Targeting the Transforming Growth Factor-β Signaling in Cancer Therapy

  • TGF-${\beta}$ pathway is being extensively evaluated as a potential therapeutic target. The transforming growth factor-${\beta}$ (TGF-${\beta}$) signaling pathway has the dual role in both tumor suppression and tumor promotion. To design cancer therapeutics successfully, it is important to understand TGF-${\beta}$ related functional contexts. This review discusses the molecular mechanism of the TGF-${\beta}$ pathway and describes the different ways of tumor suppression and promotion by TGF-${\beta}$. In the last part of the review, the data on targeting TGF-${\beta}$ pathway for cancer treatment is assessed. The TGF-${\beta}$ inhibitors in pre-clinical studies, and Phase I and II clinical trials are updated.

NFAT Transcription Factor Inhibitory Constituents from Cnidium officinale

  • Four hundred varieties of plant extracts were screened for inhibitory activity against the NFAT transcription factor which plays an important role in inducing immune response. Among them, the MeOH extract of Cnidium officinale showed potent activity, and the activity-guided separation yielded butylidenephthalide, senkyunolide A and falcarindiol as the active constituents. The $IC_{50}$ value of butylidenephthalide was $1.3{\times}10^{-4}\;M$ and was similar to that of senkyunolide A $(2.1{\times}10^{-4}\;M)$. Interestingly, falcarindiol showed higher activity $(IC_{50},\;2.6{\times}10^{-5}\;M)$ than the two phthalides.

  • Brand relationship, which refers to the bond between brands and consumers, plays an important role in marketing due to its positive or negative effects on consumer behavior. The goals of this research were 1) to identify the types of brand relationships between young female consumers and apparel brands, and 2) to examine the influence of brand relationships over consumers'favorable attitudes and strong attachment (durability) to the brands. For the purpose of this study, ten casual wear brands, which are highly perceived by and popular with high school and university female students, were chosen by a preliminary inquiry. And then focused group interviews were performed to develop a questionnaire. The main examination was performed from August 20 to September 12, 2000, using five types of surveys each of which contained two brands. SPSS 8.0 was used for factor analysis and recession analysis. Four types of brand relationships were categorized based on the results of factor analysis: self-devotion, intimacy, rejection and trust. Each factor showed significant influence on both positive altitude and durability.

  • In order to construct a successful electronic commerce system, three main essential factors must be satisfied to obtain the best effective outcomes. The three main essential factors are as follows : economic factor, effectiveness factor and convenient factor. In order to understand the role of these three factors, one must have some insight knowledge about security to assist him to implement these three factors in his construction of an electronic commerce system. This paper analyses a implementation mechanism of security systems based on the SET 1.0 standard for electronic commerce systems, thus providing an effective plan for the construction of a security system in the SET-based electronic commerce field. This paper helps to analyse the elements of security vulnerabilities in the SET 1.0 standard implementation and also helps to understand the SET 1.0 protocol.

Selection of Canonical Factors in Second Order Response Surface Models

  • A second-order response surface model is often used to approximate the relationship between a response factor and a set of explanatory factors. In this article, we deal with canonical analysis in response surface models. For the interpretation of the geometry of second-order response surface model, standard errors and confidence intervals for the eigenvalues of the second-order coefficient matrix play an important role. If the confidence interval for some eigenvalue includes 0 or the estimate of some eigenvalue is very small (near to 0) with respect to other eigenvalues, then we are able to delete the corresponding canonical factor. We propose a formulation of criterion which can be used to select canonical factors. This criterion is based on the IMSE(=Integrated Mean Squared Error). As a result of this method, we may approximately write the canonical factors as a set of some important explanatory factors.

TTF-1 Expression in PACAP-expressing Retinal Ganglion Cells

  • In mammals light input resets the central clock of the suprachiasmatic nucleus by inducing secretion of pituitary adenylate cyclase-activating polypeptide (PACAP) from retinal ganglion cells (RGCs). We previously showed that thyroid transcription factor 1 (TTF-1), a homeodomain-containing transcription factor, specifically regulates PACAP gene expression in the rat hypothalamus. In the present study we examined the expression of TTF-1 in PACAP-synthesizing retinal cells. Fluorescence in situ hybridization (FISH) showed that it is abundantly expressed in RGCs of the superior region of the retina, but in only a small subset of RGCs in the inferior region. Double FISH experiments revealed that TTF-1 is exclusively expressed in PACAP-producing RGCs. These results suggest that TTF-1 plays a regulatory role in PACAP-expressing retinal ganglion cells.

  • Generally, power factor correction (PFC) techniques play an important role in the power supply technology. Many new circuit topologies and control strategies for PFC have been proposed. Among them, the brideless PFC (BPFC) reduces the number of switching devices and the losses and improves the power density as well. Moreover, by implementing the improved topology in the discontinous conduction mode (DCM) it ensures almost unity power factor in a simple and effective manner. In the DCM operation gives additional advantages such as zero-current turn-on in the power switches, zero-current turn-off in the output diode and reduces the complexity of the control circuitry. In this paper, a new control strategy for the BPFC is proposed. Also, the performance of the proposed system is demonstrated through experiments.

Vascular Endothelial Growth Factor Upregulates Follistatin in Human Umbilical Vein Endothelial Cells

  • Vascular endothelial growth factor (VEGF), plays a key role in angiogenesis. Many endogenous factors can affect angiogenesis in endothelial cells. VEGF is known to be a strong migration, sprouting, survival, and proliferation factor for endothelial cells during angiogenesis in endothelial cells. Searching for novel genes involved in VEGF signaling during angiogenesis, we carried out differential display polymerase chain reaction on RNA from VEGF-stimulated human umbilical vein endothelial cells (HUVECs). In this study, follistatin (FS) differentially expressed in VEGF-treated HUVECs, compared with controls. Addition of VEGF (10ng/L) produced an approximately 11.8-fold increase of FS mRNA. F5 or VEGF produced approximately 1.8- or 2.9-fold increases, respectively, in matrix metalloproteinase-2 (MMP-2) secretion for 12h, compared to the addition of a control buffer. We suggest that VEGF may affect the angiogenic effect of HUVECs, through a combination of the direct effects of VEGF itself, and the indirect effects mediated via induction of FS in vitro.

A Study on the Safety Assessment of Zinc Plating Process (아연도금공정의 안전성평가)

  • There are now the plating process that have many hazardous factor cause of the using numerous noxious chemical and bad working environments. The purpose of the study is to make a selection of suitable safety evaluation method that can analyze and righteously find with numerous hazardous factor of the plating process. And another purpose is to systematically adjust the risk of plating process by comprehension of the role of process, equipment, and source material. Therefore, these studies are carried out in the following three investigations of this report. The first research understands the injurious human health and environment by analyzing hazardous material based on the MSDS. To evaluate the safety of process and compartment, the second research is proposed the security secure counterproposal by using the FMEA and the HAZOP. The final research is devoted to systematically analyze the hazard by applying for reasonable guide word and doing the HAZOP for hazardous factor in specific process.

Zerumbone, Sesquiterpene Photochemical from Ginger, Inhibits Angiogenesis

  • Here, we investigated the role of zerumbone, a natural cyclic sesquiterpene of Zingiber zerumbet Smith, on angiogenesis using human umbilical vein endothelial cells (HUVECs). Zerumbone inhibited HUVECs proliferation, migration and tubule formation, as well as angiogenic activity by rat aorta explants. In particular, zerumbone inhibited phosphorylation of vascular endothelial growth factor receptor-2 and fibroblast growth factor receptor-1, which are key regulators of endothelial cell function and angiogenesis. In vivo matrigel plug assay in mice demonstrated significant decrease in vascularization and hemoglobin content in the plugs from zerumbone-treated mice, compared with control mice. Overall, these results suggest that zerumbone inhibits various attributes of angiogenesis, which might contribute to its reported antitumor effects.
