Hydrothermal Synthesis of Nanosized Sulfated Zirconia as an Efficient and Reusable Catalyst for Esterification of Acetic Acid with n-Butanol

  • A kind of nanosized sulfated zirconia was prepared by a hydrothermal method, and full characterized by XRD, TEM, BET, TGA, and FTIR. Its catalytic activity was evaluated in the esterification reactions, including the testing of the catalytic reusability and the optimization of reaction conditions. The obtained catalyst was revealed to be highly efficient solid catalyst for the esterification of acetic acid with n-butanol, presenting the advantages of high conversion and selectivity, easy recovery, and steady reusability.

A Study on the Development of the Gear Profile Design Program (기어 치형 설계 프로그램 개발에 관한 연구)

  • In this paper, the gear design program is presented. The profile of gears is created using classical mathematic formulations. In each gear, a kinematic joint is applied and one can define the 20 contact condition between gear pairs. Initial and boundary conditions such as force, torque, velocity, acceleration, etc. can be set. Thus, it is possible to analyze dynamic characteristics of gear pairs such as reaction moment and the variation of angular velocity. In order to find the optimal profile of gear pairs, two optimization methods based on design of experiments are inserted in the program; One is the Taguchi method and the other is the response surface analysis method. To verify the program, the rack & pinion gear is created and analyzed. Simulation results show that the developed program is useful and result data is reliable.

Characteristics of SiC Whisker-Reinforced LAS Matrix Composites Fabricated by the Mixed Colloidal Route and the Sol-Gel Process (콜로이드 혼합법 및 Sol-Gel 법에 의해 제조한 SiC 휘스커 강화 LAS 기지 복합체의 특성)

  • SiC whisker-reinforced LAS matrix composites were developed by a mixed colloidal processing route. An optimization of processing conditions was made using the zeta potential data of silica, boehmite, and SiC whisker dispersions. Similarly, the SiC whisker-reinforced composites were also fabricated by the conventional sol-gel process using the hydrolysis-condensation reaction of relevant metal alkoxides. The composites fabricated by the mixed colloidal processing route were characterized by a uniform spatial distribution of SiC whisker throughout the matrix. The fracture toughness increased from 1.3 MPa.m1/2 for the LAS specimen to 5.0 Mpa.m1/2 for the hot-pressed composite (95$0^{\circ}C$ and 20 MPa for 20 min) containing 20 wt% SiC whisker. The increase in fracture toughness appears to result mainly from the crack deflection and the crack bridging by whiskers with some additional toughenings from the whisker pullout and the matrix prestressing mechanisms.

Selection of RAPD Markers for Phytophthora infestans and PCR Detection of Phytophthora infestans from Potatoes

  • For rapid and secure differentiation of P. infestans from other Phytophthora species, two fragments obtained from randomly amplified polymorphic DNA (RAPD) profiles were selected as markers. Also, primers for in polymerase chain reaction (PCR) to detect P. infestans specifically were developed by analyzing the sequences of ITSII regions in rDNA of Phytophthora species. The primers, PISP-1 and ITS3 amplified a single. Fragment 450 bp of about in P. infestans, but not in other fungal or bacterial isolates. Annealing temperatures and template DNA quantities were varied for the optimization of PCR conditions. From the result of the PCR detection study, species-specific primers were selected under annealing temperatures ranging from 55$^{\circ}C$ to 61$^{\circ}C$, and template DNA levels ranging from 10 pg to 100 ng.

  • Crosslinked agar-g-poly(acrylic acid) (x-agar-g-PAA) super absorbent with a water absorbency ($Q_{H2O}$) of approximately 660 g/g was synthesized by the copolymerization of agar with an acrylic acid monomer. KPS and MBA were used as the initiator and crosslinker, respectively. Grafting was performed in air. Infrared spectroscopy was used to identify the product of copolymerization. The optimum conditions to synthesize the x-agar-g-PAA superabsorbent were 0.1 g of agar, 0.1 g of the KPS initiator, for 15 min; 50% AA monomer, 0.005 g of the MBA crosslinker, for a propagation time of 5 min; and 1 M NaOH for 15 min to allow for saponification. The reaction temperature was $80{^\circ}C$.

Optimization of Production of Trehaolse from Maltose using Recombinant Trehalose Synthase from Thermus caldophilus GK24 (재조합 트레할로스 합성효소에 의한 맥아당으로부터 트레할로스 생산 최적화)

  • Recombinant trehalose synthase from Thermus caldophilus GK24 showed an ability to produce trehalose from maltose. The activity of the partially purified enzyme was not influenced by most metal ions at 1 mM but was inhibited by 10 mM $Co^{2+}$, $Mn^{2+}$, and $Fe^{2+}$. Enzyme activity varied during prolonged reaction due to changes in the environmental conditions. Thus, the reaction was carried out for an extended time with optimized conditions of $45^{\circ}C$ and pH 7.0. An yield of 32.9% was reached at $60^{\circ}C$ after reaction for 22 h, and, maximum trehalose conversion (69.2%) was attained at $25^{\circ}C$. The yields obtained using enzyme dosages of 10, 25, and 50 U/g were 62.3, 62.3 and 59.0 %, respectively, though the initial conversion rate was higher when the higher dose was used. Similar profiles of trehalose production yields were observed with reaction working volumes of 10 ml to 2,000 ml.

Optimization and Characteristics of Removal Condition of Livestock Wastewater Using a Photocatalytic Process (광촉매공정 적용시 축산폐수의 처리특성 및 최적화)

  • The photocatalytic degradation of livestock wastewater has been investigated over $TiO_2$ photocatalysts irradiated with a ultraviolet (UV) light. The effect of operational parameters, i.e., distance, reaction area, concentration of suspended solids(SS), and column diameter on the degradation of livestock wastewater has been performed in lab-scale. The optimal conditions for livestock wastewater were determined: distance was 3 cm (less than 7 cm), reaction area was $3.6\;m^2$, SS concentration was 40 mg/L (less than 300 mg/L) and column diameter was 5 mm (less than 10 mm). Under the optimal conditions, COD, color and coliform removal efficiencies were approximately 49%, 53% and 100%, respectively. Non-biodegradable COD removal efficiency increased with 57% using by photocatalysis process. Therefore, it is shown that photocatalysis has an effect on degradation of non-biodegradable organic matter.

  • Yeonsan Ogae has been known as supporting health and high efficacy of treatment. In recent days, as the efficacy of functional peptides has known, the optimization of oligo peptides production and its characteristics from Ogae legs has been performed. Response surface method was used to perform the optimizaion of enzyme hydrolysis. The range of processes was temperature ( 40, 50 and $60^{\circ}C$), pH( pH 6.0, 7.0 and 8.0 ), and enzyme( 1, 2 and 3% ). The degree of hydrolysis, amino acids, molecular weight of products were analyzed. The optimum process of enzyme hydrolysis were determined as temperature $58^{\circ}C$, pH 7.5, and enzyme concentration 3%. At optimum conditions, the degree of hydrolysis after 2 h reaction was 75-80%. The amino acid and were 168.131 mg/100 g, respectively. The molecular weight of products by using MALDI-TOF was ranged from 300 to 1,000 Da.

Extraction of Natural Emulsifier from Medicago sativa L. and Sapindus saponaria L.: Optimization using CCD-RSM (알팔파 및 무환자나무열매로부터 천연유화제의 추출: CCD-RSM을 이용한 최적화)

  • In this study, natural emulsifiers were extracted from Medicago sativa L. and Sapindus saponaria L. The extraction yield using CCD-RSM and the extraction process of foaming stability of the extracted product were optimized and 95% confidence interval was used to confirm the statistical reasonableness of the optimization. Herein, independent parameters were the ethanol volume and extraction temperature, whereas reaction parameters were the extraction yield and foaming stability. Under the condition of 53.5 vol% ethanol and extraction temperature (70.9 ℃), the maximum yield and foaming stability of the extracted product from Medicago sativa L were predicted as 26.2 wt% and 44.5%, respectively. In the case of the extraction from Sapindus saponaria L, the maximum yield and foaming stability were expected to be 31.9 wt% and 47.5% under the optimized conditions including 60.4 vol% of ethanol and extraction temperature (72.4 ℃). The average experimental error for validating the accuracy was about 3.4(± 0.3)% and 5.0(± 0.04)% for the extraction processes from Medicago sativa L. and Sapindus saponaria L., respectively.

Optimization of DNA Extraction and PCR Conditions for Fungal Metagenome Analysis of Atmospheric Particulate Matter (대기 입자상물질 시료의 곰팡이 메타게놈 분석을 위한 DNA 추출 및 PCR 조건 최적화)

  • Several challenges arise in DNA extraction and gene amplification for airborne fungal metagenome analysis from a particulate matter (PM) samples. In this study, various conditions were tested to optimize the DNA extraction method from PM samples and polymerase chain reaction (PCR) conditions with primer set and annealing temperature. As a result of comparative evaluation of DNA extraction under various conditions, chemical cell lysis using buffer and proteinase K for 20 minutes and bead beating treatment were followed by using a commercial DNA extraction kit to efficiently extract DNA from the PM filter samples. To optimize the PCR conditions, PCR was performed using 10 primer sets for amplifying the ITS2 gene region. The concentration of the PCR amplicon was relatively high when the annealing temperature was 58℃ with the ITS3tagmix3/ITS4 primer set. Even under these conditions, when the concentration of the PCR product was low, nested PCR was performed using the primary PCR amplicon as the template DNA to amplify the ITS2 gene at a satisfactory concentration. Using the methods optimized in this study, DNA extraction and PCR were performed on 15 filter samples that collected PM2.5 in Seoul, and the ITS2 gene was successfully amplified in all samples. The optimized methods can be used for research on analyzing and interpreting the fungal metagenome of atmospheric PM samples.
