S-haplotypes and Genetic Diversity in 'Danji' Radish (Raphanus sativus L. var. hortensis)

  • The distribution of S-haplotypes and genetic relationships were evaluated for 47 accessions of 'Danji' radish (Raphanus sativus L. var. hortensis Baker f. gigantissimus Makino) originating from Jeju Island in South Korea. A total of 22 S-haplotype-specific SCAR markers for the S locus glycoprotein (SLG) and S receptor kinase (SRK) loci were tested, and six primer sets amplified locus-specific PCR fragments from at least one 'Danji' radish accession. S5 and S21 alleles atthe SLG locus were the most frequently distributed, and detected from 87.5% and 64.6% of the accessions, respectively. The frequency of the class-II haplotype at the SLG locus was 75%, more frequent than the class-I haplotype. The S23 allele at the SRK locus was detected from 7 accessions. Grouping of the accessions based on S-allele composition revealed three major groups, while 8 accessions showed a unique allelic composition. The genetic diversity of 47 'Danji' radishes and 1 'Gwandong' radish were also evaluated with 38 RAPD primers. A total of 312 bands were scored, and showed that 138 bands (44.2%) were monomorphic among the accessions, whereas 174 (55.8%) bands were polymorphic. Polymorphism rates ranged from 0.2 to 1.0, indicating significant variations in detecting polymorphism across RAPD primers. The genetic similarity coefficients among all pairs of the 48accessions varied from 0.62 to 0.93, and 42% of the comparisons exhibited values higher than 0.85. All the cultivars could be distinguished based on the DNA fingerprints revealed by RAPD. The comparisons between the dendrograms based on S-haplotypes and RAPDs indicate an unrelated and sporadic distribution for several accessions; however, there was a tendency for accessions with the same S-allelic composition to group into the same cluster.

Growth-Promoting Effects of Vegetable Extracts on Selected Human Lactic Acid Bacteria

  • Ethanol extracts from 36 vegetable samples were assayed for their growth-promoting effects on Bifidobacterium bifidum, B. longum, and Lactobacillus casei The growth-promoting effects varied according to bacterial strain and vegetable species. In modified Gy rgy broth, extracts of Lactuca sativa, Lycopersicon esculentum and L escutentum var. cerasiforme exhibited strong growth-promoting responses toward B. longum, and significant and strong growth- promoting response toward B. bifidum was observed in extracts of Actinidia arguta, Allium cepa, A. sativum, Brassica campestris subsp. napus vats. pekinensis, Capsicum frutescens, Daucus carota var. sativa, L sativa, 1. esculentum and L. esculentum var. cerasforme, Nelumbo nucifera, Cucurbita moschata, Lackca sativa var. capitata, and Rubus coreanus. For L casei, extracts of A. fshlosum, A. hberosum, Cichorium intbus, Cucurbita moschat\ulcorner Ipomoea batatas, 1. sativa var. capitata, L. esculentum, P. brachycarpa, Raphanus sativus, R. coreanus, and S. melongena strongly enhanced the growth of this bacteria. In modified Gy rgy broth, the promoting effect was most pronounced with B. bifidum and L. casei among lactic acid bacteria used. In MRS broth, A. arguta, A. cepa, A. sativum, B. campestris subsp. napus var. pekinensis, C. frutescens, and D. carota var. sativa L. satiw var. capitata, and R. coreanus strongly enhanced the growth of B. bifidum, Growth of B. longum was strongly affected by the addition of extracts from L. sativa var. capitata. For L casei, moderate growth-promoting responses were observed in 9 vegetable extracts. The promoting effect in MRS broth was most pronounced with B. bifidum among lactic acid bacteria used.

Antioxidant Activity and Anti-inflammatory Effects of Raw and Processed Fruits and Vegetables

  • In this study we investigated antioxidant and anti-inflammatory activities of Malus Domestica (apple), Pyrus Communis L. (pear), Daucus carota L. (carrot), Brassica oleracea var. (broccoli), Brassica oleracea var. capitata (cabbage) and Raphanus sativus L. (radish), that were obtained from local market. As these are common fruits and vegetables that are widely consumed, we aimed to investigate their beneficial properties especially the antioxidant and anti-inflammatory. The samples were processed by an indirect heating method and their properties were compared to their raw forms. Based on DPPH and ABTS assay, processed samples showed better antioxidant activity compared to raw samples, and processed pear sample had the best antioxidant activity. The anti-inflammatory activities of the samples were also investigated in LPS-treated RAW 264.7 cells. The mRNA expressions of pro-inflammatory mediators and cytokines (iNOS, COX-2, $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) were assessed by RT-PCR. Processed samples exhibited better inhibition of iNOS, compared to the raw forms. Processed broccoli and cabbage samples exhibited outstanding anti-inflammatory effects. The samples did not exhibit cytotoxicity against RAW 264.7 cells up to 1mg/ mL as shown in the cell viability assay. Taken together, processed broccoli and cabbage samples exhibited the strongest anti-inflammatory properties.

FISH Karyotype and GISH Meiotic Pairing Analyses of a Stable Intergeneric Hybrid xBrassicoraphanus Line BB#5

  • xBrassicoraphanus line BB#5, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L. var. rafiphera induced by N-methyl-N-nitroso-urethane mutagenesis in microspore culture, shows high seed fertility and morphological uniformity. Dual-color fluorescence in situ hybridization (FISH) using 5S and 45S rDNA probes and genomic in situ hybridization (GISH) using B. rapa genomic DNA probe were carried out to analyze the chromosome composition and the meiosis pairing pattern compared to its parental lines. The somatic chromosome complement is 2n = 38, which consists of 17 metacentric and two submetacentric chromosomes with lengths of 2.18 to $5.01{\mu}m$. FISH karyotype analysis showed five and eight pairs of 5S and 45S rDNA loci. GISH meiosis pairing analysis showed that 19 complete bivalents were most frequent and accounted for 42% of the 100 pollen mother cells examined. Based on chromosome number, size, morphology, rDNA distribution, and meiosis pairing pattern, both parental genomes of B. rapa and R. sativus appear to exist in xBrassicoraphanus line BB#5, demonstrating its genome integrity. Such stable chromosome constitutions and meiotic pairing patterns in somatic and meiotic cells are very rare in natural and synthetic intergeneric hybrids. Chromosomal studies and genetic and phenotypic changes in allopolyploids a re discussed. The results p resented h erein will b e usef ul f or f urther g enomic s tudy o f xBrassicoraphanus lines and their improvement as promising new breeding varieties.

토양수분(土壤水分), 재배시기(栽培時期), 수확기(收穫期) 및 시비(施肥)가 무우의 질산염함량(含量)에 미치는 영향 (Effects of Soil Moisture, Seasons, Harvesting Period and Fertilizer Application on the Nitrate Content of Radish(Raphanus sativus L. var. niger(Mill.) S. Kerner))

  • 재배환경(栽培環境)(토양수분(土壤水分), 재배시기(栽培時期), 수확시기(收穫時期), 시비량(施肥量))이 무우의 $NO_3$-함량(含量)에 어떤 영향을 미치는 가를 조사(調査)코저 pot 및 온실내포장(溫室內圃場)을 이용(利用)한 실험(實驗)에서 다음과 같은 결과(結果)를 얻었다. 1) 건조상태(乾燥狀態)에서 자란 무우는 $NO_3$-함량(含量)이 높았고, 토양수분(土壤水分)이 충분(充分)한 곳에서 자란 무우는 그 함량(含量)이 낮았다. 2) 여름철에 재배한 무우는 $NO_3$-함량(含量)이 낮았고, 가을, 봄재배(栽培)로 갈수록 그 함량(含量)은 높았다. 3) $NO_3$-함량(含量)은 수확기(收穫期)가 진행(進行)됨에 따라 엽(葉)${\cdot}$엽병(葉柄) 그리고 근부(根部)에서 감소(減少)하는 경향을 보였다. 4) 질소질 비료의 증시(增施)는 $NO_3$-함량(含量)을 다소 증가시켰으나 그 증가폭(增加幅)은 시비량(施肥量) 증가(增加)에 비(比)해 아주 작았다. 5) $NO_3$-함량(含量)은 엽병(葉柄)에서 가장 높았는데 이는 뿌리에 비(比)해 2배에 해당했고 잎의 3배에 달했다. 6) Multiple regression 분석(分析)에 의하면 Mg, K, T/R ratio 그리고 일조량(日照量)과 온도(溫度)가 중요(重要)한 요인(要因)으로 $NO_3$-함량(含量)을 설명(說明)해 주었다. 7) 영양학적인 면에서, 무우 중의 $NO_3$-는 큰 의미가 없음을 검토하였다.

Antioxidant and Anti-inflammatory Properties of Raw and Processed Fruits and Vegetables

  • Reactive oxygen species (ROS) generated from metabolic reactions cause oxidative DNA damage, which results in oxidative tissue injury. Therefore, there is an increasing demand in the intake of high antioxidant sources in order to maintain a healthy environment in cells. In this study, we investigated the antioxidant and anti-inflammatory activities of Malus domestica (apple), Pyrus communis L. (pear), Daucus carota L. (carrot), Brassica oleracea var. (broccoli), Brassica oleracea var. capitata (cabbage), and Raphanus sativus L. (radish) obtained from the local market. Since these are common fruits and vegetables that are widely consumed, we aimed to investigate their beneficial properties, placing particular emphasis on their antioxidant and anti-inflammatory properties. The samples were processed via an indirect heating method and their properties were compared to their raw forms. The samples, up to 1 mg/mL concentration, did not exhibit cytotoxicity against RAW 264.7 cells as demonstrated by cell viability assays. Altogether, processed broccoli and cabbage samples exhibited the strongest anti-inflammatory properties.

무우바이러스병과 매개진딧물과의 상관관계에 관하여 (Studies on the Correlation between Virus Diseases and Aphid Vectors in Radish Fields)

  • 무우의 바이러스 이병율과 매개진딧물 비래밀도간의 상관관계를 조사하고자 전주지방에 무(Raphanus sativus L. var. Chungsu gungjung)를 파종하고 1982년 4월부터 11월 사이에 실시하였다. 진딧물 비래밀도는 황색수반으로 채집하고, 바이러스 이병율은 병징을 육안으로 조사하였다. 전주지방의 진딧물 비래밀도는 5월 하순과 10월 중순에 최성기를 나타냈으며 매개진딧물의 비래분포도 같은 경향이었다. 무우의 바이러스병 매개진딧물은 복숭아혹진딧물 (Myzus perticae), 무우테두리 진딧물(Lipaphis erysimi), 목화진딘들(Aphis gossypii), 양배추가루진딧물(Brevicoryne brassicae)이 채집되었고, 전체진딧물 대 매개진딧물의 비율은 $62.82\%(39.260/62.499)$이었다. 매개진딧물중 복숭아혹진딧물이 $82.53\%(32.401/39.260)$로서 우점종으로 나타났다. 춘계의 바이러스 이병율은 급상승하며 높은데 비하여 추계의 바이러스 이병율은 완만하면서 낮았다. 매개진딧물의 밀도상승과 바이러스 이병율간의 상관관계는 춘계는 $r=0.7414^*(\hat{Y}=8.1444+0.0551X)$, 추계는 $r=0.9117^{**}(\hat{Y}=10.2590+0.463X)$로서 고도의 유의성이 인정되었다. 또 매개진딧물의 비래와 바이러스 병징발현간에는 약 15일의 기간이 소요되는 것으로 나타났다. 바이러스병의 이병주 보존구가 이병주 제거구보다 발병율이 높게 나타났다.

Evaluating Plant Uptake of Veterinary Antibiotics with Hydroponic Method

  • Veterinary antibiotics (VAs) has been used to treat animal disease and to increase animal weight as growth promoter. However, abused usage of VAs can cause production of antibiotic resistance genes (ARGs) in the environment and additionally, residual of VAs in soil can be transferred into crops. Therefore, main objective of this research was to examine bioaccumulation of VAs in sprouts (red cabbage, Brassica Olearacea L. var. Capitata f. rubra and red radish, Raphanus sativus) with hydroponic method. Total of 7 VAs in 2 different classes of VAs (tetracyclcines: tetracycline, oxytetracycline, chlortetracycline, sulfonamides: sulfamethoxazole, sulfamethazine, sulfamethiazole, macrolides: tylosin) were evaluated and experiment was conducted with solid phase extraction (SPE)/high performance liquid chromatography tandem mass spectrometry (HPLC/MS/MS). Initial spiked concentration of 7 VAs was $5mg\;L^{-1}$ and cultivation period was 8 days. Result showed that growth of sprouts was inhibited about 23-27% when VAs was introduced. Amount of bioaccumulated VAs was also differed depending on class of VAs. The highest amount of bioaccmulated VAs was tetracycline and sulfamethoxazole in each class with a concentration of 4.05, $7.73mg\;kg^{-1}$ respectively. Calculated transfer ratio of VAs into crops was also ranged 0.38-54.27%. Overall, bioaccumulation of VAs in crops can be varied depending on crop species and class of VAs. However, further research should be conducted to verify bioaccumulation of VAs in crops in the soil environment.

A Study on the Utilization of Urban Garden Design Derived from the Traditional Farming Method Gyeonjongbeop from the Joseon Period: Focused on Imwongyeongjeji Bolliji

  • Background and objective: Traditional farming is winning recognition as a sustainable alternative farming method. As urban farming increases in South Korea, it is crucial to develop more sustainable farming techniques. Gyeonjongbeop is the traditional farming method introduced in the Joseon period. This study was conducted to propose a productive garden model suitable for urban farming through the interpretation of traditional farming methods contained in Imwongyeongjeji Bolliji and to test the model on an actual field. Methods: Using the design and cropping system of Gyeonjongbeop as the research materials, we reviewed its tillage and cultivation and examined the applicability. We proposed a modified method by extracting parts applicable to urban farming. According to the methods, we created a garden with ridges and furrows, cultivated proper vegetables, and evaluated their growth. Results: Raphanus sativus, Allium fistulosum L., Brassica juncea, and Spinacia oleracea grown on ridges showed higher growth than those grown on a conventional flat field. The growth of Hordeum vulgare var. hexastichon and Triticum aestivum L. on furrows was also slightly higher. This proved that the method could make up for the deficiencies of barley and wheat that are weak against winds and cold and are easily destroyed by the spring rains. Conclusion: Ridge and furrow cultivation derived from Gyeonjongbeop can be an efficient urban farming system compared to the conventional cultivation in flat fields. The system can use fallow lands in winter for year-round urban farming. In addition, the application of the traditional farming system can enhance the humanistic value of urban farming.

지렁이 분립이 엽채류의 생육에 미치는 영향 - 분립의 최적혼합비율의 추정 - (The Effect of Vermicast on the Growth of Vegetable Plants - Estimation of Optimum Mixture Ratios of Vermicast -)

  • 본 연구는 톱밥발효 돈분을 vermicomposting으로 처리하여 생산된 분립을 상토재의 원료인 피트모스와 혼합하였을 때 이 화학적 특성과 엽채류(근대, 열무, 시금치, 상추)의 생육에 미치는 영향을 조사하여 분립의 적정 혼합비율을 추정하였다. 이 화학적 분석결과 지렁이 분립량이 증가할수록 용적비중(bulk density), 진밀도(particle density), pH 전기전도도(electrolytic conductivity), 무기물(ash), 전질소(total nitrogen), 양이온 치환능력(cation exchange capacity), 유효인산함량(available $P_2O_5$) 및 치환성양이온향량(exchangeable cation)이 증가하였다. 근대는 분립 $40{\sim}100%$의 혼합비율에서 초장(PL), 엽면적(LA), 지상부중(SHW), 근중(RW), 근장(RL) 및 생물학적 수량(BY)이 다른 처리구보다 유의하게 높았고, 열무는 분립 $40{\sim}60%$의 혼합비율에서 지상부중(SHW), 근중(RW) 및 생물학적 수량(BY)이 다른 처리구보다 유의하게 높았다. 시금치는 분립 40%의 혼합비율에서 지상부중(SHW), 근중(RW) 및 생물학적 수량(BY)이 다른 처리구보다 유의하게 높았고, 상추는 분립 40%의 혼합비율에서 초장(PL), 엽면적(LA), 지상부중(SHW), 근중(RW), 근장(RL) 및 생물학적 수량(BY)이 다른 처리구보다 유의하게 높았다. 이상의 결과로부터 본 실험에서 분립의 첨가비율은 근대 $40{\sim}100%$, 열무 $40{\sim}60%$, 시금치와 상추 40%가 적정혼합비율이었다.
