Prediction of Listeria monocytogenes Growth Kinetics in Sausages Formulated with Antimicrobials as a Function of Temperature and Concentrations

  • This study was conducted to develop a model to describe the effect of antimicrobials [potassium sorbate (PS), potassium lactate (PL), and combined PL and sodium diacetate (SDA, PLSDA)] on the growth parameters of Listeria monocytogenes such as specific growth rate (SGR) and lag phase periods (LT) in air-dried raw sausages as a function of storage temperature (4, 10, 16, and $25^{\circ}C$). Results showed that the SGR of L monocytogenes was dependent on the storage temperature and level of antimicrobials used. The most effective treatment was the 4% PLSDA, followed by the 2% PLSDA and 4% PL and 0.2% PS exhibited the least antimicrobial effect. Increased growth rates were observed with increasing storage temperatures from 4 to $25^{\circ}C$. The growth data were fitted with a Gompertz equation to determine the SGR and LT of the L. monocytogenes. Six polynomial models were developed for the SGR and LT to evaluate the effect of PS (0.1, 0.2%) and PL (2,4%) alone and PLSDA (2, 4%) on the growth kinetics of L. monocytogenes from 4 to $25^{\circ}C$.

Growth Performance of Juvenile Red-spotted Grouper (Epinephelus akaara) ♀ × Giant Grouper (E. lanceolatus) ♂ Hybrid across Temperatures

  • The present study measured the growth performance of juvenile hybrid of red-spotted grouper ♀ × giant grouper ♂ (RGGG) across four water temperatures (19, 23, 27, and 31℃) and compared it to those of maternal purebreds (red-spotted grouper ♀ × ♂, RG) for eight weeks. The specific growth rate (SGR) and weight gain (WG) of RGGG increased as temperature increased, with greater SGR and WG at higher temperatures (27 and 31℃) than RG. The condition factor of RGGG was higher than that of RG and there were no differences between temperature groups within breeding lines. Food consumption increased at higher temperatures for both RGGG and RG, and was the highest in the 31℃ group. Similar to the SGR and WG, the food conversion rate (FCR) in RGGG decreased with increasing water temperature, with the significantly lowest value at 31℃; in RG, however, FCR progressively decreased in the 27℃ group, then increased in the 31℃ group. Furthermore, the FCR of the 31℃ group of RGGG, which had the lowest value among the RGGG groups, was lower than that of the 27℃ group of RG, which had the lowest RG value. In the analysis of nonlinear regression curves within the range of experimental temperature, the optimum temperature for SGR and FCR in RGGG was 31℃, unlike in RG. In conclusion, the growth performance of juvenile RGGG seems to have largely improved from the influence of paternal traits, hybrid with giant grouper, and it is presumed to be more profitable for commercial production in tropics/subtropics.

Methods and Applications to estimate the Conversion Factor of Resource-based Relative Value Scale for Nurse-Midwife's Delivery Service in the National Health Insurance (조산원(助産院)의 분만간호서비스에 대한 건강보험수가 산출방법과 적용방안)

  • Purpose: This paper analyzed alternative methods of calculating the conversion factor for nurse-midwife's delivery services in the national health insurance and estimated the optimal reimbursement level for the services. Methods: A cost accounting model and Sustainable Growth Rate (SGR) model were developed to estimate the conversion factor of Resource-Based Relative Value Scale (RBRVS) for nurse-midwife's services, depending on the scope of revenue considered in financial analysis. The data and sources from the government and the financial statements from nurse-midwife clinics were used in analysis. Results: The cost accounting model and SGR model showed a 17.6-37.9% increase and 19.0-23.6% increase, respectively, in nurse-midwife fee for delivery services in the national health insurance. The SGR model measured an overall trend of medical expenditures rather than an individual financial status of nurse-midwife clinics, and the cost analysis properly estimated the level of reimbursement for nurse-midwife's services. Conclusion: Normal vaginal delivery in nurse-midwife clinics is considered cost-effective in terms of insurance financing. Upon a declining share of health expenditures on midwife clinics, designing a reimbursement strategy for midwife's services could be an opportunity as well as a challenge when it comes to efficient resource allocation.

Effect of the Dietary EPA in Replacement Diets for the Culture of Brackish Flea, Diaphanosoma celebensis (기수산 물벼룩의 배양을 위한 대체 먹이원 내 EPA 영향)

  • We investigated the effect on dietary EPA (eicosapentaenoic acid) source (EP) into formulated diets for growth of brackish flea, Diaphanosoma celebensis. The highest density and specific growth rate (SGR) of brackish fleas were observed on Tetraselmis suecica (TE) trial, but these of trial were not significantly differed with that of Chlorella (CH) + Crypthecodinium sp. (CR) + EP trial, contained EPA oil (P>0.05). Contrastively, CH trial showed the lowest SGR. And in the RNA/DNA ratio, 0.08 of TE trial was the highest ratio out of whole trials, but the trial not significantly differed with that of CH + CR + EP trial (P>0.05). But, the ratio of CH trial was the lowest ratio at 0.05 out of whole trials (P<0.05). Through out results, the EPA source for flea culture was showed a positive effect through their growth and SGR. Therefore, we suggested that a usage of the source with CH could replace T. suecica as good diet for culture of the brackish flea.

Methods and Estimates of the Reimbursement for the Nurse Midwifery Center in the National Health Insurance (조산원의 건강보험수가 산출방법과 추계)

  • Purpose: The purpose of this study is to develop the optimal nursing fee for nurse-midwifery center (MC) in the national health insurance system. Methods: The three methodologies used to calculate the conversion factors for the MCs in the national health insurance include cost accounting method, sustainable growth rate (SGR) model, and index model. In this study, the macro-economic indicators and the national statistics were used to estimate the conversion factors for the MCs. Results: The optimal nursing fee for the MCs in 2011 was estimated to be an increase of 57.7% by cost accounting analysis, a decrease of 17.1% by SGR model, and a decrease of 16.1% by index model. The results from SGR model and index model could had been biased due to the upswing of medical spendings in the short-term period (2008~2009). A sensitivity analysis of pre-delivery subsidy program for OB & GYN hospitals and clinics showed that the program has substantially diminished the demand for the MC services. Conclusion: More reliable methodologies to estimate nursing fees precisely are required to prove the value of nurses' services and a government subsidy program for the MC services should be followed from a social perspective.

Effects of Temperature, pH, and Potassium Lactate on Growth of Listeria monocytogenes in Broth

  • A total of 60 growth curves were generated with combinations of temperature, pH, and potassium lactate (PL) (60% (v/v) commercial solution) to determine the lag time (LT) and specific growth rate (SGR) of L. monocytogenes in broth. LT and SGR were significantly (P<0.05) affected by temperature, pH, concentration of PL, or the combined interaction of these factors. LT was extended and SGR was reduced significantly (P<0.05) by increased concentration of PL at lower temperature and pH. Listericidal effect was observed in the broth containing 2, 3, and 4% PL at pH 5.0 and $4^{\circ}C$. The antimicrobial activity of PL against L. monocytogenes increased when the pH of the medium was decreased at all temperatures tested. The results suggest that PL has antimicrobial properties to suppress the growth of L. monocytogenes. Potassium lactate has many potential applications as an antimicrobial additive in variety of refrigerated ready-to-eat foods.

OBSERVATIONS OF $HC_3N$ TOWARD THE SGR B2 MOLECULAR CLOUD

  • We have observed the 10-9 transitions of $HC_3N$ and its $^{13}C$ substitutes ($H^{13}CCCN,\;HC^{13}CCN$, and $HCC^{13}CN$), and the vibration ally excited 12-11 ($v_r=1$) $HC_3N$ transition toward the Sgr B2 molecular cloud. The observed $HC_3N$ emission shows an elongated shape around the Principal Cloud ($\~$4.5 pc in R.A. $\times$ 7.4 pc in Decl.). The optically thin $H^{13}CCCN$ line peaks around the (N) core and we derive the total column density $N(H^{13}CCCN) = 4 {\times}10^{13} cm^{-2}$ at this position. Toward the 2' N cloud which shows the peculiar chemistry, the $HC_3N$ lines show enhancements compared to the extended envelope. The shocks of the 2' N may have resulted in the enhancement of $HC_3N$. The hot component of $HC_3N$ is strongly concentrated around the (N) core and its HPW is $\~$0.9 pc in diameter. We derive the lower limit of the abundance ratio $N(HC_3N)/N(H^{13}CCCN)$ to be larger than 40 in most regions except the (M) and (N) cores. The fractionation processes of $^{13}C $at this region may not be as effective as previously reported.

KEY SCIENCE OBSERVATIONS OF AGNs WITH THE KaVA ARRAY

  • KaVA (KVN and VERA Array) is a new combined VLBI array composed of KVN (Korean VLBI Network) and VERA (VLBI Exploration of Radio Astrometry). Here, we report the following two issues. (1) We review the initial results of imaging observations of M87 at 23 GHz following Niinuma et al. (2014). The KaVA images reveal extended outflows including complex substructures such as knots and limb-brightening, in agreement with previous VLBI observations. KaVA achieves a high dynamic range of ~1000, more than three times better than that achieved by VERA alone. (2) Based on subsequent observations and discussions led by the KaVA AGN SubWorking Group, we set monitoring observations of Sgr $A^{\ast}$ and M87 as our Key Science Project (hereafter KSP) because of the closeness and largeness of their central super-massive black holes. The main science goals of the KSP are (i) testing the magnetically-driven-jet paradigm by mapping velocity fields of the M87 jet, and (ii) obtaining tight constraints on physical properties of the radio emitting region in Sgr $A^{\ast}$. Towards KSP, we show the first preliminary images of M87 at 23 GHz and Sgr $A^{\ast}$ at 43 GHz with the bandwidth of 256 MHz.

Estimating the Reimbursing Price Level of Oriental Medical Services in the National Health Insurance (한방의료서비스의 건강보험수가 산출방법과 추정)

  • Objectives : This paper analysed the alternative methods of calculating conversion factor for oriental medicine in the National Health Insurance and estimated the conversion factor(reimbursing price level) of the oriental medical services, based on health insurance claims data and macro economic data. Methods : Comparing cost accounting method, SGR model, and index model to estimate conversion factor in the national health insurance, six empirical models were derived depending on the scope of revenue considered in financial indicators. Classifications of data and sources used in the analysis were identified as officially released by the government. Results and Conclusion : Cost accounting analysis and SGR model showed a two digit decrease in the physician fee schedule of oriental medical services in the national health insurance, while index model indicated a positive increase in the fee reimbursed. As expected, SGR model measured an overall trend of health expenditures rather than an individual financial status of medical institutions, and index model properly estimated the level of payments to oriental medical doctors. Upon a declining share of health expenditures on oriental medicine, a global budget system fixed to a flat rate of total budget could be an opportunity as well as a challenge.

  • Celebes rainbow (Marosantherina ladigesi) is one of Indonesia's exported ornamental fish commodities, but the exploitation of this fish only relies on wild catches. The rise of unlimited fishing, especially those using poison, has changed the aquatic environment, threatening sustainability and causing fish extinction. This study aimed to evaluate the effectiveness of several types of feed in improving the absolute growth rate (AGR), specific growth rate (SGR), survival rate (SR), feed conversion ratio (FCR), feed efficiency (FE), hematology, and immune response of Celebes rainbow. The fish used in this study were male ornamental Celebes rainbow (M. ladigesi) weighing 1.32 ± 0.21 g/ind, reared in 54 L-aquariums at a stocking density of 30 individuals/aquarium for six weeks. The fish were fed according to the test diet consisting of live Tubifex sp worms, dry Tubifex sp worms, Spirulina platensis, and crumble pellets. The parameters observed were AGR, SGR, SR, FCR, FE, hematology, intestinal histology, liver histology, and a challenge test with the pathogenic bacteria Aeromonas hydrophila. The results showed that fish-fed live Tubifex sp worms had better AGR, SGR, SR, FCR, FE, hematology, and disease resistance compared to all other treatments. These results indicate that live Tubifex sp worms are the best feed for rearing Celebes rainbow.
