors. 3. Countermeasures for School Related Stress according by Individual As for the countermeasures for school related stress according to the individuality, the active reacting was statistically significant difference according to the gender index, and the student's degree to react passively was statistically significant difference according to their grade. The students in different school years showed statistically significant difference reactions in both passive ways and active ways. students tended to react differently in an unreality index. Lastly, Wheather they had extra classes and physical exercises made then to react passively or actively. When the results are compiled, students are most affected by the stress related to grades, which reflected the university entrance examination oriented school life. An alternative is that the development of educational program that factors in the students' talents and aptitude is needed urgently. Moreover, character and career path related guidance and education are just as important as the academics. Given that the stress factors exert some degree of influence on the countermeasures, it is necessary to block the factors in advance and for all the teachers and parents to make an effort to react actively.

Life Stress, Social Support and Suicidal Ideation of North Korean Refugee Women in South Korea (탈북여성의 일상생활 스트레스가 자살생각에 미치는 영향과 사회적 지지의 조절효과)

  • The purpose of this study is to identify the moderating effect of social support between daily life stress and suicidal ideation of North Korean refugee women in South Korea. We survey with self-report questionnaires for 156 refugee women, conduct descriptive and regression analysis. As results, (1) almost half of the surveyed experience suicidal ideation in a year, (2) daily life stress affects suicidal ideation controlling with depression, PTSD, alcohol use, income, and age, (3) social support which North Korea refugee women perceive buffers the relationship between life stress and suicidal ideation. From these results, implications are suggested. Gender cognitive policies are needed in North Korea refugees in South Korea. Also, community mental health services and sustainable community program for North Korea refugee women are important to prevent their suicidal attempts.
