Simultaneous Determination of Cinnamaldehyde and Coumarin in Oryeong-san using HPLC with Photodiode Array Detector

  • Objectives : To develop and validate High-performance liquid chromatography-photodiode array methods for simultaneous determination of two constituents in Oryeong-san(ORS). Methods : Reverse-phase chromatography using a Gemini C18 column operating at $40^{\circ}C$, and photodiode array(PDA) detection at 280 nm, were used for quantification of the two marker components of ORS. The mobile phase using a gradient flow consisted of two solvent systems. Solvent A was $H_2O$ and solvent B was acetonitrile. Results : Calibration curves were acquired with correlation coefficient ($r^2$)>0.9999, and the relative standard deviation(RSD) values(%) for intra- and inter-day precision were not exceed 1.0%. The recovery rate of each compound was in the range of 93.01-104.16%, with an RSD less than 2.0%. The contents of two compounds in ORS were 1.10-3.72 mg/g. Conclusions : The established HPLC method will be helpful to improve quality control of ORS.

  • Acyclic polyether dicarboxylic acid have been studied as metal cation carriers in a bulk liquid membrane system. The proton-ionizable ligands feature allows the coupling of a cation transport to reverse proton transport. This feature offers promise for the effective separation and concentration of metal cations with the metal cation transport being driven by a pH gradient. Metal cation transport increased regularly with increasing hydroxide($H^-$) concentration of source phase and with proton($H^+$) concentration of receiving phase. Competitive transport by the acyclic polyether dicarboxylic acids is selective for calcium ion over other alkaline-earth cations.

Determination of Authentic Chiisanoside in Acanthopanax senticousus by High Performance Liquid Chromatography

  • High performance liquid chromatography (HPLC) was used for the analysis of chiisanoside in each stem and root of Acanthopanax senticosus collected from South Korea, North Korea, China and Russia. A reverse-phase system using a gradient of H$_{2}$O and acetonitrile as the mobile phase was developed and detection was at 210nm. The analysis was successfully carried out within 30 min. Chiisanoside was measured in the stem and root of A. senticosus collected from various countries.

  • High performance liquid chromatography (HPLC) was used for the analysis of chiisanoside in Acanthopanax species. A reverse-phase system using a gradient of H$_2$O and acetonitrile as the mobile phase was developed and a wavelength of detection was at 210 nm. The analysis was successfully carried out for 30 min. Chiisanoside was measured in the fruit, stem and root of A. sessiliflorus, A. koreanus, A. divaricatus and A. senticosus.

  • Qantitative analysis of six compounds: (+)-catechin, benzoic acid, gallic acid methyl ester, paeonol, paeoniflorin, and albiflorin from Paeonia lactiflora extracts was performed using high-performance liquid chromatography and an ultraviolet (UV) detector, following different extraction methods. A reverse-phase column was used in a gradient elution system, and UV detection was performed at 280 nm. The results showed that the quantity of paeoniflorin was the highest in ethanol and water extracts (73.89 and 57.87 mg/g, respectively) among the six compounds. This study contributes a good analysis method for the contents of P. lactiflora and would be propitious for developing medicines and functional foods.

  • Let L = -∆ + V be a Schrödinger operator, where the potential V belongs to the reverse Hölder class. In this paper, by the subordinative formula, we investigate the generalized Poisson operator PLt,σ, 0 < σ < 1, associated with L. We estimate the gradient and the time-fractional derivatives of the kernel of PLt,σ, respectively. As an application, we establish a Carleson measure characterization of the Campanato type space 𝒞𝛄L (ℝn) via PLt,σ.

  • This study explained the effects of radiation, magnetic field, and nanoparticle shape on the peristaltic flow of an Upper-Convected Maxwell nanofluid through a porous medium in an asymmetric channel for a better understanding of cooling and heating mechanisms in the presence of magnetic fields. These phenomena are modeled mathematically as a system of non-linear differential equations, that are solved under long-wavelength approximation and low Reynolds number conditions using the perturbation method. The results for nanofluid and temperature described the behavior of the pumping characteristics during their interaction with (the vertical position, thermal radiation, the shape of the nanoparticle, and the magnetic field) analytically and explained graphically. Also, the combined effects of thermal radiation parameters and some physical parameters on pressure rise, pressure gradient, velocity, and heat distribution are pointed out. Qualitatively, a reverse velocity appears with combined high radiation and Grashof number or combined high radiation and low volume flow rate. At high radiation, the spherical nanoparticle shape has the greatest effect on heat distribution.

Isolation and Identification of Inhibitory Compounds from Morus alba cv. Kuksang on α-amylase and α-glucosidase (국상(Kuksang) 뽕잎(Morus alba L.)으로부터 α-amylase와 α-glucosidase 저해 물질 분리 및 동정)

  • The objective of this research was to evaluate the inhibitory activities of phenolic compounds isolated from mulberry (Morus alba) leaves of 109 types against α-amylase and α-glucosidase. The inhibitory activity of the water extracts from Morus alba cv. Kuksang against α-amylase and α-glucosidase were determined as 93.8% and 48.7% respectively. The total phenolic content of extracts from Morus alba cv. Kuksang was 9.7±0.2 mg/g soluble in water and 14.3±0.2 mg/g soluble in ethanol. The inhibitory activity of the water extracts from Morus alba cv. Kuksang at 200 μg/ml phenolics concentration against α-amylase and α-glucosidase were determined as 100% and 82.6% respectively. The purification of inhibitory compounds was carried out by Sephadex LH-20 and MCI-gel CHP-20 column chromatography using a gradient elution procedure by nomal phase type (EtOH→distilled water) and reverse phase type (distilled water→MeOH). The quercetin was confirmed to be the chemical structure of the inhibitory compound against α-amylase and α-glucosidase by spectroscopic analysis of FAB-MS, NMR and IR spectrum.

Purification of Band 3 from the Human Erythrocyte Membrane and its Incorporation into Liposome (사람 적혈구막 Band 3의 정제 및 Liposome으로의 도입)

  • Band 3, the predominent 95,000 dalton anion transport protein, is the major intrinsic glycoprotein of the human erythrocyte membrane. This anion carrier exists as a dimer and binds the cytoskeletons such as spectrin, ankyrin and actin. And the liposomes are vesicular structures which form spontaneouly upon hydration of phospholipids. These artificial lipid vesicles have been investigated as model of the biological membranes and as a mean of improving the delivery of nucleic acids, drugs, proteins and biological substances to specific target tissues and cells. In this study, we were purified Band 3 from the human erythrocyte membrane(ghost) was prepared by hemolysis of intact human erythrocyte with weak alkali-hypotonic solution. Band 6 was removed from ghost by extracting with solution of an ionic strength of 0.15. Band 3 and Band 4 were solubilized selectively by extracting Band 6-depleted ghosts with Triton X-100 under nondenaturing conditions. Band 3 was then purified from Triton X-100 extract treated with p-chloromercuribenzoate by sucrose density gradient ultracentrifugation. This purified Band 3 was incorporated into liposomes prepared by reverse-phase evaporation. Phosphatidyl L-serine and cholesterol(1 : 1 molar ratio) were dissolved in chloroform and then chloroform was removed by rotatory evaporation under reduced pressure. Band 3 solution without Triton X-100 was introduced into a mixture of lipids and diethylether. Diethylether was subsequently removed by evaporation. This purified Band 3 and its incorporation into liposomes were confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

An Efficient Pixel Value Prediction Algorithm using the Similarity and Edge Characteristics Existing in Neighboring Pixels Scanned in Inverse s-order (역 s-순으로 스캔된 주변 픽셀들에 존재하는 유사성과 에지 특성을 이용한 효율적인 픽셀 값 예측 기법)

  • In this paper, we propose an efficient pixel value prediction algorithm that can accurately predict pixel value using neighboring pixel values scanned in reverse s-order in the image. Generally, image has similarity with similar values between adjacent pixel values, and may have directional edge characteristics. In this paper, we proposed a method to improve pixel value prediction accuracy by improving GAP(Gradient Adjacent Pixel) algorithm for predicting pixel value by using similarity between adjacent pixels and edge characteristics. The proposed method increases the accuracy of the predicted pixel value by precisely predicting the pixel value using the positional weights of the neighboring pixels. Experiments on real images confirmed the superiority of the proposed algorithm. The proposed algorithm is useful for applications such as reversible data hiding, reversible watermarking, and data compression applications.
