Development of Primary Reference Gas Mixtures for Liquid Propane (혼합 액체 프로판 표준가스 개발)

  • Liquefied Petroleum Gas is divided into liquefied gases containing propane (C3H8) and butane (C4H10). The quality of LPG varies greatly depending on the composition of the mixture, so it is important to measure the composition accurately. It is difficult to determine the composition of the mixture because liquid and gas coexist at room temperature. Therefore, the uncertainty in determining the concentration of hydrocarbons by component is high, and there are many problems that differ from the actual content standard. Therefore, it is necessary to develop a mixed liquid propane standard gas for the composition and accurate concentration of hydrocarbon substances. Mixed liquid propane standard gas is manufactured into bellows-type constant-pressure cylinders by ISO-6142 (2015). The homogeneity of the four standard gases manufactured was confirmed to be GC-FID. The manufacturer's uncertainty of expansion was 0.01 % to 0.30 % and homogeneity was 0.03 % to 0.25 %. In this mixed liquid propane standard gas, the relative expansion uncertainty of weight method, manufacturing consistency, cylinder adsorption and long-term stability was developed within 0.26 %-1.3 9% (95% of confidence level, k=2).

Establishment of a Tm-shift Method for Detection of Cat-Derived Hookworms

  • Melting temperature shift ($T_m-shift$) is a new detection method that analyze the melting curve on real-time PCR thermocycler using SYBR Green I fluorescent dye. To establish a $T_m-shift$ method for the detection of Ancylostoma ceylanicum and A. tubaeforme in cats, specific primers, with GC tail of unequal length attached to their 5' end, were designed based on 2 SNP loci (ITS101 and ITS296) of the internal transcribed spacer 1 (ITS1) sequences. The standard curve of $T_m-shift$ was established using the standard plasmids of A. ceylanicum (AceP) and A. tubaeforme (AtuP). The $T_m-shift$ method stability, sensitivity, and accuracy were tested with reference to the standard curve, and clinical fecal samples were also examined. The results demonstrated that the 2 sets of primers based on the 2 SNPs could accurately distinguish between A. ceylanicum and A. tubaeforme. The coefficient of variation (CV) of $T_m$- values of AceP and AtuP was 0.07% and 0.06% in ITS101 and was 0.06% and 0.08% in ITS296, respectively. The minimum detectable DNA concentration was $5.22{\times}10^{-6}$ and $5.28{\times}10^{-6}ng/{\mu}l$ samples of AceP and AtuP, respectively. The accuracy of $T_m-shift$ method reached 100% based on examination of 10 hookworm DNA samples with known species. In the clinical detection of hookworm in 69 stray cat fecal sample, the $T_m-shift$ detection results were consistent with the microscopic examination and successfully differentiated between the 2-hookworm species. In conclusion, the developed method is a rapid, sensitive and accurate technique and can provide a promising tool for clinical detection and epidemiological investigation of cat-derived hookworms.

A Low Jitter Delay-Locked Loop for Local Clock Skew Compensation (로컬 클록 스큐 보상을 위한 낮은 지터 성능의 지연 고정 루프)

  • In this paper, a low-jitter delay-locked loop that compensates for local clock skew is presented. The proposed DLL consists of a phase splitter, a phase detector(PD), a charge pump, a bias generator, a voltage-controlled delay line(VCDL), and a level converter. The VCDL uses self-biased delay cells using current mode logic(CML) to have insensitive characteristics to temperature and supply noises. The phase splitter generates two reference clocks which are used as the differential inputs of the VCDL. The PD uses the only single clock from the phase splitter because the PD in the proposed circuit uses CMOS logic that consumes less power compared to CML. Therefore, the output of the VCDL is also converted to the rail-to-rail signal by the level converter for the PD as well as the local clock distribution circuit. The proposed circuit has been designed with a $0.13-{\mu}m$ CMOS process. A global CLK with a frequency of 1-GHz is externally applied to the circuit. As a result, after about 19 cycles, the proposed DLL is locked at a point that the control voltage is 597.83mV with the jitter of 1.05ps.

A Study of Shelf Life about Li-ion Battery (리튬 2차 전지의 저장 수명에 관한 연구)

  • In the field of defense, one-shot devices such as missiles are stored for a long period of time after they are manufactured, so it is essential to predict their storage life. A study was conducted to find the shelf life of a Li-ion battery used in one-shot devices. To do this, a Li-ion battery that has been used in weapon systems for more than 5 years was secured. A non-functional test was performed on the battery to check for external changes or failures. After the non-functional test, a discharge test was performed to measure the performance after storing it. Through the test, the performance was checked, including the initial charging voltage, discharge time, and battery temperature, and the trend of the change was identified. An F-test, One-way ANOVA, and regression analysis were performed to verify the aging, and the shelf life of the battery was estimated by an approximation formula that was derived through a regression analysis. As a result of the ANOVA, the p-value was less than the reference value of 0.05, and the performance of the battery decreased by more than 15% after a certain period of time. This change is assumed to result from the change in physical properties of the lithium polymer cell.

The History and Development of the Marine Environment QA/QC (Quality Assurance/Quality Control) Management System (해양환경 정도관리제도 운영에 대한 고찰)

  • The Marine Environment QA/QC management system has been operated since 2010 to secure the reliability of data and improve the analysis capabilities of measurement and analysis institutions. From 2010 to 2020, the cumulative number of measurement and analysis institutions participated in the QA/QC management system was 266. And the number of certificates issued by the ministry of oceans and fisheries is 182. A total of 32 reference materials for proficiency testing and interlaboratory comparisons have been developed. They were first developed focusing on items (Nutrients, COD) commonly analyzed in marine environmental measuring network, marine pollution impact surveys, sea area utilization impact assessment, deepsea water surveys, and information network on fishing ground environments. In addition, it is time to expand the filed of the QA/QC management system, such as seawater temperature, salinity, PCBs and PAHs in sediments, which are mainly analyzed in most monitoring programs. On-site assessment has been conducted for 162 laboratories according to ISO/IEC 17025 to evaluate their conformity of the quality management system and deficiency. In terms of management and technology requirements, about 4.2% of organizations showed insufficient division of duties among employees 8.7% of them revealed the lack of employee training. By test item, about 6.3% of organizations showed the lack of standard substance management and the state of the cleaning glassware was pointed out in about 5.4% of them. The QA/QC management system should be continuously supplemented by identifying the causes of nonconformities and area for improvement.

Electrochemical Deposition Characteristics of Ca2+ on Cu Wire Electrode in CaCl2 Molten Salt (CaCl2 용융염에서 Ca2+의 Cu 전극에 대한 전기화학적 증착 특성평가)

  • With the expansion of the automobile market, the demand for Nd as an essential rare earth material for automobile motors is rapidly increasing. Research on the calcio-thermic reduction process between Nd2O3 and calcium-based alloys has been extensively studied in order to manufacture Nd. In this study, Ca-Cu, as a reducing for Nd2O3, was prepared by electrolysis in CaCl2 molten salt. Cu wire and graphite were employed as a working electrode and a counter electrode for electrolysis reaction, respectively. The reference electrode was manufactured by putting Ag wire in a mixture of AgCl and CaCl2 at a ratio of 1:99 mol%. The cyclic voltammetry results showed that the deposition of Ca2+ on the surface of working electrode was observed from a potential of -1.8 V, and the reduction potential of Ca2+ decreased as the reaction temperature increased. The diffusion coefficient of Ca2+ calculated by the chronoamperometry experiment was found to be 5.4(±6.8)×10-6 cm2/s. In addition, Ca-Cu liquid alloy was prepared by applying a constant potential to Cu electrodes. The element ratio of Ca-Cu alloy formed by applying a potential of -2.0 V was found to Ca:Cu=1:4.

Current Status of X-ray CT Based Non Destructive Characterization of Bentonite as an Engineered Barrier Material (공학적방벽재로서 벤토나이트 거동의 X선 단층촬영 기반 비파괴 특성화 현황)

  • Under high-level radioactive waste repository conditions, bentonite as an engineered barrier material undergoes thermal, hydrological, mechanical, and chemical processes. We report the applications of X-ray Computed Tomography (CT) imaging technique on the characterization and analysis of bentonite over the past decade to provide a reference of the utilization of this technique and the recent research trends. This overview of the X-ray CT technique applications includes the characterization of the bentonite either in pellets or powder form. X-ray imaging has provided a means to extract grain information at the microscale and identify crack networks responsible for the pellets' heterogeneity. Regarding samples of pellets-powder mixtures under hydration, X-ray CT allowed the identification and monitoring of heterogeneous zones throughout the test. Some results showed how zones with pellets only swell faster compared to others composed of pellets and powder. Moreover, the behavior of fissures between grains and bentonite matrix was observed to change under drying and hydrating conditions, tending to close during the former and open during the latter. The development of specializing software has allowed obtaining strain fields from a sequence of images. In more recent works, X-ray CT technique has served to estimate the dry density, water content, and particle displacement at different testing times. Also, when temperature was added to the hydration process of a sample, CT technology offered a way to observe localized and global density changes over time.

A Study of Dyeing Properties of Cotton Fabrics Under Supercritical CO2 Depending on Dyestuff : by C.I. Disperse orange 155, C.I. Disperse red 167 (초임계 유체 염색용 염료에 따른 면 섬유의 염색 특성 : C.I. Disperse orange 155, C.I. Disperse red 167)

  • In this study, the dyeing properties of supercritical fluid dyed cotton fabrics were investigated which use two types of dyes, such as C.I. Disperse orange and C.I. Disperse red 167. Dyeing temperature, pressure and leveling time were equally applied at 130 ℃, 250 bar, and 60 minutes with reference to the related literature, and experiments were performed at concentrations of 0.04, 0.1, 0.4 and 0.8 % o.w.f with different concentrations. Dyeability was confirmed through measurement of washing fastness and color coordinate, and a calibration curve of each dye was drawn up and the absorbance of the residual dye was measured to confirm the amount of residual dye and the dye exhaustion rate at the corresponding concentration. As a result of color difference measurement, as the concentration increased, the L* value decreased and the K/S value increased. However, as the concentration increased, the increase in K/S value decreased compared to the input amount, and this tendency was more obvious in C.I. Disperse red 167 than in C.I. Disperse orange 155. The dye exhaustion rate which was calculated by using the amount of residual dye in the pot was also C.I. Disperse orange 155 was 96.16 % and C.I. Disperse red 167 was 94.57 %. However, as the dyeing concentration increased, the dye exhaustion rate decreased, that C.I. Disperse orange was 95.33 % and C.I. Disperse red 167 was 90.63 %. As a result of the washing fastness test for both dyes, dyed samples of which concentrations were 0.4 and 0.8 % o.w.f decreased by 0.5 ~ 1.0 grade. This is predicted because the dye did not completely adhere to the amorphous region of the cotton fiber and the dye simply adsorbed. The fastness to rubbing also maintained at least grade 3-4 up to the 0.1 % o.w.f concentration, but at the concentration of 0.4 % o.w.f or higher, it fell to grade 1 or lower, showing a very poor friction fastness.

Design and Development of the SNIPE Bus System (초소형위성 SNIPE 본체 설계 및 개발)

  • In this paper, the contents of the design and development process of the 6U micro-satellite Snipe (SNIPE, national name Toyosat; small scale magnetospheric and Ionospheric plasma experiment ), which was developed to observe the near-global space environment through polarization flight for the first time in Korea, were described. Snipe performs transversal flight to observe the Earth's surrounding space environment in three dimensions, and aims to simultaneously observe the space plasma density and temperature in the ionosphere, as well as temporal changes in the solar magnetic field and electromagnetic waves. In this way, it was developed by dividing it into a test certification model (EQM) and a flight model (FM) to perform the actual mission for at least six months, away from developing a cube satellite for short-term space technology verification or manpower training. Currently, Snipe, which has completed the development of a total of four FM and completed all space environment tests, is scheduled to launch 2023. In this paper, we introduce the design contents and development process of the Snipe satellite body ahead of launch, and hope that it will be a useful reference for the development of 6U-class micro-satellite for full-scale mission in Korea.

Development of a lateral flow dipstick test for the detection of 4 strains of Salmonella spp. in animal products and animal production environmental samples based on loop-mediated isothermal amplification

  • Objective: This study aimed to develop loop-mediated isothermal amplification (LAMP) combined with lateral flow dipstick (LFD) and compare it with LAMP-AGE, polymerase chain reaction (PCR), and standard Salmonella culture as reference methods for detecting Salmonella contamination in animal products and animal production environmental samples. Methods: The SalInvA01 primer, derived from the InvA gene and designed as a new probe for LFD detection, was used in developing this study. Adjusting for optimal conditions by temperature, time, and reagent concentration includes evaluating the specificity and limit of detection. The sampling of 120 animal product samples and 350 animal production environmental samples was determined by LAMP-LFD, comparing LAMP-AGE, PCR, and the culture method. Results: Salmonella was amplified using optimal conditions for the LAMP reaction and a DNA probe for LFD at 63℃ for 60 minutes. The specificity test revealed no cross-reactivity with other microorganisms. The limit of detection of LAMP-LFD in pure culture was 3×102 CFU/mL (6 CFU/reaction) and 9.01 pg/μL in genomic DNA. The limit of detection of the LAMP-LFD using artificially inoculated in minced chicken samples with 5 hours of pre-enrichment was 3.4×104 CFU/mL (680 CFU/reaction). For 120 animal product samples, Salmonella was detected by the culture method, LAMP-LFD, LAMP-AGE, and PCR in 10/120 (8.3%). In three hundred fifty animal production environmental samples, Salmonella was detected in 91/350 (26%) by the culture method, equivalent to the detection rates of LAMP-LFD and LAMP-AGE, while PCR achieved 86/350 (24.6%). When comparing sensitivity, specificity, positive predictive value, and accuracy, LAMP-LFD showed the best results at 100%, 95.7%, 86.3%, and 96.6%, respectively. For Kappa index of LAMP-LFD, indicated nearly perfect agreement with culture method. Conclusion: The LAMP-LFD Salmonella detection, which used InvA gene, was highly specific, sensitive, and convenient for identifying Salmonella. Furthermore, this method could be used for Salmonella monitoring and primary screening in animal products and animal production environmental samples.
