Comparison of Inhibitory Effect of Rice Bran-extracts of the Colored Rice Cultivars on Carcinogenesis (유색미 겨 추출물의 품종간 발암과정 억제효과의 비교)

  • Anticarcinogenicity of 12 kinds of colored rice cultivars was investigated in vitro with Ilpumbyeo as a control. Pigment and hexane fractions were prepared from the rice brans through extraction with 80% ethanol containing 0.5% TFA or n-hexane to use subsequent evaluation of their anticarcinogenic effect by determining inhibitory activity against DNA strand scission and tumor promotion, as well as antimutagenic activity using SOS chromotest system. Antimutagenic activity of the pigment fractions was revealed to decrease in order of LK1B4-12-1-1 > Sanghaehanghyulla > HP833-1-3-1-1 > Chokoto14 > Jagwangdo > Jajin > Kilmheugmi > Tiwan-Tsi-C > Heugjinmi > Linsia-Shoea-Dau > Suweon425 > Suweon415. The activity of the hexane fractions decreased in order of LK1B4-12-1-1 > Sanghaehyanghyulla>Ilpunibyeo > HP833-1-3-1-1 > Chokoto14 > Jagwangdo > Jajin > Linsia-Shoea-Dau >Tiwan-Tsi-C > Heugjinmi > Suweon425, Kilimheugmi > Suweon415. Results showed that inhibitory activity against DNA strand scission in the pigment fractions decreased in order of HP833-1-3-1-1 > Tiwan-Tsi-C > Jajin > Chokoto14 > Suweon425 > Heugjinmi > LK1B4-12-1-1 > Suweon415 > Jagwangdo > Kilimheugmi > Linsia-Shoea-Dau > Sanghaehyanghyulla. On the other hand, the activity of the hexane fractions decreased in order of LK1B4-12-1-1 > Suweon 415 > Kilimheugmi > Chokoto14 > Heugjinmi > Jajin > Suweon425 > Tiwan-Tsi-C > Sanghaehyanghyulla a Linsia-Shoea-Dau > Jagwangdo > HP833-1-3-1-1 > Ilpumbyeo. Experiment using the short-term assay for promotor substance showed that inhibitory activity contained in the pigment fractions against rumor promotor TPA decreased in order of Linsia-Shoea-Dau > Kilimheugmi > HP833-1-3-1-1, Suweon425 > Heugjinmi > Jajin > Chokoto14 > Sanghaehyanghylla > Suweon 415 > LK1B4-12-1-1 > Tiwan-Tsi-C > Jagwangdo.

Genetic Diversity and Relationship by SSR Markers of Korean Soybean Cultivars (한국 콩 육성품종의 SSR마커에 의한 유전적 다양성과 유연관계)

  • Genetic diversity of 91 Korean soybean cultivars was assessed with 20 simple sequence repeat (SSR). Twenty SSR loci generated a total of 149 alleles. The number of alleles for each SSR locus ranged from 3 to 15 with a mean of 7.5 alleles. Genetic diversity estimated by PIC value of 91 cultivars was ranged from 0.424 to 0.905 with an average of 0.711. Cluster analysis based on Nei's genetic distances classified 91 soybean cultivars except Geomjeongkong 4 into 7 groups. The majority groups were I, IV, and VI which included 26, 24, and 18 cultivars, respectively. Obvious differences in genetic diversity appeared to be related with the released periods of cultivars and utilization type of cultivars, but not with breeding sites. Cultivars released in 1970's and in 1990's showed the lowest and the highest genetic diversities with 0.576 and 0.706, respectively. Soybean cultivars for vegetable and early maturity showed the lowest genetic diversity with 0.514, while those for soy sauce and tofu showed the highest genetic diversity with 0.691. Genetic distance between soybean cultivar groups developed before 1969 and during 1970's was the nearest, while genetic distance between those developed in 1970's and 1990's was the furthest. Cultivar group for vegetable and early maturity showed the furthest genetic distance with cultivar group for soy sauce and tofu, while it showed the nearest genetic distance with cultivar group for cooking with rice. Genetic distance was greater between soybean cultivar groups developed in Suwon and Iksan than between those developed in Milyang and Iksan.

Comparison of Agrobacterium-mediated Transformation Efficiency in 43 Korean Wheat Cultivars (국내 밀 43개 품종에 대한 아그로박테리움 형질전환 효율성 검정)

  • Agrobacterium-mediated transformation (AMT) is a method that allows for the stable integration of DNA fragments into the plant genome. Transgenic plants generated through AMT typically exhibit a lower copy number of the transgene compared to those induced by particle bombardment. Furthermore, AMT offers a straightforward and efficient approach for generating transgenic plants. While the transformation efficiency of wheat is comparatively lower than that of other monocot plants such as Rice (Oryza sativa L.) and Maize (Zea mays L.), the cultivars 'Bobwhites' and 'Fielder' are commonly employed for wheat transformation. To date, there have been no reported instances of successful development of transgenic plants using Korean wheat varieties through AMT. This study aims to assess the transformation efficiency of 43 Korean wheat cultivars using the GUS assay, with the goal of identifying suitable Korean wheat cultivars for AMT. The pCAMBIA1301 vector, carrying the β-glucuronidase (GUS) gene, was incorporated into Agrobacterium strain EH105. Following the inoculation of Agrobacterium into immature embryos, GUS assays were conducted 'Saeol', 'Jopum', and 'Jonong' showed 100% (the number of embryos showing GUS spots/the number of embryos used for AMT) among 43 cultivars. In addition, cultivars with more than 70% were 'Saekeumgang', 'Jojung', 'Tapdong', 'Anbaek', 'Dabun', 'Sugang', 'Keumgang', 'Jeokjung', 'Seodun', 'Joeun', 'Dajung', and 'Baekjung'. It seems that the 15 cultivars above showed the possibility of using AMT. On the other hand, 'Yeonbaek', 'Goso', 'Baekgang', and 'Johan' showed less than 20% and GUS spots were not observed in 'Gru', 'Gobun', 'Milseong', and 'Shinmichal-1'. This study explores transient GUS expression in Korean wheat cultivars seven days after AMT. The observed initial high efficiency of transient transformation suggests the potential for subsequent stable transformation efficiency. Korean wheat cultivars demonstrating elevated transient transformation efficiency could serve as promising candidates for the development of stable transgenic wheat.

Development of Proteomics-based Biomarkers for 4 Korean Cultivars of Sorghum Seeds (Sorghum bicolor (L.) Moench) (국내 수수 종자 분석을 위한 프로테오믹스-기반 바이오마커 개발)

  • BACKGROUND: Sorghum (Sorghum bicolor (L.) Moench) ranks as the 6th most planted crop in the world behind wheat, rice, maize, soybean, and barley. The objective of this study was to identify bio-marker among sorghum cultivars using proteomics approach such as two-dimensional polyacrylamide gel electrophoresis (2-DE) coupled with mass spectrometry (MS). METHODS AND RESULTS: Proteins were extracted from sorghum seed, and separated by 2-DE. Total 652 spots were detected from 4 different sorghum seed after staining of 2-DE with colloidal Coomassie brilliant blue (CBB). Among them, 8 spots were differentially expressed and were identified using MALDI-TOF/TOF mass spectrometry. They were involved in RNA metabolism (spot1, spot 4), heat shock proteins (HSPs, spot 2), storage proteins (spot 3, spot 5, and spot 6), and redox related proteins (spot 8). Eight of these proteins were highly up-regulated in Whinchalsusu (WCS). The HSPs, Cupin family protein, and Globulin were specifically accumulated in WCS. The DEAD-box helicase was expressed in 3 cultivars except for WCS. Ribonuclease T2 and aldo-keto reductase were only expressed in 3 cultivars except for Daepung-susu (DPS). CONCLUSION(S): Functions of identified proteins were mainly involved in RNA metabolism, heat shock protein (HSP), and redox related protein. Thus, they may provide new insight into a better understanding of the charactreization between the cultivars of sorghum.

Evaluations on agronomic traits of rice transgenic lines (벼 형질전환계통의 주요 작물학적 특성에 대한 고찰)

  • This study was conducted to evaluate the performances of rice transgenic lines along with their wild types in terms of agronomic traits. A total of 32 rice transgenic lines, through previously conducted collaborative researches between molecular biologists and conventional rice breeders, were selected as promising lines. As the introduced functional genes, 17 genes, which were putatively related with high yield, disease and herbicide resistance, abiotic stress tolerance, and diversifying endosperm starch components, were transformed into three Japonica cultivars, Nipponbare, Nagdongbyeo, and Dongjinbyeo. The transgenic lines exhibited significantly deviated performances from their wild types on agronomic traits such as days to heading, culm length and yield potential. Multivariate analyses on transgenic lines to the evaluated agronomic traits also indicated random manner of phenotypic deviations from their wild type in terms of deviation directions and degrees. Our results suggested that, therefore, breeding strategies to control unexpected deleterious phenotypic performances among transgenic lines would be critical as much as the functions and proper expressions of the transformed genes.

Heat stress resistance of Ilmi

  • Rice production and quality could be changed by temperature condition. Extremely high temperature event have been occurred more frequently for global warming. To increase rice quality against to global warming, breeding of heat tolerance rice cultivar is needed. Ilmi which is the one of Korean leading rice cultivar shows heat stress resistant character during ripening stage. Yield and quality (brown and milled rice) of Ilmi did not show significant different under high temperature condition compared to control condition. However, the main physiological characters for heat resistance of Ilmi have been not investigated yet. Therefore we try to investigate the heat tolerance characters of Ilmi. Two rice cultivars, Ilmi and Ilpum-which is heat susceptible rice cultivar, were cultivated under natural condition in wagnor pot until heading was appeared. After checking heading date, each material was cultivated under different temperature condition, heat($32/22^{\circ}C$) and control($26/16^{\circ}C$) condition. Anti-oxidant enzyme activity was checked during ripening stage in each material. Catalase and ascorbic peroxidase activity of leaf under heat stress condition were higher in Ilmi than Ilpum especially early ripening stage. Analyzing of stress resistance using $H_2O_2$, the flag leaf of Ilmi showed more green color than Ilpum with higher chlorophyll content than those of Ilpum. We also checked the amount of $H_2O_2$ content in young leaf of each material by treating high temperature. $H_2O_2$ content in each material was increased according to treatment time. However $H_2O_2$ content of young leaf in Ilmi was less than those in Ilpum. Also catalase and ascorbic peroxidase activity in leaf increased much faster in Ilmi than Ilpum. With those data, we confirmed that heat stress resistance of Ilmi is due to the higher anti-oxidant activity against to stress condition. We will investigate the heat tolerance characters of Ilmi more in further study to enhance the breeding effect of heat stress tolerance rice.

Genetic Analysis on the Bacterial Blight Resistance Gene from a Wild Relative, Oryza minuta (야생벼 Oryza minuta에서 유래한 수원506호의 흰잎마름병 저항성유전자에 대한 고찰)

  • Bacterial blight (BB), cuased by the vascular pathogen Xanthomonas oryzae pv. oryzae, is one of the major threats in rice fields worldwide. In Korea, two resistance genes against BB, Xa1 and Xa3 had been intensively used for developing high quality japonica rice cultivars. Those traditional resistance sources have being rapidly ified by the adopting of BB pathogen through mutations of the corresponding avr-genes, such as K3a exhibiting high compatibility to both Xa1 and Xa3. To expanding genetic resource against BB in Korea, the Suweon506, an introgression line between a Korean japonica cultivar, Hwaseong and a wild relative, Oryza minuta, was be subjected for genetic analysis owing to the BB resistance. Through association analyses between the pathotyping and genotyping results for each $F_2$ progenies, derived from a cross between Suweon506 and a Tongil type cultivar, Milyang23, a major resistant dominant gene is localized on the subterminal region of rice chromosome 4, where at least three BB resistancde genes, Xa1, Xa2, and Xa22, were reported previously.

Inhibitory Activity of the Extracts from the Pigmented Rice Brans on Inflammatory Reactions (유색미 겨 추출물의 염증반응 억제활성)

  • Effects of the extracts from bran part of the pigmented rices on inflammation was evaluated by determining their inhibitory action on the production of nitric oxides, histamines and matrix metalloproteinase(MMP) from inflammatory leukocytes. Effects on the production of nitric oxides in a macrophage cell line, RAW264.7 cells, were determined, demonstrating that any significant difference was not detected between the normal rice and the pigmented rice extracts. Inhibitory effects on the histamine-release from a basophilic cell line, RBL-2H3, were examined, showing 3.6 to 5.4-fold increase in the inhibitory activity compared to that of the normal rices. Among the pigmented rice cultivars tested, especially, inhibitory activity of LK1-3-6-12-1-1 was the greatest. Using RAW264.7 cells, we examined the effect of the pigmented rice extracts on the MMP activity. The results showed that the enzyme activity increased with the increasing concentration of the normal rice extract. However, the pigmented rice extracts, except LK1A-2-12-1-l, acted to decrease the MMP activity with their increasing concentrations. The results described above showed the superiority of the pigmented rice extracts in inhibition on release of histamine and MMP, pivotal factors for causing inflammatory responses, from the leukocytes.

Heading and Ripening Characters of Major Early Maturing Breeding Rice Lines According to Transplanting Date and Temperature Condition (이앙기 및 온도에 따른 주요 벼 조생종 교배모본의 출수 및 등숙 특성 변화)

  • Early-maturing rice type is cultivated to produce rice before the Korean Thanksgiving Day. We investigated the flowering and ripening characteristics of major breeding lines in early maturing rice type. In Jeonju, field growth days (FGD) from transplanting to heading changed about 3.3 days by 10 days of transplanting days and about 2.5 days by the change of mean temperature during growth period. As the temperature during ripening stage, 1000 brown rice weight was changed 0.2 g, the rice quality of brown and milled rice was changed 5% and 6.5%, respectively. Baegilmi and Kittake showed early heading habit suitable for harvesting before Korean Thanksgiving. Joun and Pecos showed good ripening characteristics under high temperature during the ripening stage. We expect that these characteristics might be useful for breeding new rice cultivars for harvesting before Korean Thanksgiving.

Fermentation and Quality Characteristics of Yakju According to Different Rice Varieties (쌀 품종에 따른 약주의 발효 및 품질 특성)

  • This study was performed to improve quality of traditional Yakju and compares quality of Yakju according to different rice cultivars. The pHs of Ilmi Yakju and Hanarum Yakju showed 4.14 and 4.07, respectively and the other Yakju's pHs were ranged from 3.92 to 3.98. The content of total acid of Yakju using Indica rice imported from Thailand was the highest among the samples. The major components of free sugar was glucose and the highest content of total free sugars was found in Indica Yakju. The content of reducing sugars in Yakju using Indica rice was the higher than other samples. The ethanol content of Hanarum Yakju showed higher than those Yakju's and the lowest ethanol content found for the indica Yakju. The volatile compounds from the rice Yakju were identified by GC-MS. Twenty-one volatile compounds were found in rice Yakju. And the major volatile compounds were ethanol, acetic acid, 1-methyl-1-propanol, 2-methyl-butane, 3-methyl-1-butanol, iso-amylalcohol and 1-hexanol from Yakju. As the result of sensory test, higher scores for smell and color were found for the Yakju used Hanarum Yakju. The highest score sweat and acid were found for the Anda rice Yakju in sensory test. The result of sensory evaluation indicated that Hanarum Yakju and Anda Yakju were better than the other samples, and the value of bitterness was no significant in this test.
