오미자(Schizandra chinensis Baillon) 에탄올 추출물의 항산화 활성 및 항돌연변이 활성 분석 (Analysis of Antioxidative Activity and Antimutagenic Effect of Ethanol Extract from Schizandra chinensis Baillon)

  • 본 연구에서는 오미자 에탄올 추출물의 항산화 활성과 총폴리페놀 함량 및 총 플라보노이드 함량을 측정하고, Salmonella typhimurium TA100을 이용하여 항돌연변이 활성을 분석하였다. 오미자 에탄올 추출물의 DPPH 라디칼 소거능은 $500{\mu}g$/assay의 농도에서 57%이었고, $IC_{50}$값이 $435\;{\mu}g$/assay으로 나타나 비교적 높은 항산화 활성을 가지고 있음을 보여주었다. 오미자 에탄올 추출물의 총 폴리페놀 함량과 총 플라보노이드 함량은 각각 9.53 mg/g과 3.97 mg/g으로 분석되었다. 또한 오미자 에탄올 추출물은 sodium azide(5 mg/plate) 와 4-NQO(1 mg/plate)에 대한 항돌연변이 활성이 각각 45%, 82%로 나타나 4-NQO에 대해 높은 돌연변이 억제 효과를 가지고 있는 것으로 나타났다. 본 연구를 통해 오미자 에탄올 추출물이 높은 항산화 활성과 항돌연변이 활성을 가진 것으로 나타나, 앞으로 천연 항산화제 및 항돌연변이 기능성 소재로 사용될 수 있는 가능성을 보여주었다.

오미자의 현탁배양세포로부터 Gomisin J의 생산 (Production of Gomisin J from Cell Suspension Cultures of Schisandra chinensis Baillon)

  • Cell growth and gomisin J production by suspension cultures of Schisandra chinensis Baillon were investigated under various culture media, initial sucrose concentrations, shaking speeds, and inoculum sizes. Callus was induced from in vitro cultivated leaf segments on MS medium supplemented with $1\;mg/{\ell}$ NAA. The maximum dry cell weight of 2.23 g was obtained at inoculum size of 0.5 g fresh cell weight and in MB5 medium supplemented with $1\;mg/{\ell}$ NAA, 3% sucrose after 8 weeks. The production of gomisin J in suspension cell cultures was maximized in WPM medium containing 5% sucrose. The shaking speed for maintaining maximal cell dry weight was 100 rpm while the best shaking speed for gomisin J accumulation was 140 rpm.

  • Depression is one of the most common neuropsychiatric disorders and has been associated with the neuroendocrine system and alterations in behavior. Schisandra chinensis Baillon is one of major medicinal plants used as a Korea medicine and food sources, and has been processed in the fields of various food products and medicinal herbs. The chronic mild stress (CMS) protocol is widely used to evoke depressive-like behaviors in laboratory mice or rat. The CMS procedure induced some behavioral changes that are compatible with the common expectations, i.e. 'anhedonic' behavior and can affect corticosterone level. The present study, Schisandra chinensis extract administration by daily gavage from the 3 weeks exhibited an antidepressant-like effect on CMS-induced depression in mice. Schisandra chinensis extract administration at dose of 200mg/kg significantly increased the sucrose consumption, and decreased the immobility durations in forced swim test and tail suspension test. Furthermore the corticosterone level decreased than control group. In conclusion, Schisandra chinensis extract showed antidepressant-like effects on sucrose preference test, forced swimming test and tail suspension test based on CMS model.

Studies on the Terpenoids in the Volatile Constituents of Liaoning Schisandra Chinensis Baillon

  • The terpenoids in the volatile constituents of Liaoning Shcisandra Chinesis Baillon have been determined by the analytical method of GC/MS. Thirty terpenoids molecular structure were characterized. They are 11.89% monterpenes, 4.60% monoterpene oxides, 58.74% sesquiterpene hydrocarbons, and 1.62% oxygen-containing sesquiterpenoids in the total volatile constituents quantified by chromatograph. Among them, the sesquiterpene make up the characteristic constituents. Every terpenoid constituent percent content was obtained using area normalization method of HP-59970 chemstation.

Estimating Economic Optimum Planted Area for Sustainable Schisandra chinensis Cultivation

  • This study determined the economic impact of environment-friendly cultivation and the optimal cultivation area of Omija (Korean for Schisandra chinensis Baillon) for full-time farmers by analyzing the management performance of existing Omija cultivators. The study divided the target income into urban household income and Omija farm income, and estimate the optimal cultivation area by substituting the target profit from the cost-volume-profit analysis model. The optimum cultivation area was 1.4 ha for general cultivation, 1.08 ha for organic cultivation, and 1.18 ha for pesticide-free farming cultivation considering the average urban household income as the target, and 0.81 ha for general cultivation, 0.63 ha for organic cultivation, and 0.69 ha for pesticide-free farming, considering the average 2012 farm household income as the target. Therefore, the study reached conclusion that it is necessary to secure the price of Omija farm and stable support for income increase. Therefore, the support plan for income stabilization of Omija farm should be considered. Especially, the central government should provide various policies and financial support to help the optimal cultivation area of Omija Farm.

오미자의 형질전환된 근으로부터 리그난 화합물의 검출 (Detection of Lignans from Transformed Root Cultures of Schisandra chinensis Baillon)

  • Transformed roots of Schisandra chinensis were obtained following co-cultivation of in vitro cultivated plantlet segments with Agrobaterium rhizogens ATCC15834. This root was examined for its growth and gomisin J contents under various culture conditions. Among the six basal culture media tested, WPM (Lloyd & McCown, 1980) medium supplemented with 5% sucrose was the best roots growth 6.2 (g D.W/flask) and gomisin J accumulation 1.56 $(X10^{-3}\;ug/g\;D.W)$. Initial inoculum size correlated with the yield of biomass while gomisin J contents was not affect. Gomisin J production was influenced by the initial sucrose concentration and the highest production yield was achieved at the concentration of 7%. The optimal shaking speeds for roots growth and gomisin J production was 120 and 140 rpm, respectively.

Schisandra Chinensis Baillon regulates the gene expression of phase II antioxidant/detoxifying enzymes in hepatic damage induced rats

  • BACKGROUND/OBJECTIVES: This study investigated the antioxidant activities and hepatoprotective effects of Schisandra chinensis Baillon extract (SCE) against tert-butyl hydroperoxide (t-BHP)-induced oxidative hepatic damage in rats. MATERIALS/METHODS: Sprague-Dawley (SD) rats were pretreated with SCE (300, 600, and 1,200 mg/kg BW) or saline once daily for 14 consecutive days. On day 14, each animal, except those belonging to the normal control group, were injected with t-BHP (0.8 mmol/kg BW/i.p.), and all of the rats were sacrificed 16 h after t-BHP injection. RESULTS: Although no significant differences in AST and ALT levels were observed among the TC and SCE groups, the high-dose SCE group showed a decreasing tendency compared to the TC group. However, erythrocyte SOD activity showed a significant increase in the low-dose SCE group compared with the TC group. On the other hand, no significant differences in hepatic total glutathione (GSH) level, glutathione reductase (GR), and glutathione peroxidase (GSH-Px) activities were observed among the TC and SCE groups. Hepatic histopathological evaluation revealed that pretreatment with SCE resulted in reduced t-BHP-induced incidence of lesions, such as neutrophil infiltration, swelling of liver cells, and necrosis. In particular, treatment with a high dose of SCE resulted in induction of phase II antioxidant/detoxifying enzyme expression, such as glutathione S-transferase (GST) and glutamate-cysteine ligase catalytic subunit (GCLC). CONCLUSIONS: Based on these results, we conclude that SCE exerts protective effects against t-BHP induced oxidative hepatic damage through the reduction of neutrophil infiltration, swelling of liver cells, and necrosis. In addition, SCE regulates the gene expression of phase II antioxidant/detoxifying enzymes independent of hepatic antioxidant enzyme activity.

Effects of Soil Water Potential and Nitrogen Fertilization on Characteristics of Photosynthesis and Chlorophyll Fluorescence Induction in Schisandra chinensis Baillon

  • Management of soil water and fertilization is known to primarily affect physiological properties and yield in plant. The effect of soil water potential and nitrogen application on characteristics of photosynthesis and chlorophyll fluorescence in Schisandra chinensis Baillon was investigated on a sandy loam soil. Net photosyntheis rate and transpiration rate increased as a photon flux density and was highest at -50kPa of soil water potential. Light compensation point ($1.5{\mu}molm^{-1}s^{-1}$) and dark respiration ($0.13{\mu}molCO_2m^{-1}s^{-1}$) was lowest at -50 kPa but maximum photosynthesis rate ($13.10{\mu}molCO_2m^{-1}s^{-1}$) and net apparent quantum yield ($0.083{\mu}molCO_2m^{-1}s^{-1}$) was highest at -50 kPa. As results of chlorophyll fluorescence by OJIP analysis, maximum quantum yield (Fv/Fm) of photosystem II (PSII) and PIabs was higher in treatments of -50 kPa and -60 kPa respectively, which reflects the relative reduction state of PSII. But the relative activities per reaction center such as ABS/RC and DIo/RC were low with decreasing soil water potential. Net photosyntheis rate and transpiration rate were highest at treatment of soil testing 1.0 times ($92kgha^{-1}$). Application of nitrogen resulted in high Fv/Fm, $PI_{abs}$ and low ABS/RC, DIo/RC. This result implies that -50 kPa of soil water potential and nitrogen fertilizer may improve the efficiency of photosynthesis through controlling a photosystem in Schisandra chinensis Baillon.

Effects of Fertilization and Co-Application of Compost Tea on Fruit Growth and Accumulation of Anthocyanin in Omija (Schisandra Chinensis Baillon)

  • This study was conducted to evaluate the influence of fertilization on characteristics of growth and accumulation of anthocyanin in fruit of Omija (Schisandra chinensis Baillon). Nitrogen supply mainly affected growth of fruit and the anthocyanin content in Omija respective of vegetative growth steps. The anthocyanin content in fruit was significantly low in high N supply and non-fertilization. The conjunctive supply of nitrogen and compost tea resulted in a higher anthocyanin content of fruit, total nitrogen content of leaf, and nitrate in soil. This result implies that nitrogen supply to Omija plant, affects the accumulation patterns of anthocyanin in different ways, e.g. it delays the quantitative biosynthesis at low nitrogen supply during fruit maturation or enhances anthocyanin degradation during the final maturation steps.

Schisandrol A and gomisin N from Schisandra chinensis extract improve hypogonadism via anti-oxidative stress in TM3 Leydig cells

  • BACKGROUND/OBJECTIVES: Male hypogonadism is a condition where the body does not produce enough testosterone and significantly impacts health. Age, obesity, genetics, and oxidative stress are some physiological factors that may contribute to testosterone deficiency. Previous studies have shown many pharmacological benefits of Schisandra chinensis (S. chinensis) Baillon as an anti-inflammatory and antioxidant. However, the molecular mechanism of attenuating hypogonadism is yet to be well established. This research was undertaken to study the effects of S. chinensis extract (SCE) on testosterone deficiency. MATERIALS/METHODS: S. chinensis fruit was pulverized and extracted using 60% aqueous ethanol. HPLC analysis was performed to analyze and quantify the lignans of the SCE. RESULTS: The 2,2-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) scavenging assays confirmed that the SCE and its major lignans (schisandrol A and gomisin N) inhibit oxidative stress. Effects of SCE analysis on the testosterone level under oxidative stress conditions revealed that both schisandrol A and gomisin N were able to recover the lowered testosterone levels. Through mRNA expression of TM3 Leydig cell, we observed that the SCE lignans were able to induce the enzymes involved in testosterone biosynthesis-related genes such as 3β-HSD4 (P < 0.01 for SCE, and P < 0.001 for schisandrol A and gomisin N), 17β-HSD3 (P < 0.001 for SCE, schisandrol A and gomisin N), and 17, 20-desmolase (P < 0.01 for schisandrol A, and P < 0.001 for SCE and gomisin N). CONCLUSIONS: These results support that SCE and its active components could be potential therapeutic agents for regulating and increasing testosterone production.
