이공산(異功散)의 혈관신생(血管新生) 및 암전이(癌轉移) 억제효과(抑制效果)에 관한 연구(硏究) (The Study on the Process and Quality Control of Rhus Verniciflua Stokes Extract (Nexia))

  • Ekongsan (EKS) was expected to have inhibitory effects on angiogenesis, considering the fact that its constituents such as Ginseng Radix, Glycyrrhizae Radix and Citri Pericarpium were reported to inhibit angiogenesis. Moreover, recently several metabolites transformed by the human intestinal microflora were reported to enhance effectiveness compared to their crude drugs. Based on these data, this study was designed to confirm whether the EKS metabolites (EKS-M) can significantly exert the anti-angiogenic and anti-metastatic activites. Hence, with EKS and EKS-M, viability assay, proliferation assay, in vitro tube formation assay, gelatin zymogram assay, in vitro invasion assay were carried out. EKS showed less toxicity in ECV304 and HT1080 cells than EKS-M. EKS-M inhibited the proliferation of HT1080 cells by 30% at 200 ${\mu}g/m{\ell}$ and 42% at 400 ${\mu}g/m{\ell}$ respectively. Also, EKS-M degraded the tube network at 200 ${\mu}g/m{\ell}$. EKS and EKS-M inhibited the expression of MMP-9 at 200 and 400 ${\mu}g/m{\ell}$in HT1080 cells. EKS reduced the invasive activity of HT1080 cells through matrigel coated transfilter at the concentration of 200 ${\mu}g/m{\ell}$ more effectively than EKS-M. These data suggest that EKS and EKS-M has anti-angiogenic and anti-metastatic activities.

Clostridium perfringens KCTC 5014의 성장 억제를 위한 항산화 천연산물의 효과 (The Growth Inhibiting Effect of Enterobacteria, Clostridium pefringens KCTC 5014, by the Combination of Natural Products)

  • C.perfringens의 성장은 선택되어진 천연산물의 한 종류 및 이들의 조합에 의하여 특징적으로 성장이 저해되었으며, 이것은 항산화력이 강한 천연산물들을 free radical의 제거와 동시 에 C.pefringens의 성장 억제 물질로 사용할 수 있고 이를 이용하여 기능성 항산화 제제의 제조가 가능하리라 사료된다. 한 종류의 천연산물, 백작약, 백출, 음양곽, 석창포, 오미자, 기 및 황련 등에 의하여 성장이 저해되었고, 황기는 40%의 제어율을 보여, 이 세균의 대사를 제어하는 성분이 있거나, 세포막의 수송 환경의 변화에 의한 물질의 전달이 제어되는 것으로 추정된다. 그리고 두 종류 천연산물의 조합인 황금과 감초, 황금과 상백피, 황백과 감초 및 황금과 황기등의 조합에 의해서 저해되었고, 황백과 감초의 조합에서 64%의 제어 효과를 보였다. 세 종류의 천연산물 조합인 황금과 황백 및 구기자, 황금과 황백 및 음양곽의 조합에 의하여 70%의 제어율을 보였고, 황백, 음양곽, 황기 및 감초의 조합등의 4종류의 천연산물 조합에 의해서도 성장이 저해되었으며, 75%의 제어율을 보였다. 4종류의 조합의 항산화력은 황백, 음양곽, 황기 및 감초 조합의 AUC가 581.89로써 대조군보다 4.4배 정도 강하였으며, 항산화력 이 강한 조합에서 제어가 잘되는 것으로 나타났다. Hydroxyl radical의 소거 활성은 황백, 음양곽, 황기 및 감초의 조합이 300배의 희석액에서 93.49%로서 최상의 소거율을 보였고, 방향족의 함량 및 총 phenol할량은 가공된 녹차보다 10배 정도 높은 것으로 나타났다. 이것은 항산화력이 강한 천연산물들을 이용하여 free radical의 제거와 동시에 C. pefringens의 성장 저해 물질로 사용할 수 있으며, 생체의 기능을 향상시킬 수 있는 기능성 제제로의 응용 가능성을 보였다.

  • We get the conclusion this following through bibliographic consideration about the cause of disease, disease mechanisms and remedy, prescription of tuberculosis of the cervical lymphic node. 1. The tuberculosis of the cervical Iymphic node is named the linear lump of scrofula(마도라력), the scrofula around neck(반사력), the wide-spread scrofula(류주력), the papule like lotus seed(련자력), the scrofula(라력), the subcutaneous nodes(結核), the scrofula due to disorder of Gi(기력) and the scrofula due to accumulation of phlegm(담력) according to the criterion of regions of disease, causes, rounding parts and shapes. 2. The cause of the tuberculosis of the cervical lymphic node are the stagnation of liver Gi(肝氣鬱結), the accumulation of phlegm and the stagnation of Gi(痰凝氣滯), the deficiency of vital essence of the liver and kidney(肝腎陰虛), the wind-heat of liver, gallbladder and triple warmer(肝膽三焦風熱), the dry-fire(燥火), the dryness of blood(血燥), the unwholesome diet(飮食不潔), the abundance of diet(食味之厚), the stagnation of Gi(鬱氣之積), the exhaustion syndrome(虛勞), the excessive thought(思慮過多), the toxin of wind-heat(風熱毒) and the germ of subcutaneous nodes(結核菌). 3. Symptoms of the tuberculosis of the cervical lymphic node are swelling slowly in comparison in the early days of occurrence, and are not pain, not itch, not heat, not other special symptoms. But in some cases, tuberculosis of the cervical lymphic node are quickly swelling in the early days of occurrence, and the fever and pain appear, The pyosis is accompanied with the fever and the pain at first generally, and then removing the pus from abscess is dissolved the fever and the pain in the majority of cases and representative of the general exhaustion syndrome. 4, The remedy of tuberculosis of the cervical Iymphic node is the early days, clearing the liver and relieves constraint(疏肝解鬱), phlegm and dissolving accumulation(化痰散結) the middle days, pus draining and toxin expelIing(托裏透膿) the latter days, replenish the kidney and strengthen the spleen(滋腎健脾). 5. The medication to treat the tuberculosis of the cervical lymphic node are the 益氣養榮湯(lkgiyangyoungtang)(14th), the 夏枯草散(Hagochosan)(10th) and et cetra in regular sequence. 6. The herb to treat the tuberculosis of the cervical Iymphic node are the 連翹(FRUCTUS FORSYTHlAE)(59th), the 甘草(RADIX GLYCYRRHIZAE)(51th), the 當歸(RADIX ANGELICAE GlGANTIS)(47th), the 黃芩(RADlXSCUTELLARIAE)(40th), the 夏枯草(SPICA PRUNELLAE)(23th) in regular sequence.

Effects of Kaolinite (Macsumsuk) and Herb Mixtures on the Quality and Physicochemical Properties of Pork

  • The current study was conducted to identify technology for the production of high quality pork, based on the meat consumption habits of consumers. Macsumsuk, a type of kaolinite (a clay mineral), and/or a mixture of herbs (Mori Folium, Sophorae Radix, Glycyrrhizae Radix, Citri Leiocarpae Exocarpium, and Pogostemi Herba) were added to the diets of fattening pigs. Sixty barrow pigs (4 kinds of treatment ${\times}5$ pigs/treatment ${\times}3$ replicates) were randomly assigned to either the Control (no additives), T1 (3% Macsumsuk), T2 (3% Herb mixtures), or T3 (3% Macsumsuk + 3% Herb mixtures) groups, and were fed the diets for 60 d. Dressed weights were in the order of T1 ($93.40{\pm}4.68kg$) > T2 ($91.40{\pm}6.52kg$) > Control ($88.80{\pm}1.57kg$) > T3 ($86.80{\pm}2.01kg$). Back-fat thickness of the Control animals ($23.2{\pm}1.03mm$) was significantly greater than that of the various treatment groups (p<0.01). Numeric values representing the carcass yield and quality grade were higher for all the treated groups than the Control group, thought the difference was not statistically significant. Crude fat content was significantly higher in the Control group ($2.23{\pm}0.34%$) than in the treated groups (p<0.05). The addition of both Macsumsuk and herb mixtures into the diets of the pigs significantly reduced cooking loss of the pork compared to the Control (p<0.05). No statistically significant changes were observed in the shear force (average $5.87{\pm}0.54kg/cm^2$), water holding capacity (average $54.59{\pm}3.16%$), or CIE values of the pork, whereas cholesterol levels significantly decreased (p<0.01) in those fed Macsumsuk and/or the herb mixtures. No significant changes in fatty acid composition, total saturated fatty acid (SFA), total unsaturated fatty acid (UFA), or UFA/SFA ratios were observed by any of the treatments. In conclusion, the results obtained from this study suggest that the addition of Macsumsuk and/or herb mixtures into the diets of growing and fattening pigs improves the pork quality by reducing cooking loss, decreasing cholesterol content, and enhancing sensory characteristics.

Toxicity of Eun-bi San Pharmacopuncture Injection inthe Muscle of Rats

  • Objectives: This study was carried out to analyze the single-dose toxicity of Eun-Bi San pharmacopuncture injected into the muscle of Sprague-Dawley (SD) rats. Methods: All experiments were performed at Biotoxtech, an institution certified to conduct non-clinical studies under the Good Laboratory Practice (GLP) regulations. Six week old SD rats reared by ORIENTBIO were chosen for this pilot study. The reason SD rats were chosen is that they have been widely used in safety tests in the field of medicine, so the results can be easily compared with many other databases. The Eun-Bi San pharmacopuncture was made in a clean room at the Korean Pharmacopuncture Institute (KPI, K-GMP). The constituents of the Eun-Bi San pharmacopuncture are Angelicae gigantis radix, Strychni semen and Glycyrrhizae radix. These were extracted at low temperature and low pressure in an aseptic room at the KPI. Doses of Eun-Bi San pharmacopuncture, 0.25, 0.5 and 1.0 mL, were administered to the experimental groups, and a dose of normal saline solution, 1.0 mL, was administered to the control group. This study was performed under the approval of the Institutional Animal Ethics Committee of Biotoxtech Co., Ltd. Results: No deaths or abnormalities occurred in any of the four groups. No significant changes in weight, hematological parameters or clinical chemistry between the control group and the experimental groups were observed. To determine if abnormalities existed in any organs and tissues, we used microscopy to examine representative histological sections of each specified organ; the results showed no significant differences in any of the organs or tissues. Conclusion: The results of this study suggest that treatment with Eun-Bi San pharmacopuncture is relatively safe and that its clinical use may be beneficial. Further evaluations and studies on this subject will be needed to provide more concrete evidence in support of these conclusions.

영양각산이 Thioacetamide 유발 간손상에 미치는 영향 (Effect of Youngyanggak-san against Thioacetamide Induced Acute Liver Damage in Rat)

  • Objectives : The current study is to evaluate the hepatoprotective effect of youngyanggak-san (YGS) on thioacetamide (TAA)-induced acute liver injury in rats. Methods : YGS is composed of Glycyrrhizae Radix, Asiasari Radix, Cimicifugae Rhizoma, Saigae Tataricae Cornu. While N-YGS (non-youngyanggak-san) doesn't include Saigae Tataricae Cornu. Two samples were administrated TAA together for 3 days. Thirty-six rats were divided into four groups. Rats except for the normal group were received TAA (200 mg/kg of body weight, I.P) were divided into three groups (n=9/group) : Group 1 (TAA only), Group 2 (TAA + 200 mg/kg YGS) and Group 3 (TAA + 200 mg/kg N-YGS). Acute liver damage confirmed using histological examination, The factors associated with oxidative stress and liver function activity measured in serum. Also, expressions of inflammation related proteins were investigated by western blot analysis. Results : Oxidative stress factors such as ROS and $ONOO^-$ in the Group 2 was manifested by a significant rise compared with Group 1. YGS markedly decreased the elevated ROS and $ONOO^-$. Furthermore, YGS significantly reduced the levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) The nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) activation induced by TAA led to increase both inflammatory mediators and cytokines. While YGS administration remarkably suppressed such the overexpression. In addition, the histopathological analysis showed that the liver tissue lesions were improved obviously in YGS treatment. Conclusion : YGS provided a hepatoprotective effect on acute liver damage through the suppression of oxidative stress. Especially, this effect enhanced markedly when Saigae Tataricae Cornu is included.

임신 중 코로나바이러스감염증-19 감염의 한방치료 적용 가능성에 대한 고찰 (A Review of the Applicability of Traditional Chinese Medicine on Coronavirus Disease 2019 Infection during Pregnancy)

  • Objectives: This study was performed to review the impact of Coronavirus disease 2019 (COVID-19) infection during pregnancy and the applicability of traditional Chinese medicine (TCM) on COVID-19 infection during pregnancy. Methods: We searched 6 data bases on August, 2022 and articles about the impact of COVID-19 infection during pregnancy, TCM treatment of COVID-19 infection, TCM treatment of pregnancy common cold (姙娠 感冒) or cough (姙娠 咳嗽) were reviewed. Results: COVID-19 infection can bring out negative effects both on pregnant women and fetus. In the case of COVID-19 infection during pregnancy, the symptoms and treatment are similar to those of general COVID-19 infection, but the safety of drug intervention has not been completely verified. Herbal medicine treatment can be applied according to the severity, stage and TCM syndrome types of COVID-19 infection, but the prohibited herbs list during pregnancy and its dosage should be checked carefully. Also, when it comes to pregnancy disease, the effect of "Stabilizing Fetus" should be considered in the perspective of TCM treatment strategy. Glycyrrhizae Radix et Rhizoma, Rhizoma Atractylodis Macrocephalae, Scutellariae Radix have been widely used on COVID-19 infection and the safety of them during pregnancy have been verified. In the case of acupuncture treatment, acupoints also can be selected according to the severity, symptoms, and TCM syndrome types of COVID-19, but acupoints with strong Qi sensitivity or acupoints located in lower abdomen should be avoided during pregnancy. Conclusions: TCM treatment can be applied to COVID-19 infection during pregnancy. Clinical studies and systematic reviews of the efficacy and safety of TCM treatment on COVID-19 infection during pregnancy are required.

특발성 혈소판 감소성 자반증의 중의치료에 대한 임상연구 동향 - CNKI검색을 중심으로 (A Review of Clinical Studies for Chinese Medicine Treatment of Idiopathic Thrombocytopenic Purpura Using the CNKI Database)

  • Objectives: The aim of this study was to analyze the latest clinical studies on Korean medicine treatment of idiopathic thrombocytopenic purpura (ITP) in the Chinese National Knowledge Infrastructure (CNKI) database. Methods: We searched the last 6 years of clinical studies discussing Oriental medicine-based treatments for ITP in the CNKI database. A meta-analysis of 13 RCTs was performed by synthesizing the outcomes, including the measured platelet count and total effective rate. The quality of the studies was assessed using Cochrane's risk of bias (RoB) tool. RevMan 5.4.1 software was used for data analysis. Results: Of the 15 selected studies, 1 was a non-randomized controlled trial (nRCT), 2 were case series, and 12 were randomized controlled trials (RCTs). Treatments in all studies included oral herbal medicine. The most frequently used herbal decoction was the Liangxue Jiedu prescription (凉血解毒方), and the most commonly used herb was Agrimonia pilosa (仙鶴草), Astragali Radix (黃芪), Fossilia Glycyrrhizae Radix et Rhizoma (甘草), and Rehmannia glutinosa Liboschitz ex Steudel (地黃). The meta-analysis showed significantly better improvement in platelet counts and total effective rate for ITP in the treatment group than in the control group. Conclusion: Treatment with herbal medicine was effective in treating ITP. However, the significance of this conclusion is somewhat limited due to the low quality of the available studies. Multifaceted and scientifically designed clinical studies are required to develop treatments for ITP based on Korean medicine. The results of this study could be used as basic data for further ITP studies.

Protective Effect of Isoliquiritigenin against Ethanol-Induced Hepatic Steatosis by Regulating the SIRT1-AMPK Pathway

  • Ethanol-induced fat accumulation, the earliest and most common response of the liver to ethanol exposure, may be involved in the pathogenesis of liver diseases. Isoliquiritigenin (ISL), an important constituent of Glycyrrhizae Radix, is a chalcone derivative that exhibits antioxidant, anti-inflammatory, and phytoestrogenic activities. However, the effect of ISL treatment on lipid accumulation in hepatocytes and alcoholic hepatitis remains unclear. Therefore, we evaluated the effect and underlying mechanism of ISL on ethanol-induced hepatic steatosis by treating AML-12 cells with 200 mM ethanol and/or ISL ($0{\sim}50{\mu}M$) for 72 hr. Lipid accumulation was assayed by oil red O staining, and the expression of sirtuin1 (SIRT1), sterol regulatory element-binding protein-1c (SREBP-1c), AMP-activated protein kinase (AMPK), and peroxisome proliferator-activated receptor alpha ($PPAR{\alpha}$) was studied by western blotting. Our results indicated that ISL treatment upregulated SIRT1 expression and downregulated SREBP-1c expression in ethanol-treated cells. Similarly, oil red O staining revealed a decrease in ethanol-induced fat accumulation upon co-treatment of ethanol-treated cells with 10, 20, and $50{\mu}M$ of ISL. These findings suggest that ISL can reduce ethanol induced-hepatic lipogenesis by activating the SIRT1-AMPK pathway and thus improve lipid metabolism in alcoholic fatty livers.

식품학적 가공에 의한 생약의 성분 및 활성 변화 (I) - Aspergillus kawachii 유래 조효소액 처리에 의한 감초 추출물 중 Liquiritigenin의 함량변화 (Changes in Chemical Composition and Biological Activities of Oriental Crude Drugs by Food Processing Techniques (I) - Changes in Liquiritigenin Contents in Licorice Extract Treated by the Crude Enzyme Extract from Aspergillus kawachii)

  • By treating crude enzyme extract from Aspergillus kawachii, the liquiritigenin content in the licorice (Glycyrrhizae Radix) was significantly increased. The liquiritigenin content reached its maximum level (45.7 mg/g licorice extract) after 60 min of incubation with the crude enzyme extract at $37^{\circ}C$, while the inactivated crude enzyme treated control contained trace amount (about 0.11 mg/g) of liquiritigenin. The enzyme-treated licorice extract inhibited more than 50% DPPH radical at 100 ppm and this was about two times higher activity compared to the enzyme-untreated control.
