The Characteristics of Thermophilic Fungi in Relation to Growth-Promoting Effect on the Mycelium of Pleurotus ostreatus (Pleurotus ostreatus 균사의 생장 촉진 효과를 나타내는 고온성 곰팡이의 특징)

  • The mycelial growth of Pleurotus ostreatus in compost is strongly stimulated by solid-state fermentation with thermophilic fungi which were isolated from oyster mushroom compost. The biochemical characteristics of these thermophilic fungi were investigated. Cellulase and ligninase activities were not detected by clear zone effect on CMC and lignin media. All of thermophilic fungi grew well with high mycelial density on xylan media and the growing rate of Sepedonium sp. S-2 observed very high. In results of MUF-test, extracellular enzyme activity of Sepedonium sp. S-2, and S-5 measured very high. On the compost after high temperature fermentation with Sepedonium sp. S-2 and S-5, the mycelial growing rate of Pleurotus ostreatus was increased about 50% and it also showed the inhibiting effect on mycelial growth of Trichoderma sp. SJG-51. Isolated thermophilic fungi, Sepedonium sp. S-2 and S-5 were expected as very useful organism for making oyster mushroom compost.

  • Papilla and cytoplasmic aggregates clearly formed on the epidermal cells of barley leaves in response to the primary germ-tubes of Erysiphe graminis f. sp. hordei, but their sizes were much smaller than those in response to the appressoria. Some cells of barley leaves exposed to powdery mildew for 36-48h were more deeply stained as compared to the other cells by acid fuchsin. However, the content of malondialdehydein in powdery mildewed leaves, one of the product of lipid peroxidation, did not increase by 96h after inoculation. Positive reactions for callose, protein and phenolics were recognized in the papilla and cytoplasmic aggregates at 6h after inoculation, but cutin, suberin, cellulose and lignin were not noticeable until 72h after inoculation. The total phenol content in methanol extracts increased with increasing time after inoculation. All histochemical reactions were not race-specific in barley­powdery mildew combinations tested.

Optimization of Media Composition on the Production of Melanin Bleaching Enzyme from Peniophora sp. JS17 (Peniophora sp. JS17 유래 멜라닌 탈색 효소 생산을 위한 배지 조성의 최적화)

  • Peniphora sp. JS17, isolated from forest old tree, produced extracellular enzymes that decolorized human hair melanin. The JS17 strain had laccase and manganese peroxidase activity while it did not has lignin peroxidase activity. Batch culture indicated that the melanin decolorization activity of JS17 strain originated from laccase. The culture conditions to maximize the production of melanin bleaching enzymes from Peniophora sp. JS17 mycelia were investigated. Among the tested media for the laccase production, minimal medium (2% glucose, 0.2% malt extract, 0.1% $KH_2PO_4$, 0.4% $MgSO_4{\cdot}7H_2O$) showed the highest activity of laccase. Then, to optimize the culture condition for the laccase activity, the influence of various carbon and nitrogen sources was investigated in minimal medium. Among various carbon and nitrogen sources, 2% xylose and 0.4% tryptone showed the highest production of laccase, respectively. The enzyme was purified using $(NH_4)_2SO_4$ precipitation and Hitrap Q sepharose column, and the purified enzyme showed two isoenzymatic bands with molecular masses of about 70 kDa by SDS-PAGE. The melanin decolorization activity was 77% and 55% within 48 h in the presence of 1-hydroxybenzotriazole (HBT) and syringaldehyde, respectively, whereas only about 9% melanin decolorized in case of no mediator.

  • This experiment was carried out to get the basic information on utilizing artemisia selengensis, the chemical component of artemisia selengensis and those Artemisia sp. were analyzed and compared. The results are as followed; The contents of water, crude ash and crude fiber in Artemisia selengensis were higer than in Artemisia sp. the contents of thiamin, riboflavin and ascorbic acid in Artemisia selengensis were a little lower than those in Artemisia sp. while niacin in Artemisia selengensis was a little higher than those in Artemisia sp. The content of dietary fiber were 40.91% for Artemisia selengensis and 4166.7mg% for Artemisia sp. and the contents of Na, Fe and Mn in Artemisia selengensis were over twice as a high as those in Artemisia sp. While the content of Cu was Artemisia selengensis was lower than that in Artemisia sp. The content of phenolic compound in Artemisia selengensis was a little higher than those in Artemisia sp.

  • Pseudomonas sp. PAMC 29040 was isolated from a maritime tundra soil in Antarctica for its ability to degrade lignin and subsequently confirmed to be able to depolymerize heterogeneous humic substance (HS), a main component of soil organic matter. The draft genome sequences of PAMC 29040 were analyzed to discover the putative genes for depolymerization of polymeric HS (e.g., dye-decolorizing peroxidase) and catabolic degradation of HS-derived small aromatics (e.g., vanillate O-demethylase). The information on degradative genes will be used to finally propose the HS degradation pathway(s) of soil bacteria inhabiting cold environments.

  • Cellulose degrading bacteria from koala faeces were isolated using caboxymethylcellulose-Congo red agar, screened in vitro for different hydrolytic enzyme activities and phylogenetically characterized using molecular tools. Bacillus sp. and Pseudomonas sp. were the most prominent bacteria from koala faeces. The isolates demonstrated good xylanase, amylase, lipase, protease, tannase and lignin peroxidase activities apart from endoglucanase activity. Furthermore many isolates grew in the presence of phenanthrene, indicating their probable application for bioremediation. Potential isolates can be exploited further for industrial enzyme production or in bioremediation of contaminated sites.

  • The Purpose of this study was to isolate and identify fungi found in excavated clothes dating on the 16th century. As a result of microscoping with LPCB staining, seven isolated strains were classified-Acremonium strictum, Acremonium kiliense, Fonsecaea sp., Candida sp., Trichoderma sp., Cladosporium spp., Penicillium spp.. In the process of isolation and identification of fungi by the sort of textile, more fungi were found in cotton, cotton batt and silk than in ramie and hemp, suggesting that hemp has antibacterial characteristics due to the presence of lignin. In the Comparison washed samples with unwashed ones, there were more kinds of fungi in washed samples. In the cellulase activity test, Acremonium kiliense had the highest level cellulase activity, suggesting that Acremonium killiense has an important Influence on damage of textiles.

  • This study was aimed to isolation and identification of attached bacteria on the clothes excavated from Deajeon area dating on the 16th century. From the observation with colony shape, 17 bacterial strains were isolated, and then 7 bacterial strains were identified with morphological and biochemical characteristics. Streptococcus sp., Alcaligenes faecalis, Gemella sp., Acinetobacter sp., Pseudomonas vesicularis, Aeromonas sain. salmonicida, Moraxella spp. In observation of the bacterial strains by the sort of textile, more bacterial strains were found in silk, cotton, and cotton batt than in ramie and hemp. It is suggest that hemp has antibacterial characteristics due to the presence of lignin. In the comparison washed samples with unwashed ones, there were more kinds of bacterial strains in washed samples. In the cellulase activity tests, all isolated bacteria had low level cellulase activity.

Bleaching of Kraft Pulp with Lignin - Degrading Enzymes

  • An unbleached hardwood kraft pulp was bleached in vitro with partially purified manganese peroxidase (MnP) from the fungus Phanerochaete sordida YK-624 without the addition of MnSO$_4$ in the presence of oxalate, malonate or gluconate known as manganese chelator, When the pulp was treated without the addition of MnSO$_4$, the pulp brightness increased by about 10 points in the presence of 2 mM oxalate, but the brightness did not significantly increase in the presence of 50 mM malonate. Residual MnP activity decreased faster during the bleaching with MnP without MnSO$_4$ in the presence of malonate than in the presence of oxalate. Oxalate reduced MnO$_2$ which already existed in the pulp or was produced from $Mn^{2+}$ by oxidation with MnP and thus supplied $Mn^{2+}$ to the MnP system. Thus, bleaching of hardwood kraft pulp with MnP, using manganese originally existing in the pulp, became possible in the presence of oxalate, a good manganese chelator and reducing reagent. Properties of partially purified MnPs from liquid cultures of white rot fungi, Ganoderma sp. YK-505, Phanerochaete sordida YK-624 and Phanerochaete chrysosporium were compared. MnP from Ganoderma sp. YK-505 was superior to MnPs from P. sordida YK-624 and P. chrysosporium in stabilities against high temperature and high concentration of $H_2O$$_2$. The MnP from Ganoderma sp. YK-505 differed in pH-activity profile from other MnPs. These data suggest that MnP from Ganoderma sp. YK-505 has different structure from those of other fungi. Bleaching of hardwood kraft pulp using the MnP from ganoderma sp. YK-505 is now in progress.
