Identification 4 kinds of Muxiang using Multiplex PCR (Multiplex PCR을 이용한 4 종류 목향(木香)의 감별)

  • Objectives : Aucklandiae Radix (Muxiang) one of important herbal medicines in oriental medicine, is defined as the dried root of Aucklandia lappa (Asteraceae). Owing to the similarities in the morphology and name, Inulae Radix (Tu-Muxiang) and Vladimiriae Radix (Chuan-Muxiang) as well as Aristolochiae Radix (Qing-Muxiang) originated from other medicinal plants are often used as substitutes and/or adulterants of Aucklandiae Radix. Therefore, a reliable authentication of these herbal medicines is necessarily for the public health and prevention of misuse. Methods : 32 samples of medicinal plants supplying Aucklandiae Radix, Inulae Radix, Vladimiriae Radix, and Aristolochiae Radix were collected in Korea and China. The ITS (Internal transcribed spacer) nucleotide sequences of samples were determined. The PCR primers to amply DNA marker of each herbal medicine were designed basing on the specific ITS regions showing differences in the sequences among medicinal plants. Results : Primer set Al R/IS F designed in this work amplified 220 bp PCR product only in samples of Aucklandiae Radix. In contrast, primer set Ih F/IS R, Vs R/IS F, and AcR F1/Ac R amplified 250 bp product, 356 bp prouct, and 516 bp product respectively to identify Inulae Radix, Vladimiriae Radix, and Aristolochiae Radix. Conclusions : The primers designed basing on the nucleotide sequences of ITS regions appearing differenced in the sequences among medicinal plants amplified the DNA markers for the identification of Aucklandiae Radix, Inulae Radix, Vladimiriae Radix, and Aristolochiae Radix. These herbal medicines were more efficiently identified by multiplex PCR method using all primers in a single PCR process.

Design and Comparison of the Pipelined IFFT/FFT modules for IEEE 802.11a OFDM System (IEEE 802.11a OFDM System을 위한 파이프라인 구조 IFFT/FFT 모듈의 설계와 비교)

  • In this paper, we design the IFFT/FFT (Inverse fast Fourier Transform/Fast Fourier Transform) modules for IEEE 802.11a-1999, which is a standard of the High-speed Wireless LAN using the OFDM (Orthogonal Frequency Division Multiplexing). The designed IFFT/FFT is the 64-point FFT to be compatible with IEEE 802.11a and the pipelined architecture which needs neither serial-to-parallel nor parallel-to-serial converter. We compare four types of IFFT/FFT modules for the hardware complexity and operation : R22SDF (Radix-2 Single-path Delay feedback), the R2SDF (Radix-2 Single-path Delay feedback), R2SDF (Radix-4 Single-path Delay Feedback), and R4SDC (Radix-4 Single-path Delay Commutator). In order to minimize the error, we design the IFFT/FFT module to operate with additional decimal parts after butterfly operation. In case of the R22SDF, the IFFT/FFT module has 44,747 gate counts excluding RAMs and the minimized error rate as compared with other types. And we know that the R22SDF has a small hardware structure as compared with other types.

Quality Characteristics and Antioxidant Activity of Chocolate Added with Rehmannia Radix Preparata Concentrate (숙지황 농축액 첨가 초콜릿의 품질 특성 및 항산화성)

  • The purpose of this study was to evaluate the quality characteristics and antioxidant activity of chocolate added with Rehmannia Radix Preparata (R. Radix Preparata) concentrate (0, 3, 6 and 9%). As the amount of R. Radix Preparata concentrate increased, water content significantly. The pH of chocolate decreased significantly as R. Radix Preparata concentrate increased, but the acidity of chocolate increased. Sugar concentration and reducing sugar contents also increased according to the amount of R. Radix Preparata concentrate. The L(lightness), a(redness) and b(yellowness) values significantly decreased as R. Radix Preparata concentrate increased. However, texture was not significantly different among the treatments. As the amount of R. Radix Preparata concentrate increased, DPPH radical scavenging and hydroxyl radical scavenging activities increased. In the results of the sensory evaluation, scores of color, taste, flavor, texture, and overall acceptability were the highest in chocolate added with 6% R. Radix Preparata concentrate. From these results, it was suggested that the optimum addition amount of R. Radix Preparata concentrate to chocolate is 6%.

Study on the Anti-oxidative Effects of Adenophorae Radix, Codonopsis lanceolatae Radix and Glehniae Radix Cum Rhizoma on Liver Cells Isolated from Oxidatively Stressed Rat (산화동물 모델 흰쥐 간세포에 대한 사삼(沙參), 양유근(羊乳根) 및 해방풍(海防風)의 항산화 효과 연구)

  • Objectives : The present study was purposed to compare Adenophorae Radix (henceforth AR), Codonopsis lanceolatae Radix (henceforth ClR) and Glehniae Radix cum Rhizoma (henceforth GRcR) concerning their anti-oxidant effect. Methods : We measured eythrocyte, leukocyte, thrombocyte, serum albumin, total bilirubin, LDL cholesterol, and glucose as well as SOD, GSH, catalase, NO, and MDA in the rat liver oxidatively stressed by AAPH. Results : 1. The oxidative stress-induced thrombocyte levels were significantly decreased in CIR-treated and GRcR-treated groups. 2. The oxidative stress-impaired SOD acitivities were significantly recovered in AR-treated and GRcR-treated groups. 3. The oxidative stress-reduced GSH contents were significantly increased in ClR-treated and GRcR-treated groups. 4. The oxidative stress-reduced catalase contents were significantly increased in all of the three groups. 5. The oxidative stress-induced NO productions were significantly decreased in all of the three groups. Conclusions : AR, ClR, and GRcR altogether showed the anti-oxidant effect in the rat liver oxidatively stressed by AAPH. The anti-oxidant properties of tAR, ClR, and GRcR seem to be similar even if those have different botanical properties and different medical efficacies in oriental medicine.

Comparative Study of Ginseng Radix, Rehmanniae Radix and Fermented Red-ginseng Extracts Effects on Memory Improvement in Scopolamine-induced Memory Impaired Mice (Scopolamine으로 유도된 인지장애 마우스의 기억력 개선 효과에 대한 인삼, 생지황, 발효홍삼 추출물의 비교 연구)

  • In this study, we investigated the effects of Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng extracts (FRG) on cognitive function in scopolamine-induced memory-impaired mice. We measured the effects of G, R, and FRG on the improvement of memory and cognition via behavior analysis. In addition, we measured the acetylcholinesterase (AChE) activity in the hippocampus of each group of mice. The expression of β-amyloid, Tau, and BDNF in the brain tissues were observed through immunohistochemical staining. Ginseng Radix (G) and Fermented red-ginseng (FRG) have effectively improved cognitive function in the water maze test. Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng (FRG) have improved the willingness of mice to explore the new environment, as confirmed by Y maze test. In addition, immunohistochemical staining revealed that Ginseng Radix (G) decreased the expression of β-amyloid and Tau in the hippocampus. In addition, fermented red-ginseng (FRG) increased the expression of BDNF. Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng (FRG) have decreased the concentration of acetylcholinesterase in the hippocampus as compared with the control group of mice. In conclusion, Ginseng Radix (G), Rehmanniae Radix (R), and Fermented red-ginseng (FRG) are considered to have the potential for development as candidate drugs to control the progression of Alzheimer's disease (AD).

Protective Effect of Sophorae Subprostratae Radix and Each Fractions on PC12 cell Damage Induced by Hypoxia/Reperfusion (PC12 세포의 허혈모델에 있어 광두근 분획물의 항산화효과연구)

  • This research was performed to investigate protective effect of Sophorae subprostratae Radix and each fractions against ischemic damage using PC12 cells. To observe the protective effect of Sophorae subprostratae Radix on ischemia damage, vibility and changes in activities of Superoxide dismutase (SOD), Glutathione Peroxidase (GPx), Catalase and Production of Malondialdehyde (MDA) were observed after treating PC12 cells with Sophorae subprostratae Radix during ischemic insult. Groups were divided into five groups: no treated (Normal), hypoxia chamber for 48hrs followed by 6h at normoxic chamber (H/R), Sop horae subprostratae Radix total phase treated group with H/R (Total), Sophorae subprostratae Radix water phase treated group with H/R (Water), Sophorae subprostratae Radix BuOH phase treated group with H/R (BuOH), Sophorae subprostratae Radix alkaloid phase treated group with H/R (Alkaloid). The results showed that (1) in hypoxiajreperfusion model using PC12 cell, the Sophorae subprostratae Radix has the protective effect against ischemia in the dose of 0.2 ㎍/㎖, 2 ㎍/㎖ and 20 ㎍/㎖, (2) Sophorae subprostratae Radix increased the activities of glutathione peroxidase and catalase. (3) the activity of Superoxide Diamutase(SOD) increased by ischemic damage, which might represent the self protection. This study suggests that Sophorae subprostratae Radix has neuroprotective effect against neuronal damage following hypoxiajreperfusion cell culture model using PC12 cell and dose dependency effects. In conclusion, Sophorae subprostratae Radix has protective effects against ischemic oxidative damage at the early stage of ischemia.

  • $Yongkaksan^{\circledR}$ is a powdered crude drug which is used to treat cough and recommended it as an expectorant. It is composed of Platycodi Radix (11.7 mg), Senegae Radix(0.5 mg), Armeniacae Semen(0.83 mg) and Glycyrrhizae Radix(8.3 mg). For the identification of individual ingredients in such powdery mixtures, microscopic method may be considered to be efficacious due to requirement of a small amount of specimens. In this paper, effectiveness of this method will be exemplified by the identification of the ingredients in $Yongkaksan^{\circledR}$ which contains 4 powdered crude drug ingredients.

  • Objectives : The aim of the present study is to evaluate the effects Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae extracton 3T3-L1 preadipocytes differentiation. Methods : We adopted Oil Red O staining methods to observe the formed lipid droplets. And the amount of lipids in adipocytes was measured using absorptiometric analysis. Results : The extracts of Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae stimulated the preadipocytes differentiation and lipid droplet formation. And the complex extract of the traditional herbal medicines stimulated there actions more than single extracts. Conclusions : The extract of Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae has stimulatory effects. on adipogenesis. Moreover, the complex extract of the traditional herbal medicines has more effect than single extracts.

Effects of SosokMyeongTang extract on the Regional Cerebral Blood Flow and Blood Pressure in Rats (소속명탕(小續命湯)이 백서(白鼠)의 국소뇌혈류량(局所腦血流量)과 혈압(血壓)에 미치는 영향(影響))

  • SosokMyeongTang(SMT) have been used in oriental medicine for many centuries as a therapeutic agent for cerebral disease. The effects of SMT on the vascular system is not known. The purpose of this study was to investigate the effects of SMT on the changes in blood pressure(BP) and regional cerebral blood flow(rCBF) of rats. SMT consists of the following components : Radix Ledebouriellae(防風), Radix Cocculi or Stephaniae(防己), Semen Armeniacae(杏仁), Cortex Cinnamomi(肉桂), Radix Scutellariae(黃芩), Radix Paeoniae Lactiflorae(白芍藥), Radix Ginseng(人蔘), Rhizoma Cnidll(川芎), Herba Ephedrae(麻黃), Radix Glycyrrhizae(甘草), Radix Aconiti(附子?), Fructus Zizyphi Jujubae(大棗), Rhizoma Zingiberis(生薑) and the changes of BP and rCBF were tested Leser-Doppler Flowmetry(LDF) The experimental results were as follows ; BP was not affected by SMT in rats, but rCBF was increased significantly by SMT in a dose dependent manner. SMT increased previous decreasd rCBF due to pretreatment methylene blue, but did not increased previous decreasd rCBF due to pretreatment L-NNA, indomethacin. Pretreatment with indomethacin decreased BP compaired with control group. These results suggest that SMT causes a diverse response of blood pressure and regional cerebral blood flow(rCBF). The increased rCBF is mediated by nitric oxide synthease.

Inhibitory Effects of Rehmannia glutinosa Liboschitz on Endothelial Cell Proliferation

  • Vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) are the most important angiogenic molecules associated with tumor-induced neovascularization. This study was carried out to investigate inhibitory effect of extracts from root of Rehmannia glutinosa LIBOSCHITZ (Rehmannia Radix and Rehmannia Radix Preparata) on endothelial cell proliferation. The methanol extracts from the medicinal herb were fractionated into n-hexane, ethyl acetate, n-butanol and aqueous fractions. Among the four fractions, the n-butanol fraction from R. Radix on exhibited highly effective inhibition (${\approx}79%$ inhibition) on the binding of KDR/Flk-1-Fc to immobilized $VEGF_{165}$ and then ethyl acetate fraction from R. Radix (${\approx}45%$ inhibition) at the concentration of $100\;{\mu}g/ml$. The n-butanol fraction efficiently blocked the VEGF- and bFGF-induced HUVEC proliferation in a dose-dependent manner, but did not affect the growth of HT1080 human fibrosarcoma cells. The n-butanol fraction more efficiently blocked the binding of KDR/Flk-1-Fc to immobilized $VEGF_{165}$ and VEGF- and bFGF-induced human umbilical vein endothelial cell proliferation than the fraction from R. Radix Preparata. Our results suggest that Rehmannia Radix may be used as a candidate for developing anti-angiogenic agent.
