Deducing Isoform Abundance from Exon Junction Microarray

  • Alternative splicing (AS) is an important mechanism of producing transcriptome diversity and microarray techniques are being used increasingly to monitor the splice variants. There exist three types of microarrays interrogating AS events-junction, exon, and tiling arrays. Junction probes have the advantage of monitoring the splice site directly. Johnson et al., performed a genome-wide survey of human alternative pre-mRNA splicing with exon junction microarrays (Science 302:2141-2144, 2003), which monitored splicing at every known exon-exon junctions for more than 10,000 multi-exon human genes in 52 tissues and cell lines. Here, we describe an algorithm to deduce the relative concentration of isoforms from the junction array data. Non-negative Matrix Factorization (NMF) is applied to obtain the transcript structure inferred from the expression data. Then we choose the transcript models consistent with the ECgene model of alternative splicing which is based on mRNA and EST alignment. The probe-transcript matrix is constructed using the NMF-consistent ECgene transcripts, and the isoform abundance is deduced from the non-negative least squares (NNLS) fitting of experimental data. Our method can be easily extended to other types of microarrays with exon or junction probes.

Seismic performance of RC bridge piers subjected to moderate earthquakes

  • Experimental investigation was conducted to evaluate the seismic ductility of earthquake-experienced concrete columns with an aspect ratio of 2.5. Eight circular concrete columns with a diameter of 600 mm were constructed with three test parameters: confinement ratio, lap-splice of longitudinal bars, and retrofitting with Fiber Reinforced Polymer (FRP) materials. The objective of this research is to examine the seismic performance of RC bridge piers subjected to a Quasi static test (QST), which were preliminary tested under a series of artificial earthquake motions referred to as a Pseudo dynamic test (PDT). The seismic enhancement effect of FRP wrap was also investigated on these RC bridge piers. Six specimens were loaded to induce probable damage by four series of artificial earthquakes, which were developed to be compatible with earthquakes in the Korean peninsula by the Korea Highway Corporation (KHC). Directly after the PDT, six earthquake-experienced columns were subjected to inelastic cyclic loading under a constant axial load of $0.1{f_c}^{\prime}A_g$. Two other reference specimens without the PDT were also subjected to similar quasi-static loads. Test results showed that specimens pre-damaged by moderate artificial earthquakes generally demonstrated good residual seismic performance, which was similar to the corresponding reference specimen. Moreover, RC bridge specimens retrofitted with wrapping fiber composites in the potential plastic hinge region exhibited enhanced flexural ductility.

Bidirectional Lateral Loading of RC Columns with Short Lap Splices (겹침이음 길이가 짧은 RC 기둥의 이방향 횡하중 가력 실험)

  • Reinforced concrete (RC) buildings built in the 1980s are vulnerable to seismic behavior because they were designed without any consideration of seismic loads. These buildings have widely spaced transverse reinforcements and a short lap splice length of longitudinal reinforcements, which makes them vulnerable to severe damage or even collapse during earthquakes. The purpose of this study is to investigate the impact of bidirectional lateral loads on RC columns with deficient reinforcement details. An experimental test was conducted for two full-scale RC column specimens. The test results of deficient RC columns revealed that bidirectional loading deteriorates the seismic capacity when compared with a column tested unidirectionally. Modeling parameters were extracted from the tested load-displacement response and compared with those proposed in performance-based design standards. The modeling parameters proposed in the standards underestimated the deformation capacity of tested specimens by nearly 50% and overestimated the strength capacity by 15 to 20%.

For the purpose of precise evaluation, the experimental investigation was conducted to study the seismic performance of the real size specimen, which is constructed for highway bridge piers in Korean peninsula. Since it is believed that Korea belongs to the moderate seismicity region, five test specimens were designed in accordance with limited ductility design concept. Another one test specimen was nonseismically designed according to a conventional code. Important test parameters were transverse reinforcement and lap splicing. Lap splicing was frequently used in the plastic hinge region of many bridge columns. Furthermore, the seismic design code is not present about lap splice in Korean Roadway Bridge Design Code. The results show that specimens designed according to the limited ductility design concept exhibit higher seismic resistance. Specimens with longitudinal steel lap splice in the plastic hinge region appeared to significantly fail at low ductility level.

Behavior of Solid and Hollow Rectangular RC Piers with 50% of Lap-Spliced Longitudinal Bars (50%주철근 겹침이음을 갖는 중실 및 중공 사각단면 교각의 거동특성)

  • Scale model tests were performed to investigate the seismic behavior of the solid and hollow rectangular RC piers with 50% of lap-spliced longitudinal bars in plastic hinge regions. Continuous bars and lap-spliced ones with a lap length of 39 times the bar diameter were arranged alternately in the sections. In order to clarify the influence of lap splice on a ductility the effect of axial force and lateral confinement were excluded in the test. The typical flexural failure conducting a ductile behavior were observed in both models. It is confirmed that the 50% of lap-spliced bars can be considered as an alternative of seismic detailing for longitudinal bars.

Restoration of pre-damaged RC bridge columns using basalt FRP composites

  • This study aims to identify the effect of both longitudinal reinforcement details and damage level on making a decision of repairing pre-damaged bridge columns using basalt fiber reinforced polymer (BFRP) jackets. Two RC bridge columns with improper details of the longitudinal and/or transverse reinforcement were tested under the effect of a constant axial load and increasing lateral cyclic loading. Test results showed that the lap-splice column exhibited an inferior performance where it showed rapid degradation of strength before achieving the theoretical strength and its deformation capacity was limited; however, quick restoration is possible through a suitable rehabilitation technique. On the other hand, expensive repair or even complete replacement could be the decision for the column with the confinement failure mode. After that, a rehabilitation technique using external BFRP jacket was adopted. Performance-based design details guaranteeing the enhancement in the inelastic performance of both damaged columns were addressed and defined. Test results of the repaired columns confirmed that both reparability and the required repairing time of damage structures are dependent on the reinforcement details at the plastic hinge zone. Furthermore, lap-splice of longitudinal reinforcement could be applied as a key design-tool controlling reparability and restorability of RC structures after massive actions.

Fatigue crack growth and remaining life estimation of AVLB components

  • The fatigue cracks initiate and propagate in the Armored Vehicle Launch Bridge (AVLB) components, especially like the splice doubler angle, splice plate, and bottom chord, due to the cyclic loading by repeated AVLB-launchings and tank-crossings. In this study, laboratory fatigue tests were conducted on six aluminum 2014-T6, four aluminum 7050-T76511, and four ASTM A36 steel compact-tension specimens to evaluate the crack growth behavior of the materials used for the components. The experimental results provide the relationship (Paris Law) between crack growth rate, da/dn, and stress intensity range, ${\Delta}K$, whose material dependent constants C and m can later be used in the life estimation of the components. Finite Element Method (FEM) was used to obtain the stress intensity factor, K, of the components with cracks. Because of the complexity of loading conditions and component geometry, several assumptions and simplifications are made in the FEM modeling. The FEM results, along with the results obtained from laboratory fatigue tests, are then utilized to estimate critical crack length and remaining life of the components.

  • Cable member in structure is tension systems in which the load carrying members transmit loads to support system by tensile stress with no compression or flexure allowed. Cable system have been widely used large span structure roof, air-supported structure, prestressed membrane, cable network roof, suspension structures, guyed tower, ocean platforms, suspension bridges. Cable member can transmit loads by the edge connected system such as socket, swaging, mechanical splice sleave, clip, wedge, loop splice etc. This study will shown an experimental results on the strength of connection socket of cables. In the results of experiment, most of cable connection specimen occurred the failure at the connection socket part before the cable arrived at tensile failure load.

Prediction of the bond strength of ribbed steel bars in concrete based on genetic programming

  • This paper presents the application of multi-gene genetic programming (MGP) technique for modeling the bond strength of ribbed steel bars in concrete. In this regard, the experimental data of 264 splice beam tests from different technical papers were used for training, validating and testing the model. Seven basic parameters affecting on the bond strength of steel bars were selected as input parameters. These parameters are diameter, relative rib area and yield strength of steel bar, minimum concrete cover to bar diameter ratio, splice length to bar diameter ratio, concrete compressive strength and transverse reinforcement index. The results show that the proposed MGP model can be alternative approach for predicting the bond strength of ribbed steel bars in concrete. Moreover, the performance of the developed model was compared with the building codes' empirical equations for a complete comparison. The study concludes that the proposed MGP model predicts the bond strength of ribbed steel bars better than the existing building codes' equations. Using the proposed MGP model and building codes' equations, a parametric study was also conducted to investigate the trend of the input variables on the bond strength of ribbed steel bars in concrete.
