Background: Methyl donor status influences DNA stability and DNA methylation although little is known about effects on DNA methyltransferases. The aim of this study was to determine whether methyl-donor status influences DNA methyltransferase (Dnmt) gene expression in cervical cancer cells, and if so, whether there are associated effects on global DNA methylation. Materials and Methods: The human cervical cancer cell line, C4-II, was grown in complete medium and medium depleted of folate (F-M+) and folate and methionine (F-M-). Growth rate, intracellular folate, intracellular methionine and homocysteine in the extracellular medium were measured to validate the cancer cell model of methyl donor depletion. Dnmt expression was measured by qRT-PCR using relative quantification and global DNA methylation was measured using a flow cytometric method. Results: Intracellular folate and methionine concentrations were significantly reduced after growth in depleted media. Growth rate was also reduced in response to methyl donor depletion. Extracellular homocysteine was raised compared with controls, indicating disturbance to the methyl cycle. Combined folate and methionine depletion led to a significant down-regulation of Dnmt3a and Dnmt3b; this was associated with an 18% reduction in global DNA methylation compared with controls. Effects of folate and methionine depletion on Dnmt3a and 3b expression were reversed by transferring depleted cells to complete medium. Conclusions: Methyl donor status can evidently influence expression of Dnmts in cervical cancer cells, which is associated with DNA global hypomethylation. Effects on Dnmt expression are reversible, suggesting reversible modulating effects of dietary methyl donor intake on gene expression, which may be relevant for cancer progression.

This study was conducted to examine the effects of adding phytoncide oil extracted from Korean pine nut cone by-product to the diet of dairy cows on milk yield and compositions, fatty acid characteristics, complete blood count and stress response. A total of 74 Holstein cows were used for 30 days and divided into two groups. Each group was given a basal diet (C) or an experimental diet containing phytoncide additives at 0.016% (T) in feed. The results showed that phytoncide feeding had no effect on milk yield. In addition, there were no observed effects on milk composition, but the ratio of fatty acid in milk was significantly affected by the phytoncide diet, and it showed a positive effect. Not only were the major functional fatty acids, conjugated linoleic acid and eicosapentaenoic acid increased, but also ${\omega}6:{\omega}3$ fatty acid ratio was reduced in milk of T group (p<0.05). In blood analysis, the complete blood count showed no significant difference between C and T group on all parameters. However, the cortisol concentration was significantly decreased in T group compared to control (p<0.05). Taken together, we suggest that phytoncide oil does not have a great influence on the physiological changes, but can be a potential feed additive that improves the milk fatty acid and stress resilience in dairy cows. In addition, it will contribute to the development of feed resource, a reduction in feed cost and a lessening of environmental pollution.

The protective effect of zinc against cortisol-induced cell injury was examined in rainbow trout gill epithelial cells. Cells exposed to cortisol for 24 h showed increased leakage of lactate dehydrogenase (LDH) as well as decreased cell viability in a dose-dependent manner. Treatment with zinc ($100{\mu}M$$ZnSO_4$) reduced the severity of both LDH release and cell death as well as protected cells against cortisol-induced caspase-3 activation, indicating reduction of apoptosis. Cortisol-induced cell death, leakage of LDH, and caspase-3 activation were blocked by the glucocorticoid receptor antagonist Mifepristone (RU-486), suggesting that cell injury was cortisol-dependent. In addition, we studied the effect of zinc on the expression of antioxidant genes such as metallothionein A (MTA), metallothionein B (MTB), glutathione-S-transferase (GST), and glucose-6-phosphate dehydrogenase (G6PD) during cortisol-induced cell injury. MTA, MTB, GST, and G6PD mRNA levels increased after treatment with zinc or cortisol, separately or in combination. Higher mRNA levels of MTA, MTB, GST, and G6PD were detected when cells were treated with $100{\mu}M$$ZnSO_4$ and $1{\mu}M$ cortisol in combination at the same time compared to treatment with zinc or cortisol separately. Cells treated with zinc showed increased intracellular free zinc concentrations, and this response was significantly enhanced in cells treated with cortisol and zinc. In conclusion, zinc treatment inhibited cortisol-induced cytotoxicity and apoptosis through indirect antioxidant action.

The risk assessment is the qualitative or quantitative evaluation of the risk posed to human health and the environment by the actual or potential presence or release of hazardous substances, pollutants or contaminants. The environmental impact assessment (EIA) is assessed by the environmental criteria, and risk assessment is assessed by the risk rate. Risk rate based on dose-response values may not be easy to apply on regulatory basis like EIA for uncertainty. Internationally there is an example of OECD program. Risk assessment of High Production Volume (HPV) Chemicals has started since the OECD Program with the 1990 Council Act on the Co-operative Investigation and Risk Reduction of Existing Chemicals. These HPV chemicals include all chemicals produced or imported at levels greater than 1,000 tonnes per year in at least one Member country or in the European Union region. The SIDS called the Screening Information Data Set is regarded as the minimum information needed to assess an HPV chemical to determine whether any further work should be carried out or not. All the data elements of SIDS including assessment for environment and health are prepared as three formats of the full SIDS Dossier, the SIDS Initial Assessment Report (SIAR), and the SIDS Initial Assessment Profile (SIAP) of an HPV chemical. In 1998 the global chemical industry through the International Council of Chemical Associations (ICCA) has joined to work with OECD. The OECD has assessed approximately 1,000 chemicals from 1991 through 2004 with ICCA. Till the February of 2005, 592 chemicals of those chemicals completed SIDS reports. Member countries have been targeted the goal of 1,000 new chemicals from 2005 to 2010 and Korea shared 36 chemicals from the 1,000 new chemicals. Currently Korea has completed SIDS reports of 7 chemicals among sponsored 24 chemicals. In conclusion SIDS project will be linked to national program for outputs application with more reliable production. Both the OECD and industry will carry out their commitment to complete assessments for more and the remaining chemicals assessment. The major outputs will contribute to cope with international chemical management.

Soyangin-Hyeongbangpaedok-san(SHBPDS) is a herbal formula used for the common cold or upper respiratory illness. In order to investigate the effect of SHBPDS, mice were orally administered with SHBPDS alcohol extract for 7 days followed by intravenous anti-CD3 injection. In addition, splenocytes and CD4 T cells were cultured with SHBPDS in response to anti-CD3 in vitro and cytokines and transcription factors were evaluated. In vivo treatment with SHBPDS significantly augmented the expressions of the percentage of CD4 T cells and CD 69, an indicator of early T cell activation. Serum levels of IL-4 were significantly increased but those of IFN-${\gamma}$ and IL-2 did not reach statistical significance. The expressions of IFN-${\gamma}$ and T-bet mRNA were significantly downregulated in SHBPDS treated mice while those of IL-4 and C-Maf were significantly upregulated. In vitro stimulation of splenocytes and CD4 T cells by SHBPDS resulted in a reduction in IFN-${\gamma}$ secretion and STAT4 activity. The IL-4 releases from both cells were slightly reduced, but STAT6 activity was rather increased. In conclusion, SHBPDS exerted an inhibition in the expression of IFN-${\gamma}$, T-bet and STAT4 while IL-4, C-Maf and STAT6 were increased. Further studies are required to examine its pharmacological effects using more appropriate animal experiments.
Compton suppression apparatus using the Compton scattering response, by inhibiting part of the spectrum Compton continuum Compton continuum in the area of the peak analysis of the gamma rays that enables a clearer device. In order to find out the geometry structure of high-purity germanium detector(HPGe) -NaI(TI) and to optimize the effect of movement, Monte Carlo simulation was used to grasp the behavioral characteristics of Compton suppression and compare several layout structures. And applied to the cylinder beaker used for the environmental measurement by using the efficiency according to the distance. For the low-energy source such as 81 keV, the Compton continuum is scarcely developed and the suppression effect is also insignificant because the scattering cross-section of the Compton effect is relatively low. In the spectrum for the remaining energy, it can be seen that the Compton continuum part is suppressed in a certain energy range. Compton suppression effect was not significantly different from positional shift. average reduction factor(ARF) value was about 1.08 for 81 keV and about 1.23 for 1332.4keV energy at the highest value. It can be seen that suppression over the Compton continuum region of the energy spectrum is a more appropriate arrangement. Therefore, it can be applied to various environmental sample measurement through optimized structure.
In an effort to evaluate Salmonella food safety using combinations of preservation techniques, its viabilities when exposed to HCl, acetic acid, and the oxidative agents (hydrogen peroxide and butyl hydrogen peroxide), were analyzed using sub-lethal heat-shocked Salmonella Typhimurium at $56^{\circ}C$. 2D gel electrophoresis and MALDI-TOF MS analyses were also conducted to determine the expression and repression of proteins in heat-shocked cells. Heat-shocked S. Typhimurium evidenced a reduction of viable counts by 1-2 log CFU/mL. However, viality of non heat-shocked S. Typhimurium decreased markedly by 5-6 log CFU/mL at a pH 4 in response to acid and oxidative stresses. Sub-lethal heat treatment greatly increased the resistance of S. Typhimurium against acid and oxidant agents. As for 2D gel electrophoresis and protein identification via MALDI-TOF MS, 17 major proteins in non heat-shocked S. Typhimurium were detected, and only 13 proteins among these proteins were detected in heat-shocked S. Typhimurium. The heat shock proteins such as DnaK and small heat shock proteins were included, and may be associated with the resistance of S. typhimurium against exposure to acids and oxidants. Therefore, even though the promising hurdle technology using the combined mild treatments including heat was applied to S. Typhimurium, the proper heat treatment to reduce its crossprotection activity toward the following preservative agents might be considered.

This study was conducted to investigate the effects of disregarding of heat detection on embryo production in superovulation of Hanwoo cows. Donors which showed 1 or 2 times of normal heat and had no abnormality of reproductive tract were selected The superovulation was performed injection of 2.5 mL FSH (Antorin R-10, Japan) 2 times on 5 days before next heat and continuously with the reduction of dose to 0.5 mL of first injection 2 times in a day for next 3 days. Otherwise, the donors of CIDR group were inserted CIDR plus (with the capsule of estradiol benzoate 10 mg) on Day 10 from standing heat for 9 days. On 6 days from insertion of CIDR, FSH was injected above same manners. The response according to the natural heat and CIDR were $82.2\%,\;89.7\%$, respectively. There were no difference between both treatments. The recovery rates of embryos were 7.7, 10.5, respectively and transferable embryos were 3.4, 3.8. There showed significant difference between both treatments (p<0.05). These results suggested that disregarding of heat detection in superovulation could be produced transferable embryos for embryo transfer and preserve the donors from the excess hormonal administration and maintain the economical lift span of genetically available Hanwoo donors.
Failure modes result in fracture or tearing, which may cause deterioration of resistance and reduction of inelastic deformation capacity. The potential failure modes for Special Concentrically Braced Frames (SCBFs) include fracture or tearing of the brace, net section fracture of the brace or gusset plate, fracture of the gusset plate welds, shear fracture of the bolts, block shear, excessive bolt bearing deformation, and buckling of the gusset plate. HSS tubular braces are commonly used in SCBFs, and net section fracture of the tubular brace may also occur through the brace net section at the end of the slot cut into the tube to slip over the gusset plate. This failure mode is categorized as a tension failure mode, and may cause dramatic loss of resistance and brittle behavior. Net section reinforcement is required according to AISC design specifications (AISC 2001). In this paper, the need to reinforce the net section area was discussed. Initially, the results of the net section fracture tests done by the University of California in Berkeley were presented with the modeling of these tests using FE models. To investigate the possibility of net section fracture in an actual frame, the slot end hole model was adapted to the frame FE model, and alternate near-fault histories were applied with tension-dominated cycles, since previous analyses showed that loading history was the most critical factor in net section fracture. The need for this reinforcement (cover plate) and the tension-dominated near-fault history were investigated.
Five different levels of Ethephon were treated on tobacco cultivar yellow Special A and Coker and harvested for curing. Changes in dehydration properties, yellowing, chlorophyll, nicotine, sugar and starch contents during the processes of curing were studied in order to justify feasibility of adoption an idea of 3-stage-curing method instead of the 5-stage. Ethephon-treated-tobacco leaves showed marked degradation of chlorophyll, greater dehydration and greater reduction in nicotine contents while little change in either sugar, starch and other chemical components. They showed also shorter hours of curing period allowing possible curing response at a relatively low temperature than those without treatment. Thus they undergo only 3 stages of flue-curing processes, yellowing, calor-fixing and vein-drying, and need 72 hours to finish curing, but the ordinary leaves need 100 hours for it.
