Molecular Phylogenetic Position of Abbottina springeri (Cypriniformes: Cyprinidae) Based on Nucleotide Sequences of RAG1 Gene (RAG1 유전자의 염기서열에 기초한 왜매치 Abbottina springeri (잉어목, 잉어과)의 분자계통학적 위치)

  • Partial nucleotide sequences of nuclear protein-coding recombination activating gene 1 (RAG1) gene of two Abbottina and five Microphysogobio species residing in Korea were analyzed to elucidate the molecular phylogenetic position of A. springeri Banarescu and Nalbant. In RAG1 tree A. rivularis was clearly separated from the monophyletic lineage composed of A. springeri, Biwia zezera and Microphysogobio species. Within this lineage B. zezera showed sister-group relationship to the monophyletic group composed of A. springeri and five Microphysogobio species. Thus, our phylogenetic tree revealed the polyphyletic nature of two Abbottina species from Korea, which result is well congruent with the previous phyletic assumption based on osteological features. The current classification of Abbottina and Microphysogobio based on morphological criteria, such as the presence or absence of papillae on lips and size of swim bladder with or without encapsulation, does not reflect their true evolutionary history.

Effects of Radix Angelicae Gigantis and Resina Ferulae on the Relaxation of Smooth Muscle and Expression of iNOS (당귀 및 아위가 평활근 이완과 iNOS 발현에 미치는 영향)

  • Objectives : Radix Angelicae Gigantis(RAG) and Resina Ferulae(RF) have been used in oriental medicine or folk medicine to increase stamina. The aim of this study was the characterization of the mechanism of action of RAG and RF on smooth muscle and macrophages in rats to find new substances for the treatment of erectile dysfunction, cardiovascular diseases and immune dysfunction. Methods : We investigated the effects of the water extracts of RAG and RF on phenylephrine or KCl-contracted rat endothelium-denuded aorta, the production of NO in vascular smooth muscle cell (VSMC) and the production of NO and induction of iNOS in the $IFN-{\gamma}-primed$ RAW 264.7 cells. Results : The water extracts of the RAG and RF showed significant concentration-dependent relaxation effects on phenylephrine or KCl-contracted rat endothelium-denuded aorta. It also reduced the tension of the rat endothelium denuded aorta which was contracted in $Ca^{2+}-free$ media. On the other hand, it increased production of NO in VSMC which was stimulated with $IL-{\beta}$ or $IL-{\beta}$ plus $IFN-{\gamma}$. The water extracts of RAG and RF increased production of NO and induction of iNOS in the $IFN-{\gamma}-primed$ RAW 264.7 cells. Conclusions : According to the above results, the water extracts of RAG and RF relaxed the smooth muscle effectively and increased the production of NO in VSMC and macrophages. So, these herbs can be applied to erectile dysfunction, hypertension, angina pectoris, artherosclerosis and a defense defect for virus or microbe.

RAG-based Hierarchical Classification (RAG 기반 계층 분류 (2))

  • This study proposed an unsupervised image classification through the dendrogram of agglomerative clustering as a higher stage of image segmentation in image processing. The proposed algorithm is a hierarchical clustering which includes searching a set of MCSNP (Mutual Closest Spectral Neighbor Pairs) based on the data structures of RAG(Regional Adjacency Graph) defined on spectral space and Min-Heap. It also employes a multi-window system in spectral space to define the spectral adjacency. RAG is updated for the change due to merging using RNV (Regional Neighbor Vector). The proposed algorithm provides a dendrogram which is a graphical representation of data. The hierarchical relationship in clustering can be easily interpreted in the dendrogram. In this study, the proposed algorithm has been extensively evaluated using simulated images and applied to very large QuickBird imagery acquired over an area of Korean Peninsula. The results have shown it potentiality for the application of remotely-sensed imagery.

RAG-1 and IgM Genes, Markers for Early Development of the Immune System in Olive Flounder, Paralichthys olivaceus

  • Fish larvae are immediately exposed to microbes from hatching to maturation of their lymphoid organs, therefore effective innate mechanisms is very important for survival. However, the knowledge of the development of immune system in fish is limited and in demand now. In vertebrates, recombination-activating gene 1 (RAG-1) and immunoglobulin M (IgM) have been considered as very useful markers of the physiological maturity of the immune system. In this study, the expression of the both genes was assessed throughout the early developmental stages of olive flounder larvae (5-55 dph) and used as markers to follow the development of immune system. RAG-1 and IgM mRNA expression was detectable at 5 dph and remained so until 55 dph. These patterns of expression may suggest that the olive flounder start to develop its function around 5 dph. Tissue distribution was found that both genes mRNAs are only expressed in the immune-related organ such as spleen, kidney and gill. The early detection of IgM mRNA led to the investigation of its presence in oocytes. Both RAG-1 and IgM mRNA transcripts were detected in unfertilized oocytes, suggesting that they are maternally transferred. The biological significance of such a phenomenon remains to be investigated.

Species and Hybrid Identification of Genus Coreoleuciscus Species in Hwnag-ji Stream, Nakdong River Basin in Korea (낙동강 상류 황지천에 서식하는 쉬리속(genus Coreoleuciscus) 어류 집단의 종 동정 및 잡종 판별)

  • A natural hybrid of interspecific between the Coreoleuciscus splendidus and C. aeruginos (Cypriniformes: Cyprinidae) was captured in the Hwang-ji Stream, a tributary of the Nakdong River basin in Korea. An interspecific hybrid between C. splendidus and C. aeruginos was genetically identified based on morphological characteristics and the sequence analysis of nuclear recombination activating gene 1 (RAG1) gene (1,334 bp) and mitochondrial cytochrome c oxidase subunit 1 (CO1) gene (1,551 bp). As a result of morphological variations, the natural hybrid appeared to have an intermediate character between two parental species (C. splendidus and C. aeruginos) in three variations of black array (s) on dorsal, caudal and anal fin rays. Phylogenetic analysis inferred from RAG1 and CO1 sequence data revealed that Coreoleuciscus populations from Hwang-ji stream consist of two pure Coreoleuciscus species and a hybrid individual group. The individuals were clearly identified the cross and reciprocal hybrid by CO1 gene analysis. In RAG1 gene, 13 nucleotide variation loci were detected and the hybrid individuals displayed the double peaks of sequence chromatograms at the 9 diagnostic positions. In this study, molecular data and morphological variations were clearly demonstrated that hybridization did occur between C. splendidus and C. aeruginos. However, F2 hybrid generation and reproductive capacity of F1 hybrid individuals were not demonstrated.

Development of Dental Consultation Chatbot using Retrieval Augmented LLM (검색 증강 LLM을 이용한 치과 상담용 챗봇 개발)

  • In this paper, a RAG system was implemented using an existing Large Language Model (LLM) and Langchain library to develop a dental consultation chatbot. For this purpose, we collected contents from the webpage bulletin boards of domestic dental university hospitals and constructed consultation data with the advice and supervision of dental specialists. In order to divide the input consultation data into appropriate sizes, the chunk size and the size of the overlapping text in each chunk were set to 1001 and 100, respectively. As a result of the simulation, the Retrieval Augmented LLM searched for and output the consultation content that was most similar to the user input. It was confirmed that the accessibility of dental consultation and the accuracy of consultation content could be improved through the built chatbot.

  • Alzheimer's disease(AD) is a geriatric dementia that is widespread in old age. In the future AD will be the largest problem in public health service. From old times, Much medicines have been used for treatment of dementia, but there is no medicine having obvious effect. AD is one of brain retrogression disease. So We studied on herbal medicine that have a relation of brain retrogression. From old times, In Oriental Medicine, Radix Polygalae and Rhizoma Acori Graminei have been used for disease in relation to brain retrogression. We studied of anti-Alzheimer effect on CT105-induced neuroblastoma cell lines by Radix Polygalae(RP) and Rhizoma Acori Graminei(RAG) water extract. As the result of this study, In RP and RAG group, the apoptosis in the nervous system is inhibited, the repair against the degeneration of Neuroblastoma cells by CT105 expression is induced. These results indicate that In RP and RAG, RAG possess the strongest in inhibitory effect of apoptosis in the nervous system and repair effect against the degeneration of Neuroblastoma cells by CT105 expression.

Building Recognition using Image Segmentation and Color Features (영역분할과 컬러 특징을 이용한 건물 인식기법)

  • This paper proposes a building recognition algorithm using watershed image segmentation algorithm and integrated region matching (IRM). To recognize a building, a preprocessing algorithm which is using Gaussian filter to remove noise and using canny edge extraction algorithm to extract edges is applied to input building image. First, images are segmented by watershed algorithm. Next, a region adjacency graph (RAG) based on the information of segmented regions is created. And then similar and small regions are merged. Second, a color distribution feature of each region is extracted. Finally, similar building images are obtained and ranked. The building recognition algorithm was evaluated by experiment. It is verified that the result from the proposed method is superior to color histogram matching based results.

  • The snailfishes (Scorpaenoidei: Liparidae) is a large group requiring taxonomic reviews because of a potential taxonomic confusion due to its great similarity in external morphology between species and a variation of its body color with size. Molecular phylogeny and taxonomic review were conducted for 10 species in 3 genera reported around the Korean waters, which facilitate understanding interrelationship within its family, and further management of fisheries resources. Molecular phylogeny using mitochondrial COI (mtCOI) and nuclear RAG2 (nRAG2) sequences indicates that the reciprocal monophyly of the three genera (Careproctus, Crystallichthys, and Liparis) were supported except Crystallichthys matsushimae, being closely related to Careproctus notosaikaiensis in only mtDNA COI topology. Liparis ingens Gilbert and Burke (1912) was reported as a distinct species differing from L. ochotensis. However, in the present study, comparing L. ingens from Korea with L. ochotensis from Japan and Russia, the two species were well corresponded in their morphology and molecule, therefore, we suggest L. ingens to be treated as a junior synonym of L. ochotensis.
