Melanogenesis Inhibition Effects of Nemopilema nomurai Hydrolized Extracts (노무라입깃해파리 가수분해 추출물의 멜라닌합성 저해효과)

  • This study was aimed to investigate skin whitening effects of Nomura's jellyfish (Nemopilema nomurai) hydrolyzed extracts (NHE). The extracts were prepared through the hydrolysis of N. nomurai using commercial proteolytic enzymes such as Protamex, Alcalase, Flavourzyme and Neutrase with optimum pHs (pH 5-8) at $55^{\circ}C$. Their whitening activities were examined from the inhibition of melanin synthesis using B16-F1 cell lines. Among the examined samples, Neutrase-treated extract (N-NHE) showed the most significant inhibition effect on melanin synthesis by 89.9% at a concentration of $100{\mu}g/mL$. This sample decreased the expression of tyrosinase and TRP-1 (tyrosinase related protein-1) proteins as well. These results suggested the potential application of NHE as whitening ingredients in cosmetic preparation.

Effects of Chromium Picolinate (CrP) on Growth Performance, Carcass Characteristics and Serum Traits in Growing-Finishing Pigs

  • An experiment was carried out to evaluate the effects of feeding graded levels of chromium in the form of chromium picolinate on growth performance, blood components, carcass grade, in vitro lipogenesis and lipolysis, and in vitro protein degradation and synthesis in growing-finishing pigs. There were no significant differences for daily weight gain, feed intake and feed conversion among treatments during growing phase, while in the finishing phase, feed intake was lower in groups fed diets with 200 ppb chromium than in other treatment (p < 0.1). Feed conversion was improved in the groups fed diets with chromium compared with control. Carcass weight was similar among treatments while carcass length was longer in groups fed diets with 200 ppb chromium (p < 0.05). Thinner carcass fat was found with groups fed diets with chromium compared to control. Three A grade of carcasses were from groups fed chromium compared to control. No significant differences were observed with blood glucose, insulin, total cholesterol, triglycerides and non-esterified fatty acid at 60kg body weight. While, at 100 kg body weight, blood triglyceride was lower in groups with 200 and 400 ppb chromium but higher in groups with 100 ppb chromium (p < 0.05). In vitro lipolysis and protein synthesis in adipose tissues were increased as dietary chromium was increased from 0 to 200 ppb (p < 0.1). As a result, 200 ppb chromium in a growing-finishing diet could improve feed efficiency and carcass traits; an increase to 400 ppb has no further effect.

  • The protease produced by Bacillus subtilis YG-95 was purified by precipitating with ammonium sulfate, DEAE-sepharose 6B and Sephadex G-100 column chromatogtaphies and its purified enzymological characteritics were investigated. The molecular weight of purified protease was estimated about 43kilodalton by SDS PAGE The optimum pH and temperature for the purified protease activity were pH 10.0 and $55^{\circ}C$, respectively. The enzyme was stable in broad range of pH 5.0 to 12.0. and at the below $45^{\circ}C$. The purified enzyme activty was inhibited by $Fe^{3+}$ and $Al^{3+}$. The activity was significantly inhibited more than 80% by O-Phenanthroline, PMSF and SDS. The $K_m$ value of the purified enzyme against Soy Protein Isolate as a substrate was 1.28 mg/ml.

  • This pot experiment was conducted to investigate the effects of combined micronutrient application($T_1$;control, $T_2$; Fe, $T_3$; Fe+Mn, $T_4$; Fe+Mn+Cu, $T_5$ ; Fe+Mn+Cu+Zn, $T_6$ ; Fe+Mn+Cu+Zn+Mo, $T_7$ ; Fe+Mn+Cu+Zn+Mo+B) on forage performance of pure and mixed cultures of orchardgrass and white clover. The third part was concerned with the changes in the contents and yields of N-compounds (crude/pure protein and soluble N-compounds) in forages. The results obtained are summarized as follows: 1. The contents of N-compounds(crude/pure protein and soluble N-compounds) were generally different according to the forage species, whether it was a pure or mixed culture, and additional fertilization, especially N. In orchardgrass, these contents were relatively low at the $T_3$ and $T_6$ in both pure and mixed cultures. In white clover, these contents were relatively decreased by the $T_1$, $T_3$, and $T_6$ in mixed culture. 2. The treatments influenced relatively more on the yields of crude/pure protein than on the dry matter yields of forages, and this tendency was more significant in white clover than in orchardgrass. 3. In white clover, the great differences in the yields of crude protein by the treatments occurred especially in mixed culture and at 5th cut without no additional fertilization. In white clover, the positive effects of optimum treatments on the yields of crude protein seemed to be decreased by the additional fertilization, especially N. In mixed culture, the favorable growth of white clover by the optimum treatments tended to be positively related to the favorable contents and yields of N-compounds. The changes in the yields of pure protein were similar to the tendency of crude protein

  • The laying performance of Japanese quails fed graded levels of high glucosinolate ($92.5{\mu}mole/g$) rapeseed meal (RSM) was assessed. One hundred and twenty Japanese quails aged 1 day-old were assigned at random to four dietary treatments consisting of 0, 50, 75 or 100 g/kg RSM in the diet replacing part of the soybean meal and de-oiled rice bran in a standard quail ration. 12 female representative quails from each diet were selected at random and housed in individual cages from 7-20 wk of age. The egg production, feed intake and FCR was comparable among the different dietary groups. The egg quality characteristics, organoleptic evaluation of boiled eggs as well as the haematological (haemoglobin, total erythrocyte count, total leucocyte count) and biochemical (glucose, protein, cholesterol, aspartate amino transferase, alanine amino transferase and alkaline phosphatase) constituents did not differ significantly among the groups. The gross and histopathological studies of vital organs did not reveal any appreciable changes. The feed cost was reduced by the incorporation of RSM in the diet, but only the production cost of quails fed the 75 g/kg RSM was lower in comparison to other groups. In the present study, the laying potential of Japanese quail was well-maintained up to the 100g/kg dietary level of rapeseed meal.

Effect of Protein and Energy Levels on Performance and Carcass Rate in Cross Bred Chicks (단백질 및 대사 에너지 수준이 유색 육용계의 생산성 및 도체율에 미치는 영향)

  • The experiment were protein and metabolizable energy level in performance and carcass rate in cross bred chicks. One day old, 720 male and female cross bred chick were used for the experiments, and 20 chicks were placed at each pen. The energy level of feed was maintained about 2,950, 3,000, 3,050 kcal/kg, and protein content was adjusted about 19, 20, 21% from 0 to 5 weeks and energy level was maintained about 3,100, 3,150, 3,200 kcal/kg, and protein content was adjusted about 17, 18, 19% from the 6 to 10 weeks old of the experiment. At the whole time, ME 3,100 (2,950), 3,150 (3,000) and 3,200 (3,050) kcal/kg treatment groups was not significant difference in weight gain, feed intake, but 3,150 (3,000), 3,200 (3,050) kcal/kg treated groups revealed to show improved feed conversion rate than the group treated with ME 3,100 (2,950) kcal (P<0.05). And CP 17 (19), 18 (20), 19 (21)% treatment groups did not significant difference weight gain and feed intake but CP 19 (21)% treated group wan significant difference feed conversion rate than the CP 17 (19)% treated group (P<0.05). ME level of 3,100 (2,950), 3,150 (3,000), 3,200 (3,050) kcal/kg treatment groups in carcass rate and abdominal fat rate was not signficant difference and protein of 17 (19), 18 (20), 19 (21)% treatment groups after end of experiment. Between ME and CP of dietary was significant appeared to interact at 6~10 weeks (P<0.05).

Effect of Sodium Bicarbonate Injection in Pre-rigor Porcine M. Longissimus lumborum on Pork Quality

  • Effects of sodium bicarbonate (SBC) injection on meat quality and functionality of porcine M. longissimus lumborum were investigated. Fifteen pigs ($100{\pm}5kg$) were randomly selected at a commercial slaughter plant. After slaughtering the loins were dissected from the carcass before chilling at approximately 30 minutes post mortem. The loins were divided into four cuts for sample treatment, and SBC of 0.25 M, 0.40 M and 0.75 M was injected (2% w/w) using a syringe. As SBC injection level was increased, muscle pH increased significantly (p<0.05). SBC injection decreased lightness ($L^*$) values on the surface of muscle. Moreover, with injection of SBC, drip loss %, cooking loss % and shear force were significantly (p<0.05) decreased, whereas WHC and $Na^+$ content were significantly (p<0.05) increased. From panel testing of uncooked pork loin, no significant differences (p>0.05) were found in aroma, off-flavor and drip between injection of SBC at different levels and the control, although color and acceptability were significantly lower (p<0.05) in control pork loin compare with injection of SBC at 0.75 M. In cooked pork loin from the panel test, aroma, flavour, off-flavour and juiciness were found to be similar (p>0.05) on all treatments, but tenderness and acceptability were significantly higher (p<0.05) with injection of SBC at 0.75 M than for control loin. Myofibrillar protein solubility of muscles treated with SBC was significantly (p<0.05) higher than that of the control, although no significant differences (p>0.05) were found in sarcoplasmic protein solubility between the treatments. These results indicated that SBC injection into pre-rigor porcine M. longissimus lumborum could improve ultimate pork quality characteristics such as meat color, water-holding capacity, and could inhibit muscle protein denaturation due to an increase in muscle pH.

  • The precise mechanism underlying the therapeutic efficacy of an extraction powder of Angelica gigas (AGE) for the treatment of degenerative osteoarthritis was investigated in primary cultured rabbit chondrocytes and in a monosodium-iodoacetate (MIA)-induced osteoarthritis rat model. The treatment with AGE (50 μg/mL) effectively inhibited NF-B activation. The anti-inflammatory mechanism was clarified by gelatin zymography and western blotting measurements of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) activities. The AGE (50 μg/mL) treatment significantly reduced MMP-9 activity. The constituents of AGE— decursinol, decursin, and decursinol angelate—were determined by LC-MS/MS after a 24 hr treatment of rabbit chondrocytes. The contents of the major products, decursin and decursinol angelate, were 3.62±0.47 and 2.14 ±0.36 μg/mg protein, respectively in AGE-treated (50 μg/mL) rabbit chondrocytes. An in vivo animal study on rats fed a diet containing 25, 50, and 100 mg/kg AGE for 3 weeks revealed a significant inhibition of the MMPs in the MIA-induced rat articular cartilage. The genetic expression of arthritic factors in the articular cartilage was examined by RT-PCR of collagen Type I, collagen Type II, aggrecan, and MMP (MMP3, MMP-9, MMP13). Specifically, AGE up-regulated the expression of collagen Type I, collagen Type II, and aggrecan and inhibited MMP levels at all tested concentrations. Collectively, AGE showed a strong specific site of action on MMP regulation and protected against the degeneration of articular cartilage via cellular regulation of MMP expression both in vitro and in vivo.

Dietary effect of Bacillus subtilis MD-02 on Innate Immune Response and Disease Resistance in Olive Flounder, Paralichthys olivaceus (넙치(Paralichthys olivaceus)의 비특이적 면역반응 및 병 저항성에 대한 Bacillus subtilis MD-02의 효과)

  • Among several marine-derived microorganisms isolated from the coast of Jeju Island that had antimicrobial activity against fish disease pathogens, Bacillus subtilis MD-02 was tested for its dietary effect on the innate immune response and disease resistance of olive flounder. Strain MD-02 was fed to the olive flounder at a concentration of $1.2{\times}10^4$, $1.2{\times}10^6$, or $1.2{\times}10^8CFU/100g$, respectively. Consequently, the hematocrit was higher in these three groups than that in the control group at 4 weeks, and the aspartate aminotransferase and alanine aminotransferase levels were decreased in the $1.2{\times}10^8$ and $1.2{\times}10^4CFU/100$ groups compared with the control group levels. The amylase activity and total protein were significantly increased in the $1.2{\times}10^4CFU/100g$ group at 3 weeks. The innate immune response, determined from the lysozyme and macrophage activities, was higher in the $1.2{\times}10^8CFU/100g$ group than in the control group. In addition, treatment of the olive flounders with Streptococcus parauberis at $1.2{\times}10^6CFU/ml$ confirmed the mortality rate, which was 100% in the control group and 40-60% in the groups fed B. subtilis MD-02, indicating that the fish had resistance to fish disease pathogens. Therefore, it was confirmed that when fed MD-02, olive flounder builds an innate immune response and acquires resistance to fish disease pathogens, indicating that B. subtilis MD-02 can be developed as a beneficial feed additive.

Effect of Exocytosis Factor on Spontaneous Zona Pellucida Hardening during in Vitro Culture of the Mouse Oocytes (생쥐 난자 배양시 외분비 관련 요소들이 자발적 투명대 경화 현상에 미치는 영향)

  • "Spontaneous" hardening of the zona pellucida of mouse oocytes during in vitro culture is most likely due to cortical granules exocytosis. Thus the purpose of the present study was to determine whether the exocytosis factor is involved in spontaneous zona pellucida hardening during in vitro culture of the mouse. The results obtained form these experiments were summarized as follows; 1. When a protein synthesis inhibitor(100${\mu}g$/ml puromycin) was added to the culture medium, it did not prevent spontaneous ZPH of mouse oocyte during in vitro culture. 2. Calmodulin antagonists (trifluoperazine and chlorpromazine) and calcium channel blocker (verapamil) had no inhibitory effect in spontaneous ZPH. 3. A microtubule assembly inhibitor, colcemid had some inhibitory effect on spontaneous ZPH. 4. Treatment with a microfillament formation blocker(cytochalasin-B) at 1${\mu}g$/ml concentration, resulted in the excellent inhibitory effect on spontaneous ZPH. However cytochalasin-B did not inhibit ethanol-induced ZPH.
