Effects of Different Extraction Extraction Media and Reaction Mixtures on Photosystem II Activity of Spinach Chloroplasts (시금치 엽록체의 광계의 활성에 미치는 추출용매와 반응용액의 영향)

  • This work deals with different extraction media and reaction mixtures on photosystem II activity of Spinach chloroplasts. The photoreduction rate of ferricyanide and DPIP by intact chloroplasts which extracted with four kinds of extraction media; S-Tris-N pH 7.2, 8.0, S-Tricine-N pH 7.2, 8.0, was measured in five kinds of reaction mixtures; S-Tris-N pH 7.2, 8.0, S-Tricine-N pH 7.2, 8.0, 0.05 M-Tris pH 7.2. The extraction medium which shows the highest photoreduction rate was S-Tris-N at pH 7.2 and S-Tricine-N at pH 8.0. As to the reaciton mixture, S-Tricine-N pH 8.0 showed the highest rate. On the complex effects of extraction media and reaction mixtures, the highest photordeuction rate of Hill oxidant by intact chloroplasts was obtained by S-Tris-N pH 7.2 extraction medium and S-Tricine-N pH 8.0 reaction mixture. The second highest activity was obtained by S-Tricine-N pH 8.0 extraction medium and reaction mixture.

  • For an odd prime p, n=4k and $d=((p^2k+1)/2)^2$, there are $(p^{2k}+1)/2$ distinct decimated sequences, s(dt+1), $0{\leq}l<(p^{2k}+1)/2$, of a p-ary m-sequence, s(t) of period $p^n-1$. In this paper, it is shown that the cross-correlation function between s(t) and s(dt+l) takes the values in $\{-1,-1{\pm}\sqrt{p^n},-1+2\sqrt{p^n}\}$ and their, cross-correlation distribution is also derived.

ANTIBACTERIAL EFFECT OF POLYPHOSPHATES ON MUTANS STREPTOCOCCI (Mutans streptococci에 대한 polyphosphate의 항균효과)

  • Mutans streptococci, especially S. mutans and S. sobrinus strongly implicated in pathogenesis of dental caries, the major cause of tooth loss in children. Use of an antibacterial agent controlling dental caries has been rationalized. The present study was performed to observe the antibacterial effect of inorganic polyphosphates (polyP) on S. mutans and S. sobrinus. S. mutans GS5 and S. sobrinus 6715 were grown in brain-heart infusion broth with or without polyP. Minimal inhibitory concentration (MIC) of polyP for S. mutans GS5 was determined to be 0.08% and that for S. sobrius 6715 was 0.17%. PolyP 15 added to the growing culture of S. mutans GS5 and S. sobrinus 6715 at their exponential phase was as effective in inhibiting the growth of S. mutans GS5 and S. sobrinus 6715 as polyP added at the very beginning of the culture. More than 85% of the cells lost their viability determined by viable cell count when polyP 15 was added to the culture of growing S. mutans GS5 at MIC, suggesting that polyP 15 has bacterial effect on the bacterium. And more than 99.9% of the cells lost their viability determined by viable cell count when polyP 15 was added to the culture of growing S. sobrinus 6715 at MIC, suggesting that polyP 15 has bacterial effect on the bacterium. Intracellular nucleotide release from S. mutans CS5 and S. sobrinus 6715 was increased in the presence of polyP 15 for 5h but was not really reversed by the addition of divalent cations like $Ca^{++}\;and\;Mg^{++}$. The majority of the cells appeared to be atypical in their shape, demonstrating accumulation of highly electron-dense granules and ghost cells. The overall results suggest that polyP have a strong bactericidal activity against S. mutans and S. sobrinus in which lysis in relation to chelation may not play the major role but unknown mechanism that possibly affects the viability of the bacterium may be involved. PolyP may be used as an agent for prevention of dental caries.

  • In this paper, we consider the jump number of the split P[S] of a subset S ordered set P. $For\ x\in\ P,\ we\ show\ that\ s(P)\leq\ s(P[x]\leq\ s(P)+2$ and give a necessary and sufficient condition for which s(P[x])=s(P).

Rheological Properties of Gelatinized Model Foods (모형식품의 리올로지 특성)

  • The model foods were prepared by simulating mositure, protein and starch, and they were heated for 30 mins, at $80^{\circ}C$ and then cooled at $25^{\circ}C$ in water bath. Their rheological properties were investigated by the use of Brookfield wide-gap rotational viscometer at $30{\sim}60^{\circ}C$, and the rotation speed ranged from 0.6 to 6 rpm and solid content ranged from 8% to 11%, the results obtained were as follows. 1. All the model foods ($P_1S_3$, $P_2S_2$, $P_1S_1$, $P_2S_1$, $P_3S_1$, $P_4S_0$) exhibited pseudoplastic behaviors with yeild stress and were thixotropic foods which showed time - dependent structural decays, but the starch food of 8 ~ 11 % solid content did not show the flow behavior. 2. The correlation between the rheological parameters and the protein content of model foods in various moisture content did not appeared a constant relationship. 3. The change of shear stress against shear rate in high starch foods was larger than that in high protein foods and the structure at initial shear time was decayed with a quatic equation according to the Tiu's Model and structural decay was in parallel with the increase of shear rate. 4. The temperature dependency of the apparent viscosity of $P_1S_2$, and $P_2S_1$ was fully expressed by Arrhenius equation and activation energies of their food were 2.35 and $1.34Kcal/g{\cdot}mol$, respectively.

SPAWNING AND LARVAL DEVELOPMENTS OF TWO SPECIES OF POLYCLAD WORMS, STYLOCHUS IJIMAI & KABURAKI AND PSEUDOSTYLOCHUS OBSCURUS (STIMPSON) (납작벌레류(Stylochus ijimai, Pseudostylochus obscurus)의 산란 및 유생발생에 관하여)

  • 1. Stylochus ijimai spawns from May to October with peak spawning from July to September, P. obscurus spawns from June to October with peak spawning from July to September. 2. S. ijimai spawns approximately 96,000-132,000 eggs by one spawning. 1. Just after spawnings, the eggs of S. ijimai were $102\mu$ in diameter, and those of P. obscurus were $108\mu$. 4. S. ijimai and P. obscurus have simple eggs. S. ijimai develop indirectly having Gotte larvae, while P. obscurus develop directly. 5. These two species show very similar patterns of early developments from maturation division of eggs to the gastrula stage. 6. The zygots of S. ijimai reaches mesentoblast stage 48 hours after fertilization, and Gotte larvae hatch out 7 days after fertilization. The zygots of P. obscurus reaches mesentoblast stage 72 hours after fertilization, and juveniles hatch out 14 days after fertilization. 7. S. ijimai, have 14 days of planktonic larvae stages bearing strong phototacic behavior : P. obscurus have 7 days of planktonic life without phototacic behavior. 8. Newly hatched larvae of S. ijimai and P. obscurus are $138\mu\;and\;170\mu$ in length respectively. The early creeping forms of S. ijimai and P. obscurus are $152\mu\;and\;185\mu$ in length respectively. 9. In the early creeping stage S. ijimai are characterized by testing processes and flattening of the body. In the same stage P. obscurus lost 2 eye-spots in the cerebral area. 10. The early creeping larvae of these two species were found only in mud-flat substrates.

Pro-Inflammatory Role of S1P3 in Macrophages

  • Sphingosine kinase 1 and its product, sphingosine 1-phosphate (S1P), as well as their receptors, have been implicated in inflammatory responses. The functions of receptors $S1P_1$ and $S1P_2$ on cell motility have been investigated. However, the function of $S1P_3$ has been poorly investigated. In this study, the roles of $S1P_3$ on inflammatory response were investigated in primary peritoneal macrophages. $S1P_3$ receptor was induced along with sphingosine kinase 1 by stimulation of lipopolysaccharide (LPS). LPS treatment induced inflammatory genes, such iNOS, COX-2, $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$. TY52156, an antagonist of $S1P_3$ suppressed the induction of inflammatory genes in a concentration dependent manner. Suppression of iNOS and COX-2 induction was further confirmed by western blotting and NO measurement. Suppression of $IL-1{\beta}$ induction was also confirmed by western blotting and ELISA. Caspase 1, which is responsible for $IL-1{\beta}$ production, was similarly induced by LPS and suppressed by TY52156. Therefore, we have shown $S1P_3$ induction in the inflammatory conditions and its pro-inflammatory roles. Targeting $S1P_3$ might be a strategy for regulating inflammatory diseases.

  • Eight new compounds were prepared by the addition reaction of thioglycolic acid to ${\beta}$-nitrostyrene and its derivatives. s-[2-Nitro-1-phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-methyl)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-methoxy)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-chloro)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-bromo)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(p-nitro)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(3-methoxy-4-ethoxy)phenylethyl]-thioglycolic acid; s-[2-nitro-1-(3,4,5-trimethoxy)phenylethyl]-thioglycolic acid; The structure of these compound were identified by elemental analysis, UV, IR and NMR spectral data.

Cross-reactivity and Protective Immunity of Streptococcus pneumonieae ClpP (페렴구균 ClpP의 면역 교차 반응과 방어효과)

  • ClpP is a stress-inducible protein and proteolytic subunit of the ATP-dependent Clp protease in prokaryotes and eukaryotes. Although its physiological roles in bacterial virulence were widely studied in various organsims, including Streptococcus pneumoniae, until now the immunological effect has not been investigated. Here, we have examined the cross reactivity of S. pneumoniae ClpP antibody with other organisms's cell lysate proteins. Although the protein sequence of S. pneumoniae ClpP was highly conserved among various organisms including human, the antibody rasised by S. pneumoniae ClpP was not cross-reacted with other organism's cell lysates, which were Saccharomyces cerevisiae , human lung A549 cell, Bacillus subtilis, Pseuomonas aeruginosa, E. coli, and Salmonella typhi. It was only reacted with S. pneumoniae and Lato-bacillus thermophilus. Thus we examined the immunoprotective effect of ClpP by immunizing mice with the purified ClpP. The mean survival time of mouse was significantly increased with the ClpP immunization. These results suggest that S. pneumoniae ClpP could be used as a vaccine candidate for prevention of S. pneumoniae infection.
