Effects of pine Bark Spent Liquor Prepared by Alkaline Sulfite-Anthraquinone Cooking as a Pellet Binder on Pellet Durability and Performance of Broiler Chicks or Laying Hens (소나무 수피 알칼리성 아황산염-안트라퀴논 증해폐액이 사료결착제로서 펠렛 내구성과 가금의 생산성에 미치는 영향)

  • Three experiments were conducted to investigate the Pine Bark Spent Liquor (PBSL) inclusion, prepared by alkaline sulfite-anthraquinone cooking, on Pellet durability index (PDI) of practical diets and performance of broiler chicks and laying hens. Fourteen treatments with four replications were assigned for PDI test in Experiment 1. Control, 10% Wheat(W10), 20% Wheat(W20), 0.25, 0.50, 1.00, 2.00 and 3.00% PBSL, 0.1, 0.2% commercial pellet binder A(CPB A) or B(CPB B), W10 plus 0.1% CPB A or B were used for PDI test. PDI was measured by PDI tester (Oriental Motors, Japan). The control diet was based on corn and soybean meal with no wheat or pellet binders inclusion. The PDI of the PBSL or other commercial pellet binder treatments were significantly higher than control groups(P<0.05). It was shown 95.9, 95.9, 95.8, and 95.7 in W10, 0.5% PBSL, 0.2% CPB A or B treatments, respectively. Thus, those treatments were applied to Experiments 2 and 3. In experiment 2, 200 male broiler chicks (Cobb$\times$Cobb) were allocated to the control, W10, PBSL 0.5%, 0.2% CPB A and B with four replications. Starter diets contained 3,169, 3,149 kcal/kg ME and 21% CP, and finisher diets were fed at the level of 3,192, 3,172 kcal/kg ME and 19W% CP. Weight gain, feed intake, feed conversion ratio (FCR) were weekly measured for 5 wk and the number of intestinal anaerobes were examined at the end of experiment. The weight gain of chicks fed PBSL was not significantly greater than control groups, but was significant different compared to that of W10 or 0.2% CPB A treatments (P<0.05). FCR of chicks treated with PBSL or other pellet binders tended to improve compared to that of control.

HARDNESS OF COMPOSITE RESIN CURED BY HIGH INTENSITY HALOGEN LIGHT (고강도 할로겐광으로 중합한 복합레진 수복재의 경도)

  • The purpose of this study was to compare the effect of the high intensity halogen light $(850\sim1000mW/cm^2)$ with that of the conventional halogen light $(400mW/cm^2)$ on the hardness of composite resin. Three resin composites (Z-100, 3M, U.S.A. : Tetric Ceram, Vivadent, Liechtenstein; SureFil, Dentsply, U.S.A.) were filed in the stainless steel moulds which were 4mm in diameter and 2, 3, 4, and 5mm in depth, respectively. They were cured under the four different modes : (1) conventional mode, 40 seconds at $400mW/cm^2$; (2) 'ramp' mode, 10 seconds at 100 to $1000mW/cm^2$ plus 10 seconds at $1000mW/cm^2$; (3) 'boost' mode, 10 seconds at $1000mW/cm^2$; and (4) 'standard' mode, 20 seconds at $850mW/cm^2$. The surface hardnesses of the top and the bottom of the resin samples were measured with a microhardness tester (MXT70, Matsuzawa, Japan). The top surface hardness was not significantly different among the curing modes. The bottom surface hardness was generally the highest in the conventional mode and the lowest in the high intensity boost mode. There was no significant difference in the bottom surface hardness between the conventional mode and the high intensity standard mode in 2mm depth. The results suggest that the curing time of the high intensity halogen light $(850mW/cm^2)$ should be at least 20 seconds to produce the equal level of the bottom surface hardness of 2mm resin composite as compared to the hardness produced by the conventional halogen light $(400mW/cm^2)$.

The Effect of Value-added Promotion and Retailer Uncertainty on Customers' Perceived Value of the Product (부가가치 제공 유형별 판매촉진과 유통업체 불확실성이 제품 가치 평가에 미치는 영향 - 사은품(Freebie) 제공 판매촉진을 중심으로 -)

  • The sales promotion strategy can be classified into two categories: cost-down and value-added promotions. Although many studies have been performed on sales promotion in the past, little attention has been given on cost-down promotion, and in particular on its strategy. Also there has been a renewed interest on value-added promotion strategy as means of attracting more customers and delivering value by providing a greater benefit. This study examines the effect of value-added promotion on the evaluation of promotional products. The topic is important because the value-added promotions are not always perceived as a benefit by consumers and they depend on the situational factors. We postulate three hypotheses on the basis of related literature. We conduct two studies: one employing experiment (study 1) and the other, quasi-experiment (study 2) to investigate the effect of two situational factors, namely the promotional package type and retailer uncertainty on the valuation of promotional products. Study 1 employs 2 promotional package types (bundled with freebie type vs. one-plus-one type) by 3 evaluation targets (overall package, a focal product and freebie) in experimental design. Also it is found that consumers devaluate the promotional product when the level of retailer uncertainty is high (t=-4.70, p=.000) as shown in Table 2. As depicted in Figure 2, the interaction effect of retailer uncertainty and package types on the evaluation of promotional product as a whole does not appear to be significant. However, when the level of retailer uncertainty is high, the focal product suffers from lower valuation if it is included in a bundle with freebie type package. The purpose of Study 2 is to cross-check the results of Study 1. The results of Study 2 also show that the consumers devaluate the promotional products as a whole when the level of retailer uncertainty is high. Furthermore, Study2 examines the consumers' willingness to purchase. The willingness to purchase of 2 different consumer groups divided by their reservation prices before and after exposure to a promotional stimuli shows no significant differences(t=1.911, p=.057). The results suggest that the consumers' reservation prices before the exposure to the promotion can be a reference prices of their's. But after the promotional events, the promotional price would become their reference prices. Furthermore, when the level of retailer uncertainty is high, consumers devaluate the promotional product as a whole. Because the promotional offerings activate the persuasion knowledge of consumers and make them incorporate negative inference about the firm's motives into their valuation and consequently, consumers tend to hesitate to purchase.

Cloning and Characterization of Phosphoinositide 3-Kinase γ cDNA from Flounder (Paralichthys olivaceus) (넙치에서 분리된 phosphoinositide 3-kinase γ 유전자의 클로닝 및 특성 연구)

  • Phosphoinositide 3-kinase (PI3K) plays a central role in cell signaling and leads to cell proliferation, survival, motility, exocytosis, and cytoskeletal rearrangements, as well as specialized cell responses, superoxide production, and cardiac myocyte growth. PI3K is divided into three classes; type I PI3K is preferentially expressed in leukocytes and activated by ${\beta}{\gamma}$ subunits of heterotrimeric G-proteins. In this study, the cDNAs encoding the $PI3K{\gamma}$ gene were isolated from a brain cDNA library constructed using the flounder (Paralichthys olivaceus). The sequence of the isolated $PI3K{\gamma}$ was 1341 bp, encoding 447 amino acids. The nucleotide sequence of the $PI3K{\gamma}$ gene was analyzed with that of other species, including Oreochromis niloticus and Danio rerio, and it turned out to be well conserved during evolution. The $PI3K{\gamma}$ gene was subcloned into the expression vector pET-44a(+), and expressed in the E. coli BL21 (DE3) codon plus cell. The resulting protein was expressed as a fusion protein of approximately 49 kDa containing a C-terminal six-histidine extension that was derived from the expression vector. The expressed protein was purified to homogeneity by His-tag affinity chromatography and showed enzymatic activity corresponding to $PI3K{\gamma}$. The binding of wortmannin to $PI3K{\gamma}$, as detected by anti-wortmannin antisera, closely followed the inhibition of the kinase activities. The results obtained from this study will provide a wider base of knowledge on the primary structure and characterization of the $PI3K{\gamma}$ at the molecular level.

IN VITRO EVALUATION OF ACCURACY AND CONSISTENCY OF FOUR DIFFERENT ELECTRONIC APEX LOCATORS (4종 전자근관장측정기의 정확성과 일관성에 관한 in vitro 연구)

  • The purpose of this study was to evaluate the accuracy and the consistency of four different electronic apex locators in an in vitro model. Fourty extracted premolars were used for the study. Four electronic apex locators (EAL) were Root ZX, Smarpex, Elements Diagnostic Unit (EDU), and E-Magic Finder Deluxe (EMF). After access preparation, the teeth were embedded in an alginate model and the length measurements were carried out at '0.5' and 'Apex' mark using four EALs. The file was cemented at the location of the manufacturers' instruction (Root ZX, EDU, EMF: 0.5 mark, SmarPex: Apex mark). The apical 4mm of the apex was exposed and the distance from the file tip to the major foramen was measured by Image ProPlus (${\times}100$). The distance from the file tip to the major foramen was calculated at 0.5 and Apex mark and the consistency of 0.5 and Apex mark was compared by SD and Quartile of Box plots. In this study, Root ZX and EMF located the apical constriction accurately within ${\pm}0.5 mm$ in 100%, whereas SmarPex and EDU located in 90% and in 70% respectively. For Root ZX and EMF, there was no significant difference between the consistency of 0.5 and Apex mark. However, for the EDU and SmarPex, Apex mark was more consistent than 0.5 mark. From the evaluation of the consistency in this study, for Root ZX and EMF, both 0.5 and Apex mark can be used as a standard mark. And for EDU and SmarPex, the Apex mark can be recommended to be used as a standard mark.

  • The 106 buildings of 200 meters' height or greater were completed around the world in 2015 (CTBUH, The Council on Tall Buildings and Urban Habitat). They beat every previous year on record, including the previous record high of 99 completions in 2014. This brings the total number of 200-meter-plus buildings in the world to 1,040, exceeding 1,000 for the first time in history and marking a 392% increase from the year 2000, when only 265 existed. South Korea recorded three completions during 2015 - improving slightly over 2014, in which it had one. This study focused on the fact that high-rise building development project risks have not reduced in Korea in spite of numerous studies and measures. And it attempted to examine whether existing studies and measures have been presented on the basis of the accurate analysis of existing studies and measures and classify and analyze the characteristics of each phase' s risk factors in the hope that its results would be one reference point as to the measure to prevent high-rise building development project risks in the future. A high-rise building development project is the high risk project as compared with the low-rise project. Because a high-rise development project takes long and is very sensitive to the changing environment. Therefore, in order to succeed the project it becomes necessary to effectively manage the risk involved in the process of the high-rise building development project. The result of this study can be used as the guideline to make the risk management system for the high-rise development project.

Effects of Supplemental LED Lighting on Productivity and Fruit Quality of Strawberry (Fragaria × ananassa Duch.) Grown on the Bottom Bed of the Two-Bed Bench System (2단 베드 시스템의 하단부에서 자란 딸기의 생산성 및 과일 품질에 미치는 보광 LED의 효과)

  • The aim of this study was to confirm that effects of supplemental LED illumination on a strawberry yield and fruit quality when strawberry grown on a bottom bed to be deficient ambient light due to shading of a upper bed during cultivation by a two-bed bench system. A strawberry was cultivated as a drip irrigation system in the two-bed bench system filled with a strawberry exclusive media from October 2015 to January 2016. The upper and the bottom bed without LED illumination for growth of a strawberry were using as a control. For LED light treatments, from 10 am to 4 pm, we illuminated LEDs as $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ of light intensity by using blue, red, and mixing LED (blue plus red) on the strawberry plants of the bottom bed. In the yield of strawberry fruit, the strawberry grown on the bottom bed treated with the blue LED significantly increased compared with that of the bottom bed part control, and increased to by near 90% of the strawberry output of the upper bed part control. The soluble sugar content of strawberry fruit grown on the upper bed part control and on the bottom bed illuminated with blue or mixed LED was higher than that of red LED and the control of the bottom bed. The content of anthocyanin was the highest increased in the strawberry grown on the upper bed part control that received a lot of ambient light, however when comparing only the bottom bed, strawberry fruits grown on all LED treatments were higher than that of the control. Therefore, we considered that using of the blue LED light on the bottom bed of two-bed bench system during strawberry cultivation is advantageous for the increase of yield and improvement of fruit quality.

Effects of Mixture Lactic Acid Bacteria and Sea Tangle on Anti-obesity and Gut Microflora in Rats (흰 쥐에서 복합 유산균과 다시마가 항비만 및 장내 미생물에 미치는 영향)

  • The present study was undertaken to investigate the effects of dietary provision of lactic acid bacteria (LB) and sea tangle (ST) on the obesity-associated intestinal microbiota in rats with obesity induced by a high-fat diet. Forty-eight 8-wk-old Sprague-Dawley rats were fed a basal diet (CON), a high fat diet (HFD; CON supplemented with 10% lard), HF supplemented with LB [HFL; $5{\times}10^8cfu$ of each of Lactobacillus rhamnosus, Lactobacillus johnsonii, Bifidobacterium longum and Bifidobacterium lactis], or HFL containing 10% ST (HFLS), with 4 replicates (cages) of 3 rats per dietary treatment, for 6 wk, and the intestinal microbiota were determined by pyrosequencing. The HFL and HFLS groups exhibited reduced rates of weight gain than the HF group, and the former groups had smaller ratios of Firmicutes and greater ratios of Bacteriodetes, with decreased Firmicutes/Bacteroidetes ratios, than the latter at the level of the phylum. Compared with the results for the HF group, HFL and HFLS had reduced ratios of the families of Roseburia, Mollicute, Erysipelotrichi, and Oscillibacter within Firmicutes associated with obesity and increased ratios of the families of Prevotella, Alistipes and Bacteroides within the Bacterioidetes phylum known to have an anti-obesity effect. The content of butyric acid in feces was greater in the HFLS group vs. HF and HFL. In conclusion, the present results suggest that dietary provision of LB plus ST has an anti-obesity effect and induced changes in intestinal microorganisms, and enhanced the content of butyric acid, which is an intestinal metabolite.

The Studies on In Vivo Embryo Production using Sex-Sorted Sperm in Korean Brindle Cattle (성 감별 정자를 이용한 칡소 체내수정란 생산에 관한 연구)

  • The objective of this study was to investigate the result of in vivo embryo collection and pregnancy rate after embryo transfer using sex-sorted sperm of Korean brindle cattle. Donor Korean brindle cattle superovulation treated by decreasing dose of FSH injection. Embryos were recovered on 7 days after the third artificial insemination. Control group semen straw used artificial insemination contained 20 million sperm. Sex-sorted semen straws contained 4 million sperm or 10 million sperm. As for the result of the recovery of the in vivo embryos derived from sex-sorted sperm, the number of transferable embryos was significantly highly recovered to be $6.20{\pm}2.28/donor$ from the control group and was significantly lowly recovered to be $1.57{\pm}1.72/donor$ from the group treated at a sperm concentration of $10{\times}10^6$ (p<0.05). The number of unfertilized embryo was $0.8{\pm}1.30/donor$ in control group which was significantly lower than the group treated at a sperm concentration of $4{\times}10^6$ (p<0.05). However, there was no significant difference in the number of undeveloped ova between control and treatment groups. Pregnancy rate after embryo transfer was shown to be 35.00% in control group and 12.50% in treatment group. The karyotype analysis of the calf derived from sex-sorted sperm resulted in a similar chromosomal distribution pattern (2n=60, XX) compared to those of common Korean native cattle.

Effect of Different Light Emitting Diode (LED) Lights on the Growth Characteristics and the Phytochemical Production of Strawberry Fruits during Cultivation (파장별 LED광이 딸기의 생장 특성과 생리 활성 물질 형성에 미치는 효과)

  • Recent unusual weather due to global warming causes shortage of daily sunlight and constitutes one of the primary reasons for agricultural damages. LED light sources are frequently utilized to compensate for the shortage of sunlight in greenhouse agriculture. The present study is aimed at evaluating formations of phytochemicals as well as growth characteristics of mature strawberry fruits ('Daewang' cultivar) during cultivation in a closed growth chamber equipped with artificial LED light as a sole light source. Each LED light of blue (448 nm), red (634 and 661 nm) or mixed blue plus red (blue:red = 3:7) was separately supplied and the intensity of each light was adjusted to $200{\pm}1{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at plant level with a photoperiod consisted of 16 hours light and 8 hours darkness. Strawberries grown under mixed LED light of blue and red wavelengths showed a higher production of fruits than those grown under other LED treatments. Fructose, one of the free sugars, increased in mixed LED light-grown fruits. Anthocyanin contents were elevated remarkably in the mixed LED light-grown fruits compared with those in other LED treatments. Contrastingly, contents of total phenolics and flavonoids were not of much different from one another among the fruits treated with various LED lights. On the other hand, ripening of strawberry fruits was found to be faster when grown under blue LED light compared with other LED treatments. Moreover, antioxidant activities of blue or red LED light-grown fruits, respectively, were significantly higher than those of mixed LED light-grown fruits. We suggest that when daylight is in shortage during cultivation in a greenhouse, supplementation of sunlight with LED light, which is composed of blue and red wavelengths, could be useful for the enhancement of productivity as well as of free sugar content in strawberry fruits. In addition, for the strawberry culture in the plant factory, selective adoption of LED light wavelength would be required to accomplish the purpose of controlling fruit maturation time as well as of enhancing contents of sugars and antioxidants of fruits.
