Expression Profiles of Cellular Retinol-binding Protein, Type II (CRBP II) in Erlang Mountainous Chickens

  • Cellular retinol-binding protein II (CRBP II) belongs to the family of cellular retinol-binding proteins and plays a major role in absorption, transport, and metabolism of vitamin A. In addition, because vitamin A is correlated with reproductive performance, we measured CRBP II mRNA abundance in erlang mountainous chickens by real-time PCR using the relative quantification method. The expression of CRBP II showed a tissue-specific pattern and egg production rate-dependent changes. The expression was very high (p<0.05) in jejunum and liver, intermediate in kidney, ovary, and oviduct, and lowest (p<0.05) in heart, hypothalamus, and pituitary. In the hypothalamus, oviduct, ovary, and pituitary, CRBP II mRNA abundance were correlated to egg production rate, which increased from 12 wk to 32 wk, peaked at 32 wk relative to the other time points, and then decreased from 32 wk to 45 wk. In contrast, the expression of CRBP II mRNA in heart, jejunum, kidney, and liver was not different at any of the ages evaluated in this study. These data may help to understand the genetic basis of vitamin A metabolism, and suggest that CRBP II may be a candidate gene to affect egg production traits in chickens.

A Feedback Control Model for ABR Traffic with Long Delays (긴 지연시간을 갖는 ABR 트래픽에 대한 피드백제어 모델)

  • Asynchronous transfer mode (ATM) can be efficiently used to transport packet data services. The switching system will support voice and packet data services simultaneously from end to end applications. To guarantee quality of service (QoS) of the offered services, source rateot send packet data is needed to control the network overload condition. Most existing control algorithms are shown to provide the threshold-based feedback control technique. However, real-time voice calls can be dynamically connected and released during data services in the network. If the feedback control information delays, quality of the serviced voice can be degraded due to a time delay between source and destination in the high speed link. An adaptive algorithm based on the optimal least mean square error technique is presented for the predictive feedback control technique. The algorithm attempts to predict a future buffer size from weight (slope) adaptation of unknown functions, which are used fro feedback control. Simulation results are presented, which show the effectiveness of the algorithm.

  • GPS(Global Positioning System) location tracking is a method for taking the precise coordinates after the coordinates are obtained by a GPS receiver, and displaying them on the map. In this paper with WAVE(Wireless Access for Vehicular Environment) simulation, we show that various services such as vehicle tracking service, real-time road conditions service and logistics can go tracking service, control and operation services according to the vehicle position and the traveling direction by using the GPS position data. A vehicle tracking system using GPS is automatically able to manage multiple RCP when exchanging data between RMA and the RCP, and it provides rapid requests and responses. To verify that multiple sessions between RMA and RM, as well as multiple sessions between RMA and RCP are able to be implemented, we take RMA as a RCP application on an OBU, until the RMA is receiving data response from corresponding RM. As a result of this experiment, we show that the response speeds of single session between RMA and RM using 1, 2, 3, and 4 kbyte unit data are similar, 62.32ms, 62.65ms, 63.02ms, and 63.48ms, respectively. Likewise, those of 128 muliple sessions using 1, 2, 3, and 4 kbyte unit data are not much more time difference, 298.08ms, 302.21ms, 322.85ms, and 329.62ms, respectively.

  • The overlay multicast that has been recently presented as IP alternative for the IP multicast has been getting much persuasion by the computing power of the hardware and the advancement of the network techniques to enforce Routing in application-level. In an overlay multicast, the system resource and the network bandwidth must be utilized efficiently to service real-time HDTV images. Specifically, the system must consider the delay and the jitter that can be incurred at the application-level. In this paper, we implement a server and a client to broadcast HDTV, in the session composed by the existing overlay multicast protocol. The broadcasting server performs the service using a TV tuner, An HDTV camcorder, and files, clients constituting a multicast group relay the received data to other clients. At this time, the information that the clients report periodically, including their delay and the network state, to the server is used as an important information to maintain an overlay session. The implementation is based on the DirectX and its performance is evaluated by the LAN test bed that has been set.

Feasibility study of β-ray detection system for small leakage from reactor coolant system

  • Because existing reactant coolant system (RCS) leakage detection mechanisms are insensitive to small leaks, a real-time, direct detection system with a detection threshold below 0.5 gpm·hr-1 was studied. A beta-ray detection system using a silicon detector with good energy resolution for beta rays and a low gamma-ray response was proposed. The detection performance in the leakage condition was evaluated through experiments and simulations. The concentration of 16N in the coolant corresponding to a coolant leakage of 0.5 gpm was calculated using the analytic method and ORIGEN-ARP. Based on the concentration of 16N and the measurement of the silicon detector with 90Sr/90Y, the beta-ray count rate was estimated using MCNPX. To evaluate the effect of gamma rays inside the containment building, the signal-to-noise ratio (SNR) was calculated. To evaluate the count rate ratio, the radiation field inside the containment building was simulated using MCNPX, and response evaluation experiments were performed using beta and gamma rays on the silicon detector. The expected beta-ray count rate at 0.5 gpm leakage was 7.26 × 105 counts/sec, and the signal-to-background count rate ratio exceeded 88 for a transport time of 10 s, demonstrating its suitability for operation inside a reactor containment building.

Regulation of S100G Expression in the Uterine Endometrium during Early Pregnancy in Pigs

  • Calcium ions play an important role in the establishment and maintenance of pregnancy, but molecular and cellular regulatory mechanisms of calcium ion action in the uterine endometrium are not fully understood in pigs. Previously, we have shown that calcium regulatory molecules, transient receptor potential vanilloid type 5 (TRPV6) and calbindin-D9k (S100G), are expressed in the uterine endometrium during the estrous cycle and pregnancy in a pregnancy status- and stage-specific manner, and that estrogen of conceptus origin increases endometrial TRPV6 expression. However, regulation of S100G expression in the uterine endometrium and conceptus expression of S100G has been not determined during early pregnancy. Thus, we investigated regulation of S100G expression by estrogen and interleukin-$1{\beta}$ (IL1B) in the uterine endometrium and conceptus expression of S100G during early pregnancy in pigs. We obtained uterine endometrial tissues from day (D) 12 of the estrous cycle and treated with combinations of steroid hormones, estradiol-$17{\beta}$ ($E_2$) and progesterone ($P_4$), and increasing doses of IL1B. Real-time RT-PCR analysis showed that $E_2$ and IL1B increased S100G mRNA levels in the uterine endometrium, and conceptuses expressed S100G mRNA during early pregnancy, as determined by RT-PCR analysis. To determine if endometrial expression of S100G mRNA during the implantation period was affected by the somatic cell nuclear transfer (SCNT) procedure, we compared S100G mRNA levels in the uterine endometrium from gilts with SCNT-derived conceptuses with those from gilts with conceptuses derived from natural mating on D12 of pregnancy. Real-time RT-PCR analysis showed that levels of S100G mRNA in the uterine endometrium from gilts carrying SCNT-derived conceptuses was significantly lower than those from gilts carrying conceptuses derived from natural mating. These results showed that S100G expression in the uterine endometrium was regulated by estrogen and IL1B of conceptus origin, and affected by the SCNT procedure during early pregnancy. These suggest that conceptus signals regulate S100G, an intracellular calcium transport protein, for the establishment of pregnancy in pigs.

Toxicity and Behavioral Changes of Medaka (Oryzias latipes) by Brine Exposure (송사리(Oryzias latipes)를 이용한 고염해수의 생태독성 및 단기적 행동변화에 관한 연구)

  • Acute toxicity test and behavioral change analysis of seawater acclimated Japanese medaka were conducted to identify the brine effects on fish by seawater desalination. 7 day acute toxicity test of brine revealed linear concentration-response relationship from 40.0~80.0 psu treatment groups. There was no significant brine effect for 30-40 psu groups and mass mortality was observed from >50 psu exposure (7-day $LC_{50}$=51.4 psu). Images from the real time camera system were analyzed to observe the changes in behavioral patterns of medaka exposed to various salinity. 40.0 and 50.0 psu exposed groups were stabilized in behavioral patterns after 3.1 and 4.6 hours, respectively and 60.0 psu group showed sharp increase in activity during first 12 hours and 50% mortality thereafter. Similar patterns were observed to 70 and 80 psu groups and both experimental groups showed 100% mortality within 12 hours. Acute toxicity test and behavioral patterns showed very similar toxicity results which revealed the increases in mortality and behavioral activities from 50.0 psu. This critical salinity for fish impacts must be implemented to brine discharge strategy by seawater desalination into the coastal area. Also, we recommend that real time camera monitoring system must be a useful tool for early warning of fish toxicity for other applications.   • BIS is an arrival guidance system which is able to supply passengers with bus service condition via Kiosks at a bus stop, internet and mobile service through pinpointing bus location in real time. It is very significant to improve the quality of traffic information by quality control of GIS-based bus network so as to maintain navigational information and to implement reliable BIS. Therefore this study aims to build criteria to quantitatively evaluate data quality of the product in accordance with the process in bus network data and to suggest guideline of quality control. To achieve this, we have categorized geometric and logical errors occurring during constructing bus network database by giving a specific case study on TAGO and set up sectional guideline and procedures to examine database for systematic and coherent quality control management. Proceeding from what has been said above, the outcome of our research leads to quality guarantee for objective and reliable bus network database and is fully expected to bring benefit of providing a more accurate public transportation information and improving reliability of BIS through preventing a variety of errors in system operation in advance.

The Lipid Efflux Effects of Dichloromethane Extract from Orostachys japonicus in 3T3-L1 Adipocyte Cells (3T3-L1 지방세포에 대한 와송 디클로로메탄 추출물의 지질 대사 개선에 관한 연구)

  • This study investigated the improved lipid metabolism effect of 3T3-L1 cells induced by adipocytes using the dichloromethane (DCM) fraction in the organic solvent extract of Wassong (Orostachys japonicus). To confirm the cell cytotoxicity, each of 6 fractions of organic solvent extracts (EtOH, Hexane, DCM, EtOAc, BuOH, and H2O) was examined using MTS assay. As a result, it was confirmed that the DCM extract was stable over the whole range of concentrations, and a DCM fraction was used to confirm the improved lipid metabolism effect. Lipid excretion was measured to confirm the change of lipid metabolism. 3T3-L1 cells induced by adipocytes were treated with DCM extract and stained with oil-red O to evaluate lipid accumulation. As a result, it was confirmed that the lipid efflux was significantly improved. In order to confirm the mechanism of lipid efflux, the mRNA expressions of ABCA1 and ABCG1, which are lipid transport proteins, were confirmed by real-time PCR. Therefore, the present study confirmed that the DCM extract from Orostachys japonicus has the effect of improving the lipid metabolism on 3T3-L1 adipocytes. In addition, the results of this study will be used as the basis for the development of functional foods using Orostachys japonicus and also for conducting research on the detailed mechanisms.
