Studies on the Pathogenicity of Aphelenchoides sp. and Rhabditis sp. attacking Cultivated Mushroom, Agaricus bisporus(Lange) Sing (Aphelenchoides sp.와 Rhabditis sp.의 양송이에 대한 병원성에 관한 연구)

  • Four species of nematodes attacking mushroom beds were found in samples taken from 35 mushroom farms throughout Korea. These were Rhabditis sp., Aphelenchoides sp., Ditylenchus sp. and Aphelenchus sp. ,Rhabditis sp. was found from compost and casing from all mushroom farms and the frequencies of Aphelenchoides sp. was 31.4% in the both compost and casing. Both Ditylenchus sp. and Aphelenchus sp. showed 2.7% of frequencies in the compost, none in casing. Temperature and moisture content of compost affected pathogenicity of Aphelenchoides sp. on mushroom mycelia grown in compost. The higher temperature and moisture content the sooner the damage became apparent, and the more rapid was subsequent destruction of mycelia. There was no mycelial destruction at the lowest temperature of $10^{\circ}C$. Rhabditis sp. completely disintegrated mycelia grown in the compost, in the early stage, the numbers of Rhabditis sp. rose gradually and then increased suddenly to reach a peak but soon declined. At first, the pH of Rhabditis-infested spawned compost declined but then rose gradually as mycelia was disintegrated by nematodes. The trend in pH of infested unspawned compost was similar to those of uninfested, unspawned compost. Cultures inoculated with surface-disinfected dead Rhabditis sp. and with tap water used in the nematode extraction procedures showed no mycelial injury associated microorganisms containing within or outside the nematodes even though added by artificial wounding of the mycelia. Cultures artificially wounded showed no injury away from the wounds without the presence of living Rhabditis sp., such wounded mycelia slowly regenerated. On the other hand, artificial wounding accelerated the breakdown of mycelia in the presence of living Rhabditis sp.

  • Five cases infected with Rhabditis sp. were detected in a survey to examine the stool specimens from rural primary school children. A large number of the larvae of Rhabditis sp. detected by the direct cellophane thick smear were cultured by the alter paper method. The eBamination was carried out in April through June, 1980 in Tangjeong-Myon, Ahsan.Gun, and in September 1983 in Sandong-Eup, Yeongwol-Gun. The results obtained in this study were as follows: 1. Prevalence: Out of 925 children, 5(0.54%) children were found to be positive. The number of the detected larvae was 110/0.1gm of feces in case 1, 35 in case 2, 130 in case 3, 86 in case 4 and 62 in case 5. 2. Larvae: The larvae were prepared by means of the direct fecal smear and measured by a micrometer equipped in the microscope. Twelve (12) through 15-day old larvae in culture were $197.1{\mu}{\textrm{m}}$ long in average, and the maximum size of the matured stage larvae was $884.0{\times}25.9{\mu}{\textrm{m}}$. However, the length variation was ranged as 173.0 to $884.0{\mu}{\textrm{m}}$. 3. Adults: The size of clubbed adult female was $1, 357{\mu}{\textrm{m}}(1, 176~1, 419)$ in length and $80{\mu}{\textrm{m}}(79~82)$ in width. Length of buccal cavity was $33{\mu}{\textrm{m}}$. A long cylindrical esophagus($273{\mu}{\textrm{m}}$) of the worms with a valved posterior cardiac bulb and with median bulbar swelling was morphologically indicated. Ditance from mouth to vulva was occupied 58% of body length. Male worm was $1, 006{\mu}{\textrm{m}}(890~1, 148)$ in length and $49{\mu}{\textrm{m}}(48~49)$ wide. Caudal alee of bursa and spicules ($75{\mu}{\textrm{m}}$ in length) were well developed. 4. Eggs: The oval shaped eggs in the female uterus, when cultured, were $66{\times}56{\mu}{\textrm{m}}$ in sixte, and the eggs laid by the adult could not be detected. So, reproduction might be thought to be ovoviviparity. 5. The five cases were re-examined during the period from the 1st to the 3rd week after stool examinations, but Rhabditis sp. were detected again in 4 cases in 1st week. When they were examined in 3rd week, larvae could not be detected. So, it was thought that the infection of Rhabditis sp. to humans was facultative.

Decrease of Nematode Population by Introduction of Nematophagous Fungi into The Soil as Affected by Inoculum Concentration and Temperature in Vitro (선충 기생 전적 진균의 접종원 농도와 온도조건에 따른 성충감염 및 집단 감소효과)

  • Five nematophagous fungi were evaluated for their nematicidal effect in vitro on Rhabditis sp. and Meloidogyne hapla in soil. Inocula of Arthrobotrys arthrobotryoides, A. conoides, A. oligospora, Dactylella lobata, and Fusarium oxyaporum were grown in moistened corn-sandy soil and chopped potato-sandy soil media, and incubated at 26$^{\circ}C$ for one week. The prepared inocula were incorporated in autoclaved sandy soil, mixing thoroughly at rates equ-invalent to 1:50, 1:100, 1:200, and 1:400, repectively, before 80g of the mixture carrying 100 Rhabditis sp. was put into petri plates. Nematophagous fungi effectively teduced the popuation of Rhabditis sp. in soil in a week or two following treatment of the incula at concentration of 1:50 and 1:100. The optimum was at $25^{\circ}C$ for nematicidial effect as high as 80-100%. The at the rate of 1:100 prepared incula were incorporated in auto-claved soil, where 100 Juveniles M. hapla were introduced per 80% soil. All fungi infected the M. hapla effectively in soil, caysing more than 90% mortality within one week. This result indicated the potential value of these fungi as promising biocontrol agents.

A Survey of Dung Beetles Infected with Larval Nematodes with Particular Note on Copris lunaris Beetles as a Vector for Gongylonema sp. in Iran

  • Dung beetles (family Scarabaeidae) are one of the largest families of beetles worldwide. Due to biological behavior of these arthropods, they are considered to play an important role in the life cycle of some helminths. In the present study, dung beetles collected from cattle pastures in rural areas of Ardabil province, north-west of Iran were examined for infection with larval stages of helminths. According to the results, nematodes of 2 genera were identified including Rhabditis and Gongylonema. The more common species was Rhabditis sp. which was found in 9 species of beetles. Out of 15 different species of dung beetles, Copris lunaris was the only scarabaeid to be found naturally infected with the larval stages of Gongylonema sp. Our new findings introduce C. lunaris as a potential biological vector for transmission of Gongylonema sp. to vertebrates in the surveyed region.

  • 최근 농작물에 대한 각종 선충류의 피해가 주목되어 있는 실성에 비추어 필자는 선충류연구의 기초적 과제로서 온실내의 화훼류 근부에서 발견되는 선충류 중 농업상 비교적 중요하다고 인정되는 종류와 그밖에 몇 종류에 대하여 형태적인 특징을 기술하였다. 즉 취급된 종류들은 다음과 같다. 1. Meloidogyne arenaria Chitwood 2. Scutellonema coheni Andrassy 3. Paurodontus gracilis Thorne 4. xiphinema americaum Cobb 5. Rhabditis sp 6. Cephalobus sp 7, Diplogaster sp 그리고 이상 7종류의 기주식물 및 기생정도를 보면 Meloidogyne arenaria Chitwood는 Begonia에 만 기생하였고 그밖에 화훼류에는 기생을 볼수 없었다. Scutellonema choeni Andrassy는 꽃치자, 소펄, 이마리 리스, 제브리나에 가장 많이 발견되었고, Paurodonthus gracilis Thorne는 동백나무, 청목. 지내고사리, 카네이션, 충에서 가장 많이 발견되었다. 그리고 Xiphinema americanum Coob는 산호수, 서향, 코피나무, 시크라맨, 용설란, 동백 등에서만 발견되었으나 그중에 용설란, 동백에서 가장 많이 볼 수 있었다. Rhabditis sp.는 초본류 화훼류의 대부분에서 발견되었으나 그중에도 산세배리아에서 가장 많았다 Diplogaster sp는 대부분의 화훼류에서 발견되었으며 그중에 크로톤에 비교적 많았다. Cephalobus sp.는 전반적으로 발견되었으며 발견정도는 모두 비슷하였다. Monhystera app. , Mononchus spp. , Dorylaimus app. 등도 조사란 과반수의 화훼류들에서 발견되었으나 특히 많이 발견되는 화훼류는 없었다. 이상 선충류중 근류를 형성하여 피해가 가장 주목되는 Mceloidogyne arenaris Chitwood에 대해서 온실내에서 그의 유일한 기주식물인 Begonia의 품종별 기생정도를 비교조사한 결과 semperflorens 품종에 가장 기생률이 높았고 metarica 품종은 중정도였으며 erytholopylla와 argento-guttata 품종들에는 기생이 없었다. 이 점은 매우 의의있는 사실로서 앞으로 더욱 연구할 과제로 삼았다. 끝으로 Sanceberia에서 많이 발견되는 Rhabditis sp를 $18^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$의 동온도별의 incubator에서 사육한 결과 본 종의 생육적온이 $30^{\circ}C$라는 사실을 알았다.

Screening of Plant Extracts for Nemastatic Activity (선충 억제 활성 식물 추출물의 선발)

  • A total of 2,714 methanol extracts of 1,677 plant species in 178 families were screened for nemastatic activity against Rhabditis sp. by 96-well microplate bioassay. The plant extracts with a concentration of $5,000{\mu}g/ml$ were mixed with aqueous nematode solution containing about 20 Rhabditis sp. and their activity was examined daily for 7 days. Out of 2,714 plant extracts examined in this test, 2,362 (87.0%) showed no negative influence on the nematode activity, while 187 (6.9%) inhibited nematode activity about 50%, 95 (3.5%) inhibited nematode activity over 90%, and 70 (2.6%) rather enhanced nematode activity. Among those showing over 50% nemastatic activity, 25 extracts were randomly selected for further screening with Rhabditis sp. and with juveniles of Meloidogyne incognita. The screening revealed that 11 extracts (44%) were consistently nemastatic. The 282 extracts with nemastatic activity could be of use in future studies on nematicidal plants.

Screening of Tissue Papers for Nematode Extraction for the Baermann Funnel Method (Baermann Funnel법의 선충 분리 여과지 선발)

  • The Baermann funnel method requires approximately four Kimwipe tissues for research a nematode count under a stereo microscope. To select more efficient and economical nematode extraction paper for nematode extraction, 15 different kinds of tissue papers were tested and compared with Kimwipe tissues. Nematode species used in the extraction efficiency tests include juvenile (J2) of Heterodera sp., J2 of Meloidogyne sp., Pratylenchus sp., Rhabditis sp., Acrobeloides sp., Panagrolaimus sp., Poikilolaimus sp. and Diplogasterida. The extraction efficiency varied between 42.0 to 88.8%. Considering costs, extraction efficacy, and clarity, the Pulling Kitchen Towel (Monalisa Co., Korea) is the best tissue, with clarity A, isolation efficiency of 69.4% (not significantly different compared to Kimwipe 1 ply 88.8%), and 1/2 costs per isolation of Kimwipe 2 ply.

Comparison of Predacity of Nematode Predatory Fungi against Meloidogyne incognita (국내 분리 포식성곰팡이들의 고구마뿌리혹선충에 대한 포식 능력 비교)

  • Fifty-two nematode predatory fungi were isolated from 37 soil samples collected from eight provinces in Korea. Isolated fungi were tested their predacity against Rhabditis sp. and Meloidogyne incognita in petri dish, and against M. incognita in greenhouse pot experiments. Fifty isolates had trapping organ of adhesive networks and two isolates had adhesive column or adhesive knob. In petri dish experiments, 5 1 isolates against Rhubditis sp. and 26 isolates against M. incognita showed over 91 % of predacity; in greenhouse experiments, however, only three isolates showed over 81% of predacity. These results imply that the results from the laboratory experiments are not consistent with those from the greenhouse experiments. Therefore, to select a promising biocontrol predatory fungi for plant-parasitic nematodes, the screening experiment should be conducted in conditions close to nature.

Status of intestinal parasite infections among children in Bat Dambang, Cambodia

  • A survey was conducted to determine the extent of intestinal parasite infection in Bat Dambang, Cambodia in March 2004. A total of 623 fecal specimens was collected from kindergarten and schoolchildren and examined using the formalin-ether sedimentation technique. The overall infection rate of intestinal parasites was 25.7% (boys, 26.2%; girls, 25.1%), and the infection rates of intestinal helminthes by species were as follows: Echinostoma sp. 4.8%, hookworm 3.4%, Hymenolepis nana 1.3%, and Rhabditis sp. 1.3%. The infection rates of intestinal protozoa were; Entamoeba coli 4.8%, Giardia lamblia 2.9%, Iodamoeba butschlii 1.4%, Entamoeba polecki 1.1 %, and Entamoeba histolytica 0.8%. There were no egg positive cases of Ascaris lumbricoides or Trichuris trichiura. All children infected were treated with albendazole, praziquantel, or metronidazole according to parasite species. The results showed that intestinal parasites are highly endemic in Bat Dambang, Cambodia.

  • A survey was made to find the extent of intestinal parasite infection in Kampongcham, Cambodia in February 2002. A total of 251 fecal specimens were collected from Tonlebat primary school children and examined by formalin-ether sedimentation technique. The overall infection rate of intestinal parasite was 54.2% (males, 57.3%; females, 50.8%). The infection rate of intestinal helminths by the species were as follows: Ascaris lumbricoides 26.3%, Echinostoma sp. 15.6%, hookworm 6.4%, Opisthorchis sp. 4.0%, Rhabditis sp. 2.4%, and Trichuris trichiura 0.4%. The infection rate of intestinal protozoa were as follows: E. coli 7.6%, G. lamblia 3.2%, I. butschlii 3.2%, and E. histolytica 0.8%. More than two different kinds of parasites were found in 16.7% of the stool samples. All the children infected were treated with albendazole, praziquantel and metronidazole according to parasite species. The results showed that intestinal parasites are highly endemic in this area.
