  • This paper examines the effects of IT technology capital and R&D stock's variation on the growth of Korea's industries with the empirical approaches. We analyze the Granger causality and Impulse response function analysis among the Korea's industrial real output, IT technology capital, and R&D stocks. When it comes to this research conclusion, we know that IT technology capital and R&D stock's shocks affect the growth of Korea's industrial sector in terms of increasing in the real output growth rate.

  • We are made up to the variation of the structure modeling of the stratum corneum by the skin on the hydration. The given skin structure is consist of the control level of the intercellular route and transcellular route, and is confirmed to the variation of the modeling for algorithm. Hydration condition is consisted to modeling by the size, form and combination, that skin impedance is appeared to result the value of measurement by the several layer such as forms of ${\xi}-R-SC-RH$, ${\xi}-R-SL-RH$, ${\xi}-R-SG-RH$, ${\xi}-R-SS-RH$ and ${\xi}-R-SB-RH$. Hydration condition is showed to the alteration difference value of S-Corneum, S-Lucidum, S-Granulosum, S-Spinosum and S-Basale. And, composition condition was constructed with the alteration modeling that was established to separate the division parts for conversion system. We will be possible to progress the improvement effectiveness of the skin and to consist of the continuous hydration control system for functional on the skin.   • Restriction fragment length polymorphism (RFLP) of a DNA has been a useful tool for analyzing genetic variation. This research was performed to establish an RFLP analytic method on the mitochondrial DNA (mtDNA) of the beet armyworm, Spodoptera exigua (Hiibner). To do this, total size of the mtDNA was measured and polymerase chain reaction (PCR) primers were selected. Its mitochondrial genome size was ca. 16kb. From a serial PCR test of 29 primers refered to the compilation of Simon et al. (1994), 22 primers were selected to amplify its mtDNA fragments. These primers resulted in short (300-700 bp) or long (1000-2000 bp) DNA products which represented a total or partial sequence of each of CO-I, CO-11, Cyt-B, ND-1, 12s rRNA, 16s rRNA, and some tRNAs. PCR-RFLP was performed in some variable mtDNA regions with 8 kinds of 4bp recognizing restriction enzymes. Different populations from Andong, Kyungsan, and Sunchun did not show any restriction site polymorphisms but had some length variation in certain regions of mtDNA.

An Analysis of Genetic Variation and Divergence on Silk Fibre Characteristics of Multivoltine Silkworm (Bombyx mori L.) Genotypes

  • The nature of genetic variation and diversity among the 65 multivoltine silkworm genotypes was evaluated for 16 post cocoon characters. The components of genetic variation revealed higher PCV (60.487%) and GCV (44.56%) for evenness (variation 1) followed by cohesion (PCV=55.38%, GCV=40.36%) and non-broken filament length (PCV=32.05%, GCV=31.28%). The higher heritability ($h^2$ in broad sense) was observed for boil-off loss (95.6%) followed by non-broken filament length (95.22%). The both genotypic and phenotypic correlation indicated significant positive correlation of filament length with non-broken filament length, silk recovery, raw silk, neatness, and low neatness; and negative correlation with denier, renditta and silk waste. The principal component analysis (PCA) revealed 75.381 % of total variance from the five principal components extracted. On the basis of Mahalonobis' $D^2$ values (Ward's minimum variance), the sixty-five multivoltine silkworm genotypes were classified in to 9 clusters with substantial inter and intra cluster distances. Number of genotypes included in different clusters varied from 3 to 17. The results indicated that the optimum distance obtained in cluster VII (15.059) along with higher cluster mean values especially for filament length, non broken filament length, renditta, silk recovery, silk waste, and raw silk emphasized the utilization of these genotypes in the conventional silkworm breeding programme for improvement of multivoltine silk fibre quality. The possibility of exploiting genetic variation in post cocoon traits for efficient breeding programme is discussed.

Tissue Specific Gene Regulation of The Anthocyanin Synthesis Regulator Gene R in Maize (옥수수의 색소 발현에 관련된 조직 특이성 조절유전자 R locus에 관하여)

  • The R locus of maize in one of several genes that regulate the anthocyanin pigments throughout the body of the plant and seed. The R gene product may regulate pigment deposition by controlling the expression of the flavonoid biosynthetic gene pathway in a tissue-specific manner. To understand the basis for tissue specific regulation and allelic variation at R, the molecular study has been done by cloning a portion of the R complex by transposon tagging with Ac. R specific probe were cloned from the R-nj mutant induced by Ac insertion mutagenesis. From southern analysis of R-r complex using the R-nj probe, the structure of R-r was proposed that R-r containes the three elements, (P)(Q)(S). These elements may organize as the inversion triplication model which (S) sequence was inverted in relation to (P) and (Q). The R-sc derivated from R-mb or R-nj was cloned with R-nj probe, and molecular genetical data showed that R-sc containes tissue specific and tissue nonspecific area, and the sequencing of R-sc are progressed now.

Localization of 5S and 25S rRNA Genes on Somatic and Meiotic Chromosomes in Capsicum Species of Chili Pepper

  • The loci of the 5S and 45S rRNA genes were localized on chromosomes in five species of Capsicum, namely, annuum, chacoense, frutescens, baccatum, and chinense by FISH. The 5S rDNA was localized to the distal region of one chromosome in all species observed. The number of 45S rDNA loci varied among species; one in annuum, two in chacoense and frutescens, and chinense, and four in baccatum, with the exceptions that 'CM334' of annuum had three loci and 'tabasco' of frutescens gad one locus. 'CM334'-derived BAC clones, 384B09 and 365P05, were screened with 5S rDNA as a probe, and BACs 278M03 and 262A23 were screened with 25S rDNA as a probe. Both ends of these BAC clones were sequenced. FISH with these BAC probes on pachytenes from 'CM334' plant showed one 5S rDNA locus and three 45S rDNA loci, consistent with the patterns on the somatic chromosomes. The 5S rDNA probe was also applied on extended DNA fibers to reveal that its coverage measured as long as 0.439 Mb in the pepper genome. FISH techniques applied on somatic and meiotic chromosomes and fibers have been established for chili to provide valuable information about the copy number variation of 45S rDNA and the actual physical size of the 5S rDNA in chili.

Analysis of Bacterial Community Structure in Gossi Cave by Denaturing Gradient Gel Electrophoresis (DGGE) (DGGE를 이용한 동굴 생태계 세균 군집 구조 분석)

  • The bacterial community of water stream, soil and guano in Gossi cave was examined by using PCR amplified the 16S rDNA-denaturing gradient gel electrophoyesis (DGGE). In this study, the genetic diversity and the similarity of bacterial community between open area and non - open area toy cave tour were investigated, and the seasonable variation pattern was compared each other. DGGE is attractive technique, as it sepayate same length dsDNA according to sequence variation typical 16S rDNA genes. The diversity and similarity of bacterial community in cave was analyzed by GC341f and PRUN518r primer sets foy amplification of V3 region of eubacteria 16S rDNA. The specific DGGE band profile of the cave water gives the possibility that the specific bacterial cell can be adapting to the specific cave environment and living in the cave. The DGGE band profiles of all samples with guano were compared and analyzed by image analyzer, in which mutual band profile was compared to be and the band intensity of guano was the highest. From these result, it is thought that the guano was main nutrient source and influenced on the community structure of the cave environment where is nutritionally limited. Pseudomonas sp. NZ060, Pseudomonas pseudoalcaligenes, uncultured Variovorax sp. and soli bacterium NS7 were identified to be on some sample from analysing DNA sequence of some DGGE band.

Measurements of Diameter Variation in Optical Fiber-Core. (광섬유 코어 Diameter-Variation 측정에 관한 연구)

  • The principal reason for the backscattering in an optical fiber is the Rayleigh scattering which is caused by non-uniform density of structure material of an optical fiber and diameter variations of the optical fiber-core along an optical fiber axis. The backscattering signal which is detected by the optical time domain reflectometer system(O.T.D.R) conatains information about both tha actual decay of power and the diameter variation along the optical fiber. In this paper, the O.T.D.R. system with 2x2 fiber directional coupler, timing control unit and gated integrator is used to measure diameter variations of an optical fiber.
