Study of stabilizing and efficacy evaluation in human of Oleanoic acid with poly-glyceryl nano emulsion system (올레아노익산의 폴리글리세릴계 나노에멀젼에서의 안정화 및 인체적용 유효성평가에 대한 연구)

  • Oleanolic acid is known as which anti-cancer, anti-sinhaeng angiogenic, anti-inflammatory, antioxidant and anti-wrinkle effects. We focused on the antioxidant activity of oleanolic acid was separated from the natural plant and It was confirmed that the whitening effect. In this study, oleanolic acid was stabilized by polyglyceryl surfactant which from natural origin with only a simple stirring operation, and compared with lecithin liposome that was manufactured with high cost facility. The transdermal transition rate of 0.4% oleanolic acid polyglyceryl nanoemulsion was 95%, and it was simillar with lecithin liposome of 92%. 65% of 3hr transdermal transition rate of polyglyceryl nanoemulsion indicate charistiristcs of quick release, compared with 45% of lecithin liposome's 3hr transdermal transition rate. In the in-vivo clinical trial test, polyglyceryl nanoemulsion of 0.4% oleanolic acid was higher 25% in 2nd week, 58% in 4th and 8th weeks than non-added oleanolic acid emulsion.

Effect of Cassiae semen extract on ovalbumin-induced allergic asthma in mice (결명자 추출물의 난알부민 감작으로 천식이 유발된 마우스에서의 개선 효과)

  • Objectives : In this study, we investigated the effect of Cassia obtusifolia Linne (Cassiae Semen; CS) extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : CR was extracted with 70% ethanol. For in vitro study, HMC-1, human mast cells were treated with CS extract at 0.2 and $0.5mg/m{\ell}$ for 1 h, and then stimulated with compound (C) 48/80 for 30 min. Primary spleenocytes were isolated from the spleen of mice, treated with CS extract for 1 h, and then stimulated with ConA for 24 h. For in vivo study, mice were sensitized at day 0, 7 and 14 with 0.2% OVA and then airway challenged using neublizer at day 21, 23, 25, and 27 to induced allergic asthma. CS extract at doses of 100 and 300 mg/kg body weight was orally administered during OVA challenge once per a day. The levels of allergic mediators such as histamine, OVA-specific IgE, IL-4, and $IFN-{\gamma}$ were measured in the sera of mice or culture supernatants by EIA and ELISA, respectively. The expression of IL-4 and $IFN-{\gamma}$ gene was determined by RT-PCR. The histopathological change of lung tissues was observed with hematoxylin and eosin (H&E) and Periodic acid Schiff (PAS) staining. Results : The treatment of CS extract in HMC-1 cells significantly inhibited C48/80-induced degranulation, and histamine release. The treatment of CS extract in spleenocytes suppressed the expression of IL-4 and $IFN-{\gamma}$ mRNA. The administration of CS extract in OVA-induced asthmatic mice significantly decreased the levels of OVA-specific IgE, and IL-4 in a dose-dependent manner with OVA-control group. In addition, CS extract inhibited the infiltration of inflammatory cells and bronchiolar damage with epithelial thickening in lung tissues of OVA-induced asthma mice, and also mucin accumulation. Conclusions : These results indicate that CS extract prevents asthmatic damage through regulating the allergic immune response.

Power Generating Characteristics of Zinc Oxide Nanorods Grown on a Flexible Substrate by a Hydrothermal Method

  • This paper describes the power generating property of hydrothermally grown ZnO nanorods on a flexible polyethersulfone (PES) substrate. The piezoelectric currents generated by the ZnO nanorods were measured when bending the ZnO nanorod by using I-AFM, and the measured piezoelectric currents ranged from 60 to 100 pA. When the PtIr coated tip bends a ZnO nanorod, piezoelectrical asymmetric potential is created on the nanorod surface. The Schottky barrier at the ZnO-metal interface accumulates elecntrons and then release very quickly generating the currents when the tip moves from tensile to compressed part of ZnO nanorod. These ZnO nanorods were grown almost vertically with the length of 300-500 nm and the diameter of 30-60 nm on the Ag/Ti/PES substrate at $90^{\circ}C$ for 6 hours by hydrothermal method. The metal-semiconductor interface property was evaluated by using a HP 4145B Semiconductor Parameter Analyzer and the piezoelectric effect of the ZnO nanorods were evaluated by using an I-AFM. From the measured I-V characteristics, it was observed that ZnO-Ag and ZnO-Au metal-semiconductor interfaces showed an ohmic and a Schottky contact characteristics, respectively. ANSYS finite element simulation was performed in order to understand the power generation mechanism of the ZnO nanorods under applied external stress theoretically.

  • Drug delivery technologies are patent protected formulation technologies that modify drug release profile, absorption, distribution, and elimination for the benefit of improving product efficacy and safety, as well as patient convenience and compliance. The most commonly used transdermal system is the skin patch using various types of technologies. Compared with other method of dosage, it is possible to use for a long term. It is also possible to stop the drug dosage are stop if the drug dosage lead to side effect. Polysaccharide, such as karaya gum and locust bean gum(LBG)/water-soluble chitosan oligomer(WSCO) were selected as base materials of TDS. Also, these polymers were characterized in terms of enhancers, tacrine contents. Among these polysaccharide, the permeation rate of karaya gum matrix was fastest in tacrine such as lipophilic drug in vitro. We used glycerin, PEG 400, and PEG 800 as enhancers. Therefore, transdermal absorption of tacrine could be improved by changing vehicle composition or by using penetration enhancers. Especially it would be anticipated that the high permeation efficacy could be obtained by using vehicle that has enhancing effect for itself and by adding enhancers to it.

  • The effect of equivalence ratio and fuel/air mixing quality on the phase-resolved gas temperatures at different phases of the oscillating pressure cycle was experimentally investigated. An atmospheric pressure, optically accessible and laboratory-scale dump combustor operating on methane with heat release rate of 1.59kW was used. Temperature measurements were made using coherent anti-Stokes Raman spectroscopy (CARS) at several spatial locations for typical unstable combustion conditions. Analysis was conducted using parameters such as phase-resolved averaged temperature, normalized standard deviation and temperature probability distribution functions (PDFs). Also the probability on the occurrence of high temperature (over 1900K) was investigated to get the information on the perturbation of equivalence ratio and NOx emission characteristics. It was shown that most of temperature histograms exhibit Gaussian profile which has short breadth of temperature fluctuation at equivalence ratio of 0.6, while beta profile was predominant for the cases of other equivalence ratios (${\Phi}$=0.55, 0.50). The characteristics on the occurrence of high temperature also displayed periodic wave form which is very similar to the pressure signal. And the amplitude of this profile goes larger as the fuel/air mixing quality become poorer. These also provided additional information on the perturbation of equivalence ratio at flame as well as NOx emission characteristics.

The Anti-Inflammatory Effect of Pegmatite by in Vivo and in Vitro Study (In vivo 및 in vitro 시험을 통한 페그마타이트의 항염증 효과)

  • Purpose: This work aimed to elucidate the anti-inflammatory effect of pegmatite in vitro and in vivo. Methods: Author evaluated the suppressive effects of pegmatite on lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production, TNF-${\alpha}$ and IL-6 release in the RAW 264.7 murinemacrophages. Results: Treatment of RAW 264.7 cells with pegmatite significantly reduced LPS-stimulated NO production and inflammatory cytokine such as TNF-${\alpha}$ and IL-6 secretion in a concentration-dependent manner. Also pegmatite showed topical anti-inflammatory activity in the arachidonic acid (AA)-induced ear edema and acetic acid-induced increase in capillary permeability assessment in mice. It was also found that pegmatite (10 mg per ear in DW) inhibited arachidonic acid induced edema at 24 h more profoundly than 1 h by topical application. Furthermore, the vascular permeability increase induced by acetic acid was significantly reduced in mice that received pegmatite in 50 mg per mouse. Conclusion: Therefore the results of the present study suggest that pegmatite is a potent inhibitor of the LPS-induced NO and inflammatory cytokine in RAW 264.7 macrophages and showed anti-inflammatory activities in vivo animal model.

  • TALP-32 is highly basic protein with a molecular weight of 32 kDa purified from human term placenta. Some basic proteins such as defensins and cecropins are known to induce cell death by increasing membrane permeability and some of them are under development as an anticancer drug especially targeting multi-drug resistant cancers. Therefore, we investigated cytotoxic effect and mechanism of TALP-32 When HeLa cell was incubated with TALP-32, cytotoxicity was increased in time and dose dependent manner. As time goes by, HeLa cells became round and plasma membrane was ruptured. Increase of plasma membrane permeability was determined with LDH release assay. Also in transmission electron microscopy, typical morphology of necrotic cell death, such as cell swelling and intracellular organelle disruption was observed, but DNA fragmentation and caspase activation was not. And necrotic cell death was determined with Annexin V/Pl staining. The cytotoxicity of TALP-32 was minimal and decreased or RBC and Hep3B respectively. These data suggests that TALP-32 induces necrosis on rapidly growing cells but not on slowly growing cells implicating the possibility of its development of anticancer peptide drug.

Antiallergic Effect of Two Variants of Artemisia princeps Pampanini

  • Antiallergic activities of two variants of Artemisia princeps Pampanini SJ-1 (named as Sajabalssuk) and SS-1 (named as Sajuarissuk) cultivated in Ganghwado, which contain high content of eupatilin compared to those cultured by other places, were investigated to evaluate the possibility as inhibitors against allergic diseases. Ethanol and supercritical fluid extracts of SJ-1 and SS-1 inhibited the release of ${\beta}-hexosaminidase$ from RBL-2H3 cells, although their water extracts were inactive. These extracts potently inhibited lipopolysaccharide-induced NO production of RAW264.7. However, these extracts almost did not scavenge free radicals. Oral administration of these extracts to mice inhibited passive cutaneous anaphylaxis reaction induced by IgE, and acute dermatitis induced by 12-O-tetradecanoylphorbol-13-acetate. However, these extracts did not inhibit chronic dermatitis. Scratching behaviors, vascular permeability, and writhing syndromes were weakly inhibited by these extract at a dose of 50 mg/kg. Based on these findings, we believe that SJ-1 and SS-1 can improve IgE-induced allergic diseases such as rhinitis and asthma.

Effects of Na-K Pump Inhibition on Contractility of Resistant Arteries in the Rabbit (저항동맥의 수축성에 대한 연구)

  • Recently endogenous digitalis-like substances were found in the blood of various cardiovascular diseases and they have been considered one of the causes of evoking hypertension. However, the mechanism of endogenous digitalis-like substances-induced hypertension is not clarified yet. Therefore, the effects of Na-K pump inhibition on the contractility of vascular smooth muscle[conduit and resistant artery were investigated, using organ bath and bioassay experiment. Aortic and carotid arterial rings[conduit artery and the branches of brachial and superior mesenteric artery[resistant artery were used to find the effect of Na-K pump inhibition. The results obtained were as followes;The magnitudes of contractions induced by norepinephrine, serotonin, or acetylcholine in all these arteries were significantly increased by the inhibition of Na-K pump. The increased contractile responses to these agonists, especially to serotonin, were much more prominant in resistant arteries. Nitroprusside-induced relaxations were attenuated by Na-K pump inhibition and there were no significant differences in the effects of Na-K pump inhibition on nitroprusside-induced relaxations of these blood vessels. Endothelium-dependent relaxation was suppressed by the inhibition of Na-K pump, especially by the administration of ouabain, and this inhibitory effect was much more prominent in the branches of superior mesenteric artery, compared with other arteries. In the branches of superior mesenteric arteries, endothelium-dependent relaxation was completely blocked by ouabain. The release of EDRF was partially suppressed by Na-K pump inhibition.From the above results, it is suggested that the hypertension due to the increase in vascular resistance can be evoked by the inhibition of Na-K pump and endogenous digitalis-like substances induce hypertension through this mechanism.

Cartilage and Bone Protection Effect of AIF, a water extract from three herbs (세가지 생약복합추출물, AIF의 연골 및 뼈 보호 효능)

  • AIF has been formulated using three herbs known to have anti-inflammatory and anti-osteolytic effects. In this study, the potential therapeutic effects of AIF for osteoarthritis were assessed in vitro and in vivo. The effects of AIF on the cartilage and bone protection (MMP-13 expression, GAG degradation, OPG release) were examined, in vitro. In addition, the therapeutic effect of AIF was evaluated using a chemical-induced osteoarthritis rat model. Rats were injected with iodoacetate intraarticularly in one knee joint and treated with the oral administration of 100 mg/kg AIF-glucosamine once a day for 3 weeks. And then, destruction of cartilage and bone was evaluated by histopathological assessment. AIF significantly inhibited the production of MMP13 and GAG in a dose dependent manner in vitro. Also, AIF increased the production of OPG. In OA rat model, the AIF-glucosamine treated group reduced cartilage destruction, compared to vehicle or glucosamine treated group. AIF showed potent protective effects for the destruction of cartilage and bone, in vitro and in vivo. These results suggest that AIF contains effective compound(s) which may modify the progression of arthritis.
