Plant cell culture strategies for the production of natural products

  • Plants have evolved a vast chemical cornucopia to support their sessile lifestyles. Man has exploited this natural resource since Neolithic times and currently plant-derived chemicals are exploited for a myriad of applications. However, plant sources of most high-value natural products (NPs) are not domesticated and therefore their production cannot be undertaken on an agricultural scale. Further, these plant species are often slow growing, their populations limiting, the concentration of the target molecule highly variable and routinely present at extremely low concentrations. Plant cell and organ culture constitutes a sustainable, controllable and environmentally friendly tool for the industrial production of plant NPs. Further, advances in cell line selection, biotransformation, product secretion, cell permeabilisation, extraction and scale-up, among others, are driving increases in plant NP yields. However, there remain significant obstacles to the commercial synthesis of high-value chemicals from these sources. The relatively recent isolation, culturing and characterisation of cambial meristematic cells (CMCs), provides an emerging platform to circumvent many of these potential difficulties.

Baseline Sensitivity to Mandipropamid Among Isolates of Phytophthora capsici Causing Phytophthora Blight on Pepper

  • Sensitivity to the new carboxylic acid amide fungicide, mandipropamid, of Phytophthora capsici causing pepper Phytophthora blight was determined on 187 isolates collected in Korea over 3 years, from 2005 to 2007. All isolates were sensitive to mandipropamid, with $EC_{30}$ values for growth of mycelia ranging from 0.001 to $0.037\;{\mu}g/ml$. Among the isolates, 147 (79.0%) isolates were sensitive to metalaxyl, whereas others were resistant to this fungicide. Mandipropamid had the same effect on mycelium growth of both metalaxyl-sensitive and metalaxyl-resistant isolates, indicating an absence of cross-resistance between these two fungicides. Comparison of the sensitivities of P. capsici isolates showed a positive correlation between sensitivity to mandipropamid and dimethomorph ($r^2$=0.8533). The results of this study indicate that there is no evidence for development of resistance to mandipropamid in this population of P. capsici isolates collected in Korea.

  • For design of early childhood children's footwear, this study was to analyze the properties of their foot. This study was performed in 200 male and 200 female early childhood children from 3 and 6 years old lived in susan urban area. There were measured 17 items of the foot and calculated 4 indexs for analysis. The result was as follows; 1 The growth rate differs from the sites of a foot. According to the growth of age, early childhood children's foot are more slender. 2. The average of foot print angle is $15~27^{\circ}$ and it is flat. Among the age groups, the numbers of 4 kinds of foot print angle are different siginificantly. 3. The average of metatarso phalanx angle is $174~178^{\circ}$ that is higher than adults'. 4. There are no significant difference between male children's foot and female childen's one except tarsal circumference and tarsal height. Tarsal part of male children is higher and ticket than female children's. 5. Because some of 3~6 age groups belong to the one foot length group, we should consider the age properties of foot in order to design footwears. When the size of footwear is divided by only foot length, the ranges of another measurements are larger than the range of foot length.

Engineering Geological Analysis for the Quarry Located at the Construction Site of the New Susan Harbor (부산 신항만 건설현장의 채석장에 대한 지질공학적 고찰)

  • A quarry is operated for filling-up the New Busan Harbor which is under construction, but actual rock quality is something different from the primary design. Therefore, object of this study is to classify whole rock quality for the quarry through site investigation and laboratory analyses because unexpected large amount of wasted rock is produced. For this object, various analyses were performed such as surveying, Schimidt Hammer test. joint spacing investigation and laboratory analyses using DIPS & RockWorks programs for evaluating joint sets and sizes of rock fragments after blasting. As a result, it is expected that large amount of wasted rock under ${\Phi}100mm$ is produced after blasting because of high joint density.

Meteorologically Adjusted Ozone Trends in the Seoul and Susan Metropolitan Areas (서울과 부산지역 기상의 영향을 제거한 오존농도 추세)

  • Surface ozone concentrations are highly sensitive to meteorological variability. Therefore, in order to reveal the long-term changes in ozone due to the changes in precursor emissions, we need to remove the effects of meteorological fluctuations on the annual distribution of surface ozone. In this paper, the meteorologically adjusted trends of daily maximum surface ozone concentrations in two major Korean cities (Seoul and Busan) are investigated based on ozone data from 11 (Seoul) and 6 (Busan) sites over the period 1992 ∼ 2000. The original time series consisting of the logarithm of daily maximum ozone concentrations are splitted into long-term, seasonal and short-term component using Kolmogorov-Zurbenko (KZ) filter. Meteorological effects are removed from filtered ozone series using multiple linear regression based on meteorologcial variables. The long-term evolution of ozone forming capability due to changes in precursor emission can be obtained applying the KZ filter to the residuals of the regression. The results indicated that meteorologically adjusted long-term daily maximum ozone concentrations had a significant upward trend (Seoul: + 3.02% yr$^{-1}$ , Busan: + 3.45% yr$^{-1}$ ). These changes of meteorologically adjusted ozone concentrations represent the effects of changing background ozone concentrations as well as the more localized changes in emissions.

Antibiofilm and Anti-β-Lactamase Activities of Burdock Root Extract and Chlorogenic Acid against Klebsiella pneumoniae

  • Small phytochemicals have been successfully adopted as antibacterial chemotherapies and are being increasingly viewed as potential antibiofilm agents. Some of these molecules are known to repress biofilm and toxin production by certain bacterial and yeast pathogens, but information is lacking with regard to the genes allied with biofilm formation. The present study was performed to investigate the inhibitory effect of burdock root extract (BRE) and of chlorogenic acid (CGA; a component of BRE) on clinical isolates of Klebsiella pneumoniae. BRE and CGA exhibited significant antibiofilm activity against K. pneumoniae without inflicting any harm to its planktonic counterparts. In vitro assays supported the ${\beta}$-lactamase inhibitory effect of CGA and BRE while in silico docking showed that CGA bound strongly with the active sites of sulfhydryl-variable-1 ${\beta}$-lactamase. Furthermore, the mRNA transcript levels of two biofilm-associated genes (type 3 fimbriae mrkD and trehalose-6-phosphate hydrolase treC) were significantly downregulated in CGA- and BRE-treated samples. In addition, CGA inhibited biofilm formation by Escherichia coli and Candida albicans without affecting their planktonic cell growth. These findings show that BRE and its component CGA have potential use in antibiofilm strategies against persistent K. pneumoniae infections.

  • The aim of this study is to) represent the relationship of organizational commitment and its antecedent variable and show the influence of them in the libraries. For this, We selected librarian\`s personal characteristics, job characteristics, organizational characteristics and work experiences as an independent variables and value commitment, commitment to stay as a dependent variables. Collected data was derived from questionnaire of the 128 librarians of 17 public libraries at susan. 1)The women\`s level of commitment was higher than man. 2)Centralization was negative to the organizational commitment. 7)The rank that influence on the organizational commitment was work experience first, and then organizational characteristics, and job characteristics was the last.

Solution Structure of a Prion Protein: Implications for Infectivity

  • Prions cause neurodegenerative diseases in animals and humans. The scrapie prion protein (PrPSc) is the major-possibly only-component of the infectious prion and is generated from the cellular isoform (PrPC) by a conformational change. Limited proteolysis of PrPSc produces an polypeptide comprised primarily of residues 90 to 231, which retains infectivity. The three-dimensional structure of rPrP(90-231), a recombinant protein resembling PrPC with the Syrian hamster (SHa) sequence, was solved using multidimensional NMR. Low-resolution structures of rPrP(90-231), synthetic peptides up to 56 residues, a longer (29-231, full-length) protein with SHa sequence, and a short here further structure refinement of rPrP(90-231) and dynamic features of the protein. Consideration of these features in the context of published data suggests regions of conformational heterogeneity, structural elements involved in the PrPC\longrightarrowPrPSc transformation, and possible structural features related to a species barrier to transmission of prion diseases.

Accuracy Assessment of Annual Energy Production Estimated for Seongsan Wind Farm (성산 풍력발전단지의 연간발전량 예측 정확도 평가)

  • In order to examine how accurately the wind farm design software, WindPRO and Meteodyn WT, predict annual energy production (AEP), an investigation was carried out for Seongsan wind farm of Jeju Island. The one-year wind data was measured from wind sensors on met masts of Susan and Sumang which are 2.3 km, and 18 km away from Seongsan wind farm, respectively. MERRA (Modern-Era Retrospective Analysis for Research and Applications) reanalysis data was also analyzed for the same period of time. The real AEP data came from SCADA system of Seongsan wind farm, which was compare with AEP data predicted by WindPRO and Meteodyn WT. As a result, AEP predicted by Meteodyn WT was lower than that by WindPRO. The analysis of using wind data from met masts led to the conclusion that AEP prediction by CFD software, Meteodyn WT, is not always more accurate than that by linear program software, WindPRO. However, when MERRA reanalysis data was used, Meteodyn WT predicted AEP more accurately than WindPRO.

Association Between Socioeconomic Status and Obesity in Adults: Evidence From the 2001 to 2009 Korea National Health and Nutrition Examination Survey

  • Objectives: The present study examined relationships between socioeconomic status (SES) and obesity and body mass index (BMI) as well as the effects of health-related behavioral and psychological factors on the relationships. Methods: A cross-sectional population-based study was conducted on Korean adults aged 20 to 79 years using data from the 2001, 2005, and 2007 to 2009 Korea National Health and Nutrition Examination Survey. Multivariate logistic and linear regression models were used to estimate odds ratios of obesity and mean differences in BMI, respectively, across SES levels after controlling for health-related behavioral and psychological factors. Results: We observed significant gender-specific relationships of SES with obesity and BMI after adjusting for all covariates. In men, income, but not education, showed a slightly positive association with BMI (p<0.05 in 2001 and 2005). In women, education, but not income, was inversely associated with both obesity and BMI (p<0.0001 in all datasets). These relationships were attenuated with adjusting for health-related behavioral factors, not for psychological factors. Conclusions: Results confirmed gender-specific disparities in the associations of SES with obesity and BMI among adult Korean population. Focusing on intervention for health-related behaviors may be effective to reduce social inequalities in obesity.
