인간 간조직세포(HepG2 Cells)에 대한 한약조성물 KOCO-P1의 효과 연구 (Study on Biological Effect of Multi-Herbal Drug KOCO-P1 on Human Hepatocytes (HepG2))

  • Objectives : The purpose of this study is to investigate the biological Effect of multi-herbal drug 'KOCO-P1' on human hepatocyte HepG2 cells. Methods : Multi-herbal drug 'KOCO-P1' was composed of Ginseng Radix, Astragali Radix, Polygonati Rhizoma, Liriopis Tuber, and Scrophulariae Radix. Cytotoxicity and cytoprotective activity of KOCO-P1 was verificated by MTT assay. And antioxidative effect of KOCO-P1 against EtOH, Nicotine was inspected by Hydroperoxide assay. Results : KOCO-P1 showed no cytotoxicity on HepG2 cells for 24, 48 hours. KOCO-P1 at 50 ${\mu}g/mL$ reduced the production of H2O2 in HepG2 cells by EtOH. KOCO-P1 at 50 ${\mu}g/mL$ reduced the production of $H_2O_2$ in $HepG_2$ cells by Nicotine. Conclusions : KOCO-P1 at the low concentration could be supposed to have antioxidative effect on human hepatocyte with no cytotoxicity.

Effect of Xingyo-tang on Learning and Memory Performances in Mice

  • The effects on memory and learning ability of the Korean herbal medicine, Xingyo-tang(XGT, 神交湯), which consists of Ginseng Radix(人蔘) 4 g, Liriopis Tuber(麥門冬) 40 g, Morindae Officinalis Radix(巴戟天) 40 g, Biotae Semen(柏子仁) 20 g, Dioscoreae Rhizoma(山藥) 40 g, Euryales Semen(?實) 20 g, Scrophulariae Radix(玄蔘) 40 g, Salviae Miltiorrhizae Radix(丹蔘) 12 g, Poria(茯神) 12 g, Cuscutae Semen(免絲子) 40 g, was investigated. The effects of XGT on learning and memory performance were examined in normal or memory impaired mice by using avoidance tests, Pentobarbital -induced sleep test, fear conditioning task, novel object recognition task, and water maze task. Hot water extract from XGT was used for the studies. Learning ability and memory are based on modifications of synaptic strength among neurons that are simultaneously active. Enhanced synaptic coincidence detection leads to better learning and memory. The XGT-treated (30 mg/100 g and 60 mg/100 g, p.o.) mice exhibit superior ability in learning and memorizing when performing various behavioral tasks. XGT did not affect the passive avoidance responses of normal mice in the step through and step down tests, the conditioned and unconditioned avoidance responses of normal mice in the shuttle box, lever press performance tests, and the ambulatory activity of normal mice in normal condition. In contrast, XGT produced ameliorating effects on the memory retrieval impairment induced by ethanol. XGT also improved the memory consolidation disability induced by electric convulsive shock (ECS). XGT extended the sleeping time induced by pentobarbital dose-dependently, suggesting its transquilizing or antianxiety action. These results suggest that XGT has an improving effect on the impaired learning through the effects on memory registration and retrieval.

유통 한약재 중 벤조피렌 함량 모니터링 및 안전성 평가 (Monitoring and Risk Assessment of Benzo(a)pyrene Content in Medicinal Herbs)

  • In this study, to investigate of safety for Benzo(a)pyrene in medicinal herb, 8 kinds of selected commercial herbal medicines (Rehmanniae Radix, Rehmanniae Radix Preparata, Amomi Tsao-Ko Fructus, Cimicifugae Rhizoma, Cyperi Rhizoma, Magnoliae Cortex, Scutellariae Radix, Scrophulariae Radix) were analysed using the high performance liquid chromatography with fluorescence detector and assessed the health risk. The levels of benzo(a)pyrene were from non-detection to $28.1{\mu}g/kg$, and the average was $3.6{\mu}g/kg$. Based on a nationwide survey of the consumption of medicinal herb by the Korean population, we estimated the potential risk from the ingestion of benzo(a)pyrene. The estimated daily intake of benzo(a)pyrene was 1.6 ng/kg b.w./day for group only know the daily average intake of medicinal herb. The MOE (margin of exposure) of benzo(a)pyrene for estimate of health risk was $1.93{\times}10^5$. Therefore, health risk from benzo(a)pyrene through intake of herbal medicine was considered negligible.

소음인(少陰人)으로 오진(誤診)한 소양인(少陽人) 결흉(結胸) 치험례(治驗例) (A case of patient diagnosed as and treated with Hyeongbangdojeoksan)

  • One woman, Sixty three years old, suffered from chest pain, abdominal pain, nausea and vomiting for about three months. Although many examinations, brain magnetic resonance imaging(MRI), chest computed tomography(CT), abdominal CT, gastric endoscope, abdominal ultrasonography, and so forth, was performed from every point of view, those were non-significant. At first, I wrongly diagnosed her case as disease of Soumin and gave Soumin Gwakhyangjeongkisan. But she complained of diarrhea and chest discomfort. I re-diagnosed her case as Soyangin Gyeolhyung and treated with Hyeongbangdojeoksan, herbal medicine, consisted of Rhizoma Rehmanniae(生地黃), Lignum Akebiae(木通), Radix Scrophulariae(玄蔘), Semen Trichosanthis(瓜蔞仁), etc. Three weeks have passed from first visit and she did not suffered from pain. I experienced the excellent effect of Hyeongbangdojeoksan to Soyangin Gyeonlhyung.

현삼(玄蔘)이 NC/Nga mice에서 유발된 피부염에 미치는 영향 (Inhibitory Studies of Scrophulariae Radix on Development of Dermatitis in NC/Nga Mice)

  • Objectives The purpose of this study is to examine of the effect of SPAR medicines on the atopy eruption control Methods This experiment is about the expression of IgE, IL-4, IL-6, IL_13, IgM, IgG2a, IgG2b, IgG1 level in serum, and $IFN-{\gamma}$ production by SPAR medicines. We assayed for $CD3e^+/CD69^+$, $CD044^+/CD19^+$ positive cells by flow cytometry in splenocytes and observed the revelation of $CD3e^+/CD69^+$, $CD4^+/CD8^+$, $CD44^+/CD19^+$ marker in PBMC, spleen and DLN. We also observed the outturn of IL-4, IL-5, CCR3, $IFN-{\gamma}$ in skin of a NC/Nga mice. We also analyzed NC/Nga mice's ear and neck-back skin after biopsy and dye by H&E staining method, measured about epidermis and dermis part in comparison with control group. Results SPAR medicines as treatment result to a NC/Nga mice, clinical skin severity score decreased remarkably than the ontrol group. Specially, experiment was results by measuring IgE and IL-6 content in serum 8 weeks, 10 weeks, 12 weeks, 16 weeks, 20 weeks respectively, and it was decreased remarkably than the control group. After experiment ended, the result that observed the revelation CD3e, CD4, CD8, CD19, CD69, CD11a marker in lymph node establishment were observed and that B/T rate becomes recover as normal with political background. In addition to that, the control group was decreased in the measured value of IL-4, IL-5, IL-13, IgM, IgG2a, IgG1's level in serum, and $IFN-{\gamma}$' production secreted in Th1 cell displayed increase by SPAR medicines. IL-4, IL-5, CCR3, and $IFN-{\gamma}$'s gene revelation amount displayed marked decrease than the control group in result that observe effect that get in skin of a NC/Nga dermatitis mouse. Moreover in culture supernatant which cultivate for 14 days after separate skin cell, IL-13 and IL-6 production, and $CD69^+/CD3e^+$, $CD44^+/CD19^+$ expression cell number was decreased than the control group's number. Course inflammation immunocyte permeated of result that effect that SPAR medicines get to NC/Nga mice's skin establishment analyzes ear and neck-back skin after biopsy, and dye by H&E method decreased about epidermis and inflammation of dermis part remarkably than the control group. Conclusions Th1 cell and Th2 cell observe to be shifted by secretion amount of IL-4 and $IFN-{\gamma}$ by SPAR medicines could know that SPAR medicines can be use for treatung allergy autoimmune disease.

  • Authors investigated the pathogenesis and treatment of dennatosurgical diseases in the ImJeungJiNamUiAn(臨證指南醫案). 1.   • On the basis of Imjeungjinamuian(臨證指南醫案), authors investigated the pathogenesis and treatment of ophthalmotolaryngobgic diseases from the viewpoint of Onbyeong(溫病). 1. The symptoms and diseases investigated according to department were as follows;. 1) Ophthalmology : blepharitis, blepharedema, lacrimal hypersecretion, hyperemia, ophthalmalgla, photopsia, visual disturbance, mydriasis 2) Otology : full-feeling, otorrhea, otalgla, mastoiditis, tinnitus, hearing disturbance, vertigo 3) Rhinology : rhinorrhea, nasal obstruction, sinusitis, epistaxis 4) Laryngology : sore throat, hoarseness 5) The Others : headache, cough, asthma 2. The pathogenesis and treatment of ophthalmotolaryngologic diseases were as follows. 1) When the pathogenesis of hyperemia, otorrhea, otalgia, mastoiditis, hearing disturhance. epistaxis, sore throat, headache and cough are wind-stagnanc(風鬱), wind-warm(風溫), wind-fire(風火), wind-dryness(風燥), dryness-heat(燥熱), the treatment of pungent-cool-evaporating(辛凉解表) with Dajosan(茶調散), Mori Folium(桑葉), Lonicerae Flos(金銀花), Forsythiae Fructus(連翹), Viticis Fructus(蔓荊子), Prunellae Spica(夏枯草), Arctii Fructus(牛蒡子), etc can be applied. 2) When the pathogenesis of hoarseness, cough and asthma are cold(寒), cold with endogenous heat(寒包熱, 外冷內熱), water retention(水邪), fluid retention(伏飮), impairment of YangKi by overexertion(勞傷陽氣), the treatment of pungent-warm-evaporating(辛溫解表) with Mahaenggamseoktang(麻杏甘石湯), Socheongryongtang(小靑龍湯), Jeongryeokdaejosapyetang(정력대조사폐탕), Gyejitang(桂枝湯), Armeniacae Amarum Semen(杏仁), etc can be applied. 3) When the pathogenesis of photopsia, otorrhea, otalgia, rhinorrhea, sinusitis, epistaxis, sore throat, hoarseness, headache and cough are stagnancy-induced heat(鬱熱), wind-dryness(風燥), wind-heat(風熱), summer heat(暑熱), summer wind(暑風), insidious summer heat(伏暑), autumn heat(秋暑), autumn wind(秋風), autumn dryness(秋燥), dryness-heat(燥熱), heat in Ki system(氣分熱), insidious warm(溫伏), brain discharge by fire in Ki system(氣火 腦熱), heat in stomach(胃熱), endogenous fire by deficiency of Yin(陰虛內火), deficiency of Yin in stomach(胃陰虛), the treatment of Ki-cooling(淸氣) with Bangpungtongseongsan(防風通聖散), Ikweonsan(益元散), Gyejibaekhotang(桂枝白虎湯), Geumgwemaekmundongtang(금궤맥문동탕), Gyeongokgo(瓊玉膏), Sojae Semen Praeparatum(두시), Scutellariae Radix(黃芩), Phyllostachys Folium(竹葉), Adenophorae Radix(沙參), Mori Cortex(桑白皮), Fritillariae Cirrhosae Bulbus(貝母), etc can be applied. 4) When the pathogenesis of blepharitis, hyperemia, ophthalmalgia, full-feeling, otorrhea, otalgia, tinnitus, hearing disturbance, sinusitis, hoarseness, headache and cough are fire in liver(肝火), fire in gallbladder(膽火), ministerial fire in Soyang system(少陽相火), wind-stagnancy(風鬱), stagnancy-induced fire(鬱火), brain discharge by phlegm-fire(痰火 腦熱), the treatment of mediation(和解) with Gardeniae Fructus(梔子), Moutan Cortex(牧丹皮), Saigae Tataricae Cornu(羚羊角), Artemisiae Annuae Herba(靑蒿), Cyperi Rhizoma(香附子), Poria(적복령), etc can be applied. 5) When the pathogenesis of blepharedema and cough are dampness in both spleen and lung(脾肺濕) damp-heat(濕熱), damp-phlegm(濕痰), the treatment of dampness-resolving(化濕) with Poria(백복령), Coicis Semen(薏苡仁), Tetrapanacis Medulla(通草), Armeniacae Amarum Semen(杏仁), Talcum(滑石), etc can be applied. 6) When the pathogenesis of vertigo and cough are deficiency of Yong(營虛), heat in Yong, system(營熱), the treatment of Yong-cooling(淸營) with Rehmanniae Radix(生地黃), Liriopis Tuber(麥門冬), Biotae Semen(柏子仁), Lilii Bulbus(百合), Phyllostachys Folium(竹葉), etc can be applied. 7) When the pathogenesis of epistaxis are heat in blood system of heart(心血熱), reversed flow of fire(火上逆), overexertion(努力), the treatment of blood-cooling(凉血) with Rhinoceri Cornu(犀角), Rehmanniae Radix(生地黃), Moutan Cortex(牧丹皮), Salviae Miltiorrhizae Radix(丹參), Scrophulariae Radix(玄蔘), etc can be applied. 8) When the pathogenesis of nasal obstruction is pathogen-stagnancy(邪鬱), the treatment of resuscitation(開竅) with Sosang(少商, LU11) acupuncture can be applied. When the pathogenesis of hoarseness is evil Ki(穢濁), the treatment of resuscitation(開竅) with Arctii Fructus(牛蒡子), Lasiosphaera Seu Calvatia(馬勃), Curcumae Radix(鬱金), etc can be applied. When the pathogenesis of headache is stasis of both Ki and blood(氣血瘀痺), the treatment of resuscitation(開竅) with Cnidii Rhizoma(川芎), Asari Herba Cum Radice(細辛), Scorpio(全蝎), moxibustion(灸), etc can be applied. 9) When the pathogenesis of lacrimal hypersecretion, visual disturbance, mydriasis, tinnitus, hearing disturbance, sinusitis, epistaxis, hoarseness and cough are deficiency of Yin(陰虛), deficiency of kidney(腎虛), deficiency of both liver and kidney(肝腎虛), deficiency of both heart and kidney(心腎虛), brain discharge by deficiency of Yin(陰虛 腦熱), exuberance of Yang in liver(肝陽上亢), overexertion(勞損), the treatment of Yin-replenishing(滋陰) with Yukmijihwanghwan(六味地黃丸), Hojamhwan(虎潛丸), Jeobutang(猪膚湯), Lycii Fructus(枸杞子), Polygoni Multiflori Radix(何首烏), Rehmanniae Radix(生地黃), Schizandrae Fructus(五味子), Liriopis Tuber(麥門冬), Asini Gelatinum(阿膠), etc can be applied. 10) When the pathogenesis of ophthalmalgia, mydriasis, vertigo and headache are deficiency of Yin in liver(肝陰虛), exuberance of Yang in liver(肝陽上亢), endogenous wind(內風), excess in upper and deficiency in lower part(上實下虛), the treatment of Yin-replenishing(滋陰) and endogenous wind-calming(熄風) with Rehmanniae Radix Preparat(熟地黃), Lycii Fructus(枸杞子), Polygoni Multiflori Radix(何首烏), Paeoniae Radix Alba(白芍藥), Ostreae Concha(牡蠣), Saigae Tataricae Cornu(羚羊角), Chrysanthemi Flos(菊花), etc be applied. 11) When the pathogenesis of mydriasis, sinusitis, hoarseness, headache, cough and asthma are exhaustion of vital essence(精氣無收藏), brain discharge(腦髓不固), floating Yang(陽虛浮), exsanguination(失血), deficiency of both Yin and Yang(陰陽不足), overexertion(勞損), deficiency of Yang in kidney(腎陽虛), the treatment of Yang-restoring and exhaustion-arresting(回陽固脫) with Yangyeongtang(養營湯), Cheonjinhwan(天眞丸), Bokmaektang(복맥탕), Geonjungtang(建中湯), Dogihwan(都氣丸), Singihwan(腎氣丸), Jinmutang(眞武湯), Ostreae Concha(牡蠣), Nelumbinis Semen(蓮子肉), etc can be applied. 12) When the pathogenesis of lacrimal hypersecretion, vertigo and headache are deficiency of stomach and endogenous wind(胃虛內風), endogenous wind with phlegm(內風挾痰), liver check of stomach(肝木橫擾), the treatment of concomitant-treating of both liver and stomach(肝胃同治) with Paeoniae Radix Alba(白芍藥), Uncariae Ramulus Et Uncus(釣鉤藤), Gastrodiae Rhizoma(天麻), Astragali Radix(황기), Pinelliae Rhizoma(半夏), etc can be applied. When the pathogenesis of asthma is failure of kidney to promote inspiration(腎不納氣), the treatment of kidney-tonifing and inspiration-promoting(補腎納氣) with Singihwan(腎氣丸), Psoraleae Fructus(補骨脂), Juglandis Semen(胡桃), Aquilariae Resinatum Lignum(沈香), etc can be applied. When the pathogenesis of asthma is deficiency of Ki(氣虛), the treatment of Ki-reinforcing(補氣) with Sagunjatang(四君子湯), Insamgeonjungtang(人參建中湯), etc can be applied.

벤조피렌 기준 미설정 한약재의 오염도 조사 및 위해도 결정 (Investigation and Risk Characterization on the Contamination Level of Herbal Medicines Without Legal Benzo(a)pyrene Specification)

  • By analysing the benzo(a)pyrene concentration using HPLC-FLD and LC-MS/MS, pollution levels of herbal medicines without permitted benzo(a)pyrene specification were investigated. Average benzo(a)pyrene concentrations were 38.30, 37.46 and 21.22 ㎍/kg for Amomi Tsao-ko Fructus, Mume Fructus and Coptidis Rhizoma, respectively, which are 4 to 7 times higher than maximum permitted benzo(a)pyrene concentration of Rehmanniae Radix Preparata and Rehmanniae Radix, i.e. 5.0 ㎍/kg. Proportion of detected samples exceeding 5.0 ㎍/kg benzo(a)pyrene concentration was 22% for Cimicifugae Rhizoma and Scrophulariae Radix, 44% for Forsythiae Fructus, 67% for Mume Fructus, 100% for Amomi Tsao-ko Fructus and Coptidis Rhizoma, and collectively 29% (36 out of 125 samples) in average. In terms of risk characterization results, human exposure of benzo(a)pyrene were 7.96, 3.49 and 1.61 ng/kg b.w./day and the margin of exposure(MOE) were 1.25 × 104, 2.86 × 104 and 6.20 × 104 for Mume Fructus, Amomi Tsao-ko Fructus and Coptidis Rhizoma, respectively. MOE banding of those herbal medicines was categorized to 'low concern'. However, considering that human exposure of benzo(a)pyrene for food is legislated to 1.4~2.5 ng/kg b.w./day, it was urgent to set up the guideline of benzo(a)pyrene in herbal medicines.

소갈 치료 처방 가감이 3T3-L1 Adipocytes에서 인슐린 유사성과 인슐린 민감성에 미치는 영향 (Insulin-like and Insulin Sensitizing Effects of Modified Anti-diabetic Remedies in 3T3-L1 Fibroblasts)

  • Based on the data from our previous studies, four new diabetic remedies were composed with the addition of Coicis Semen into Okchun-san (OCH), Commelinae Herba into Gangsim-tang (GST) , Scrophulariae Radix into Sunki-san (SKS), and Erythrinae Cortex into Yukmijihuang-hwan (YMG). The water extracts of these new remedies were treated in 3T3-L1 fibroblasts and adipocytes in order to investigate insulin-like substances and insulin sensitizers, respectively. With and without differentiation inducers, unmodified SKS (SKS-O) treatment induced 3T3-L1 fibroblasts into adipocytes more than the control. However, without inducers, YMG treatment, but not SKS, induced the differentiation more than the control among modified remedies. Without inducers, SKS, OCH as well as YMG increased the induction of differentiation from 3T3-L1 fibroblasts into adipocytes, compared to the control. The treatment of OCH and YMG with 1 ng/mL insulin increased glucose uptake much more than only insulin 1 ng/mL treatment. Thus, OCH and YMG contained increased insulin actions. In conclusions, the modified remedies, OCH and YMG, contained insulin-like substances and insulin sensitizers, and they can be improved the hypoglycemic effects.

황약자가 갑상선기능항진 유발 백서에 미치는 영향 (The Effects of Dioscorea bulbifera L. on Hyperthyroidism of Rats)

  • Objective: The aim was to study experimentally the effects of Dioscorea bulbifera L. induced by sodium levothyroxine. Methods: Sprague-Dawley rats were separated into four groups. Except for the normal group, the other three were treated with sodium levothyroxine $160{\mu}g/kg$ for 5 days. Among the three groups, two (except the control) were treated with Dioscorea bulbifera L. extract of 0.75g/kg (sample A) or 1.5g/kg (sample B). $T_3-uptake,\;T_3,\;T_4$, TSH, total cholesterol, glucose, ALP, AST, ALT, tree fatty acids, $\beta-lipoprotein$, and change in the serum of rats were measured after medication with solid extract of Radix Scrophulariae. Results: $T_4$ concentration and $T_3-uptake$ level decreased in sample A and B compared to controls. $T_3$ concentration decreased but not statistically meaningful. Considering lipid metabolism, only free fatty acid level was significantly reduced glucose level also significantly decreased in sample B. Serum AST, ALT and ALP levels were slightly elevated but there was no meaning statistically, which doesn't mean Dioscorea bulbifera L. has liver toxicity because the quantity of the extract perhaps couldn't reach the minimal dose causing liver toxicity, which according to an article was 60g. Body weight significantly increased in comparison to controls. Conclusion: These results suggest that Dioscorea bulbiftra L. lowers excess thyroid hormone and increased metabolism, resulting in improvement of hyperthyroid state.
