Objective : Hwagi-Jokyeong-Tang (化氣調經湯, HJT) was described in DongeuiBogam(東醫寶鑑). This remedy has been used to treat patients with Naryeok, which is similar as tuberculous cervical lymphadenitis in western medicine. Methods : In this study, the present author investigated the effects of HJT on on Human HaCaT keratinocyte and malignant melanoma cells such as SK-MEL-2 and B16F10 in terms of cell viabilities, proliferations, DPPH free radical scavenging activities, oxygen free radical productions and inhibitory action on elastase activities. Results : HJT acceleated proliferation of HaCaT keratinocytes dose-dependantly. HJT also prevented cell death of HaCaT induced by Hydrogen peroxide, which products oxygen free radicals. On the contrary, HJT did not affect proliferations of SK-MEL-2 or B16F10. In addition, HJT was shown to have DPPH free radical scavenging activities and also have inhibitory effects on elastase activities too. On the fluorescent examinations, the present author know that HJT did not affect production levels of oxygen free radicals in malignant melanoma cell, SK-MEL-2. Conclusions : These results suggest that HJT has possibilities of usage for functional cosmetics which have skin regeneration or prevention from skin tissue injury.
Objectives : This study is to investigate the anti-inflammatory and anti-oxidant effects of Tongbi-san extract (TS) on RAW264.7 macrophages using by cell cytotoxicity, Nitric Oxide (NO) and Prostaglandin $E_2$ ($PGE_2$) production and 1,1-diphenyl-2-picryl ghdrazyl (DPPH) free radical scavenging capability. Methods : Cell cytotoxicity was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The production of NO was measured by Griess assay. The production of $PGE_2$ was measured by immunoassay. And the anti-oxidant activity was measured by the DPPH method. Results : TS did not increased significantly compared to the TS untreated group in the cell cytotoxicity. TS inhibited NO and $PGE_2$ production in lipopolysaccharide-stimulated RAW 264.7 cells. TS had the DPPH free radical scavenging capability. Conclusion : The anti-inflammtory and anti-oxidant effects of TS may be use for a treatment of anti-inflammatory diseases.
Background : At high glucose levels in $\beta$-cells, cell viability and insulin secretion are decreased by glucotoxicity. Sopyung-tang(SPT) had an effect on blood glucose level decrease and antioxidant enzyme activities in streptozotocin-induced diabetic rats. Objectives : This study performed a series of experiment to verify the effects of SPT extract on the cell viability, antioxidant enzyme activities, insulin secretion and insulin mRNA expression at hyperglycemic states of RIN-m5F. Methods : After treatment at various concentrations of SPT added to the RIN-m5F cells, cell viability by MTT assay, free radical-scavenging activity, SOD activity and insulin secretion were measured. Additionally, insulin-related gene expression was measured using real-time RT-PCR. Results : Compared to the control group, SPT extract showed considerable effects on RIN-m5F cell viability, DPPH radical-scavenging activity, superoxide dismutase (SOD) activity, insulin secretion and insulin-related gene expression. Conclusions : This study showed that SPT extract has an effect on $\beta$-cell cell viability, insulin secretion and insulin-related gene expression. Thus, SPT extract may be used for treatment of diabetes and its complications. Further mechanism studies of SPT seem to be necessary on the glucotoxicity and oxidative stress.
Objectives : Peroxynitrite (ONOO-), superoxide anion radical (?O2-) and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging process, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate Sabohwan's activity for scavenging ONOO- and its precursors. NO and ?02-. Methods : For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4.5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Results : Sabohwanblocked tert-butylhydroperoxide (t-BHP)-induced cell death in a dose-dependent fashion. It scavenged t-BHP-induced ONOO-, NO and ?O2- in YPEN cells. Sabohwan inhibited the generation of ONOO-, NO and ?O2- in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondria both in vitro and in vivo. The lipid peroxide level increased and glutathione level decreased in the LPS-treated mice, whereas the ones in the Sabohwanadministered group among the LPS-treated mice reversed toward their natural levels. Conclusions : These results suggest that Sabohwanis an effective ONOO-, ?O2- and NO scavenger, and thereby it might have a potential role as a therapy against the aging process and age-related diseases.
Objective: The purpose of this study is to observe the effects of Persicae Semen herbal-acupuncture at GB34(Yangneungcheon) on hyperlipidemia of rats caused by high-fat diet. Methods: The author performed several experiments to measure the DPPH radical scavenging activity of PS-HAS,to analyze various components and enzymes in serum and liver, and to observe the histological changes of liver and aorta. Results: 1. Persicae Semen herbal-acupuncture solution increased the DPPH radical scavenging activity in rat's liver cells. 2. In the Persicae Semen herbal-acupuncture group, compared with control group, the level of total cholesterol in serum and the ratio of total cholesterol to HDL-cholesterol decreased significantly. 3. In the Persicae Semen herbal-acupuncture group, compared with the control group, the level of glutathione in liver cells increased significantly. 4. In the Persicae Semen herbal-acupuncture group, compared with the control group, the pathological changes in liver and aorta were reduced. Conclusion: The above results suggest that Persicae Semen herbal-acupuncture at GB34 has good effects on preventing and treating hyperlipidemia.

The antioxidative properties of lactobacilli originating from humans (Lactobacillus acidophilus KCTC 3111, Lactobacillus jonsonii KCTC 3141, Lactobacillus acidophilus KCTC 3151, and Lactobacillus brevis KCTC 3498) were investigated using in vitro methods, including inhibition of lipid peroxidation, resistance to hydrogen peroxide and hydroxyl radical, hydroxyl radical scavenging activity, and glutathione peroxidase (GPX) activity. L. acidophilus KCTC 3111 showed the highest inhibition of lipid peroxidation in both intact cells (49.7%) and cell lysate (65.2%). This strain exhibited resistance to hydrogen peroxide and hydroxyl radical, which was viable for 7 h in the concentration of 1.0 mM hydrogen peroxide. In addition, this strain showed high hydroxyl radical scavenging activity. In the GPX activity assay, the highest activity was measured in L. brevis 3498. GPX activity of L acidophilus 3111 was lower than that of L. brevis 3498.
Objectives : The aerial parts of Rumex japonicus Houtt. (RF) is used by traditional clinics to treat parasite infection in East asia. This study aims a verification of anti-oxidative and anti-inflammatory effects of RF methanol extract. Methods : Anti-oxidative effects of RF were measured by scavenging activities of DPPH, superoxide, nitric oxide (NO) and peroxynitrite radicals. And also scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cells were measured. The inhibitory effects against the production of inflammatory mediators including NO, prostaglandin $E_2$ ($PGE_2$), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and the translocation of nuclear factor (NF)-${\kappa}B$ in LPS-stimulated RAW 264.7 cells by RF were tested. Results : RF scavenged DPPH, superoxide, NO and peroxynitrite radicals, and RF (at $200{\mu}g/m{\ell}$) reduced the inflammatory mediators definitely. Conclusions : These results indicate that RF may be a potential drug source for oxidative stress related inflammatory diseases.
Objectives: This study was perfomed to investigate the antioxidant activity and serotonin activity of Gami-guibi-tang on P815 Mast Cell. Methods: The effects of Gami-guibi-tang on activation of TPH-1 mRNA and AAADC mRNA in P815 mast cell were investigated. The effect of Gami-guibi-tang on content of serotonin in P815 mast cell was investigated. The effects of Gami-guibi-tang on activation of DPPH radical scavenging and SOD in P815 mast cell were investigated. Results: 1. The Gami-guibi-tang increased SOD activity and DPPH radical scavenging activity. 2. The Gami-guibi-tang increased the intracellular concentration of serotoninin 60 ${\mu}g/ml$, 80 ${\mu}g/ml$ experiment group. 3. The Gami-guibi-tang increased menaingful the manifestation TPH mRNA. 4. The manifestation of AAADC and MAO mRNA have not made menaingful changes on Gami-guibi-tang. Conclusions: This experiment shows that Gami-guibi-tang had significant anti-oxidative effect. And Gami-guibi-tang increased the intracellular concentration of serotonin. Therefore, Gami-guibi-tang can be used by the medication of major depression disorder. But Study on mechanism of increased serotonin and clinical research of Gami-guibi-tang is suggested for future research.
This study was performed to determine the optimal composition of muffins added with dropwort powder, egg and sugar. The experiment was designed based on BBD (Box-behnken design), and evaluation was carried out by means of RSM (response surface methodology), consisting of 13 experimental points with three replicates each for three independent variables dropwort powder, eggs and sugar. Using F-test, height, moisture, 1,1-diphenyl-2picriylhydrazyl (DPPH) radical scavenging activity, L-value, a-value, springiness, cohesiveness, chewiness, brittleness, color, and appearance acceptability were expressed as a linear model, whereas pH, b-value, flavor, taste, and texture were expressed as a quadratic model. After swallow and overall acceptability were expressed as a 2FI model. As dropwort powder content increased, 1,1-diphenyl-2picriylhydrazyl (DPPH) radical scavenging activity increased. Increasing amounts of dropwort powder led to reduction of sensory scores for color, appearance, flavor, taste, and texture quality. The optimum formulation determined by the numerical and graphical methods were similar: dropwort powder 6 g, egg 77 g, and sugar 60 g.
Background: We investigated the optimal aqueous extraction conditions for recovery of high yields of total phenolic compounds from roots of Arctium lappa L. (burdock, Asteraceae), and we compared their antioxidant capacity. Methods and Results: The antioxidant activity of the extracts was tested using 2,2-diphenyl-1-picrylhydrazyl, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt, and oxygen radical absorbance capacity assays. In addition, the major phenolic compounds present in the extracts were determined by high performance liquid chromatography analysis. Our results suggest that the roasted burdock $100^{\circ}C$, 15 min extract exhibited the strongest radical scavenging activity and possessed the highest concentration of phenolic compounds. The polyphenol content of both dried burdock and roasted burdock significantly increased with increase in the extraction temperature and time. Conclusions: These results indicated a relationship between phenolic compound levels in burdock and their free radical scavenging activities. This suggests that phenolic compounds significantly increase the antioxidant potential of burdock extracts.
