인천 연안에서 분리한 원유 분해 미생물의 특성 연구 (Characterization Study of Crude Oil Degrading Microbiology Isolated from Incheon Bay)

  • 토착 미생물은 친환경적 정화에 중요한 역할을 한다. 원유(crude oil)를 분해하는 80균주를 인천 연안에서 분리하고 oil film collapsing방법을 이용하여 유화능이 있는 12균주를 선별하였다. 이들 균주에 대해 ${\rho}$-nitrophenyl butylrate를 기질로 이용하여 리파아제(lipase)활성과 n-hexanedecane을 기질로 이용하여 유화(emulsification)활성을 측정하여 원유 분해 활성이 좋은 Incheon9를 선별하고 gas chromatography (FID)로 paraffine계 탄화수소를 감소시키는 것을 확인하였다. 이 균주의 16s rRNA유전자 분석을 통해 Acinetobacter sp.로 동정하고 NCBI에 등록하여 accession code (KF548540)를 부여 받았다. Acinetobacter sp. Incheon9의 성장과 유화능이 최적 배양 조건은 $20^{\circ}C$, pH 7, 1% NaCl였으며 대수증식기 기간에 가장 높은 유화능을 보였으며 탄화수소가 짧은 trybutyrin에서 분해능력이 좋았다. 이번 연구결과는 환경오염에 활용 가능한 미생물자원군의 확보를 위한 연구였으며 추후 활용을 위해서는 실제 환경에서 동일한 활성을 가지는지 여부에 대한 연구가 추가로 진행되어야 할 것이다.

Bacillus sp.의 생장과 인 제거능에 대한 단일 및 2종 혼합 중금속의 독성 평가 (Toxicity Evaluation of Single and Binary Mixture of Heavy Metals on the Growth and Phosphorus Removal Ability of Bacillus sp.)

  • In this study, the effects of single and binary heavy metals toxicity on the growth and phosphorus removal ability of Bacillus sp.. known as be a phosphorus-removing microorganism, were quantitatively evaluated. Cd, Cu, Zn, Pb, Ni were used as heavy metals. As a result of analysis of variance of the half of inhibition concentration and half of effective concentration for each single heavy metal treatment group, the inhibitory effect on the growth of Bacillus sp. was Ni < Pb < Zn < Cu < Cd. And the inhibitory effect on phosphorus removal by Bacillus sp. was Ni < Pb < Zn < Cu < Cd. When analyzing the correlation between growth inhibition and phosphorus removal efficiency of a single heavy metal treatment group, a negative correlation was found (R2 = 0.815), and a positive correlation was found when the correlation between IC50 and EC50 was analyzed (R2 = 0.959). In all binary heavy metal treatment groups, the interaction was an antagonistic effect when evaluated using the additive toxicity index method. This paper is considered to be basic data on the toxic effects of heavy metals when phosphorus is removed using phosphorus removal microorganisms in wastewater.

  • A new sphingosine (1) was isolated from the MeOH extract of a marine sponge Haliclona (Reniera) sp. by bioactivity-guided fractionation. The 1D and 2D NMR, and MS spectroscopic analyses were used to establish the planar structure of 1. The stereochemistry of the compound was defined on the basis of modified Mosher's method, comparison of optical rotation and NMR data with those of the reported. Compound 1 was mildly cytotoxic to a panel of five human solid tumor cell lines.

  • Matrix Assisted Laser Desorption ionization Time of Flight (MALDI-TOF) was used for enzymes identification related to B -1,3-glucan synthesis. Agrobacterium sp. ATCC31750 was cultivated with two stage Continuous Stirrer Tank Reactor (CSTR) and the cells were harvested and their protein profiles were analysed by two dimensional electrophoresis. The specific enzyme spot was treated with trypsin and ana lysed by MALDI-TOF to get peptide molecular weight. The peptide molecular weights were matched with Agrobacterium tumefacience's Data Base from the matrix science site, then could identify the avaliable key enzymes. In this study, we identified key metabolite of synthesis of beta-1,3-glucan, such as glucose-6-phosphate isomerase, phosphoglucomutase, B-1,3-glucan synthase and glucokinase, and we also identified uracil phosphoribocyl transferase and Ribosome recycling factor also.

Micromonospora sp. SA-246 균주가 생산하는 Isochromanequinone계 항생물질

  • In the course of screening for new bioactive compounds from oligotrophs in soil, a microorganism, designated as SA-246 and now identified as Micromonospora sp., has been shown to produce a strong antibacterial compound. The active compound was purified from broth filtrate by ethylacetate extraction, silica gel column chromatography, preparative TLC and HPLC, and was identified as crisamicin A based on mass and NMR spectral data. The compound SA- 246 exhibited not only strong antibacterial activity against Gram-positive bacteria but also cytotoxicity against cancer cell lines such as A549 (lung), SK-OV-3 (ovarian), SK-MEL-2 (melanoma), XF498 (central nervous system) and HCT15 (colon).

  • This study analyzed children's personality types with the use of the Murphy-Meisgeier Type Indicator for Children(MMTIC: adapted for Korea by Kim, 1993) and examined their relationship to the Torrance Tests of Creative Thinking(TTCT-figural, form A) and the Creative Personality Test(CPT: Yim,1997). Subjects were 210 third- and 218 sixth-grade students. Data were analyzed by one-way ANOVA and t-test. Personality types Sensation/Judgment(SJ), Sensation/Perceiving(SP), iNtuition/Thinking(NT) and iNtuition/Feeling(NF) differentiated the eleboration, originality, and flexibility dimensions of creative thinking. There were gender differences in the relation of personality type NF to elaboration and fluency. There were age differences in personality types SP and NF in relation to elaboration. Personality types differentiated between the independence, openness, attachment and adventurousness dimensions of the CPT. Age differences in personality type SP differentiated independence and type NT differentiated attachment dimensions of creative personality.

  • Dehydroaltenusin (1) was isolated from the chloroform extract of the culture filtrate of a Streptomyces sp. and its structure was determined from spectral data as well as by comparison with published values. In an XTT-based in vitro anti-HIV assay, dehydroaltenusin effectively inhibited the cytopathic effects of HIV infection at a concentration of $1-5\;{\mu}g/mL$.

진도견 정세관의 정세포와 Sertoli 세포내 glycogen의 분포 (Glycogen distribution of germ cells and Sertoli cells of seminiferous tubules in Jindo dog)

  • In an effort to obtain basic data of carbohydrate metabolism during spermiogenesis of the sexually-matured Jindo dog, the glycogen distribution in the testis was investigated by light and transmission electron microscopy. Periodic acid thiocarbohydrazide silver proteinate physical development(PA-TCH-SP-PD) staining method provided better results in the detection of glycogen granules from Sertoli cells and germ cells than the periodic acid schiff(PAS) staining method did. Pre-treatment of the tissue sections with ${\alpha}$-amylase elicited a significant decrease in PA-TCH-SP-PD stained granules, which suggested that the stained granules were of glycogen origin. High concentration of the glycogen granules were observed in the Sertoli cells, especially in its column, sheet-like processes, club-like processes, and tubular processes. The glycogen granules were unevenly distributed in some Sertoli cell columns. These results strongly indicated that the Sertoli cells of Jindo dogs showed vigorous activity of carbohydrate metabolism.

  • The taxonomic position of actinomycete strain SCNU5$\^$T/, isolated from extensive foam in the aeration basin of an activated sludge process, was clarified by phenotypic, chemotaxonomic and phylogenetic analyses. The strain possesses wall chemotype IV, MK-9(H$\^$0/), as the major menaquinone, and contains saturated, monounsaturated and 10-methyl branched fatty acids. The G+C content of its DNA is 68.1 mol%. Phenotypic data and DNA relatedness to known species indicate that the strain SCNU5$\^$T/ represents a new species within the genus Tsukamurella, for which we propose the name Tsukamurella sunchonensis SP. NOV. The type Strain Of T. sunchonensis is SCNU5$\^$T/ (=KCTC 9827$\^$T/).

Scalarane-type Sesterterpenes from the Philippines Sponge Hyrtios sp.

  • The marine sponge Hyrtios sp. collected in the Philippines was extracted and partitioned. The resulting organic layer was purified by C18 reversed-phase column chromatography and HPLC to achieve the separation of nine scalarane-type sesterterpenes, including one new compound with eight known scalarane analogs. The chemical structures of the isolated compounds 1-9 were elucidated by 1D and 2D NMR and MS data analysis. All nine compounds were evaluated for their antibacterial activities against three Gram-positive and three Gram-negative bacteria. The compound 3 exhibited potent antibacterial activities against Bacillus subtilis and Micrococcus luteus. The compounds 7 and 9 displayed considerable activities against Bacillus subtilis and the others had moderate results.
