가공식품 중 새우의 검출을 위한 샌드위치 ELISA의 개발 (Development of Sandwich ELISA for the Detection of Shrimp in Processed Foods)

  • 가공식품 중 새우의 검출을 위한 샌드위치 ELISA(sELISA)의 조건을 확립하기 위해 홍다리 새우의 tropomyosin에 대한 특이항체를 이용하여 하였다. 이때, 새우의 검출범위는 1-100 ppm (${\mu}g/mL$)이며, 검출한계는 0.3 ppm이었다. 특이항체의 교차반응 결과, 홍다리새우, 흰다리 새우, 칵테일 새우, 바다가재, 꽃게에 대한 반응성은 각각 100, 73, 155, 18, 0%를 나타내었으며 새우에 대한 특이성이 매우 높았다. 열처리한 시료와 항체간의 반응성은 $100^{\circ}C$까지는 121-221%로 안정하였으나 $121^{\circ}C$에서는 반응성이 급격히 감소하였다(7.8%). 크림스프, 이유식, 소시지, 어묵, 소스에 대한 spike test에서 새우의 분석회수율은 각각 397, 639, 168, 234, 0%로 소스를 제외하고 매우 높게 나타났다. sELISA에 의하여 14점의 시판시료 중 새우의 함유 유무를 조사한 결과, 정성적으로 원료의 표시사항과 일치하는 비율은 79%이었다.

감초분말첨가 김치의 품질 (Quality of Licorice (Glycyrrhiza uralensis) Powder Added Kimchi)

  • 본 연구의 목적은 김치의 맛을 증진시키기 위하여 감미료로 첨가되는 설탕 대신에 분말상태의 감초를 첨가하여 김치의 품질특성을 개선하고자 하는 것이다. 김치 제조원료에 설탕 또는 감초를 첨가하여 젖산균의 생육과 산생성 및 관능성 특성에 미치는 영향을 조사하였다. 김치에 첨가된 감초의 영향은 다음과 같다. pH는 비숙성시료와 1일 숙성시료는 감초의 첨가에 따라 차이가 없었으나, 2일 숙성시료와 3일 숙성시료의 경우는 감초의 첨가에 따라 pH가 다소 높아지는 경향을 보였다. 산도는 비숙성시료와 1일 숙성시료는 감초의 첨가농도가 증가함에 따라 유의적으로 증가하였으나(p<0.05), 2일 숙성시료와 3일 숙성시료의 경우는 반대로 감초의 첨가농도가 증가함에 따라 유의적으로 감소하였다(p<0.05). 감초의 첨가에 의하여 젖산균수는 변화가 없었다. 관능검사의 결과를 보면 1-3일 숙성된 시료의 전반적인 기호도와 맛은 감초 0.1% 또는 0.2% 첨가시료가 표준시료와 같거나 다소 우수하였고, 1.0% 첨가시료가 가장 저조하였다. 냄새와 조직감은 1.0% 첨가시료가 다른 시료보다 저조하였고, 색상은 시료 사이에 차이가 없었다.

Wine Production Using Osmotic Solution from Dried Mango Process

  • This study evaluated the potential of utilizing the osmotic solution from dried mango processing as alternative raw material for mango wine making. Fermentation was carried out using two kinds of yeast strains Saccharomyces bayanus, Lalvin EC-1118 and Saccharomyces cerevisiae, Lalvin D-47 at 20$^{\circ}C$ for 28 days. Physicochemical analysis during fermentation was performed for each treatment and the resulting wine samples were analyzed for color, volatiles and sensory properties. Results of physicochemical analysis between the two fermenting samples as well as the wine samples show almost similar results regardless of the yeast strains. Wine color of sample wines after storage were not significantly different at p<0.05 and when compared with a commercial mango wine. From the volatile analysis, esters and alcohols constituted majority of the compounds. Production of several esters, alcohols, acids and terpenes were affected by yeast strain used in fermentation. Results of sensory analysis showed that wines fermented by S. bayanus EC-1118 strain was more acceptable although sensory scores between the treatments and the reference wine showed significant differences in all the attributes evaluated, except for bitterness. The utilization of osmotic solution from dried mango process could produce similar properties with existing commercial mango wines although there is still need for further work on the improvement of some sensory attributes of the mango wines.

  • $^{137}$Cs 용액을 추적자로 사용하여 동시와 역동시 계수방법에 의해 $^{137}$Cs 용액의 방사능을 측정하였다. 본 연구에서의 측정선원은 $^{137}$Cs과 추적자를 혼합하여 제작하였으며 혼합 비율은 동시와 역동시 계수 방법에서 각각 1/3과 1/5로 하였다. $^{137}$Cs용액의 비 방사능은 $\beta$채널 저에너지 문턱 준위 변화와 효율외삽 방법에 의해 산출되었다. 동시와 역동시 계수 방법에 의해 산출된 결과는 기준일 현재 552.78kBq/g과 554.32kBq/g이었으며 합성오차는 각각 1.60kbq/g과 1.51kBq/g이었다. 두 개의 결과는 오차범위 내에서 잘 일치함을 보여준다.

Polarization Phase-shifting Technique for the Determination of a Transparent Thin Film's Thickness Using a Modified Sagnac Interferometer

  • We propose a polarization phase-shifting technique to investigate the thickness of $Ta_2O_5$ thin films deposited on BK7 substrates, using a modified Sagnac interferometer. Incident light is split by a polarizing beam splitter into two orthogonal linearly polarized beams traveling in opposite directions, and a quarter-wave plate is inserted into the common path to create an unbalanced phase condition. The linearly polarized light beams are transformed into two circularly polarized beams by transmission through a quarter-wave plate placed at the output of the interferometer. The proposed setup, therefore, yields rotating polarized light that can be used to extract a relative phase via the self-reference system. A thin-film sample inserted into the cyclic path modifies the output signal, in terms of the phase retardation. This technique utilizes three phase-shifted intensities to evaluate the phase retardation via simple signal processing, without manual adjustment of the output polarizer, which subsequently allows the thin film's thickness to be determined. Experimental results show that the thicknesses obtained from the proposed setup are in good agreement with those acquired by a field-emission scanning electron microscope and a spectroscopic ellipsometer. Thus, the proposed interferometric arrangement can be utilized reliably for non-contact thickness measurements of transparent thin films and characterization of optical devices.

Accuracy of intraoral scans of edentulous jaws with different generations of intraoral scanners compared to laboratory scans

  • PURPOSE. Purpose of this in vitro study was to determine the accuracy of different intraoral scans versus laboratory scans of impressions and casts for the digitization of an edentulous maxilla. MATERIALS AND METHODS. A PEEK model of an edentulous maxilla, featuring four hemispheres on the alveolar ridges in region 13, 17, 23 and 27, was industrially digitized to obtain a reference dataset (REF). Intraoral scans using Cerec Primescan AC (PRI) and Cerec AC Omnicam (OMN), as well as conventional impressions (scannable polyvinyl siloxane) were carried out (n = 25). Conventional impressions (E5I) and referring plaster casts were scanned with the inEOS X5 (E5M). All datasets were exported in STL and analyzed (Geomagic Qualify). Linear and angular differences were evaluated by virtually constructed measurement points in the centers of the hemispheres (P13, P17, P23, P27) and lines between the points (P17-P13, P17-P23, P17-P27). Kolmogorov-Smirnov test and Shapiro-Wilk test were performed to test for normal distribution, Kruskal-Wallis-H test, and Mann-Whitney-U test to detect significant differences in trueness, followed by 2-sample Kolmogorov-Smirnov test to detect significant differences in precision (P < .008). RESULTS. Group PRI showed the highest trueness in linear and angular parameters (P < .001), while group E5I showed the highest precision (P < .001). CONCLUSION. Intraoral scan data obtained using Primescan showed the highest trueness while the indirect digitization of impressions showed the highest precision. To enhance the workflow, indirect digitization of the impression itself appears to be a reasonable technique, as it combines fast access to the digital workflow with the possibility of functional impression of mucosal areas.

Hyperspectral imaging technique to evaluate the firmness and the sweetness index of tomatoes

  • The objective of this study was to evaluate the firmness and the sweetness index (SI) of tomatoes with a hyperspectral imaging (HSI) technique within the wavelength range of 1000 - 1550 nm. The hyperspectral images of 95 tomatoes were acquired with a push-broom hyperspectral reflectance imaging system, from which the mean spectra of each tomato were extracted from the regions of interest. The reference firmness and sweetness index of the same sample was measured and calibrated with their corresponding spectral data by partial least squares (PLS) regression with different preprocessing methods. The calibration model developed by PLS regression based on the Savitzky-Golay second-derivative preprocessed spectra resulted in a better performance for both the firmness and the SI of the tomatoes compared to models developed by other preprocessing methods. The correlation coefficients ($R_{pred}$) were 0.82, and 0.74 with a standard error of prediction of 0.86 N, and 0.63, respectively. Then, the feature wavelengths were identified using a model-based variable selection method, i.e., variable importance in projection, from the PLS regression analyses. Finally, chemical images were derived by applying the respective regression coefficients on the spectral image in a pixel-wise manner. The resulting chemical images provided detailed information on the firmness and the SI of the tomatoes. The results show that the proposed HSI technique has potential for rapid and non-destructive evaluation of firmness and the sweetness index of tomatoes.

다층 퍼셉트론 기반 고해상도 위성영상의 상대 방사보정 (Relative Radiometric Normalization for High-Spatial Resolution Satellite Imagery Based on Multilayer Perceptron)

  • 다중시기의 위성영상에 대해 일관성 있는 변화탐지 결과를 획득하기 위해서는 전처리 과정이 필수적으로 이루어져야 한다. 특히, 분광값과 관련된 전처리 과정은 방사보정으로 수행될 수 있으며, 일반적으로 상대 방사보정이 활용되고 있다. 하지만, 대부분의 상대 방사보정은 두 영상간의 관계를 선형으로 가정하며, 생태학적 차이와 같은 비선형적인 분광특성은 고려되지 않는다. 따라서, 본 연구에서는 방사 및 생태학적 특성에 대한 복합적인 보정을 수행할 수 있는 비선형적인 관계를 가정한 상대 방사보정을 제안하였다. 제안된 방법은 입력영상 및 참조영상을 선정하고, no-change method를 통해 radiometric control set samples를 추출하였다. 또한, 충분한 정보를 고려하기 위하여 화소값뿐만 아니라 분광지수들이 추출되었고, 비선형적인 관계의 모델링은 다층 퍼셉트론을 통해 수행되었다. 최종적으로 기존의 상대 방사보정기법과 비교 분석을 수행하였고, 시각적 및 정략적으로 평가한 결과 제안된 방법이 기존의 상대 방사보정보다 우수한 것을 확인하였다.

  • 본 연구에서는 건설현장에 설치된 표지판을 측색하여 색기준에 대한 적합성 검토와 현장여건에 적합한 관리 방안을 제시하고자 하였으며, 현장에 설치된 안전표지판은 관련기준에 적합한 정도가 적색 33%, 황색 22%, 청색 3%, 녹색 28%로 나타나 각각의 현장에 설치된 법적인 기준에 대부분 적합하지 않은 것으로 나타났으며, 청색 안전표지판에 대하여 보다 적극적인 제조생산과 관리에 대한 관련기관의 감독이 필요하다.

Western dietary pattern is associated with higher risk of lower lean muscle mass in Korean postmenopausal women: data from the Korea National Health and Nutrition Examination Survey 2008-2011

  • BACKGROUND/OBJECTIVES: In a healthy person, from 35 years of age, there is an annual loss of muscle mass at the rate of 1-2% and is associated with a decline in the quality of life. This study aimed to identify the particular dietary patterns associated with the risk of lower lean muscle mass in Korean postmenopausal women. SUBJECTS/METHODS: The Korea National Health and Nutrition Examination Survey (KNHANES) is a population-based, continuous cross-sectional annual survey. The participants of the KNHANES IV (2008-2009) and V (2010-2011) were considered for this study. The study sample consisted of 1548 postmenopausal women, aged 45-86 years. Lower lean muscle mass was defined as having appendicular skeletal muscle mass corrected for body weight less than 1 standard deviation of the young reference group aged 20 to 39 years in KNHANES IV and V. To identify the dietary pattern using factor analysis, 24-h recall data was used. RESULTS: The prevalence of lower lean muscle mass was 31.3% in this study population. Four dietary patterns were identified by factor analysis; 'Diverse', 'Western', 'Traditional', and 'Snacks and beverages'. The 'Western' pattern, highest factor loadings for flour and bread, potatoes, red meat, processed meat, eggs, and cheese, was significantly associated with a high (60%) risk of lower lean muscle mass (odds ratio [95% confidence interval] = 1.60 [1.07-2.39], P for trend = 0.01) after adjustments for potential covariates. The other 3 dietary patterns were not associated with lower lean muscle mass. CONCLUSIONS: The study findings suggest that the 'Western' dietary pattern that includes flour and bread, potatoes, red meat, processed meat, eggs, and cheese, may be associated with a higher risk of lower lean muscle mass in Korean postmenopausal women.
