Effects of Saururus chinensis Baill on Lipid Metabolism Against TCDD Damage (다이옥신 저해능에 대한 삼백초의 지질대사에 미치는 효과)

  • This study was carried out to investigate the inhibitive effect of Saururus chinenesis Baill (SCB) on lipid peroxidation in Sprague-Dawely rat (SD-rat) accutely exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Saururus chinenesis Baill (200 mg/kg) was administered into rats intraperitoneally for four weeks, seven days after the injection of TCDD (1 ${\mu}$g/kg). we examined th lipid parameters by measuring the levels of Total Cholestero, Triglyceride(TG), HDL-Cholestero, and LDL-Cholesterol in serum and Malone Dialdehyde (MDA) in liver tissue of rats. Cholesterol was siginificantly elevated in TCDD-treated abnormal group (TTA). The higher level of HDL-Cholesterol was found in Saururus chinenesis Baill and TCDD administered group (STT), which showed the lower levels of Total-Cholesterol and LDL-Cholesterol. TG content in the TTA was 34.84% increased compared to Non TCDD-treated group (NTT). That of STT was inhibited by 18.90% compared to TTA. MDA content in the TTA was 44.72% increased compared to NTT. That of STT was inhibited by 17.14% compared to TTA.

Effect of Anti-oxidant, Anti-inflammatory and Anti-invasive of PMA-induced Matrix Metalloproteinase (MMP-2) and MMP-9 Activities of Water Extract and Solvent Fractions of Saururus Chinensis (삼백초 물 추출물과 유기용매 분획물의 항산화, 항염증 및 PMA에 의해 유도된 MMP-2 및 MMP-9활성 침윤 억제 효과)

  • Saururus chinensis is a perennial plants, its flavonoid compound is known to exhibit anti-oxidative activity. This study was aimed to investigate the effect of Water Extract and Solvent Fractions of Saururus chinensis on antioxidant, anti-inflammatory and anti-invasive of Phorbol 12-myristate 13-acetate (PMA)-induced matrix metalloproteinase (MMP-2) and MMP-9 activities. Plant samples were fractionated into hexane, CHCl3, ethyl acetate, butanol, and water fractions, and each of these was assayed individually. The water fraction showed the highest extraction yield at 9.25%(w/w). Anti-oxidative activity was analyzed by DPPH assay. Cell viability was detected by the MTS assay. Anti-inflammatory activity was assayed by the nitric oxide (NO) production in mouse macrophage Raw 264.7 cells. The activity and mRNA expression of MMP-2 and MMP-9 in human oral squamous carcinoma YD-10B cells were examined by zymography and RT-PCR. As results, MMP-2/-9 activation was increased in PMA induced YD-10B cells. In PMA-treated YD-10B cells, the increased mRNA expression and protein activation of MMP-2/-9 were significantly inhibited in the ethyl acetate fraction. The ethyl acetate fraction showed the highest anti-oxidative activity at 73.38%. The ethyl acetate fraction at non-cytotoxic concentrations significantly exhibited the anti-inflammatory activity of Raw 264.7 cells in dose-dependent manner. In conclusion, these findings demonstrate that the ethyl acetate fraction obtained from a chinensis water extract potentiates a promising therapeutic anti-invasive agent and, therefore, as an anti-cancer drug for cancer prevention and therapy in oral cancer.

Saururus chinenesis Extracts Scavenge Reactive Oxygen Species and Modulate Nitric Oxide Production in Raw 264.7 Macrophages

  • Saururus chinensis Baill has been used in Korean folk medicine for the treatment of various diseases such as edema, Jaundice, and furuncle. The components of this plant were extracted into four fraction. Among the four fraction, hexane and ethyl acetate fraction were highly toxic to 3T3 mouse embryo fibroblast and Raw 264.7 mouse macrophage, but n-butanol and residue fraction did not show any toxic effect to those cell lines. n-Butanol and residue fraction exhibited antioxidant effects on hydro-gen peroxide, hydroxyl radical, and superoxide anion directly in vitro and in the 3T3 fibroblasts. All the four fractions inhibited lipid peroxidation measured by thiobarbituric acid reactive substances (TBARS) formation. In addition, n-butanol and residue fraction showed inhibitory effects on lipopolysaccharide (LPS)-induced nitric oxide production, and also down-regulated inducible nitric oxide synthase (iNOS) mRNA transcription 6 h after LPS stimulation in Raw 204.7 cells. Only n-butanol fraction, which mainly consists of flavonoids, inhibited NF-kB activation by decreasing IkBa degradation 90 min after LPS stimulation. horn the results, it is suggested that this plant could be a good candidate material for drug development based on its antioxidant and/or anti-inflammatory constituents.

The Dyeability and Antimicrobial Activity of Saururus Chinensis(I) (삼백초의 염색성 및 항균성(I))

  • Saururus-chinensis was used for the natural due stuff extraction and concentration to produce the condensed dye liquid. Using this liquid, the silk and the cotton fabrics were dyed. And then the K/S value, color difference were measured in order to determine the dye ability and the antimicrobial activity. The K/S values of silk were much higher than those of cotton. In terms of the silk fabric, the Al, Cr-premordanting and Fe, Cu-simmordanting method was most efficient of the mordanting method. The various suface colors on the dyed fabric were resulted according to used mordants and mordanting methods. The dye fastness was significantly improved when mordants were added. Among them, Al-mordant improved more than 1 level. The Cu, Fe-mordants showed the greatest antimicrobial activity on both of mordant treat silk and cotton.

Saucerneol B with Hepatoprotective Effect of the Roots of Saururus chinensis

  • In order to find the new hepatoprotective agents from natural products, the isolation and identification of biological active components of the roots of Saururus chinensis has been carried out. A MeOH extract of this plant showed the significant hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells. Five lignans including sauchinone, manassantin A, manassantin B, saucerneol B, and di-O-methyltetrahydrofuriguaiacin B were isolated and identified by spectroscopic evaluation. (omitted)

  • Sauchinone, a known lignan, was isolated from the root of Saururus chinensis as an active principle responsible for inhibiting the production of NO in LPS-stimulated RAW264.7 cells by activity-guided fractionation. Sauchinone dose-dependently inhibited not only the production of NO, but also the expression of iNOS mRNA and protein in LPS-stimulated RAW 264.7 cells. Furthermore, sauchinone prevented LPS-induced NF-kB activation, which is known to play a critical role in iNOS expression, assessed by a reporter assay under the control of NF-kB. (omitted)

  • In our search for NF-kB inhibitors from natural resources, we have previously identified two structurally related dilignans, manassantin A and B as specific inhibitors of NF-kB activation from Saururus chinensis. However, their molecular mechanism of action remains unclear. We here demonstrate that manassantin A and B are potent inhibitors of NF-kB activation by the suppression of transciptional activity of RelA/p65 subunit of NF-kB. These compounds significantly inhibited the induced expression of NF-kB reporter gene by LPS or TNF-a in a dose-dependent manner. (omitted)

  • In the course of our search for intercellular adhesion molecule-1 (ICAM-1)/leukocyte function-associated antigen-1 (LFA-1) mediated cell adhesion inhibitors from natural sources, new type of cell adhesion inhibitors were isolated from the MeOH extract of Saururus chinensis roots. On the basis of spectral evidence, the structures of the active compounds were identified as manassantin A and B. Manassantin A and B inhibited phorbol 12-myristate 13-acetate (PMA)-induced homotypic aggregation of the human promyelocytic leukemia HL-60 cells without cytotoxicity with MIC value of 1.0 and 5.5 nM, respectively.
