In Vitro 반추위 발효를 통한 볏짚 분해와 섬유소 박테리아 군집 변화의 관계 연구 (Study on the Relationships between Rice Straw Degradation and Changes of Fibrolytic Bacteria Population by in Vitro Rumen Fermentation)

  • 본 연구는 반추동물이 볏짚을 소화하는 과정 중에 주요 섬유소 분해 박테리아의 분포 변화를 연구하여 반추위내 볏짚이용효율을 증진시키기 위한 진보적 연구 자료를 제공하고자 실시하였다. 본 연구를 수행하기 위하여 분쇄 볏짚을 기질로 사용한 In vitro 반추위 발효를 실시하였으며, 배양 0. 4, 8, 12, 48시간에 볏짚 건물 소화율 및 섬유소 분해 박테리아 균수 변화를 조사하였다. 섬유소 분해 박테리아 균수는 F. succinogenes. R. albus와 R. flavefaciens를 대상으로 Quantitative real-time PCR을 이용하여 16S rDNA의 copy No의 log값(Log copy No)을 측정하였다. 섬유소 분해 박테리아의 총 균수는 F. succinogenes, R. albus와 R. flavefaciens의 볏짚 표면 부착 및 배양액내 부유 박테리아의 총합으로 배양 시간에 따라 증식을 하여 24시간에서 최고 군집(29.0 Log copy No)을 형성하고, 이후 감소하였다. 이에 볏짚 소화율도 24시간까지 점점 큰 폭으로 증가하였고, 이후에는 점점 둔화되는 경향을 나타냈다. F. succinogenes, R. flavefaciens 및 R. albus는 배양 시작 즉시 볏짚 표면에 부착이 발생됨을 발견하였으며, 이때 이들의 부착 군집의 비율은 각각 34.5%, 84.4% 및 67.9%로 차지하였다. 그리고 이들 섬유소 분해 박테리아의 볏짚 부착 비율은 모든 균수가 공히 배양 4시간째부터 94.7% 이상 최고 수준에 이르렀고, 부착 균수는 균종에 따라 각각 배양 12시간 또는 24시간째에 최고 수준을 나타냈다. F. succinogenes, R. flavefaciens 및 R. albus의 최고 수준의 볏짚 표면 부착 속도는 배양 12시간에 시작되어 배양 24시간에 각각 10.33, 9.28 및 8.30 Log copy No/h/g DM에 도달하였다. 그리고 섬유소 분해 속도는 표면 부착 속도보다 좀 늦은 배양 24시간에 0.95% DM/h로 최고 정점에 도달하였다. 따라서 본 연구결과는 반추위 발효 진행 과정에서 섬유소 분해 박테리아 총균의 증식에 따라 조사료의 소화가 증가되며, 상당히 많은 섬유소 분해 박테리아들이 조사료 유입과 동시에 표면 부착이 발생하고, 이들 부착 섬유소 분해 박테리아의 활성화가 먼저 진행된 후에 섬유소 분해 활성화가 이루어져야 조사료 소화의 극대화를 가져올 수 있다는 사실을 제시하여 주었다.

Effects of Methylcellulose on Cellulolytic Bacteria Attachment and Rice Straw Degradation in the In vitro Rumen Fermentation

  • An in vitro experiment was conducted to evaluate the effect of methylcellulose on the attachment of major cellulolytic bacteria on rice straw and its digestibility. Rice straw was incubated with ruminal mixture with or without 0.1% methylcellulose (MC). The attachment of F. succinogenes, R. flavefaciens and R. albus populations on rice straw was measured using real-time PCR with specific primer sets. Methylcellulose at the level of 0.1% decreased the attachment of all three major cellulolytic bacteria. In particular, MC treatment reduced (p<0.05) attachment of F. succinogenes on rice straw after 10 min of incubation while a significant reduction (p<0.05) in attachment was not observed until 4 h incubation in the case of R. flavefaciens and R. albus. This result indicated F. succinogenes responded to MC more sensitively and earlier than R. flavefaciens and R. albus. Dry matter digestibility of rice straw was subsequently inhibited by 0.1% MC, and there was a significant difference between control and MC treatment (p<0.05). Incubated cultures containing MC had higher pH and lower gas production than controls. Current data clearly indicated that the attachment of F. succinogenes, R. flavefaciens and R. albus on rice straw was inhibited by MC, which apparently reduced rice straw digestion.

Effect of feeding a by-product feed-based silage on nutrients intake, apparent digestibility, and nitrogen balance in sheep

  • Background: Literature is lacking on the effects of feeding by-product feed (BF)-based silage on rumen fermentation parameters, nutrient digestion and nitrogen (N) retention in sheep. Therefore, this study was conducted to determine the effect of replacing rye straw with BF-based silage as a roughage source on ruminal parameters, total-tract apparent nutrient digestibility, and N balance in sheep. Methods: The by-product feed silage was composed of spent mushroom substrate (SMS) (45 %), recycled poultry bedding (RPB) (21 %), rye straw (11 %), rice bran (10.8 %), corn taffy residue (10 %), protected fat (1.0 %), bentonite (0.6 %), and mixed microbial additive (0.6 %). Six sheep were assigned randomly to either the control (concentrate mix + rye straw) or a treatment diet (concentrate mix + BF-based silage). Results: Compared with the control diet, feeding a BF-based silage diet resulted in similar ruminal characteristics (pH, acetate, propionate, and butyrate concentrations, and acetate: propionate ratio), higher (p < 0.05) ruminal NH3-N, higher (p < 0.05) ether extract digestibility, similar crude protein digestibility, lower (p < 0.05) dry matter, fiber, and crude ash digestibilities, and higher (p < 0.05) N retention (g/d) Conclusion: The BF-based silage showed similar energy value, higher protein metabolism and utilization, and lower fiber digestion in sheep compared to the control diet containing rye straw.

  • The normal feeding approach of goats might be due to their precise anatomical and physiological characteristics of entity, which permit them to be highly selective, to eat legume silages and wild green grass. This review has been designed to consider the grazing behavior, fodder selection, and feed composition of goats. Various herbs and corns consumed by goats have numerous nutritive resources. Based on the general herbaceous intake activities and behavior of goats, they prefer wild grass such as grass grown in the steep hills than soft grass. Because the digestion capacity of cellulose feed has higher digestion level compared to other non-ruminants within rumen and it is advantageous to use wild forest or mountain grass which comprises high proportion of cellulose feed for goat. In South Korea, there are abundant feed resources for goats because of occupying large areas of mountains. Thus, goat production and feeding costs could be reduced if plants are used from the wild forest as a feed for goats relative to grassland grazing. Also, it is expected to contribute in improvement of goat farming with harmonious relationship between the grassland and wild forest while satisfying animal welfare and physiological desires of livestock.

혼합 메탄균과 반추위 섬유소 분해균 첨가가 메탄발생에 미치는 영향 (Effects of Supplementation of Mixed Methanogens and Rumen Cellulolytic Bacteria on Biochemical Methane Potential)

  • 본 연구는 메탄생성에 직접적으로 관여하는 혼합 메탄균과 셀롤로스 등의 고분자 물질의 가수분해 반응에 활성이 뛰어난 반추위 내 혐기성 섬유소분해균 중에서 대표적인 Fibrobacter succinogenes, Ruminococcus flavefaciens 및 Ruminococcus albus를 biochemical methane potential (BMP) 시험에 첨가하였을 때 메탄 발생에 미치는 영향을 조사하고자 수행되었다. BMP시험은 멸균증류수를 첨가한 control과 각각의 미생물 배양액을 첨가한 혼합 메탄균 첨가구 (M), F. succinogenes 첨가구 (FS) R. flavefaciens 첨가구 (RF), R. albus 첨가구 (RA) 및 RA+FS 혼합첨가구와 M+RA+FS 혼합 첨가구로 총 7개 처리구로 각 처리구별 3반복으로 진행되었다. 미생물 배양액의 첨가량은 식종액과 기초혐기배지 (anaerobic basic medium) 혼합액 50 mL에 1% (0.5 mL), 3% (1.5 mL) 및 5% (2.5 mL) 씩 첨가 하였고 배양을 위한 기질로는 cellulose ($2.0g\;VS\;L^{-1}$)이 이용되었다. BMP 시험을 위해 40일간 배양이 지속되었고 중온소화를 위해 $38^{\circ}C$의 배양기에서 수행되었다. 실험의 결과 총 바이오가스 및 메탄 발생량은 5% FS에서 다른 처리구와 비교하여 각각 10.4~22.7% 및 17.4~27.5% 높았다 (p<0.05). 총고형물 (TS) 분해율도 가스발생 결과와 유사하였는데, 전반적으로 FS가 높게 나타났으며, 5% FS에서 64.2%로 가장 높았다. 휘발성 고형물 (VS) 분해율은 5% FS와 5% RF가 각각 68.4 및 71.0%로 가장 높았다. BMP 종료시 배양액내 pH는 모든 처리구가 6.4이상으로 메탄발효에 큰 영향을 주지 않았음을 알 수 있었다. 결론적으로 본 실험의 결과 혐기소화에 대한 회분식 배양에서는 메탄생성단계보다는 가수분해단계에서 특히, F. succinogenes 배양액의 첨가량이 증가할수록 메탄의 생성량을 증가시킴을 알 수 있었다.

Effect of Triticale Dried Distillers Grains with Solubles on Ruminal Bacterial Populations as Revealed by Real Time Polymerase Chain Reaction

  • Real time PCR was used in this study to determine the effect of triticale dried distillers grains with solubles (TDDGS) as a replacement for grain or barley silage in finishing diets on the presence of six classical ruminal bacterial species (Succinivibrio dextrinosolvens, Selenomonas ruminantium, Streptococcus bovis, Megasphaera elsdenii, Prevotella ruminicola and Fibrobacter succinogenes) within the rumen contents of feedlot cattle. This study was divided into a step-wise adaptation experiment (112 days) that examined the effects of adaptation to diets containing increasing levels of TDDGS up to 30% (n = 4), a short-term experiment comparing animals (n = 16) fed control, 20%, 25% or 30% TDDGS diets over 28 days, and a rapid transition experiment (56 days) where animals (n = 4) were rapidly switched from a diet containing 30% TDDGS to a barley-based diet with no TDDGS. It was found that feeding TDDGS as replacement for barley grain (control vs. 20% TDDGS) decreased 16S rRNA copy numbers of starch-fermenting S. ruminantium and S. bovis (p<0.001 and p = 0.04, respectively), but did not alter 16S rRNA copy numbers of the other rumen bacteria. Furthermore, feeding TDDGS as a replacement barley silage (20% vs. 25% and 30% TDDGS) increased 16S rRNA copy numbers of S. ruminantium, M. elsdenii and F. succinogenes (p<0.001; p = 0.03 and p<0.001, respectively), but decreased (p<0.001) the 16S rRNA copy number of P. ruminicola. Upon removal of 30% TDDGS and return to the control diet, 16S rRNA copy numbers of S. ruminantium, M. elsdenii and F. succinogenes decreased (p = 0.01; p = 0.03 and p = 0.01, respectively), but S. dextrinosolvens and S. bovis increased (p = 0.04 and p = 0.009, respectively). The results suggest that replacement of TDDGS for grain reduces 16S rRNA copy numbers of starch-fermenting bacteria, whereas substitution for barley silage increases 16S rRNA copy numbers of bacteria involved in fibre digestion and the metabolism of lactic acid. This outcome supports the contention that the fibre in TDDGS is highly fermentable.

Effects of purified lignin on in vitro rumen metabolism and growth performance of feedlot cattle

  • Objective: The objectives were to assess the effects of purified lignin from wheat straw (sodium hydroxide dehydrated lignin; SHDL) on in vitro ruminal fermentation and on the growth performance of feedlot cattle. Methods: In vitro experiments were conducted by incubating a timothy-alfalfa (50:50) forage mixture (48 h) and barley grain (24 h) with 0, 0.25, 0.5, 1.0, and 2.0 mg/mL of rumen fluid (equivalent to 0, 2, 4, 8, and 16 g SHDL/kg diet). Productions of $CH_4$ and total gas, volatile fatty acids, ammonia, dry matter (DM) disappearance (DMD) and digestion of neutral detergent fiber (NDF) or starch were measured. Sixty Hereford-Angus cross weaned steer calves were individually fed a typical barley silage-barley grain based total mixed ration and supplemented with SHDL at 0, 4, 8, and 16 g/kg DM for 70 (growing), 28 (transition), and 121 d (finishing) period. Cattle were slaughtered at the end of the experiment and carcass traits were assessed. Results: With forage, SHDL linearly (p<0.001) reduced 48-h in vitro DMD from 54.9% to 39.2%, NDF disappearance from 34.1% to 18.6% and the acetate: propionate ratio from 2.56 to 2.41, but linearly (p<0.001) increased $CH_4$ production from 9.5 to 12.4 mL/100 mg DMD. With barley grain, SHDL linearly increased (p<0.001) 24-h DMD from74.6% to 84.5%, but linearly (p<0.001) reduced $CH_4$ production from 5.6 to 4.2 mL/100 mg DMD and $NH_3$ accumulation from 9.15 to $4.49{\mu}mol/mL$. Supplementation of SHDL did not affect growth, but tended (p = 0.10) to linearly reduce feed intake, and quadratically increased (p = 0.059) feed efficiency during the finishing period. Addition of SHDL also tended (p = 0.098) to linearly increase the saleable meat yield of the carcass from 52.5% to 55.7%. Conclusion: Purified lignin used as feed additive has potential to improve feed efficiency for finishing feedlot cattle and carcass quality.

가열온도, 가열시간 및 부형제의 첨가량이 도축 반추위 내용물의 자일란, 셀룰로오스 및 전분 분해효소 활성에 미치는 영향 (Effects of Temperature and Time for Heating and Filler Content on the Activities of Xylanase, Cellulase and Amylase in Slaughterhouse Rumen Content)

  • 본 연구는 도축장에서 폐기되는 도축 반추위 내용물을 사일리지 혹은 TMR (total mixed ration) 사료 첨가용 효소제로 개발하기 위한 목적으로 수행되었다. 도축 반추위 내용물에는 상당한 수분이 함유되어 있어, 적절한 건조과정을 거치지 않고서는 그 활용이 원활하지 않다. 그러나 효소는 열에 민감한 단백질로 구성되어 있기 때문에 적절한 건조 조건의 개발이 필요하다. 본 연구에서는 통계적 방법을 이용하여 가열온도(60, 75, $90^{\circ}C$), 가열시간(12, 30, 48 시간) 및 부형제의 비율(12, 22.5, 33%)이 도축 반추위 내용물에 함유된 다양한 효소활성에 미치는 효과를 분석하였다. 총 3가지 효소, xylan 분해효소, 셀룰로오스 분해효소 및 전분 분해효소를 검토하였고, 각 효소활성들에 대한 각 요인들의 효과가 매우 다양하게 나타났다(p<0.05). 셀룰로오스 분해효소와 전분 분해효소의 활성은 가열온도가 증가함에 따라 감소하였으며(p<0.05), xylan 분해해소는 열에 대한 안전성이 다른 효소들에 비하여 우수하였다. 자일란, 셀룰로오스, 전분 분해효소를 증가시키는 적정 부형제의 비율은 22.5, 12 그리고 33%로 나타났다. 비록 3가지 효소들의 제형화에 있어 공통적으로 적용될 수 있는 최적점을 도출하지는 못하였으나, 본 연구결과에서 얻어진 효소들의 반응 값들을 이용하여 목적하는 효소에 따라서 다양한 방법으로 적용이 가능할 것으로 판단된다.

Effects of fermentation on protein profile of coffee by-products and its relationship with internal protein structure measured by vibrational spectroscopy

  • Objective: To our knowledge, there are few studies on the correlation between internal structure of fermented products and nutrient delivery from by-products from coffee processing in the ruminant system. The objective of this project was to use advanced mid-infrared vibrational spectroscopic technique (ATR-FT/IR) to reveal interactive correlation between protein internal structure and ruminant-relevant protein and energy metabolic profiles of by-products from coffee processing affected by added-microorganism fermentation duration. Methods: The by-products from coffee processing were fermented using commercial fermentation product, called Saus Burger Pakan, consisting of various microorganisms: cellulolytic, lactic acid, amylolytic, proteolytic, and xylanolytic microbes, for 0, 7, 14, 21, and 28 days. Protein chemical profiles, Cornell Net Carbohydrate and Protein System crude protein and CHO subfractions, and ruminal degradation and intestinal digestion of protein were evaluated. The attenuated total reflectance-Ft/IR (ATR-FTIR) spectroscopy was used to study protein structural features of spectra that were affected by added microorganism fermentation duration. The molecular spectral analyses were carried using OMNIC software. Molecular spectral analysis parameters in fermented and non-fermented by-products from coffee processing included: Amide I area (AIA), Amide II (AIIA) area, Amide I heigh (AIH), Amide II height (AIIH), α-helix height (αH), β-sheet height (βH), AIA to AIIA ratio, AIH to AIIH ratio, and αH to βH ratio. The relationship between protein structure spectral profiles of by-products from coffee processing and protein related metabolic features in ruminant were also investigated. Results: Fermentation decreased rumen degradable protein and increased rumen undegradable protein of by-products from coffee processing (p<0.05), indicating more protein entering from rumen to the small intestine for animal use. The fermentation duration significantly impacted (p<0.05) protein structure spectral features. Fermentation tended to increase (p<0.10) AIA and AIH as well as β-sheet height which all are significantly related to the protein level. Conclusion: Protein structure spectral profiles of by-product form coffee processing could be utilized as potential evaluators to estimate protein related chemical profile and protein metabolic characteristics in ruminant system.

Relative Palatability to Sheep of Some Browse Species, their In sacco Degradability and In vitro Gas Production Characteristics

  • A study was conducted to estimate the nutritive value of some selected acacia forages using palatability index, in sacco degradability and in vitro gas production characteristics. Ten wethers (mean wt. $18{\pm}3.5kg$) were offered Acacia tortilis, Acacia nilotica, Acacia mellifera, Acacia brevispica, Acacia Senegal and Leucaena leucocephala (control) using a cafeteria system to determine the species preference by the animals. The acacia species were rich in nitrogen and showed variable palatability pattern. Significant (p<0.05) differences in relative palatability index (RPI) were detected among the species with the following ranking: brevispica > leucaena > mellifera > tortilis > Senegal > nilotica. Acacia nilotica appeared to be of low relative palatability with RPI of 24% and this was attributed to relatively high phenolic concentrations. The DM potential degradability (B) and rate of degradation (c) of the species were significantly (p<0.05) different, ranging from 40.1 to 59.1% and 0.0285 to 0.0794/h respectively. Acacia species had moderate levels of rumen undegradable protein, much higher than that in leucaena. In vitro gas production results indicated the effect of polyphenolic compounds on the fermentation rate, with lower gas production recorded from A. nilotica and tortilis. Based on RPI, A. brevispica and mellifera were superior to the rest and comparable to L. leucocephala. Long-term feeding trials are required with the superior species when used as protein supplements to poor quality diets.
